The field metabolic rates (FMR) and rates of water flux were measured in two species of varanid lizards over five periods of the year in tropical Australia. The energetics of these species were further investigated by directly measuring activity (locomotion) and body temperatures of free-ranging animals by radiotelemetry, and by measuring standard metabolic rate (over a range of body temperatures) and activity metabolism in the laboratory. Seasonal differences in the activity and energetics were found in these goannas despite similar, high daytime temperatures throughout the year in tropical Australia. Periods of inactivity were associated with the dry times of the year, but the onset of this period of inactivity differed with respect to habitat even within the same species. Varanus gouldii, which inhabit woodlands only, were inactive during the dry and late dry seasons. V. panoptes that live in the woodland had a similar seasonal pattern of activity, but V. panoptes living near the floodplain of the South Alligator River had their highest levels of activity during the dry season when they walked long distances to forage at the receding edge of the floodplain. However, during the late dry season, after the floodplain had dried completely, they too became inactive. For V. gouldii, the rates of energy expenditure were 196 kJ kg–1 day–1 for active animals and 66 kJ kg–1 day–1 for inactive animals. The rates of water influx for these groups were respectively 50.7 and 19.5 ml kg–1 day–1. For V. panoptes, the rates of energy expenditure were 143 kJ kg–1 day–1 for active animals and 56 kJ kg–1 day–1 for inactive animals. The rates of water influx for these two groups were respectively 41.4 and 21.0 ml kg–1 day–1. We divided the daily energy expenditure into the proportion of energy that lizards used when in burrows, out of burrows but inactive, and in locomotion for the two species during the different seasons. The time spent in locomotion by V. panoptes during the dry season is extremely high for a reptile (mean of 3.5 h/day spent walking), and these results provide an ecological correlate to the high aerobic capacity found in laboratory measurements of some species of varanids.  相似文献

The esterification of lauric acid with geraniol catalyzed by the commercially immobilized lipase preparation from Mucor miehei, Lipozyme(R), was studied in well-stirred flasks. The enzyme support was characterized in terms of its internal and external surface area, protein location, and protein content. It was found that the enzyme was mainly located on the external surface of the support, therefore, internal diffusional limitations were not important. It was also shown that the protein content of the support depends on the size of the particle, with smaller particles containing higher amounts of protein per unit weight. Under the conditions studied, the reaction was not under external mass transfer limitations, and the initial reaction rate depended on the size of the support particles. This was mainly due to the different protein contents on the support as a function of particle size and not to internal or external mass transfer limitations. Also, it was found that the inhibition exerted by water was predominantly a physical effect due to its accumulation around the enzyme. The physiological states with respect to cell growth and ethanol production in a yeast fed-batch culture expressed in linguistic form could be recognized on-line by fuzzy inferencing based on error vectors. The error vector was newly defined here in a macroscopic elemental balance equation. The physiological states for cell growth and ethanol production were characterized by error vectors using many experimental data from fed-batch cultures. Fuzzy membership functions were constructed from the frequency distributions of the error vectors and state recognition was performed by fuzzy inferencing. In particular, an unusual physiological state for a yeast cultivation, in which aerobic ethanol production was accompanied by very low cell growth, could be recognized accurately. According to the results of the state recognition, an energy parameter, the P/O ratio in the metabolic reaction model was adaptively estimated, and the cell growth was successfully evaluated with the estimated P/O. Seeds of cauliflower cv. Hipop and Brussels sprouts cv. Asmer Aries were aged at 20% moisture content for 24 h; all seeds retained a germination of over 70% after ageing although the mean germination time increased. Prolonged aerated hydration for up to 32 h at 20°C followed drying resulted in improved performance of both unaged and aged cauliflower seeds and aged Brussels sprouts. Thus, all seed showed reductions in the mean germination time to the extent that after 32 h hydration the aged cauliflower seeds performed as well as high quality unaged seed. The improvement of aged seeds was also revealed an increase in germination after the controlled deterioration test following up to 24 h (cauliflower) or 32 h (Brussels sprouts) aerated hydration. This increase was indicative of a decrease in the extent of deterioration present after aerated hydration. Deleterious effects of prolonged hydration were observed in Brussels sprouts after 32 h although these may be explained desiccation injury after treatment since radicle emergence had occurred during hydration. The improvements in seed performance may be explained the activation of metabolic repair occurring during the early part of the hydration period therereducing the extent of deterioration that has been sustained during ageing, with further improvements due to the advancement of the germination process.  相似文献

1. 1. The thermoregulatory responses to manipulations of photoperiod in wood mice (Apodemus sylvaticus), which were drawn from a population living at a high latitude (57°N) were studied.
2. 2. Mice captured in spring were acclimated to two different photoperiod regimes 16L:8D and 8L:16D at a constant ambient temperature of 24°C, for 3 weeks.
3. 3. Daily rhythms of body temperature, oxygen consumption and body temperature at various ambient temperatures, nonshivering thermogenesis (the response to a noradrenaline injection) and body mass were measured. Minimal overall thermal conductance was calculated for both groups.
4. 4. Acclimation to long photophase increased the thermoregulatory abilities at relatively high ambient temperatures while that of long-scotophase increased thermoregulatory abilities at low ambient temperatures.
5. 5. Changes in photoperiod may therefore be used as cues for seasonal acclimatization of thermoregulatory mechanisms in this population of wood mice.

Metabolic profiling by capillary liquid chromatography-electrospray mass spectrometry was used to monitor shifts in the proferrioxamine profiles of Erwinia amylovora in response to externally supplied potential proferrioxamine precursors, selected stable-isotope-labeled precursors and atypical precursors. Based on the qualitative and quantitative shifts in the proferrioxamine profiles, lysine and arginine are unambiguous, and agmatine, ornithine, diaminobutyric acid and the corresponding C3–5 diamines are highly likely precursors for proferrioxamine biosynthesis in E. amylovora. 5-Hydroxylysine (Hyl), a recently discovered growth inhibitor for E. amylovora, suppresses proferrioxamine production. The Hyl-induced growth inhibition can be reversed by basic amino acids. Summary In order to quantify female and male fitness values of clones in a Pinus sylvestris L. seed orchard, multilocus-genotypes of parental clones were compared with those of open pollinated seeds in the bulked orchard crop. Female and male contributions to individual seeds were distinguished by observing enzyme gene loci active in both endosperm and embryo tissue. Seed probes from two successive flowering periods were surveyed. The female and male fitnesses of five parental clones measured relative to the population mean were derived. The contributions of four clones were found to be sexually asymmetric. One clone, for instance, made exclusively female contributions in one flowering period. Variations existed in fitness values between clones. Deviations in sex specificity occurred between flowering periods: one clone contributed asymmetrically in both periods, but in sexually reversed proportions. A method to comprehensively quantify and illustrate the observed phenomena is proposed.  相似文献

The East Usambara Mountain forests constitute what is probably one of the richest biological communities in Africa in terms of plant and animal species numbers and endemic taxa. This review presents brief accounts of the flora and of three invertebrate and four vertebrate groups and shows the percentage endemic taxa to vary from 2% (mammals) to 95% (millipedes) as a proportion of the true forest species. Notes are given on the geology, soils, climate and present land use of the Usambaras. Biological richness is considered to be due to long periods of isolation and geological stability coupled with periods of species immigration during times of re-establishment of a continuous forest cover. The nature of the endemic elements is briefly discussed. Evidence is given to show that the forests are subject to increasing pressure from legal and illegal encroachment due to agriculture (tea, cardamon, subsistence) and forestry timber operations. Air photograph analysis shows a forest decrease of some 50% in the vicinity of Amani from 1954 to 1976. The low conservation status of most forest reserves and the lack of detailed knowledge on the distribution, status and biology of the endemic species means present conservation efforts are poor and haphazard. This review calls for greatly increased research inputs and a complete halt to all exploitation of natural forest areas until a long term conservation land use plan can be implemented.  相似文献

Behm C. A. and Bryant C. 1982. Phosphoenolpyruvate carboxykinase from Fasciola hepatica. International Journal for Parasitology12: 271–278. The kinetic properties of a partially purified preparation of phosphoenolpyruvate carboxykinase (PEPCK) from F. hepatica were examined. The pH optimum for the carboxylation reaction is 5.8–6.2. The enzyme is more active with Mn2+ than Mg2+ and the Mn2+ saturation curve was sigmoid. Apparent Km values for the substrates GDP, IDP, PEP and HCO3? were determined and found to be in the same range as those reported for other helminths except that the enzyme is less sensitive to low PEP concentrations. GTP and ATP at 0.5 and 1.0 mM inhibit the enzyme; the GTP inhibition was greater in the presence of Mg2+ than Mn2+ and was competitive with GDP. It was concluded that the activity of PEPCK from F. hepatica is controlled by the concentration of reactants and the ambient pH, that the accumulation of GTP is a sensitive mechanism for inhibiting the carboxylation reaction and that PEPCK activity in the cytosol is likely to be favoured over that of pyruvate kinase except when pH is high and PEP concentration low.  相似文献

The rubidium content of whole blood was estimated by instrumental neutron activation analysis. In 46 healthy children it amounts to {ie193-1} g/g dry weight. There was no difference between the values found for infants, toddlers, and school children. In 29 dietetically treated patients with phenylketonuria and maple-syrup-urine disease the values were significantly lower than in healthy children. During the first three months of diet therapy the rubidium levels remained in the lower range of the normal values, decreasing to about 60% of the mean of normal values later on. With increasing length of diet therapy these values tended to decrease. It remains questionable whether these decreased levels reflect only an induced biochemical phenomenon without biological importance, or whether they are the first signs of a deficiency syndrome.  相似文献
