The medial forebrain bundle (MFB) was partially lesioned with 6-hydroxydopamine (6-OHDA) in order to investigate the effect of deficient central noradrenergic regulation on thyrotropin (TSH) secretion in the rat. 6-OHDA injection into the MFB significantly reduced the noradrenaline (NA), dopamine (DA) and serotonin (5-HT) content of the whole hypothalamus. NA and 5-HT concentrations were also significantly decreased in the paraventricular nucleus (PVN). The MFB lesion did not affect the clonidine (250 g/kg, i.p.) induced stimulation of TSH release or the isoproterenol (1 mg/kg i.p.) induced decrease in TSH levels. Thyrotropin releasing hormone (TRH, 5 g/kg i.v.) caused a similar significant stimulation of TSH secretion in lesioned and non-lesioned rats. The present results do not support the hypothesis that the blunted TSH response to TRH observed in depressed patients results from a deficiency in noradrenergic neurotransmission.  相似文献

Summary Cytoplasmic streaming in the freshwater, coencytic green alga,Dichotomosiphon tuberosus, is regulated by light. Conspicuous changes are observed in the number of microtubules cross-linked together in bundles when the cytoplasmic streaming is modulated by light. In an attempt to identify the cross-linker, we stainedD. tuberosus cells with antibodies specific for several different microtubules-associated proteins (MAPs) from vertebrates. Antibodies raised against bovine adrenal 190 kDa MAP stained the algal cells, and the pattern of staining was quite similar to that obtained with tubulin-specific antibodies. Examination by immunoelectron microscopy revealed that the antibodies specific for the 190 kDa microtubule-associated protein (MAP) were located along the microtubules. Western blotting demonstrated that the antibodies crossreacted with a peptide fromD. tuberosus with a molecular weight of about 90 kDa. This peptide was heat-stable, a property shared by the bovine 190 kDa MAP. Moreover, this 90 kDa peptide, crossreacted with antibodies raised against a synthetic peptide, identical to the tubulin-binding domain found in the 190 kDa MAP and in a tau protein. Partially purified 90 kDa protein fromD. tuberosus has the ability to bundle microtubules when mixed with a tubulin fraction fromD. tuberosus, in the presence of taxol. 1. Hypertension can be induced by some types of stress in the rat. The aim of the present work was to study the putative implication of brain norepinephrine (NE) in blood pressure increase due to social deprivation stress. 2. The effects of 6-hydroxydopamine (6-OHDA) lesions of the ventral noradrenergic bundle (VNEB) on the hypertensive response induced by brief social deprivation stress in young Wistar rats were examined. NE, dopamine (DA), and epinephrine (EPI) levels were measured by HPLC coupled with electrochemical detection in two brain areas (hypothalamus and medulla oblongata) relevant for blood pressure regulation. 3. VNEB lesions prevented the hypertensive response produced by isolation. Twelve or 20 days after 6-OHDA administration, NE and EPI but not DA levels decreased in the hypothalamus of the lesioned rats. In contrast, no catecholamine changes were detected in medulla oblongata. 4. These data suggest that the VNEB plays a role in the triggering of the hypertensive response induced by social deprivation stress in young Wistar rats.  相似文献

Summary We have previously shown that pertussis toxin (PTX) stimulates delayed-onset, [Ca2–] a -dependent catecholamine (CA) release from bovine chromaffin cells. We now show that this effect of PTX is inhibited in part (50%) by dihydropyridine Ca2–-channel antagonists niludipine and nifedipine, and is potentiated by the dihydropyridine Ca2+-channel agonist Bay K-8644. We and others have shown that pretreatment of chromaffin cells with PTX results in enhanced catecholamine secretion in response to high [K] a , nicotine and muscarine, and here we extend these observations by showing that toxin pretreatment also enhances the secretory response to [Ba2+] a . All these data are consistent with the concept that PTX may act on Ca2– channels. To examine the possibility of a direct action of the toxin on the voltage-gated L-type Ca2+ channel known to be present in these cells, we studied the effects of the toxin on whole cell Ca2+ currents. We found and report here that spontaneous electrical activity was considerably increased in PTX-treated cells. Our measurements of whole cell inward Ca2+ currents indicate that the underlying mechanism is a marked shift of the activation curve of the L-type Ca2+ current along the voltage axis towards more negative potentials. While treatment of the cells with PTX had no effect on L-type Ca2+-channel conductance (6 nS/cell at 2.6mm [Ca2+] a ). PTX evoked the activation of a new class of Ca2+-selective channels (5 pS in 25mm [Ca2+]pipet), which are rather insensitive to membrane potential. We have termed theseG-type calcium channels. These data suggest that treatment with PTX not only increases the probability of L-type Ca2+-channel activation at more negative potentials, but also increases the probability of opening of an entirely new, voltage-independent, Ca2+ channel. These actions of PTX should promote Ca2+ entry and might explain the stimulation by the toxin of CA secretion from medullary chromaffin cells in culture.  相似文献

Abstract: Catecholamine secretion has been measured with electrochemical techniques from isolated, single adrenal medullary chromaffin cells with carbon-fiber microelectrodes. The electrode tip, which is of similar dimensions to the cell, is placed adjacent to the cell to enable the measurement of local secretion. Secretion is caused by exposing the cell to nanoliter volumes of solution containing nicotinic receptor agonists or depolarizing agents. The identification of secreted substances is made with cyclic voltammetry at both bare electrodes and electrodes coated with a perfluorinated cationexchange polymer. Catecholamine secretion is induced by nicotine (10–500 μ M ), carbamylcholine (1 m M ), and K+ (60 m M ). All agents that induce secretion lead to a broad envelope of secreted catecholamines on which sharp concentration spikes are superimposed. The concentration spikes can be monitored with a time resolution of tens of milliseconds when the electrodes are used in the amperometric mode. Release induced by nicotine and K+ is inhibited by Cd2+ (0.5 m M ), and hexamethonium selectively blocks the nicotineinduced secretion. The actions of nicotine are found to continue for a longer period of time than those of the other secretagogues tested.  相似文献

Based on the heavy-atom coordinates determined by the electron microscopy for the seven main helical regions of bacteriorhodopsin with the all-trans retinal isomer, energy optimizations were carried out for helix bundles containing the all-trans retinal and 13-cis retinal chromophores, respectively. A combination of simulated annealing and energy minimization was utilized during the process of energy optimization. It was found that the 7-helix bundle containing the all-trans isomer is about 10 kcal/mol lower in conformational energy than that containing the 13-cis isomer. An energetic analysis indicates that such a difference in energy is consistent with the observation that absorption of a 570-nm proton is required for the conversion of a bacteriorhodopsin from its all-trans to 13-cis form. It was also found that the above conversion process is accompanied by a significant conformational perturbation around the chromophore, as reflected by the fact that the beta-ionone ring of retinal moves about 5.6 A along the direction perpendicular to the membrane plane. This is consistent with the observation by Fodor et al. (Fodor, S.P.A., Ames, J.B., Gebhard, R., van der Berg, E.M.M., Stoeckenius, W., Lugtenburg, J., & Mathies, R.A., 1988, Biochemistry 27, 7097-7101). Furthermore, it is interesting to observe that although the retinal chromophore undergoes a significant change in its spatial position, the orientation of its transition dipole changes only slightly, in accord with experimental observations. In other words, even though orientation of the retinal transition dipole is very restricted, there is sufficient room, and degrees of freedom, for the retinal chromophore to readjust its position considerably. This finding provides new insight into the subtle change of the retinal microenvironment, which may be important for revealing the proton-pumping mechanism of bacteriorhodopsin. The importance of electrostatic and nonbonded interactions in stabilizing the 7-helix bundle structure has also been analyzed. Electrostatic interactions favor an antiparallel arrangement among adjacent helices. Nonbonded interactions, however, drive most of the closely packed helices into an arrangement in which the packing angles lie around -160 degrees, a value very near the -154 degrees value computed earlier as the most favorable packing arrangement of two poly(Ala) alpha-helices (Chou, K.-C., Némethy, G., & Scheraga, H.A., 1983, J. Phys. Chem. 87, 2869-2881). The contribution of an omega-conotoxin GVIA (omega Cgtx)-sensitive Ca2+ influx pathway to the effects of angiotensin II (AII) receptor activation was examined in bovine adrenal medullary (BAM) cells. Pretreatment of BAM cells with 10(-6) M omega Cgtx blocked stimulation of exocytosis by the degradation-resistant analogue, sarcosine1-angiotensin II (S1-AII). In contrast, omega Cgtx had no effect on basal secretion, nor did it inhibit [3H]norepinephrine and [32P]ATP release in response to bradykinin, another phospholipase C-linked receptor agonist. Similarly, omega Cgtx pretreatment inhibited the stimulation of 45Ca2+ uptake by S1-AII, but did not affect the response to bradykinin. This selective inhibition did not appear to be due to blockade of AII receptors by omega Cgtx, as the accumulation of 3H-labeled inositol phosphates in response to S1-AII was not inhibited. The peak S1-AII-stimulated increase in the intracellular free Ca2+ concentration (Cai) in fura 2-loaded BAM cells also was not significantly reduced by omega Cgtx (or by stimulating in nominally Ca(2+)-free buffer), indicating that this response is dependent on intracellular Ca2+ pools. However, a small omega Cgtx-sensitive Cai response was detected after depletion of intracellular Ca2+ pools with ionomycin. This study shows that AII receptors, but not bradykinin receptors, are linked to an omega Cgtx-sensitive Ca2+ influx pathway in BAM cells.  相似文献

Summary The kinetic and steady-state characteristics of calcium currents in cultured bovine adrenal chromaffin cells were analyzed by the patch-clamp technique. Whole cell inward Ca2+ currents, recorded in the presence of either 5.2 or 2.6mm Ca2+ exhibited a single, noninactivating component. To analyze the effects of Ca2+ and Bay K-8644 on the kinetics of the Ca2+ currents, we used a modified version of the Hodgkin-Huxley empirical model. At physiological [Ca2+] (2.5mm) the midpoint of the steady-state Ca2+-channel activation curve lay at –6.9 mV. Increasing the [Ca2+] to 5.2mm shifted the midpoint by –4.3 mV along the voltage axis. At the midpoint, changes in potential of 7.8 mV (for 5.2mm Ca2+) and 9.2 mV (for 2.5mm Ca2+) induced ane-fold change in the activation of the current. Increasing [Ca2+]0 from 2.5 to 5.2mm induced a marked increase in the rate constant for turning on the Ca2+ permeability. Conductances were estimated from the slope of the linear part of the currentvoltage relationships as 8.7 and 4.2 nS in the presence of 5.2 and 2.5mm Ca2+, respectively. Incubation of the cells in the presence of Bay K-8644 at increasing concentrations from 0.001 to 0.1 m increased the slope conductance from 4.2 to 9.6 nS. Further increases in the concentration of Bay K-8644 from 1 to 100 m induced a marked reduction in the conductance to 1.1 nS. In the presence of Bay K-8644 (0.1 m) the midpoint of the activation curve was shifted by 6.1 mV towards more negative potentials, i.e., from –6.9 to –13 mV. At the midpoint potential of –13 mV, a change in potential of 6.9 mV caused ane-fold change in Ca2+ permeability. The kinetic analysis showed that Bay K-8644 significantly reduced the size of the rate constant for turning off the Ca2+ permeability.  相似文献

Electrical stimulation of the medial forebrain bundle, in a manner that augmented the release of dopamine in the forebrain, rapidly increased the striatal content of preproenkephalin (but not preprotachykinin) mRNA. This effect was mimicked by administration of either the indirect (dopamine-releasing) agonist methamphetamine or by the D-2 dopamine receptor agonist quinpirole, but not by the D-1 agonist SKF 38393. These data suggest that D-2 receptors, which mediate a stimulatory effect on enkephalin gene expression, may be subsaturated under basal conditions and, therefore, responsive to increases in synaptic dopamine.  相似文献

The loading and transport functions of vascular bundles in maize (Zea mays L.) leaf strips were investigated by microautoradiography after application of 14CO2. The concentrations of 14C-contents in thin-walled sieve tubes of individual bundles in the loading and transport regions were determined by digital image analysis of silver-grain density over the sieve tubes and compared. In the loading region, relatively high concentrations of 14C-contents were found in the thin-walled sieve tubes of small bundles and in the small, thin-walled sieve tubes of the intermediate bundles; the concentration of 14C-label in large bundles was very low. In the transport region, at a transport distance of 2 cm, all of the small bundles contained 14C-assimilates, but generally less than the same bundles did in the loading region; by comparison, at that distance intermediate and large bundles contained two-to threefold more 14C-assimilates than the same bundles in the loading region. The lateral transfer of assimilates from smaller to larger bundles via transverse veins could be demonstrated directly in microautoradiographs. A reverse transport from larger to smaller bundles was not found. At a transport distance of 4 cm, all large and intermediate bundles were 14C-labeled, but many of the small bundles were not. Although all longitudinal bundles were able to transport 14C-asimilates longitudinally down the blade, it was the large bundles that were primarily involved with longitudinal transport and the small bundles that were primarily involved with loading.  相似文献
