Fractional rates (% · day–1) of synthesis and degradation were determined by measuring the output of N-methylhistidine (MeHis) in the excreta at 4 and 8 weeks of age in the chicken. At 4 weeks of age, the fractional rate of synthesis of the meat-type stock was twice that of the egg-type stock (White Leghorn), but the fractional rates of synthesis at 8 weeks of age were similar (4.1–5.1% · day–1) among stocks. The fractional rate of degradation (1.3–1.5% · day–1) of the meat-type stock at 8 weeks of age was less than half the rate of the egg-type stock (2.9% · day–1). The fractional rates of synthesis and degradation at 4 weeks of age in the Satsuma native fowl were relatively high compared with those in the other stocks. In particular, the rate of degradation (8.6% · day–1) at 4 weeks of age was approximately twice that of other stocks. These results show that fractional rates of synthesis and degradation of muscle protein in the chicken differ among genetically diverse groups. Quinolizidine alkaloids formed in the leaves of Lupinus albus L. are translocated via the phloem to the other plant organs, especially the maturing fruits. Compared with amino-acid transport in the phloem, the alkaloids contribute about 8% to the overall nitrogen being exported from the leaf. Since it is likely that the alkaloids are subsequently degraded in the target tissues a minor role of quinolizidine alkaloids might be nitrogen transport. A marked diurnal fluctuation of alkaloids was observed in the leaves, the phloem sap, the roots and the fruits with an increase during the day and an amplitude of several hundred percent thus providing evidence for a rapid turnover of endogenous alkaloids.Abbreviations QA

A method for the estimation of chlorophyll turnover in wheat leaves is presented. This is based on the inhibition of chlorophyll synthesis by treatment of the cut leaves with laevulinic acid (LA), a competitive inhibitor of δ-aminolaevulinic acid dehydratase. The turnover of chlorophyll in young, greening leaves, given short periods of light was a relatively rapid process. Summary A field experiment on N turnover in rotations of groundnut, sorghum and pigeonpea crops was conducted in an Indian Alfisol during 1978–80.15N-labelled urea N was applied @ 40 kg ha–1 in 1978. In the first year, the groundnut utilized 19.5% of the applied labelled N, sorghum 25.5%, and pigeon pea 10.3%. More fertilizer N was removed through weeding than by crop uptake in sorghum and pigeon pea. The fertilizer N left behind in soil upto 40 cm depth was 44.4% in groundnut plots, 35.1% in sorghum plots and 40.1% in pigeon pea plots.The uptake in 1979 of the residual fertilizer N in the soil was 8.9% in sorghum, 8.3% in groundnut and 2.8% in pigeon pea. In 1980, it declined to less than 2% for pigeon pea and groundnut and to about 4% for sorghum.A balance sheet drawn at the end of each rotation showed that about 51.3% of the applied labelled N was accounted for in groundnut-sorghum-pigeon pea rotation, 70.9% in sorghumpigeon pea-groundnut, and 43.5% in pigeon pea-groundnut-sorghum. 相似文献

Photosynthetically fixed 14C was analyzed in various chemical fractions from leaves and stems of cottonwood (Populus deltoides Bartr. ex. Marsh.) during dormancy induction. Dormancy was induced by 8-h photoperiods and 20/14°C temperature regimes. Within 4 weeks under short days, terminal buds were set and leaf expansion and stem elongation had stopped. 14C2 was fed to a leaf at Leaf Plastochron Index 7 for 30 min. Either after this 30 min feeding period or after a 48-h translocation period the plants were sampled, freeze-dried, extracted and analyzed for14C. 14C-fixation decreased during dormancy induction from 60% to 17% of the 3.7 MBq 14C applied at 0 week and 8 weeks, respectively. Percentage distribution of 14C in chemical fractions of source leaves reflected leaf age and translocation inhibition. In rapidly growing plants, considerable 14C was incorporated into leaf protein while most of the soluble14C-sugars were either metabolized or translocated out of the leaf. After terminal bud set, the percentage of 14C in the protein and residue fractions decreased rapidly and that in the sugar fraction increased. Percent distribution in stems closely reflected changing metabolic pathways of carbon flow as influenced by dormancy induction. For example, the 14C in structural carbohydrates decreased in 5 weeks under short days from 65 to less than 10% of the 14C recovered in the chemical fractions, thus indicating cambium inhibition. At the same time the percentage of 14C in starch and sugar increased indicating storage. Short term (after 30 min) incorporation of 14C into the protein and starch fractions of leaves changed relatively little throughout the 8-week induction period. Summary Large areas of photoreceptor membrane are synthesized in the retinula cells of the crab Leptograpsus variegatus at dusk. Initially, new membrane differentiates from rough endoplasmic reticulum (ER) as large tubules of smooth ER. These tubules transform to concentric ellipsoids of closely apposed pairs of membranes (doublet ER), sometimes passing through an intervening crenate form. The new membrane is transported through bridges of cytoplasm that cross the palisade to the rhabdom region, from which the remains of the rhabdomeres that were built during the previous dusk have been dissolved. The degradation of the old microvilli of one rhabdomere is accomplished without affecting neighbouring rhabdomeres of other cells. New microvilli are assembled in situ from sheets of doublet ER, which are converted to tubules oriented in the same direction as the future microvilli. The cytoplasmic face of the ER remains the cytoplasmic face of the tubules, which become progressively narrower, partly by further longitudinal division, until the final diameter of the microvillus is reached. A central core is often seen in transverse sections of mature microvilli. It may be involved in the final consolidation, but rhabdomeric microvilli are not formed in the same manner as those of intestinal brush border cells. There is no evidence that new membrane passes through the Golgi compartment before incorporation into the rhabdom, as is the case for rod outer segment membrane in vertebrate photoreceptors. 相似文献

Exogenously supplied bovine haemin, fed to etiolated barley leaves, inhibited chlorophyll synthesis in leaves exposed to light. Haemin inhibited the regeneration of protochlorophyllide (P650) and the conversion of exogenously supplied δ-aminolaevulinate (ALA) to protochlorophyll (P630). The effect of haemin on chlorophyll production was overcome by incubating the leaves in water in the dark before light treatment, suggesting the operation of a rapid haem destruction mechanism in leaves. Protohaem turnover in dark-grown leaves was between 8 and 9 hr, based on the rate of degradation of erogenous haemin and the rate of protohaem breakdown in laevulinic acid (LA) treated leaves. The rate constant for haem destruction was 85 pmol/nmol/hr in the dark and 45 pmol/nmol/hr after 4 hr light. There was no evidence that light affects the synthesis of protohaem. It appears that the regulation of endogenous levels of protohaem is by breakdown and it is this mechanism which is under light control. Haem considerably decreased the incorporation of radioactivity from glycollate-[14C], glycine-[14C] and glutamate-[14C] into accumulated ALA in the presence of LA. 相似文献

Citrate and other polyanion binding to ferricytochrome c partially blocks reduction by ascorbate, but at constant ionic strength the citrate-cytochrome c complex remains reducible; reduction by TMPD is unaffected. At a constant high ionic strength citrate inhibits the cytochrome c oxidase reaction competitively with respect to cytochrome c, indicating that ferrocytochrome c also binds citrate, and that the citrateferrocytochrome c complex is rejected by the binding site at high ionic strength. At lower ionic strengths, citrate and other polyanions change the kinetic pattern of ferrocytochrome c oxidation from first-order towards zero-order, indicating preferential binding of the ferric species, followed by its exclusion from the binding site. The turnover at low cytochrome c concentrations is diminished by citrate but not the Km (apparent non-competitive inhibition) or the rate of cytochrome a reduction by bound cytochrome c. Small effects of anions are seen in direct measurements of binding to the primary site on the enzyme, and larger effects upon secondary site binding. It is concluded that anion-cytochrome c complexes may be catalytically competent but that the redox potentials and/or intramolecular behaviour of such complexes may be affected when enzyme-bound. Increasing ionic strength diminishes cytochrome c binding not only by decreasing the ‘association’ rate but also by increasing the ‘dissociation’ rate for bound cytochrome c converting the ‘primary’ (T) site at high salt concentrations into a site similar kinetically to the ‘secondary’ (L) site at low ionic strength. A finite Km of 170 μM at very high ionic strength indicates a ratio of of about 5000. It is proposed that anions either modify the E10 of cytochrome c bound at the primary (T) site or that they perturb an equilibrium between two forms of bound c in favour of a less active form. 相似文献

肌醇磷脂代谢与V-mos癌基因转化细胞的相关性,迄今为止未见报导。本文用6m2细胞(Moloney鼠类肉瘤病毒(含V-mos)温度敏感突变株(MoMuSVts110)转化的NRK细胞)为模型,探讨了肌醇磷脂代谢与细胞转化的相关性。在33℃ (转化型温度)时,细胞内PIP(磷脂酰肌醇-4-磷酸)含量明显高于39℃(正常型温度),显示出转化型6m2细胞中存在一个提高的PI激酶活性。同时可见DG(二酰甘油)和IP_3(肌醇三磷酸)含量和蛋白激酶C(PKC)活性均明显高于正常型细胞。当细胞由39℃转至33℃10min,PIP、DG、IP_3含量和PKC活性均明显增加,并伴随有PKC活性由胞质向质膜上的转移。实验结果表明肌醇磷脂代谢参与了6m2细胞转化过程。文中对其作用机理进行了讨论。 相似文献
