Articular cartilage lubricates the contact surfaces in human joints and provides a shock-absorbing effect which protects the joint under dynamic loading. However, this shock-absorbing effect is gradually reduced as the result of normal wear, tear and aging-related cartilage loss. Thus, with the increasing average human life expectancy, the issue of joint health has attracted significant interest in recent decades. In developing new materials for the repair or regeneration of damaged articular cartilage, it is essential that the difference in the mechanical properties of healthy and damaged cartilages is well-understood. In the present study, the hardness and Young's modulus of damaged and healthy porcine articular cartilage samples are evaluated via a quasi-static nanoindentation technique. A dynamic mechanical analysis method is then applied to determine the viscoelastic properties of the two samples. The results presented in this study provide a useful insight into the mechanical properties of articular cartilage at the mesoscale, and therefore fill an important gap in the literature. 相似文献

During the 5 s of the acrosome reaction of Limulus polyphemus sperm, a 60-microm-long bundle of scruin-decorated actin filaments straightens from a coiled conformation and extends from the cell. To identify the motive force for this movement, we examined the possible sources of chemical and mechanical energy and show that the coil releases approximately 10-13 J of stored mechanical strain energy, whereas chemical energy derived from calcium binding is approximately 10-15 J. These measurements indicate that the coiled actin bundle extends by a spring-based mechanism, which is distinctly different from the better known polymerization or myosin-driven processes, and that calcium initiates but does not power the reaction. 相似文献

Begomoviruses of the Geminiviridae are usually transmitted by whiteflies and rarely by mechanical inoculation. We used tomato leaf curl New Delhi virus (ToLCNDV), a bipartite begomovirus, to address this issue. Most ToLCNDV isolates are not mechanically transmissible to their natural hosts. The ToLCNDV-OM isolate, originally identified from a diseased oriental melon plant, is mechanically transmissible, while the ToLCNDV-CB isolate, from a diseased cucumber plant, is not. Genetic swapping and pathological tests were performed to identify the molecular determinants involved in mechanical transmission. Various viral infectious clones were constructed and successfully introduced into Nicotiana benthamiana, oriental melon, and cucumber plants by Agrobacterium-mediated inoculation. Mechanical transmissibility was assessed via direct rub inoculation with sap prepared from infected N. benthamiana. The presence or absence of viral DNA in plants was validated by PCR, Southern blotting, and in situ hybridization. The results reveal that mechanical transmissibility is associated with the movement protein (MP) of viral DNA-B in ToLCNDV-OM. However, the nuclear shuttle protein of DNA-B plays no role in mechanical transmission. Analyses of infectious clones carrying a single amino acid substitution reveal that the glutamate at amino acid position 19 of MP in ToLCNDV-OM is critical for mechanical transmissibility. The substitution of glutamate with glycine at this position in the MP of ToLCNDV-OM abolishes mechanical transmissibility. In contrast, the substitution of glycine with glutamate at the 19th amino acid position in the MP of ToLCNDV-CB enables mechanical transmission. This is the first time that a specific geminiviral movement protein has been identified as a determinant of mechanical transmissibility. 相似文献

植物可以利用体内次生代谢产物的变化来抵御昆虫取食和机械损伤.芥子油苷是拟南芥的主要次生代谢产物.通过剪刀剪取叶片(40%面积)对温室培养的拟南芥幼苗莲座叶进行机械损伤处理,观察机械损伤后8个时间点拟南芥叶片中不同种类芥子油苷含量和组合模式的变化.结果表明机械损伤后3 h叶片中芥子油苷总含量开始明显上升,脂肪族和吲哚族芥子油苷含量在损伤后3 h也都显著高于损伤前.在检测到的12种芥子油苷中,4-甲基亚磺酰丁基芥子油苷(4-methylsulphinylbutyl GS,4MSOB)的含量最多,占芥子油苷总量的48.5%,并且在损伤3 h后含量增加.4MSOB含量的变化成为影响莲座叶中芥子油苷组合模式的主导因素.其它各种芥子油苷在损伤后不同时间点的变化也存在差异. 相似文献

The fluorescent dye retrograde tracing technique, using fast blue in combination with fluorogold, was used to examine thalamocortical projections from the ventrobasal complex to primary somatosensory cortex in chronic spinal cats that sustained T12 cord transection at 2 weeks of age. Following cord transection at this age, it has been shown that forelimb afferents can excite the deprived hindlimb projection zone, in addition to the region of somatosensory cortex that they normally occupy (McKinley et al, 1987). These two regions of cortex are separated by over 10 mm, thus facilitating the determination of whether the forelimb representation in “hindlimb cortex” is derived from the sector of the ventrobasal complex of the thalamus representing the forelimb, hindlimb, or both. Injections of the two dyes into separate regions of the cortex that were excited by the same peripheral forelimb receptive fields produced single labeling of two nonoverlapping clusters of thalamic neurons. This finding suggests that the projections for these two areas are independent and distinct, and indicates that altered thalamocortical projections do not contribute the critical component underlying reorganizational changes observed at the cortical level after spinal cord transection. It is hypothesized that the degree of reorganization required to achieve the magnitude of change observed in the cortex must occur below the level of the thalamocortical relay. 相似文献

Exclusion effects on vegetation characteristics and their correlation to soil factors in the semi-arid rangeland of Mu Us Sandland, China 总被引:1,自引:0,他引:1
Exclusion has been applied as a main measure for re-vegetation all over the world. This paper, by comparing the results of
year-round exclusion, seasonal exclusion, and non-exclusion, quantified the vegetation variations under three different exclusion
measures and their correlation to soil factors. The analysis results for community species component and plant diversity using
multi-response permutation procedures (MRPPs) showed that exclusion did change the species component and increase plant diversity
remarkably, while the period of exclusion had no significant influence on these two community features. The indicator species
analysis and calculation of similarity indices indicated that community for year-round exclusion were becoming xerophytization
and unpalatability, and showed highly spatial heterogeneity of plant species distribution, whereas community for seasonal
exclusion was under stable non-equilibrium condition. Detrended correspondence analysis (DCA) and detrend canonical correspondence
analysis (DCCA) results of relationship between plant species and soil variables demonstrated that soil moisture was a controlling
factor for plant species component, microbiotic soil crust cover, soil organic matter, and soil bulk density had significant
effects on soil moisture, among which microbiotic soil crust was a leading factor owing to its limitation to rainfall infiltration
on the one hand, and its constraints to entrance of herbaceous seeds into soil or to germination of soil seeds on the other
hand. As a result of long-term removal of animal grazing, crust kept intact in year-round exclusion community, which was a
main reason of community xerophytization. It was also obvious from ordination results that some important environmental factors,
such as tempo-spatial change of rainfall and corresponding tempo-spatial change of soil moisture, were neglected during direct
gradient analysis. In addition, biodiversity was close related to soil nutrients as well as to soil moisture condition (soil
water content and crust cover), and it had positive relation to available N, and negative relation to available P. Higher
soil N had advantage to non-leguminous plants growth on nutrition-poor sand land definitely. The impact of P to community
component was unclear and should be studied from plant physiology. Further researches on non-equilibrium theory in semi-arid
rangeland will provide a scientific and flexible animal development paradigm for being implementing livestock fen-raising
and grazing-forbidden policies in China.

Objective: To conduct a clinical and economic evaluation of outpatient weight loss strategies in overweight and obese adult U.S. women. Research Methods and Procedures: This study was a lifetime cost‐use analysis from a societal perspective, using a first‐order Monte Carlo simulation. Strategies included routine primary care and varying combinations of diet, exercise, behavior modification, and/or pharmacotherapy. Primary data were collected to assess program costs and obesity‐related quality of life. Other data were obtained from clinical trials, population‐based surveys, and other published literature. This was a simulated cohort of healthy 35‐year‐old overweight and obese women in the United States. Results: For overweight and obese women, a three‐component intervention of diet, exercise, and behavior modification cost $12,600 per quality‐adjusted life year gained compared with routine care. All other strategies were either less effective and more costly or less effective and less cost‐effective compared with the next best alternative. Results were most influenced by obesity‐related effects on quality of life and the probabilities of weight loss maintenance. Discussion: A multidisciplinary weight loss program consisting of diet, exercise, and behavior modification provides good value for money, but more research is required to confirm the impacts of such programs on quality of life and the likelihood of long‐term weight loss maintenance. 相似文献

Materials and methods: Forty specimens (40 × 8 × 4 mm) of each material were made, and FT was assessed after polymerisation (control of reliners), after 48 ± 2 h in water at 37°C (control of denture base resin) and after storage in water at 37°C for 7, 90 or 180 days (all materials). Data (MPa.m1/2) were analysed by two‐way anova and Games–Howell test (p = 0.05). Results: Resin L exhibited the highest FT mean values. After 180 days of storage, FT mean values of L (3.37), UH (1.53) and K (1.20) were higher than those of the other periods. FT mean values of NT decreased from control (1.63) to 7 days (1.30) and then remained constant. FT mean values of TR (1.13) were similar in all periods of analysis. Conclusion: The denture base resin L showed higher FT mean values than the reline resins. Long‐term water storage increased the FT of L, UH and K, reduced the FT of NT and did not influence the FT of TR. 相似文献

A novel procedure for the separation of the cellular thiol pool according to the molecular weight and localization of compounds with sulphydryl groups is presented. This simple and rapid method allows the differentiation of thiols into three major fractions-low molecular weight (LMT, primarily glutathione and free cysteine), protein-bound (TPT) and pellet-bound (PBT, associated with cell walls and broken organelles). Moreover, determination of the ratio between surface (readily reactive) thiols (ATG) and those that are more or less buried in the protein structure (BTG) can be achieved. In intact pea leaves, the amounts of the total thiols (LMT+PBT+TPT) varies from 2.5 to 4.8 micromol/g of fresh material. The data for LMT, PBT and TPT were related to each other in the approximate ratio 1:2:7. Treatments of pea plants with high temperature, salinity and low amounts of atrazine affect these sulphydryl types differently. For a greater understanding of the applicability of this method to physiological research, the main mechanisms leading to alterations in the cellular thiol pool are discussed. Furthermore, it is suggested that the proportion of available to buried thiols (ATG/BTG) in proteins could be used as a convenient marker for stress impacts. 相似文献

Human skin heals more slowly in aged vs. young adults, but the mechanism for this delay is unclear. In humans, eccrine sweat glands (ESGs) and hair follicles underlying wounds generate cohesive keratinocyte outgrowths that expand to form the new epidermis. Here, we compared the re‐epithelialization of partial‐thickness wounds created on the forearm of healthy young (< 40 yo) and aged (> 70 yo) adults. Our results confirm that the outgrowth of cells from ESGs is a major feature of repair in young skin. Strikingly, in aged skin, although ESG density is unaltered, less than 50% of the ESGs generate epithelial outgrowths during repair (vs. 100% in young). Surprisingly, aging does not alter the wound‐induced proliferation response in hair follicles or ESGs. Instead, there is an overall reduced cohesiveness of keratinocytes in aged skin. Reduced cell–cell cohesiveness was most obvious in ESG‐derived outgrowths that, when present, were surrounded by unconnected cells in the scab overlaying aged wounds. Reduced cell–cell contact persisted during the repair process, with increased intercellular spacing and reduced number of desmosomes. Together, reduced outgrowths of ESG (i) reduce the initial number of cells participating in epidermal repair, (ii) delay wound closure, and (iii) lead to a thinner repaired epidermis in aged vs. young skin. Failure to form cohesive ESG outgrowths may reflect impaired interactions of keratinocytes with the damaged ECM in aged skin. Our findings provide a framework to better understand the mediators of delayed re‐epithelialization in aging and further support the importance of ESGs for the repair of human wounds. 相似文献
