Attempts to estimate photosynthetic rate or gross primary productivity from remotely sensed absorbed solar radiation depend on knowledge of the light use efficiency (LUE). Early models assumed LUE to be constant, but now most researchers try to adjust it for variations in temperature and moisture stress. However, more exact methods are now required. Hyperspectral remote sensing offers the possibility of sensing the changes in the xanthophyll cycle, which is closely coupled to photosynthesis. Several studies have shown that an index (the photochemical reflectance index) based on the reflectance at 531 nm is strongly correlated with the LUE over hours, days and months. A second hyperspectral approach relies on the remote detection of fluorescence, which is a directly related to the efficiency of photosynthesis. We discuss the state of the art of the two approaches. Both have been demonstrated to be effective, but we specify seven conditions required before the methods can become operational.  相似文献

In the present study, we documented the promising role of thyroid hormones status in animals in modulation of Na+–Pi transport activity in intestinal brush border membrane vesicles (BBMV) which was accompanied with alterations in BBM lipid composition and fluidity. Augmentation of net Pi balance in hyperthyroid (Hyper-T) rats was fraternized with accretion of Pi transport across BBMV isolated from intestine of Hyper-T rats as compared to hypothyroid (Hypo-T) and euthyroid (Eu-T) rats while Na+–Pi transport across BBMV was decreased in Hypo-T rats relative to Eu-T rats. Increment in Na+–Pi transport in intestinal BBMV isolated from Hyper-T rats was manifested as an increase in the maximal velocity (Vmax) of Na+–Pi transport system. Furthermore, BBMV lipid composition profile in intestinal BBM from Hyper-T was altered to that of Hypo-T rats and Eu-T rats. The molar ratio of cholesterol/phospholipids was higher in intestinal BBM from Hypo-T rats. Fluorescence anistropy of diphenyl hexatriene (rDPH) and microviscosity were significantly decreased in the intestinal BBM of Hyper-T rats and decreased in Hypo-T rats as compared to Eu-T rats which corroborated with the alteration in membrane fluidity in response to thyroid hormone status of animals. Therefore, thyroid hormone mediated change in membrane fluidity might play an important role in modulating Na+–Pi transport activity of intestinal BBM. (Mol Cell Biochem 278: 195–202, 2005)  相似文献

Antecedent studies have suggested that lipid composition and fluidity of cellular membranes of various organs are altered in response to thyroid hormone status. To date, the effects of thyroid hormone status on these parameters have not been examined in rat renal apical membrane in regard to sodium-dependent phosphate transport. In the present study, we determined the potential role of alterations in cortical brush-border membrane lipid composition and fluidity in modulation of Na+–Pi transport activity in response to thyroid hormone status. Thyroid hormone status influences the fractional excretion of Pi, which is associated with alteration in renal brush-border membrane phosphate transport. The increment in Na+–Pi transport in renal BBMV isolated from Hyper-T rats is manifested as an increase in the maximal velocity (Vmax) of Na+–Pi transport. Further, the cholesterol content was significantly increased in renal BBM of Hypo-T rats and decreased in Hyper-T rats as compared to the Eu-T rats. The molar ratio of cholesterol/phospholipids was also higher in renal BBM from hypo-T rats. Subsequently, fluorescence anisotropy of diphenyl hexatriene (rDPH) and microviscosity were significantly decreased in the renal BBM of the Hyper-T rats and increased in the Hypo-T rats as compared to Eu-T rats. The result of this study, therefore, suggest that alteration in renal BBM cholesterol, cholesterol/phospholipid molar ratio, and membrane fluidity play an important role in the modulation of renal BBM Na+–Pi transport in response to thyroid hormone status of animals. Plant growth, chlorophyll (Chl) content, photosynthetic gas exchange, ribulose-1,5-bisphosphate carboxylase (RuBPCO) enzyme activity, and Chl fluorescence in radish (Raphanus sativus var. longipinnatus) plants were examined after turnip mosaic virus (TuMV) infection. Plant fresh mass, dry mass, Chl content, net photosynthetic rate (P N), transpiration rate (E), stomatal conductance (g s), and RuBPCO activity were significantly lower in infected plants after 5 weeks of virus infection as compared to healthy plants. The 5-week virus infection did not induce significant differences in intercellular CO2 concentration (C i, photochemical efficiency of photosystem 2, PS2 (Fv/Fm), excitation capture efficiency of open PS2 reaction centres (Fv'/Fm'), effective quantum efficiency of photosystem 2 (ΔF/Fm'), and photochemical quenching (qP), but non-photochemical quenching (qN) and alternative electron sink (AES) were significantly enhanced. Thus the decreased plant biomass of TuMV-infected plants might be associated with the decreased photosynthetic activity mainly due to reduced RuBPCO activity.  相似文献

Photosynthetic irradiance response of vegetative and reproductive structures of the green-flowered deciduous perennial green hellebore was studied by the comparative use of chlorophyll (Chl) fluorescence techniques and gas exchange measurements. All the Chl-containing organs (leaves, sepals, stalks, and fruits) examined were photosynthetically active showing high intrinsic efficiencies of photosystem 2 (Fv/Fm: 0.75–0.79) after dark adaptation. Even in the smaller fertile and sterile parts of the flower (nectaries and anthers) a remarkable photosynthetic competence was detected. With increasing photon flux densities (PFD) electron transport rates, actual quantum yields, and photochemical quenching coefficients of the main photosynthetic organs decreased in the order: leaf>sepal>fruit>stalk. At moderate to high PFDs the sepals achieved maximum electron transport rates corresponding to about 80 % of concomitant mature leaves. In contrast, maximum net photosynthetic rate of the sepals [2.3 mol(CO2) m–2 s–1] were less than one fourth of the leaves [10.6 mol(CO2) m–2 s–1]. This difference is explained by a 70–80 % lower stomatal density of sepals in comparison to leaves. As the basal leaves emerge late during fruit development, the photosynthetically active sepals are a major source of assimilates, contributing more than 60 % of whole-plant CO2 gain in early spring. The ripening dehiscent fruits are characterized by an effective internal re-fixation of the respirational carbon loss and thus additionally improve the overall carbon budget.  相似文献

Chromosomal locations of 19 horse immunity-related loci (CASP1, CD14, EIF5A, FCER1A, IFNG, IL12A, IL12B, IL12RB2, IL1A, IL23A, IL4, IL6, MMP7, MS4A2, MYD88, NOS2A, PTGS2, TFRC and TLR2) were determined by fluorescence in situ hybridization. For IFNG and PTGS2, this study is a confirmation of their previously reported position. In addition, microsatellite (HMBr1) was localized in the same region as IFNG. All genes were assigned to regions of conserved synteny and the data obtained in this study enhance the comparative human-horse map. Cytogenetic localization of IL6 to ECA4q14-q21.1 suggested a new breakage point that changes the order of loci compared with HSA7. The map assignments of these loci serve as anchors for other loci and will aid in the search for candidate genes associated with traits in the horse.  相似文献

The aggregation of alpha-synuclein is believed to be a critical step in the etiology of Parkinson's disease. A variety of biophysical techniques were used to investigate the aggregation and fibrillation of alpha-synuclein in which one of the four intrinsic Tyr residues was replaced by Trp, and two others by Phe, in order to permit fluorescence resonance energy transfer (FRET) between residues 39 (Tyr) and 125 (Trp). The mutant Y125W/Y133F/Y136F alpha-synuclein (one Tyr, one Trp) showed fibrillation kinetics similar to that of the wild-type, as did the Y125F/Y133F/Y136F (one Tyr, no Trp) and Y39F/Y125W/Y133F/Y136F (no Tyr, one Trp) mutants. Time-dependent changes in FRET, Fourier transform infrared, Trp fluorescence, dynamic light-scattering and other probes, indicate the existence of a transient oligomer, whose population reaches a maximum at the end of the lag time. This oligomer, in which the alpha-synuclein is in a partially folded conformation, is subsequently converted into fibrils, and has physical properties that are distinct from those of the monomer and fibrils. In addition, another population of soluble oligomers was observed to coexist with fibrils at completion of the reaction. The average distance between Tyr39 and Trp125 decreases from 24.9A in the monomer to 21.9A in the early oligomer and 18.8A in the late oligomer. Trp125 remains solvent-exposed in both the oligomers and fibrils, indicating that the C-terminal domain is not part of the fibril core. No FRET was observed in the fibrils, due to quenching of Tyr39 fluorescence in the fibril core. Thus, aggregation of alpha-synuclein involves multiple oligomeric intermediates and competing pathways.  相似文献

Proteins containing the STAR RNA-binding domain fulfill vital roles in RNA biogenesis, yet a detailed understanding of STAR domain RNA binding specificity is lacking. In Caenorhabditis elegans, the STAR protein GLD-1 directly binds the 28 nucleotide recognition element TGE within the 3' untranslated region of tra-2 mRNA. The GLD-1:TGE interaction promotes translational silencing of tra-2 mRNA, marking a pivotal event in the spermatogenesis to oogenesis switch in C.elegans hermaphrodites. By measuring the binding affinities of both GLD-1 and TGE mutants, we have explored the molecular determinants of STAR domain specificity. Site-directed GLD-1 mutants were guided by sequence homology with human splicing factor 1 (SF1), for which an RNA:protein complex structure is available in the work done by Liu et al. The RNA binding affinity of 11 mutant GLD-1 proteins was measured, and their binding specificity was assessed with a series of TGE RNAs containing natural or modified nucleotides. This combinatorial analysis of both RNA and protein mutants revealed a diverse array of specificities of individual nucleotide-binding pockets along the interface. At nucleotide position 18, adenosine appears to be specified by the overall shape of a pocket lined with aliphatic side-chains. At position 19, the high preference for cytidine is dependent on both the length of an amino acid side-chain and the identity of terminal functional groups. The nucleotide 21 binding pocket exhibits low discrimination for cytidine, and accommodates most nucleobases. The highly hydrophobic binding interface and apparent small number of hydrogen bonding read-out interactions at these positions is consistent with our finding that few amino acids seem to function individually in establishing binding specificity. Rather, specificity is conferred by the shape of the nucleotide-binding pocket. Our data provide the first detailed, quantitative analysis of the STAR domain, and highlight features of STAR:RNA recognition that are distinct among single-stranded RNA-binding proteins.  相似文献
