A character analysis reveals a clearly intermediate position of the tetraploidV. persica (2n = 28) between the two diploid speciesV. polita andV. ceratocarpa (both 2n = 14) which are morphologically rather different and have been placed by several authors in different sections of the genus.V. ceratocarpa is native to subhumid deciduous forests of the Caucasus and of the Elburz mountains (N. Iran);V. polita has its centre of variation in the Elburz range where it grows in therophyte habitats. Three other closely related species,V. bungei, V. siaretensis, andV. francispetae, are endemic to the Elburz range which is the main centre of diversity and variability of theV. agrestis group. This comprises all the above mentioned species and also two more European weeds:V. agrestis andV. opaca. Veronica polita, was probably originally native to open places in deciduous mountain forests, before becoming a weed in neolithic times and migrating to Europe; nowadays it has an almost world-wide distribution. The allotetraploidV. persica combines the ecological characters of its parents, the slightly xerophyticV. polita and the more mesophyticV. ceratocarpa, thus being preadapted to become a highly successful weed with a large ecological range. Plants of Taraxacum sellandii Dahlst., a microspecies adapted to fertile, and Taraxacum nordstedtii Dahlst., adapted to infertile soils, were cultured hydroponically, either on a complete nutrient solution or on one deprived of nitrogen, phosphorus, or potassium ions. For all four treatments, the growth and internal mineral concentration of the plants was monitored. For plants cultured on a complete nutrient solution, the uptake rates of nitrate, phosphate, and potassium ions were determined. Luxury consumption of the three macronutrients was computed as the excess of ion absorption over the ion uptake rates minimally required to sustain maximum growth. In these calculations the critical N, P, or K+ concentrations, earlier derived, were used as parameters describing the mineral status minimally required to allow maximum growth. Efficiency in use of the three macroelements at various levels of mineral accumulation was also computed. Finally, the response to phosphate starvation as related to phosphate uptake capacity and the accumulation of P was investigated.
The physiological properies investigated provide a causal background for the superior adaptation of T. nordstedtii as compared to T. sellandii to infertile sites. Taraxacum nordstedtii had a higher relative luxury consumption of NO3, H2PO-4, and K+, a higher efficiency in N and P use at N– and (severe) P-deficiency, respectively; and, after phosphate starvation, a relatively high preservation of phosphate uptake capacity and an enlargement of P storage. In combination with the low potential growth, luxury consumption will be particularly effective in T. nordstedtii in preventing or minimizing mineral deficiency. The distribution of minerals between cytoplasm and vacuoles as a factor in mineral use efficiency is discussed.  相似文献

The effect of HCO 3 - on ion absorption by young corn roots was studied in conditions allowing the independent control of both the pH of uptake solution and the CO2 partial pressure in air bubbled through the solution. The surface pH shift in the vicinity of the outer surface of the plasmalemma induced by active H+ excretion was estimated using the initial uptake rate of acetic acid as a pH probe (Sentenac and Grignon (1987) Plant Physiol. 84, 1367). Acetic acid and orthophosphate uptake rates and NO 3 - accumulation were slowed down, while 86Rb+ uptake and K+ accumulation rates were increased by HCO 3 - . These effects were similar to those induced by 4-(2-hydroxyethyl)-1-piperazineethane sulfonic acid/2-amino-2-(hydroxymethyl)-1,3-propanediol (Hepes-Tris). They were more pronounced when the H+ excretion was strong, were rapidly reversible and were not additive to those of Hepes-Tris. The hypothesis is advanced that the buffering system CO2/H2CO3/HCO 3 - accelerated the diffusion of equivalent H+ inside the cell wall towards the medium. Amid discussions of an agricultural crisis and the failure of largescale, mechanized, capitalintensive development schemes in Nigeria, the Kofyar of Plateau State provide a case study of farmers spontaneously expanding food crop production for the market, using indigenous lowenergy technology. Temporary, followed by permanent, migration from the Jos Plateau homeland to frontier settlements on the fertile Benue plains has been accompanied by a change from initial shifting cultivation in forest clearings to permanent, intensively tilled and fertilized homestead fields. Labor is organized primarily in households that have grown in size and complexity. Cooperative and exchange work groups are also important for meeting seasonal bottlenecks and providing the careful, disciplined cultivation that intensive agriculture requires. Kofyar now devote up to 50% of their labor to cash crops, and they purchase considerable quantities of manufactured goods and medical services. Their uncoerced adaptation to an environment of new land resources and market incentives suggests both the advantages of indigenous development with a minimum of state control or interference and the limitations of a conventional dependency theory perspective.  相似文献

The engineering considerations common to large scale chromatographic purification of proteins are reviewed. A discussion of the industrial chromatography fundamentals is followed by aspects which affect the scale of separation. The separation column geometry, the effect of the main operational parameters on separation performance, and the physical characteristics of column packing are treated. Throughout, the emphasis is on ion exchange and size exclusion techniques which together constitute the major portion of commercial chromatographic protein purifications. In all cases, the state of current technology is examined and areas in need of further development are noted.

The physico-chemical advances now underway in chromatographic separation of biopolymers would ensure a substantially enhanced role for these techniques in industrial production of products of new biotechnology.  相似文献

Summary The toxicity of chromium and tin on growth, photosynthetic carbon-fixation, oxygen evolution, heterocyst differentiation and nitrogenase activity ofAnabaena doliolum and its interaction with bivalent cations has been studied. Some interacting cations, viz. Ca2+, Mg2+ and Mn2+, substantially antagonised the toxic effects of chromium and tin with reference to growth, heterocyst differentiation and nitrogenase activity in the following hierarchal sequence: Ca2+ > Mg2+ > Mn2+. However, the sequence of hierarchy was Mg2+ > Ca2+ > Mn2+ for carbon fixation and Mn2+ > Mg2+ > Ca2+ for photosynthetic oxygen evolution. Synergistically inhibitory patterns were noticed for all the parameters, viz. growth,14CO2 uptake, oxygen evolution, heterocyst differentiation and nitrogenase activity ofA. doliolum when Ni2+, Co2+ and Zn2+ were combined with the test metals in the growth medium. These cations followed the following sequence of synergistic inhibition: Ni2+ > Co2+ > Zn2+. Among all the interacting cations, Ca2+, Mg2+ and Mn2+ exhibited antagonistic effects which relieved the test cyanobacterium from metal toxicity. In contrast to this, Ni2+, CO2+ and Zn2+ showed synergistic inhibition which potentiating the toxicity of test metals in the N2-fixing cyanobacteriumA. doliolum. It is evident from the present study that bivalent cations, viz. Ca2+, Mg2+, Mn2+, Ni2+, Co2+ and Zn2+, may appreciably regulate the toxicity of heavy metals in N2-fixing cyanobacteria if present in aquatic media.  相似文献

Ethylcholine mustard aziridinium ion (ECMA) was infused intracerebroventricularly (icv) to rats followed by measurement of two markers of presynaptic cholinergic neurons, choline acetyltransferase (ChAT) activity and high affinity choline transport (HAChT), in the hippocampus and cortex. Bilateral icv administration of 1, 2, or 3 nmol of ECMA per side produced dose-dependent reductions in each marker in the hippocampus, but not in the cortex, one week after treatment. Reductions of 52% and 46% for ChAT activity and HAChT, respectively, were produced in the hippocampus by 3 nmol ECMA. Measurement of these two markers at different times after icv infusion of 2 nmol ECMA/ventricle revealed that the activity of ChAT was reduced to a greater extent than was HAChT in the hippocampus 1 day and 1, 2, 4, and 6 weeks after treatment. The maximal reductions of ChAT activity and HAChT (61% and 53%, respectively) were reached between 1 and 2 weeks after ECMA administration. There was no evidence of regeneration of either marker at 4 or 6 weeks posttreatment. HAChT and ChAT activity in the cortex were not altered at any of the posttreatment times examined.ECMA-induced deficits in hippocampal ChAT activity and HAChT were not counteracted by the following treatments: (i) daily administration of GM1 ganglioside (10 mg/kg, intraperitoneally (ip)) from the day prior to infusion of ECMA until 2 weeks later; (ii) daily administration of GM1 ganglioside between 2 and 6 weeks after infusion of ECMA; and (iii) icv administration of nerve growth factor (NGF) twice per week for 2 weeks after ECMA treatment. Since similar treatments with NGF and GM1 ganglioside ameliorate lesions induced by other methods, these results indicate that the mechanism of lesion formation and the surviving cellular components influence the functional effects of neurotrophic factors. In contrast to the above results, treatment with vitamin E significantly attenuated ECMA-induced deficits of ChAT activity and HAChT. Further studies of the effects of vitamin E on the development of ECMA-induced deficits may help to elucidate the mechanism action of ECMA.  相似文献

The interactions of eight piperidine derivatives with nicotinic receptor complexes fromTorpedo californica electric organ were studied using [125I]alpha-bungarotoxin ([125I]BGT) as a probe for the acetylcholine binding site and [3H]perhydrohistrionicotoxin ([3H]H12-HTX) as a probe for a site associated with the receptor-gated ion channel.Cis- andtrans-2-methyl-6-n-undecanyl piperidines (MUP), major constituents of fire ant venom, had a high-affinity for [3H]H12-HTX binding sites (Ki=0.08–0.24 M), but had no affect on receptor binding. MUP affinity for [3H]H12-HTX binding sites was approximately doubled in the presence of 1 M carbamylcholine. Introduction of a 2-hydroxyl group to the undecanyl side channel had little effect on activity of the alkaloid. The analog 2,6- (but not 3,5-) dimethylpiperidine was a moderately active inhibitor of [3H]H12-HTX binding (K i-8.8 M). 2-Methylpiperidine was considerably less active (K i=600 M), although it was more potent than either 3- or 4-methylpiperidine. The affinities of 2,6-dimethylpiperidine and 2-methylpiperidine for [3H]H12-HTX binding sites were decreased in the presence of 1 M carbamylcholine. Carbamylcholine affinity for the receptor was increased by up to 7 fold in the presence of 10 and 32 M MUP, but was decreased in the presence of 2,6-dimethylpiperidine and 2-methylpiperidine. Thecis- andtrans-isomers of MUP were equipotent in producing each of its effects. In these actions, MUP resembles a variety of other compounds derived from 2,6-disubstituted piperidines, including histrionicotoxins, gephyrotoxins and pumiliotoxins. These studies establish the importance of alkyl substitutions in theortho position of the piperidine ring in conferring ion channel specificity, and the importance of substantial alkyl side chains in conferring the ability of channel blockers to stabilize the nicotinic receptor complex in high affinity, desensitized conformations.  相似文献

Synopsis Fish migration may be viewed as the product of two processes; the selection and tracking of optimal environmental conditions through time and space, and the use of predictive information about environmental structure to bias movements towards a goal. The establishment and maintenance of directional bias is based on the interaction of experience and instinct. The preoccupation of much fish orientation research with innate fixed patterns of behavior on one hand and hydrodynamics on the other has led us to underestimate the possibility that orientation is a flexible process relying on developmental sequences, calibration of the motor-sensory interaction based on experience and the learning of environmental pattern. Evidence illustrating how experience and learning may influence the direction of movement and how the goal is recognized is presented according to two general categories: (a) imprinting and early experience and (b), spatial learning, including the social transmission of migratory routes and directions. In the first category, the olfactory hypothesis of salmon homing is briefly reviewed and new data presented describing olfactory imprinting in Atlantic salmon,Salmo salar. In the second category, evidence is presented demonstrating the modifiability of sun-compass orientation and the ability of some fish species to learn the spatial distribution of landmarks. The role of social transmission in the migration of coral reef fishes is reviewed. The possible role of these learning phenomena in the formation of familiar area maps, route-based and location-based navigation and the critical distance factor is considered. The relationship between life history and the nature of learning in migratory orientation is discussed  相似文献
