The presence of Ca2+-ATPase activities with high-affinity sites for Ca2+ in brush border as well as basolateral plasma membranes of rat duodenal epithelium has been reported previously (Ghijsen, W.E.J.M. and van Os, C.H. (1979) Nature 279, 802–803). Since both plasma membranes contain alkaline phosphatase (EC 3.1.3.1), which also can be stimulated by Ca2+, the substrate specificity of Ca2+-induced ATP-hydrolysis has been studied to determine whether or not alkaline phosphatase and Ca2+-ATPase are two distinct enzymes. In basolateral fragments, the rate of Ca2+-dependent ATP-hydrolysis was greater than that of ADP, AMP and p-nitrophenylphosphate at Ca2+ concentrations below 25 μM. At 0.2 mM Ca2+ the rates of ATP, ADP, AMP and p-nitrophenylphosphate hydrolysis were not significantly different. In brush border fragments the rates of ATP, ADP and AMP hydrolysis were identical at low Ca2+, but at 0.2 mM Ca2+, Ca2+-induced hydrolysis of ADP and AMP was greater than either ATP or p-nitrophenylphosphate. Alkaline phosphatase in brush border and basolateral membranes was inhibited by 75% after addition of 2.5 mM theophylline. Ca2+-stimulated ATP hydrolysis at 1 μM Ca2+ was not sensitive to theophylline in basolateral fragments while the same activity in brush border fragments was totally inhibited. At 0.2 mM Ca2+, Ca2+-induced ATP hydrolysis in both basolateral and brush border membranes was sensitive to theophylline. Oligomycin and azide had no effect on Ca2+-stimulated ATP hydrolysis, either at low or at high Ca2+ concentrations. Chlorpromazine fully inhibited Ca2+-stimulated ATP hydrolysis in basolateral fragments at 5 μM Ca2+, while it had no effect in brush border fragments. From these results we conclude that, (i) Ca2+-ATPase and alkaline phosphatase are two distinct enzymes, (ii) high-affinity Ca2+-ATPase is exclusively located in basolateral plasma membranes, (iii) alkaline phosphatase activity, present on both sides of duodenal epithelium, is stimulated slightly by low Ca2+ concentrations, but this Ca2+-induced activity is inhibited by theophylline and shows no specificity with respect to ATP, ADP or AMP.  相似文献

Summary A detailed stereological analysis has been made of the organelle content of rabbit acinar cells during the restoration of granule stores following extensive degranulation with isoprenaline (IPR). Rabbits were sacrificed 2, 4, 8, 12 and 16 h after IPR administration and the volumes and proportions of intracellular organelles were compared with those of untreated glands.At 2 h only 5% of cell volume was occupied by secretion granules, but there was already evidence of nascent granule formation. The volume per cell of secretion granules increased in sigmoid fashion and by 16 h amounted to 350 m3/cell, 36% of cell volume, which are values similar to those of the control replete glands. IPR treatment caused some initial swelling of the cells, and there were transient increases in the volumes of several compartments. However, the volume of mitochondrial and lysosomal compartments had returned to control levels by 4 h and that of the nucleus by 8 h. The greatest increase was in the volume of the rough endoplasmic reticulum which had increased by nearly 70% by 2 h and remained enlarged throughout the period of restitution. However, neither the volume nor the proportion of the smooth membraned compartment varied throughout the period of analysis.The results are analysed in the light of the overall response of the cells to IPR and the interaction of the organelles during the synthetic phase of the secretory cycle. They are presented as a basis for ensuing studies of the granule populations and the membrane composition of the cells during the restoration of granule stores.  相似文献

Summary The fate of spermatozoa that do not participate in fertilization was investigated by electron microscopy. After artificial insemination, we observed several spermatozoa between the fibers of the outer layer of the vitelline membrane of the ovum. One or more spermatozoa were also found in a phagocytic vesicle of macrophages located in the intercellular space of the mucosal epithelium of the infundibulum or in the outer layer of the vitelline membrane.From these observations, we assume that the superfluous spermatozoa in the lumen of the anterior part of the oviduct might be removed by inclusion into the outer layer of the vitelline membrane and by phagocytosis by macrophages.The authors are greatly indebted to Assoc. A web-like reticulum underlying the human erythrocyte membrane was studied at a resolution of 5–10 nm by means of a scanning electron microscope. The network was visualized in isolated membranes (ghosts) torn open to reveal their interior space and in residues derived from ghosts extracted with Triton X-100. It formed a continuous (rather than patchy) cover over the entire cytoplasmic surface, except where lifted off or torn away. Filaments (5–40 nm in diameter), annular figures (40–60 nm in diameter), and nodes (30–100 nm in diameter) were prominent in different networks. The dimensions of the filaments and the interstices in the reticulum varied with conditions, suggesting that the network has elastic properties. This reticulum is probably related to the erythrocyte membrane proteins spectrin and actin.  相似文献

Evidence is presented that Polian vesicles of the sea cucumber, Holothuria cinerascens, a member of an echinoderm class considered close to the vertebrate evolutionary line, are organs of inflammatory (including immunologic) responsiveness. Summary Intracellular microelectrode techniques were employed to study the mechanism by which amphotericin B induces a transient mucosa-negative transepithelial potential (V ms) in the gallbladder ofNecturus. When the tissue was incubated in standard Na-Ringer's solution, the antibiotic reduced the apical membrane potential by about 40 mV, and the basolateral membrane potential by about 35 mV whereas the transepithelial potential increased by about 5 mV. The electrical resistance of the apical membrane fell by 83%, and that of the basolateral membrane by 40%; the paracellular resistance remained unchanged. Circuit analysis indicated that the equivalent electromotive forces of the apical and basolateral membranes fell by 35 and 11 mV, respectively. Changes in potentials and resistances produced by ionic substitutions in the mucosal bathing medium showed that amphotericin B produces a nonselective increase in apical membrane small monovalent cation conductance (K, Na, Li). In the presence of Na-Ringer's on the mucosal side, this resulted in a reduction of the K permselectivity of the membrane, and thus in a fall of its equivalent emf. During short term exposure to amphotericin B,P Na/P Cl across the paracellular pathway did not change significantly, whereasP K/P Na doubled. These results indicate that V ms is due to an increase of gNa across the luminal membranes of the epithelial cells (Cremaschiet al., 1977,J. Membrane Biol. 34:55); the data do not support the alternative hypothesis (Rose & Nahrwold, 1976.J. Membrane Biol. 29:1) that V ms results from a reduction in shuntP Na/P Cl acting in combination with a rheogenic basolateral Na pump.  相似文献

Interactions between spectrin and the inner surface of the human erythrocyte membrane have been implicated in the control of lateral mobility of the integral membrane proteins. We report here that incubation of “leaky” erythrocytes with a water-soluble proteolytic fragment containing the membrane attachment site for spectrin achieves a selective and controlled dissociation of spectrin from the membrane, and increases the rate of lateral mobility of fluorescein isothiocyanate-labeled integral membrane proteins (> 70% of label in band 3 and PAS-1). Mobility of membrane proteins is measured as an increase in the percentage of uniformly fluorescent cells with time after fusion of fluorescent with nonfluorescent erythrocytes by Sendai virus. The cells are permeable to macromolecules since virus-fused erythrocytes lose most of their hemoglobin. The membrane attachment site for spectrin has been solubilized by limited proteolysis of inside-out erythrocyte vesicles and has been purified (V). Bennett, J Biol Chem 253:2292 (1978). This 72,000-dalton fragment binds to spectrin in solution, competitively inhibits association of 32P-spectrin with inside-out vesicles with a Ki of 10?7M, and causes rapid dissociation of 32P-spectrin from vesicles. Both acid-treated 72,000-dalton fragment and the 45,000 dalton-cytoplasmic portion of band 3, which also was isolated from the proteolytic digest, have no effect on spectrin binding, release, or membrane protein mobility. The enhancement of membrane protein lateral mobility by the same polypeptide that inhibits binding of spectrin to inverted vesicles and displaces spectrin from these vesicles provides direct evidence that the interaction of spectrin with protein components in the membrane restricts the lateral mobility of integral membrane proteins in the erythrocyte.  相似文献
