目的对ICU住院患者鲍曼不动杆菌感染的流行状况及耐药情况进行调查,为临床合理选用抗菌药物提供参考。方法对2012年7月至2013年12月ICU住院患者送检的693份各种临床标本进行病原菌的分离鉴定,对分离出的鲍曼不动杆菌采用K-B法做体外药敏试验。结果 693份临床标本共分离出110株鲍曼不动杆菌,其中多重耐药菌株有103株(93.6%),感染的标本来源以痰液最为常见(86.4%)。鲍曼不动杆菌对常用抗菌药物耐药广泛,对氨苄西林、头孢唑啉100%耐药,对哌拉西林、头孢曲松、复方新诺明、氨曲南、环丙沙星、头孢吡肟、哌拉西林/他唑巴坦及庆大霉素等多种抗菌药物的耐药率大于80.0%。结论 ICU患者鲍曼不动杆菌感染多由多重耐药菌株引起,感染部位以下呼吸道为主,其对抗菌药物耐药情况相当严峻,加强耐药性监测及合理使用抗菌药物,对减少耐药菌株的产生具有重要意义。 相似文献

The small chaperone protein Hsp27 confers resistance to apoptosis, and therefore is an attractive anticancer drug target. We report here a novel mechanism underlying the tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) sensitizing activity of the small molecule , an inactive analog of the phosphoinositide 3-kinase inhibitor inhibitor LY303511, in HeLa cells that are refractory to TRAIL-induced apoptosis. On the basis of the fact that LY294002 is derived from LY303511, itself derived from quercetin, and earlier findings indicating that quercetin and LY294002 affected Hsp27 expression, we investigated whether LY294002 sensitized cancer cells to TRAIL via a conserved inhibitory effect on Hsp27. We provide evidence that upon treatment with LY303511, Hsp27 is progressively sequestered in the nucleus, thus reducing its protective effect in the cytosol during the apoptotic process. LY303511-induced nuclear translocation of Hsp27 is linked to its sustained phosphorylation via activation of p38 kinase and MAPKAP kinase 2 and the inhibition of PP2A. Furthermore, Hsp27 phosphorylation leads to the subsequent dissociation of its large oligomers and a decrease in its chaperone activity, thereby further compromising the death inhibitory activity of Hsp27. Furthermore, genetic manipulation of Hsp27 expression significantly affected the TRAIL sensitizing activity of LY303511, which corroborated the Hsp27 targeting activity of LY303511. Taken together, these data indicate a novel mechanism of small molecule sensitization to TRAIL through targeting of Hsp27 functions, rather than its overall expression, leading to decreased cellular protection, which could have therapeutic implications for overcoming chemotherapy resistance in tumor cells. The increase in severity of droughts associated with greater mortality and reduced vegetation growth is one of the main threats to tropical forests. Drought resilience of tropical forests is affected by multiple biotic and abiotic factors varying at different scales. Identifying those factors can help understanding the resilience to ongoing and future climate change. Altitude leads to high climate variation and to different forest formations, principally moist or dry tropical forests with contrasted vegetation structure. Each tropical forest can show distinct responses to droughts. Locally, topography is also a key factor controlling biotic and abiotic factors related to drought resilience in each forest type. Here, we show that topography has key roles controlling biotic and abiotic factors in each forest type. The most important abiotic factors are soil nutrients, water availability, and microclimate. The most important biotic factors are leaf economic and hydraulic plant traits, and vegetation structure. Both dry tropical forests and ridges (steeper and drier habitats) are more sensitive to droughts than moist tropical forest and valleys (flatter and wetter habitats). The higher mortality in ridges suggests that conservative traits are not sufficient to protect plants from drought in drier steeper habitats. Our synthesis highlights that altitude and topography gradients are essential to understand mechanisms of tropical forest''s resilience to future drought events. We described important factors related to drought resilience, however, many important knowledge gaps remain. Filling those gaps will help improve future practices and studies about mitigation capacity, conservation, and restoration of tropical ecosystems. 相似文献

AIMS: To determine the effect of diet acidification and an in-feed antibiotic growth promotant (Tylosin, Ty) on selected culturable bacterial populations in the gastrointestinal tract (GIT) of mice. METHODS AND RESULTS: Female C57Bl mice were given a standard diet supplemented with Acid Pak (AP) or Ty in the drinking water. After 21 days, lumen and adherent populations of Enterobacteriaceae, enterococci/streptococci, and lactic acid bacteria (LAB) from the ileum, caecum, colon and faeces were enumerated. General intestinal health was assessed by the frequency of haemolytic bacteria in the different intestinal compartments. Contrary to expectations, AP and Ty significantly increased haemolytic bacteria in the lumen of the caecum and colon (P<0.05). The small but significant growth-enhancing effect of Ty (P<0.05) was associated with decreases in enterococci/streptococci and surprisingly, LAB, as well as increases in coliforms. AP, which failed to improve growth rates, reduced coliforms, had limited effects on enterococci/streptococci, and specifically failed to promote the growth of LAB populations in all intestinal compartments. Ty supplementation was also associated with a significant increase in macrolide-resistant enterococci throughout the GIT. CONCLUSIONS: Dietary acidification is less effective than Ty in modulating the population dynamics of selected culturable populations of enteric bacteria. SIGNIFICANCE AND IMPACT OF THE STUDY: The mouse can provide a useful experimental model to examine the effects of new dietary supplements, formulations or regimes on changes in microbial population dynamics, including monitoring for antibiotic resistance. 相似文献

The gene para in Drosophila melanogaster encodes an α subunit of voltage-activated sodium channels, the presumed site of action of DDT and pyrethroid insecticides.
We used an existing collection of Drosophila para mutants to examine the molecular basis of target-site resistance to pyrethroids and DDT. Six out of thirteen mutants tested
were associated with a largely dominant, 10- to 30-fold increase in DDT resistance. The amino acid lesions associated with
these alleles defined four sites in the sodium channel polypeptide where a mutational change can cause resistance: within
the intracellular loop between S4 and S5 in homology domains I and III, within the pore region of homology domain III, and
within S6 in homology domain III. Some of these sites are analogous with those defined by knockdown resistance (kdr) and super-kdr resistance-associated mutations in houseflies and other insects, but are located in different homologous units of the channel
polypeptide. We find a striking synergism in resistance levels with particular heterozygous combinations of para alleles that appears to mimic the super-kdr double mutant housefly phenotype. Our results indicate that the alleles analyzed from natural populations represent only
a subset of mutations that can confer resistance. The implications for the binding site of pyrethroids and mechanisms of target-site
insensitivity are discussed.

Analysis of total aromatic amino acid (free and bound) in some cucumber accessions selected previously for resistance to western flower thrips, Frankliniella occidentalis (Pergande) [Thysanoptera: Thripidae], indicated that low concentrations of these essential nutrients, relative to total leaf protein, were correlated with a reduction in damage by the insect. Further analysis of samples of four important horticultural crops (lettuce, tomato, pepper and cucumber) with unknown levels of resistance to thrips showed a significant genotypic variation in the concentrations of total aromatic amino acids relative to the total leaf protein. Accessions from each crop with low or high concentrations of aromatic amino acids in proteins were exposed to thrips larvae. Regression analysis showed a highly significant positive correlation between aromatic amino acid concentration in leaf protein and thrips damage, regardless of crop species. It is concluded that higher concentrations of aromatic amino acids in plant proteins are important for successful thrips development. These results provide plant breeders with a promising tool for indirect selection without using undesirable insect bioassays. 相似文献
