We have developed an online analytical method that combines A431 cell membrane chromatography (A431/CMC) with high performance liquid chromatography and mass spectrometry (LC/MS) for identifying active components from Radix Caulophylli acting on human EGFR. Retention fractions on A431/CMC model were captured onto an enrichment column and the components were directly analyzed by combining a 10-port column switcher with an LC/MS system for separation and preliminary identification. Using Sorafenib tosylate as a positive control, taspine and caulophine from Radix Caulophylli were identified as the active molecules which could act on the EGFR. This A431/CMC-online-LC/MS method can be applied for screening active components acting on EGFR from traditional Chinese medicines exemplified by Radix Caulophylli and will be of great utility in drug discovery using natural medicinal herbs as a source of novel compounds.  相似文献

Parental effects are a major source of phenotypic plasticity and may influence offspring phenotype in concert with environmental demands. Studies of “environmental epigenetics” suggest that (1) DNA methylation states are variable and that both demethylation and remethylation occur in post‐mitotic cells, and (2) that remodeling of DNA methylation can occur in response to environmentally driven intracellular signaling pathways. Studies of mother‐offspring interactions in rodents suggest that parental signals influence the DNA methylation, leading to stable changes in gene expression. If parental effects do indeed enhance the “match” between prevailing environmental demands and offspring phenotype, then the potential for variation in environmental conditions over time would suggest a mechanism that requires active maintenance across generations through parental signaling. We suggest that parental regulation of DNA methylation states is thus an ideal candidate mechanism for parental effects on phenotypic variation.  相似文献

Cannibalism is widespread among many different organisms, and can have both negative and positive fitness consequences. Avoiding eating relatives can minimize negative fitness consequences of cannibalism. Such avoidance requires kin discrimination, but evidence for this ability among cannibals is limited with little data that address the cues used in such discrimination. We examined whether larvae of the ladybird beetle Harmonia axyridis avoid eating their relatives. We further manipulated environmental factors to create individuals that had similar or dissimilar rearing environments to begin to test for endogenous versus exogenous recognition cues. In our experiments, third-instar larvae were much less likely to cannibalize if they were interacting with a relative. Larvae that did cannibalize kin required more encounters, and significantly delayed cannibalism, compared to larvae cannibalizing unrelated individuals. Acquired cues were less important. Even though the different rearing environment resulted in significantly different phenotypic effects, similarity or dissimilarity of rearing environment had no effect on cannibalism, and there was no interaction between environments and relatedness in cannibalism. We suggest that H. axyridis has a well-developed kin discrimination system, and that kin recognition in this ladybird beetle is based on endogenous rather than exogenous cues. We also argue that these cues reflect either direct or indirect genetic effects on larval phenotypes.  相似文献

Neuromedin U (NMU) mediates various physiological functions via NMUR1 and NMUR2 receptors. NMUR2 has been considered a promising treatment option for diabetes and obesity. Although NMU-8, a shorter peptide, has potent agonist activity for both receptors, it is metabolically unstable. Therefore, NMU-8 analogs modified with long-chain alkyl moieties via a linker were synthesized. An octadecanoyl analog (17) with amino acid substitutions [αMePhe19, Nle21, and Arg(Me)24] and a linker [Tra-γGlu-PEG(2)] dramatically increased NMUR2 selectivity, with retention of high agonist activity. Subcutaneous administration of 17 induced anorectic activity in C57BL/6J mice. Owing to its high metabolic stability, 17 would be useful in clarifying the physiological role and therapeutic application of NMU.  相似文献

Enset is a large, single‐stemmed perennial herbaceous plant domesticated as a staple food crop only in Ethiopia. Khat is a perennial plant cultivated for its economically important leaves and twigs that are the sources of stimulant when chewed. We address the issue of yield estimation of both crops, as they are important for the livelihoods of smallholders in the home garden systems in Southern Ethiopia and have received little attention so far. The objective of this study was to develop linear allometric models for estimating the edible (food and feed) and commercial yields of enset and khat plants, respectively. Data were collected from 20 enset and 100 khat plants. Diameter at 50‐cm height (d50), pseudostem height (hp) and their combination were good predictor variables for the food products of enset with adjusted R2 values above 0.85, while d50, hp, edible pseudostem height (hep), total height (ht) and their combination were good predictor variables for the feed products of enset with adjusted R2 values above 0.70. For dwarf khat plants crown area (ca) combined with total height (ht) resulted in the best prediction with an adjusted R2 of 0.77, while the leaf and twig dry weight for tall khat plants was best predicted by ca with adjusted R2 of 0.43. A large body of theory predicts that populations diffusing in heterogeneous environments reach higher total size than if non‐diffusing, and, paradoxically, higher size than in a corresponding homogeneous environment. However, this theory and its assumptions have not been rigorously tested. Here, we extended previous theory to include exploitable resources, proving qualitatively novel results, which we tested experimentally using spatially diffusing laboratory populations of yeast. Consistent with previous theory, we predicted and experimentally observed that spatial diffusion increased total equilibrium population abundance in heterogeneous environments, with the effect size depending on the relationship between r and K. Refuting previous theory, however, we discovered that homogeneously distributed resources support higher total carrying capacity than heterogeneously distributed resources, even with species diffusion. Our results provide rigorous experimental tests of new and old theory, demonstrating how the traditional notion of carrying capacity is ambiguous for populations diffusing in spatially heterogeneous environments.  相似文献

The present experiment was carried out in Luotong Mountain Natural Reserve in Jilin Province of China in 2007. We recorded and analyzed the echolocation calls of Rhinolophus ferrumequinum in different habitats by using Avisoft Bioacoustics USG 116 and Avisoft-SASLAB PRO (Avisoft Bioacoustics, Berlin, Germany). Our results showed that R. ferrumequinum foraged in diverse habitats in the study area, and their echolocation calls were significantly variable in different habitats (One-Way ANOVA, P < 0.05). Vegetative, climatic and topographical factors were selected by using the principal component analysis and the correlations between the parameters of echolocation calls and these environmental factors were analyzed. The results indicated that although R. ferrumequinum always emitted FM/CF/FM echolocation calls in different habitats, the parameters of echolocation calls varied with variable environmental factors. Significant negative correlation existed between FM1 bandwidth and arbor height (r = ?0.948, P < 0.05), FM2 bandwidth and arbor height (r = ?0.825; P < 0.05), FM1 starting frequency and canopy area (r = ?0.967, P < 0.05), FM2 ending frequency and canopy area (r = ?0.958, P < 0.05), FM1 starting frequency and air relative humidity (r = ?0.776, P < 0.05), FM2 ending frequency and air relative humidity (r = ?0.875, P < 0.05), peak frequency and air relative humidity (r = ?0.794, P < 0.05), pulse duration and average shrub height (r = ?0.911, P < 0.05), and inter-pulse interval and average shrub height (r = ?0.990, P < 0.05). Significant positive correlation existed between peak frequency and number of plants (r = 0.756, P < 0.05), and pulse duration and height below the canopy (r = 0.870, P < 0.05). Our results suggested that many kinds of ecological factors (such as vegetation factor, climatic factor and topographical factor) affected the structure of echolocation calls and made them diverse in different habitats, i.e., echolocation calls of bats had phenotypic flexibility and eco-adaptability. These characteristics determined the degree of available habitats and natural resources for R. ferrumequinum.  相似文献

Effect of increased temperature during egg maturation on the mass of single eggs produced by the offspring was investigated experimentally in Atlantic salmon Salmo salar. Mass of eggs produced by next‐generation females was larger when their mothers experienced warmer water during the last two months of egg maturation, relative to those that experienced unheated river water. There was no similar trans‐generational paternal effect on offspring egg mass.  相似文献

增强UV-B辐射会对植物生长和生理生化过程产生有害效应。克隆植物中,相连的克隆分株对经常共享资源和激素,然而鲜有关于异质性UV-B辐射下UV-B辐射方向对克隆整合的影响及克隆植物形态结构变化的报道。模拟同质(克隆分株片段均处于自然背景辐射)和异质(克隆分株一端处于自然背景辐射,另一端处于补加的UV-B辐射)UV-B辐射,以克隆植物白三叶为材料,进行连接和隔断处理,研究UV-B辐射方向对克隆整合强度变化、叶片形态结构特化及生理可塑性的影响。结果表明:异质性UV-B辐射下,15N同位素标记端保留的15N百分比高于同质UV-B辐射处理,转移到无标记相连端的15N含量则降低,紫外辐射处理和同位素标记是否处于同一分株端对结果无显著性影响,说明克隆植物白三叶生理整合存在但整合强度降低,辐射方向与克隆整合强度无关;隔断处理组气孔长度增加,栅栏组织增厚,但连接处理组却无此变化,表明生理整合在白三叶叶片形态结构特化中发挥作用。UV-B辐射下,最小荧光、电子传递速率及光化学淬灭系数降低但非光化学淬灭系数升高,而生理整合却使结果相反;叶绿素和紫外吸收物可在异质性UV-B辐射相连的两端运输分享。以上均表明异质UV-B辐射环境中,UV-B辐射胁迫端克隆分株通过生理整合从非胁迫端获益,并以此提高胁迫环境中克隆植物对资源的利用效率。  相似文献
