In Hermissenda type-B photoreceptors, the spike is generated in the axon and back-propagated to the soma, resulting in smaller somatic spikes. Experimentally, blocking the A-type K+ current (IK,A) results in broadening of somatic spikes. Similarly, in a compartmental model of the photoreceptor, reducing the maximum A-type K+ conductance (gK,Amax) results in broadening of somatic spikes. However, simulations predict that little or no broadening of axonal spikes occurs when gK,Amax is reduced. The results can be explained by the voltage-dependent properties of IK,A and the different potential ranges that the somatic and axonal spike traverse. Because of the steeper I-V curve and faster activation of the K+ channels at higher potentials, the recruitment of additional K+ channels in the axon is able to compensate for the decrease in K+ conductance, yielding less spike broadening. These results also support the idea that spike duration in the axon may not be reliably inferred based upon recordings collected from the soma. Action Editor: Jonathan D. Victor  相似文献

During flight, the wings of Drosophila melanogaster beat nearly 200 times per second. The indirect flight muscle fibers that power this movement have evolved to resist the repetitive mechanical stress that results from the 5-ms wing beat cycle at a strain amplitude of 3.5%. In order to understand how this is achieved at the sarcomere level, we have analyzed the mechanical properties of native thick filaments isolated from indirect flight muscle. Single filaments adsorbed onto a solid support were manipulated in physiological buffer using an atomic force microscope. Images taken after the manipulation revealed that segments were stretched, on average, to 150%, with a maximum at 385% extension. The lateral-force-versus-displacement curve associated with each manipulation contained information about the bending and tensile properties of each filament. The bending process was dominated by shearing between myosin dimers and yielded a shear modulus between 3 and 13 MPa. Maximum tension along the stretched filaments was observed at ∼ 200% extension and varied between 8 and 17 nN. Based on current models of thick filament structure, these variations can be attributed to cross-links between myosin dimers distributed along the filament.  相似文献

Only a limited number of studies have addressed the reliability of extracellular markers of cardiac repolarization time, such as the classical marker RTeg defined as the time of maximum upslope of the electrogram T wave. This work presents an extensive three-dimensional simulation study of cardiac repolarization time, extending the previous one-dimensional simulation study of a myocardial strand by Steinhaus [B.M. Steinhaus, Estimating cardiac transmembrane activation and recovery times from unipolar and bipolar extracellular electrograms: a simulation study, Circ. Res. 64 (3) (1989) 449]. The simulations are based on the bidomain - Luo-Rudy phase I system with rotational fiber anisotropy and homogeneous or heterogeneous transmural intrinsic membrane properties. The classical extracellular marker RTeg is compared with the gold standard of fastest repolarization time RTtap, defined as the time of minimum derivative during the downstroke of the transmembrane action potential (TAP). Additionally, a new extracellular marker RT90eg is compared with the gold standard of late repolarization time RT90tap, defined as the time when the TAP reaches 90% of its resting value. The results show a good global match between the extracellular and transmembrane repolarization markers, with small relative mean discrepancy (?1.6%) and high correlation coefficients (?0.92), ensuring a reasonably good global match between the associated repolarization sequences. However, large local discrepancies of the extracellular versus transmembrane markers may ensue in regions where the curvature of the repolarization front changes abruptly (e.g. near front collisions) or is negligible (e.g. where repolarization proceeds almost uniformly across fiber). As a consequence, the spatial distribution of activation-recovery intervals (ARI) may provide an inaccurate estimate of (and weakly correlated with) the spatial distribution of action potential durations (APD).  相似文献

Net photosynthetic rate (P N) measured at the same CO2 concentration, the maximum in vivo carboxylation rate, and contents of ribulose-1,5-bisphosphate (RuBP) carboxylase/oxygenase (RuBPCO) and RuBPCO activase were significantly decreased, but the maximum in vivo electron transport rate and RuBP content had no significant change in CO2-enriched [EC, about 200 μmol mol−1 above the ambient CO2 concentration (AC)] wheat leaves compared with those in AC grown wheat leaves. Hence photosynthetic acclimation in wheat leaves to EC is largely due to RuBP carboxylation limitation.  相似文献

嘉陵江小三峡硅藻群落结构及水环境   总被引:2,自引:0,他引:2  
为了研究嘉陵江小三峡硅藻群落结构及水质状况,按枯水期(2006年11月—2007年3月)、平水期(2007年4—6月)和丰水期(2007年7—9月)分别采集6个样点的硅藻样品,分析其物种组成、细胞密度、多样性和均匀度指数及硅藻商等,并结合理化指标对水质进行评价。结果表明:嘉陵江小三峡共有硅藻78种(含变种和变型),隶属2纲、10科、20属;硅藻细胞密度为0.53×104~3.58×104 ind·L-1,总平均1.55×104 ind·L-1;多样性指数值(H′)为1.62~2.96,总平均2.33;均匀度指数值(E)为0.28~0.56,总平均0.42;嘉陵江小三峡水质总体上属β-中污水体,沙溪、北温泉水质较好,属微污水体,碚石、牛石沱水质次之,为β-中污水体,盐井、毛背沱水质最差,为α-中污水体。  相似文献

Aims:  Leptospirosis is a public health problem worldwide. Traditionally, microscopic agglutination test (MAT) and cross-agglutinin absorption test (CAAT) are used to identify leptospires. However, these techniques are laborious and time-consuming, requiring the maintenance of a collection of more than 200 reference strains and correspondent rabbit antisera. The purpose of this study was to evaluate the pulsed-field gel electrophoresis (PFGE) method for discrimination of Leptospira serovars.
Methods and Results:  Fourteen clinical isolates of Leptospira spp. were analysed by MAT before being characterized by PFGE. The isolates were compared with a library of 206 different reference Leptospira serovars. All the isolates gave clear profiles with high resolution. PFGE and MAT results were in agreement for all clinical isolates evaluated. Twelve isolates were classified as serovar Icterohaemorrhagiae/Copenhageni by PFGE. By MAT, these isolates were classified as serogroup Icterohaemorrhagiae with titres ranging from 3200 to 25 600. Two isolates were classified as serovar Canicola by PFGE, and as serogroup Canicola by MAT with titres higher than 3200.
Conclusions:  PFGE offers the advantages of simple, reliable and reproducible results.
Significance and Impact of the Study:  PFGE provides a convenient tool for the identification of clinical isolates.  相似文献

Dose‐intensive chemotherapy results in an obligatory period of severe neutropenia during which patients are at high risk of infection. While patient support with donor neutrophils is possible, this option is restricted due to donor availability and logistic complications. To overcome these problems, we explored the possibility of large scale ex vivo manufacture of neutrophils from hematopoietic progenitor cells (HPC). CD34+ HPC isolated from umbilical cord blood (UCB) and mobilized peripheral blood (mPB) were expanded in serum‐free medium supplemented with stem cell factor, granulocyte colony stimulating factor, and a thrombopoietin peptide mimetic. After 15 days of cultivation a 5,800‐fold expansion in cell number was achieved for UCB, and up to 4,000‐fold for mPB, comprising 40% and 60% mature neutrophils respectively. Ex vivo expanded neutrophils exhibited respiratory burst activity similar to that for donor neutrophils, and were capable of killing Candida albicans in vitro. These yields correspond to a more than 10‐fold improvement over current methods, and are sufficient for the production of multiple neutrophil transfusion doses per HPC donation. To enable clinical scale manufacture, we adapted our protocol for use in a wave‐type bioreactor at a volume of 10 L. This is the first demonstration of a large scale bioprocess suitable for routine manufacture of a mature blood cell product from HPC, and could enable prophylactic neutrophil support for chemotherapy patients. Although these molecules are distinct entities, with no proven physical interaction, their presence and activity in many cells appears to be coordinated. The major objective of this study is to understand the upper thermal limits and potential impact of temperature elevation on copepods caused by coastal power plants. Laboratory experiments were designed to evaluate the upper incipient lethal temperature (UILT) and critical thermal maximum (CTMax) of eight coastal copepod species collected from a subtropical bay in spring and summer. The 48h-UILT of copepods acclimatized at 16.0, 20.0, 28.0 °C were 26.4-29.1, 27.3-30.1, 32.9-36.9 °C, respectively. And the CTMax of copepods acclimatized at 28.0 °C was 35.80-41.03 °C. The UILT of copepods increased significantly with rising acclimatization temperatures, but the difference values between UILT and acclimatization temperatures decreased, which indicated that the seawater temperature elevation induced the growing mortalities of copepods with increasing natural seawater temperatures from the thermal addition of power plants. The results also showed that estuarine copepods had more tolerances to the thermal stress than those from other more stable marine environments. As to the calanoid copepod species, there was a significant negative correlation between the CTMax and body length (p < 0.01). So it seemed that the copepod species with large body size were more sensitive to the thermal addition than the smaller ones. Thus, owing to the temperature increase, the copepod species diversity might reduce and the composition of copepod communities might tend to be small-sized in natural sea areas close to the coastal power plants.  相似文献
