Cell cultures are indispensable to develop and study efficacy of therapeutic agents, prior to their use in animal models. We have the unique ability to model well differentiated human airway epithelium and heart muscle cells. This could be an invaluable tool to study the deleterious effects of toxic inhaled chemicals, such as chlorine, that can normally interact with the cell surfaces, and form various byproducts upon reacting with water, and limiting their effects in submerged cultures. Our model using well differentiated human airway epithelial cell cultures at air-liqiuid interface circumvents this limitation as well as provides an opportunity to evaluate critical mechanisms of toxicity of potential poisonous inhaled chemicals. We describe enhanced loss of membrane integrity, caspase release and death upon toxic inhaled chemical such as chlorine exposure. In this article, we propose methods to model chlorine exposure in mammalian heart and airway epithelial cells in culture and simple tests to evaluate its effect on these cell types.  相似文献

Global climate change affects the distributions of ectotherms and may be the cause of several conservation problems, such as great displacement of climatic suitable spaces for species and, consequently, important reductions of the extent of liveable places, threatening the existence of many of them. Species exposure (and hence vulnerability) to global climate change is linked to features of their climatic niches (such as the relative position of the inhabited localities of each species in the climatic space), and therefore to characteristics of their geographic ranges (such as the extent of the distributions or altitudinal range inhabited by the species). In order to analyze the pattern of response of Argentine reptiles to global climate change, we ran phylogenetic generalized least squares models using species exposure to global climate change as a response variable, and (i) niche properties (breadth and position of the species in the climate space) and (ii) general features of the distribution of species (maximum latitude, altitudinal range, maximum elevation, distributional range and proximity to the most important dispersal barrier) as predictors. Our results suggest that the best way to explain climate change exposure is by combining breadth and position of climatic niche of the species or combining geographic features that are indicators of both niche characteristics. Our best model shows that in our study area, species with the narrowest distributional ranges that also inhabit the highest elevations are the most exposed to the effects of global climate change. In this sense, reptile species from Yungas, Puna and Andes ecoregions could be especially vulnerable to the effects of climate change. We believe that these types of models may represent an interesting tool for determining species and places particularly threatened by the effects of global climate change, which should be strongly considered in conservation planning.  相似文献

目的:探讨负压封闭引流技术(VSD)对兔颅骨外露缺损创面愈合的治疗效果。方法:选取成年新西兰大白兔76只,平均分为四组并建立兔颅骨外露实验模型。其中,A组(19只):于兔颅骨上方制作直径为2.0cm的圆形创面,保留骨膜,采用-120mmHg负压引流和常规换药治疗;B组(19只):实验动物处理同A组,仅采用常规换药治疗;C组(19只):在兔颅骨上制作直径2.0cm的圆形创面,剔除骨膜,治疗方法同A组;D组(19只):实验动物处理同C组,治疗方法同B组。每组各抽取10只,观察创面愈合率和创面愈合时间;其余9只分别在第7天、10天、20天、30天进行取材检测,分析疗效机制。结果:A组创面愈合时间为19.40±1.65天,B组为24.00±2.31天;C组为25.40±4.43天,D组为30.00±5.50天。运用VSD治疗和常规治疗创面愈合时间比较有统计学意义(P0.05)。结论:VSD治疗兔骨外露缺损创面能有效缩短创面愈合时间,促进血管再生,胶原蛋白合成。  相似文献

应用小波熵分析大鼠脑电信号的动态变化特性   总被引:19,自引:0,他引:19  
应用小波熵(一种新的信号复杂度测量方法)分析大鼠在不同生理状态下脑电复杂度的动态时变特性。采用慢性埋植电极记录自由活动大鼠的皮层EEG,使用多分辨率小波变换将EEG信号分解为δ、θ、α和β四个分量,求得随时间变化的小波熵。结果表明:在清醒、慢波睡眠和快动眼睡眠三种生理状态下,EEG的小波熵之间存在显著差别,并且在不同时期其值与各个分解分量之间具有不同的关系,其中,慢波睡眠期小波熵还具有较明显的变化节律,反映了EEG微状态中慢波和纺锤波的互补性。由此可见,小波熵既能区别长时间段EEG复杂度之间的差别,又能反映EEG微状态的快速变化特性。  相似文献

Activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione reductase (GR), glutathione-S-transferase (GST), as well as total glutathione (tGSH) concentration were analyzed in the hemolymph and fat body of the European corn borer Ostrinia nubilalis Hubn. and the Mediterranean borer Sesamia cretica Led. (Lepidoptera, Noctuidae). Controls were maintained at 8°C while experimental groups of larvae were exposed to –3°C for ten days and then to –12°C for 23 days (only for Ostrinia). Cold exposure significantly increased fat body SOD, GR, and GST activities of Ostrinia larvae. Only GST activity and tGSH levels increased significantly in Ostrinia larval hemolymph on cold exposure. In Sesamia larvae after cold exposure, hemolymph CAT activity was significantly lower, while fat body tGSH increased. The antioxidant defense systems of these two species show differences, probably influenced by their respective cold-hardiness metabolism. According to its antioxidant profile, the response of Ostrinia suggests a significant physiological alteration in its metabolism during cold exposure, indicating a compensatory mechanism. Abstract

The effects of no. 20 diesel oil exposure, 2,4-dichlorophenol (2,4-DCP) exposure and combined exposure on the antioxidant defences of Daphnia magna have been studied systematically for the first time. Daphnia magna was exposed for 1 day or 10 days to several concentrations of 0, 0.005, 0.01, 0.05, 0.1, 0.5 or 1.0 mg L?1 solutions. Antioxidant defences consisting of the levels of reduced glutathione (GSH) and glutathione disulfide (GSSG) and activities of superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GR), selenium dependent glutathione peroxidase (Se-GPx) and glutathione S-transferase (GST) of daphnids were determined to evaluate their protective roles and to analyse the occurrence of oxidative stress. The possible antioxidant defence mechanisms are discussed. Furthermore, GST can be a potential biomarker and an early-warning index for the pollutants in waters in that GST responded sensitively to 1 day and 10 days of exposure to diesel oil and 2,4-DCP and 10 days of combined exposure. Crossover comparisons showed an antagonistic action about the no-observed-effect concentration (NOEC) against Daphnia magna, which needs further studies.  相似文献

Human health risk estimates for sites with contaminated soils are often based on the assumption that the outdoor soil sieved to < 250 μm is a reasonable surrogate for predicting exposures via incidental soil ingestion. In vitro bioaccessibility tests are also increasingly used to “improve” ingestion exposure predictions of contaminants at different sites. However, when considered in terms of factors that influence desorption of contaminants from particles and uptake into humans, available studies indicate that current “standards of practice” with respect to assays of oral bioaccessibility have a number of significant shortcomings, at least in Canada. These shortcomings are discussed and various factors that influence the assessment of bioaccessibility of contaminants in soils are examined. We finish with proposing some minimum data submission requirements to support the application and relevance of bioaccessibility assays at contaminated sites.  相似文献

通过分析2003—2006年连续3个冬季北京城区和城郊越冬长耳鸮的食团,研究了长耳鸮食物组成的时间与空间变化特征。每月收集1次长耳鸮食团,通过其内容物分析长耳鸮食物中的猎物种类和数量构成。结果表明:城区和城郊两地长耳鸮3种主要猎物中蝙蝠的被捕食率分别为41.6%和43.0%,鼠类为27.5%和39.4%,鸟类为30.7%和18.9%;城区和城郊长耳鸮的食物构成在2004—2005年与2005—2006年的冬季存在显著差异,初冬和冬末的食物构成差异也呈显著水平;同区域的长耳鸮食物构成在年度间和不同越冬时期皆存在显著差异;两地越冬长耳鸮捕食猎物比例的差异与生存环境中猎物资源的可获得性有关。  相似文献

Mature mouse oocytes were exposed prior to in vitro fertilization to visible light during 1, 2, or 4 hr at an intensity of 4,000 lux. Compared to controls cultured under identical conditions but protected from light, exposed eggs did not show any significant modification of cleavage speed and rate. After transfer of blastocysts obtained in vitro in uteri of pseudopregnant females, the implantation rate and the proportion of normal fetuses were not found to be different in relation to preliminary light exposure of oocytes fertilized and cultured in vitro.  相似文献
