Vegetation treatments have been widely implemented in efforts to enhance conditions for wildlife populations. Yet the effectiveness of such efforts often lack rigorous evaluations to determine whether these practices are effective for targeted species. This is particularly important when manipulating wildlife habitats in ecosystems that are faced with multiple stressors. The sagebrush (Artemisia spp.) ecosystem has been altered extensively over the last century leading to declines of many associated species. Wyoming big sagebrush (A. tridentata wyomingensis) is the most widely distributed subspecies, providing important habitats for sagebrush-obligate and associated wildlife. Sagebrush often has been treated with chemicals, mechanical treatments, and prescribed burning to increase herbaceous forage species released from competition with sagebrush overstory. Despite many studies documenting negative effects of sagebrush control on greater sage-grouse (Centrocercus urophasianus) habitat, treatments are still proposed as a means of improving habitat for sage-grouse and other sagebrush-dependent species. Furthermore, most studies have focused on vegetation response and none have rigorously evaluated the direct influence of these treatments on sage-grouse. We initiated a 9-year (2011–2019) experimental study in central Wyoming, USA, to better understand how greater sage-grouse respond to sagebrush reduction treatments in Wyoming big sagebrush communities. We evaluated the influence of 2 common sagebrush treatments on greater sage-grouse demography and resource selection. We implemented mowing and tebuthiuron application in winter and spring 2014 and evaluated the pre- (2011–2013) and post-treatment (2014–2019) responses of sage-grouse relative to these management actions. We evaluated responses to treatments using demographic and behavioral data collected from 620 radio-marked female greater sage-grouse. Our specific objectives were to evaluate how treatments influenced 1) sage-grouse reproductive success and female survival; 2) sage-grouse nesting, brood-rearing, and female resource selection; 3) vegetation responses; and 4) forbs and invertebrates. Our results generally suggested neutral demographic responses and slight avoidance by greater sage-grouse in response to Wyoming big sagebrush treated by mowing and tebuthiuron. Neither mowing nor tebuthiuron treatments influenced nest survival, brood survival, or female survival. Selection for nest and brood-rearing sites did not differ before and after treatments. Females selected habitats near treatments before and after they were implemented; however, the strength of selection was lower after treatments compared with pre-treatment periods, which may be explained by a lack of response in vegetation and invertebrates following treatments. Perennial grass cover and height varied temporally yet did not vary systematically between treatment and control plots. Forb cover and species richness varied annually but not in relation to either treatment type. Perennial grass cover and height, forb cover, and forb species richness did not increase within mowed or tebuthiuron-treated areas that received 2 or 6 years of grazing rest compared with areas that received no grazing rest. Finally, forb and invertebrate dry mass did not differ between treated plots and control plots at mowing or tebuthiuron sites in any years following treatments. Results from our study add to a large body of evidence that sage-grouse using Wyoming big sagebrush vegetation communities do not respond positively to sagebrush manipulation treatments. Management practices that focus on the maintenance of large, undisturbed tracts of sagebrush will best facilitate the persistence of sage-grouse populations and other species reliant on the sagebrush steppe.  相似文献

Theoretical computations (molecular dynamics and combined quantum chemical and molecular mechanical geometry optimizations) have been performed on horse liver alcohol dehydrogenase. The results provide evidence that Glu-68, a highly conserved residue located 0.47 nm from the catalytic zinc ion, may intermittently coordinate to the zinc ion. Structures with Glu-68 coordinated to the zinc ion are almost as stable as structures with Glu-68 at the crystal position and the barrier between the two configurations of Glu-68 is so low that it can readily be bypassed at room temperature. There is a cavity behind the zinc ion that seems to be tailored to allow such coordination of Glu-68 to the zinc ion. It is suggested that Glu-68 may facilitate the exchange of ligands in the substrate site by coordinating to the zinc ion when the old ligand dissociates.  相似文献

Plants exploit an array of defences against insect herbivores based on chemical and biomechanical properties. There is increasing evidence that plant toughness comprises a particularly effective defence against herbivory, yet studies to date have focussed exclusively on leaf toughness and folivore behaviour. The relationship between root mechanical properties and the chewing behaviour of a root‐feeding insect, the Agriotes spp. wireworm (Coleoptera: Elateridae L.), feeding on tobacco (Nicoiana tabacum) is investigated. Root toughness is manipulated using introduced transgenes for the down‐regulation of key enzymes in the lignin biosynthesis pathway: cinnamoyl‐CoA reductase (CCR line) and caffeate O‐methyltransferase and cinnamyl alcohol dehydrogenase (CO line). Two biomechanical analyses (i.e. conventional cutting and notched tensile) are conducted to quantify root toughness on both lines. Roots from the CO line are significantly tougher than those of the CCR line in terms of fracture toughness and fracture energy, although not for cutting energy or stiffness. Bioassays that compel wireworms to chew through roots demonstrate that only 30% can penetrate roots of the CO line compared with 90% on the CCR line. It takes wireworms over twice as long to penetrate roots from the CO line (8 h) compared with CCR roots (3.5 h). There is a statistically significant positive relationship between penetration time and fracture toughness evaluated with tensile tests, although not with cutting energy from cutting tests. Using this exploratory model system, it is concluded that root toughness derived from tensile tests is a practical indicator of the ability of root‐feeding insects to penetrate roots.  相似文献

Magnetic therapy as a self‐care intervention has led to the conduct of numerous human trials and animal experiments. Results concerning the analgesic efficacy of magnetic exposure, however, are inconsistent. By using a magnetic device generating an inhomogeneous static magnetic field (iSMF), here we studied how the whole‐body exposure to iSMF may influence the mechanical withdrawal threshold (MWT) of the hind paw in different stages of neuropathic pain evoked by partial ligation of the sciatic nerve in mice. It was found that iSMF exposure did not prevent the decrease of MWT in the first postoperative week. A 2‐week long iSMF treatment that was started just after the nerve ligation elevated MWT values to a modest extent. However, the effectiveness of a daily exposure to iSMF was much more prominent when it was applied between postoperative days 15 and 28. In this case, MWT was already noticeably increased after the first treatment and it practically reached the control values by the end of the 2‐week long exposure period. The results suggest that exposure to iSMF cannot prevent the development of mechanical allodynia, but can inhibit processes that maintain the increased sensitivity to mechanical stimuli in neuropathic pain. Colobines are similar in their exploitation of a high percentage of leaf matter. However, this observation obfuscates interesting differences among genera of Southeast Asian colobines in morphology and behavior that may be reflected in the degree to which they rely on mastication or gut volume and gut retention time when ingesting and digesting leaves. We detail the use of a laboratory-based method to measure the mechanical properties of foods selected and processed by 4 captive species of Southeast Asian Colobinae —Pygathrix nemaeus, Pygathrix cinerea, Trachypithecus delacouri, and Trachypithecus laotum hatinhensis— at the Endangered Primate Rescue Center (EPRC), Vietnam. We also detail a field method that quantifies chewing rates and chewing behavior via a consumer-grade video camera and laptop computer. Observations in the captive setting permit a degree of experimental control that is not possible in the wild, and the location of the EPRC in the primates’ habitat country permitted us to provide leaves that they encounter and eat in the wild. We collected toughness data with a portable tester designed by Lucas et al. The average toughness of selected leaves does not differ among the taxa, nor does the length of time spent chewing foods. However, there are differences in feeding rate, with Trachypithecus spp. chewing foods twice as fast as Pygathrix spp. Our findings suggest that Trachypithecus spp. emphasize comminution of food by mastication, while Pygathrix spp. emphasize the comminution of leaf matter in the stomach. The hypothesis is supported by data on molar size, gut mass, and gut morphology. We provide new insights into dietary variation among primate species and detail methods that are typically conducted only in a laboratory setting. We augment the findings with additional data on activity, feeding rates, and tooth morphology.  相似文献

Here we report on a method that extends the study of the mechanical behavior of single proteins to the low force regime of optical tweezers. This experimental approach relies on the use of DNA handles to specifically attach the protein to polystyrene beads and minimize the non-specific interactions between the tethering surfaces. The handles can be attached to any exposed pair of cysteine residues. Handles of different lengths were employed to mechanically manipulate both monomeric and polymeric proteins. The low spring constant of the optical tweezers enabled us to monitor directly refolding events and fluctuations between different molecular structures in quasi-equilibrium conditions. This approach, which has already yielded important results on the refolding process of the protein RNase H (Cecconi et al. in Science 309: 2057-2060, 2005), appears robust and widely applicable to any protein engineered to contain a pair of reactive cysteine residues. It represents a new strategy to study protein folding at the single molecule level, and should be applicable to a range of problems requiring tethering of protein molecules.  相似文献

1. Silicon (Si) has received increased attention as a nutrient capable of providing some measure of defence for plants against fungal pathogens, and insect and mammalian herbivores. 2. On the basis of a study including two generalist insect folivores and a phloem feeder, Massey, Ennos & Hartley (2006; Journal of Animal Ecology, 75, 595-603) have drawn attention to a putative distinction between the effects of plant Si in defending against folivorous and phloem-feeding insects. On the basis of their results they imply that phloem feeders are less likely to be adversely affected by increased plant Si than folivores. 3. However, in making this suggestion, Massey et al. have ignored many previous studies demonstrating a clear effect of plant Si on a range of phloem-feeding and some xylem-feeding insects, and that this effect stems not only from leaf mechanical properties based on opaline silica, but also from induced chemical defences seemingly mediated by soluble Si. 4. Furthermore, Massey et al. cannot claim that their study was the first demonstration of a direct effect of Si on insect herbivore preference and performance; there have been numerous earlier studies demonstrating this from folivores, stem borers, and phloem and xylem feeders. 5. We contend that current evidence indicates that Si is likely to be involved to a similar extent in enhancing resistance to all four insect feeding guilds and that any conclusion to the contrary is, at this stage, premature.  相似文献
