农杆菌介导的高效玉米遗传转化体系的建立   总被引:7,自引:0,他引:7

为了建立玉米高频再生及高效遗传转化体系, 对影响玉米胚性愈伤组织诱导的11个因素及影响胚性愈伤分化的9个因素用正交实验方法进行研究。结果显示, 基因型对胚性愈伤诱导有极显著影响。6-BA、培养基、AgNO3、2,4-D、ABA对胚性愈伤诱导的影响达到显著水平。多重比较分析显示ABA 2 mg/L每间隔1代添加对胚性愈伤诱导率有显著影响。在影响分化的因素中, 基因型和6-BA浓度表现出极强的主效应, NAA、培养基、KT、2,4-D对分化产生显著影响。Southern blotting 分析表明, 25 mg/L潮霉素选择压下抗性愈伤率作为转化体系优化指标是可靠的。在影响转化效率的因素中, acetosyringone (AS)使用浓度因基因型不同而表现出敏感度差异, 共培养温度24~25℃、农杆菌浓度和浸泡时间0.7 OD×15 min, 以及pH值5.5~6.2是最高转化率的优选组合。在整合后的玉米遗传转化体系中, 黄早4和综31自交系以抗性愈伤率为指标的GUS基因稳定转化率分别达到48.6%和46.2%。  相似文献

既要保证氮肥在优质高效农业生产中的促进作用,又要保证农业和环境的可持续发展,如何提高氮肥利用率、减轻氮肥污染成了解决问题的关键.从氮素在土壤中的生物化学转化过程入手,通过抑制剂的施用来调控氮素的转化,减缓硝化过程的进行,是解决该问题的一种有效措施.本文从硝化抑制剂的定义、筛选硝化抑制剂的条件、国内外较受关注的硝化抑制剂品种以及硝化抑制机理等几个方面对目前的研究进展进行综述,并提出目前研究中存在的问题及今后的研究方向.  相似文献

Aims Supplying optimal quantities of mineral nutrients to growing crop plants is one way to improve crop yields. Nutrients need to be used rationally in order to avoid a negative ecological impact and undesirable effects on the sustainability of agricultural production systems. Excessive application of nutrients also affects the farmer's economy. In order to calculate the amount of fertilizer to be applied to crops, it is necessary to develop recommendation programmers that adjust nutrient rates to crop requirements.Methods Experiments in two successive seasons were conducted to investigate the effect of K fertilization and foliar application of Zn and P on yield and fiber properties of cotton cv. Giza 86. Potassium (0.0 and 47.4 kg of K ha-1) was soil applied, while chelated zinc (0.0 and 57.6 g of Zn ha-1, applied twice at 70 and 85 days after sowing 'DAS') and phosphorus (0.0, 576, 1?152 and 1?728 g of P ha-1, applied twice at 80 and 95 DAS) were applied to the foliage.Important findings Dry matter yield, total chlorophyll concentration, K, Zn and P uptake per plant, number of opened bolls per plant, boll weight, seed index, lint index, seed cotton yield per plant, seed cotton and lint yield ha-1 and earliness of harvest increased with the application of K, Zn and P. Treatments generally had no significant effect on lint percentage and fiber properties, with exceptions, for micronaire reading and flat bundle strength, and uniformity ratio, where the mean values of these characters were significantly increased over the untreated control by applying K, and for the micronaire reading in the first season, when applying P at 1?728 g ha-1, and uniformity ratio in the second season, when applying P at 1?152 and 1?728 g ha-1, where the mean values of these characters were significantly increased over the untreated control by applying P. Under the conditions of this study, applying K fertilization at 47.4 kg ha-1 combined with spraying cotton plants with zinc at 57.6 g ha-1 and also with P at 1?728 g ha-1 improved growth and yield of Egyptian cotton.  相似文献

在大田条件下,研究了不同追氮时期对小麦旗叶叶绿素荧光特性、光合速率及籽粒产量的影响.结果表明,拔节期追肥较起身期或挑旗期追肥,改善了小麦旗叶PSⅡ的活性(Fv/Fo)、光化学最大效率(Fv/Fm)、光化学猝灭系数(qP)、实际量子产量(ΦPSⅡ)及光合速率,降低了籽粒灌浆中前期非辐射能量耗散,有利于叶片所吸收的光能较充分地用于光合作用,提高了籽粒灌浆后期非辐射能量的耗散,减缓了叶片光抑制程度和衰老进程.拔节期追肥可显著增加穗粒数和千粒重,提高产量.  相似文献

对两个菜用枸杞品种的主要生长特性、营养成分及食用性研究表明,大叶品种嫩芽平均产量显著高于小叶品种;大叶品种较耐高温,小叶品种较耐低温;生产上可进行大、小叶品种搭配种植,能有效提早和延长采收期,从而提高单位面积产量和经济效益。此外,小叶品种嫩芽的蛋白质、VitC含量高于大叶品种。两个品种风味各具特色,其适宜的消费群体亦有所差异。  相似文献

Legume cover crops are a potential means for overcoming N depletion in the derived savanna of West Africa. A 3-year trial was, therefore, conducted near Ibadan, southwestern Nigeria to measure the N contribution of 13 legume cover crops as compared to urea –N, using a N fertilizer replacement index for a maize test crop. Two series of trials involved the following legume cover crop species: Aeschynomene histrix, Centrosema brasilianum, Centrosema pascuorum, Chamaecrista rotundifolia, Cajanus cajan, Crotalaria verrucosa, Crotalaria ochroleuca, Lablab purpureus, Mucuna pruriens, Psophocarpus palustris, Pseudovigna argentea, Pueraria phaseoloides and Stylosanthes hamata. Trials were undertaken using a complete block design. Cover crops were planted in 1994 (Series 1) and 1995 (Series 2) in separate sites and each series was subsequently slashed and planted for one season with maize (Zea mays) in 1995 and 1996. At the 50% flowering stage, N concentration of above-ground vegetation of cover crops ranged from 21 to 38 g N kg–1. Nitrogen accumulated by 4.5-month old cover crops ranged from 14 to 240 kg N ha–1, depending on species and year. Cover crops increased grain yield of the subsequent maize crop by 25–136% over the control without N application. Nitrogen uptake by the maize crop was higher following cover crops than after maize or natural grass. The N fertilizer replacement index of cover crops for maize ranged from 11 (A. histrix) to 96 kg N ha–1 (C. cajan) in Series 2. Perennial (C. brasilianum, S. hamata, C. cajan, P. phaseoloides and C. verrucosa) and annual (C. rotundifolia, M. pruriens, C. ochroleuca and L. purpureus) species could potentially save 50 to 100 kg N ha–1 for maize crops. The cover crops accumulated more N in the wetter than in the drier year. However, the N fertilizer replacement index was higher for subsequent maize grown in the drier year. The cover crop-N recovery in maize was also higher than the urea-N uptake in the drier year. The N fertilizer replacement indexes can be predicted using the above-ground biomass amount of cover crops at 20 weeks after planting (drier year) or the N concentration at that stage (wetter year).  相似文献

Complementarity in the distribution of tree and crop root systems is important to minimise competition for resources whilst maximising resource use in agroforestry systems. A field study was conducted on a kaolinitic Oxisol in the sub-humid highlands of western Kenya to compare the distribution and dynamics of root length and biomass of a 3-year-old Grevillea robusta A. Cunn. ex R. Br. (grevillea) tree row and a 3-year-old Senna spectabilis DC. (senna) hedgerow grown with Zea mays L. (maize). Tree roots were sampled to a 300 cm depth and 525 cm distance from the tree rows, both before and after maize cropping. Maize roots were sampled at two distances from the tree rows (75–150 cm and 450–525 cm) to a maximum depth of 180 cm, at three developmental stages. The mean root length density (Lrv) of the trees in the upper 15 cm was 0.55 cm cm−3 for grevillea and 1.44 cm cm−3 for senna, at the start of the cropping season. The Lrv of senna decreased at every depth during the cropping season, whereas the Lrv of grevillea only decreased in the crop rooting zone. The fine root length of the trees decreased by about 35% for grevillea and 65% for senna, because of maize competition, manual weeding, seasonal senescence or pruning regime (senna). At anthesis, the Lrv of maize in the upper 15 cm was between 0.8 and 1.5 cm cm−3. Maize root length decreased with greater proximity to the tree rows, potentially reducing its ability to compete for soil resources. However, the specific root length (m g−1) of maize was about twice that of the trees, so may have had a competitive uptake advantage even when tree root length was greater. Differences in maize fine root length and biomass suggest that competition for soil resources and hence fine root length may have been more important for maize grown with senna than grevillea. The efficacy of Fe3+ complexes of polyethers with 8-quinolinol (8QOH) chelating groups attached to the polymer chain at different positions of the aromatic ring or having different chain length for remedying the iron-deficient maize plants was evaluated. The efficacy of chelates of polymers having terminal 8QOH residues was compared with that of complexes of ethylenediaminetetraacetic acid, 8QOH, mixtures of commercial polyethers with isopropylamino end-groups and 8QOH or FeCl3.6H2O. It was found that at 30/25 °C (day/night) and photosynthetic photon flux density 1100–1300 μmol m−2 s−1, the chlorotic maize plants recovered for 4 days of iron re-supply. An increase in the fresh and dry weight, leaf area, net photosynthetic CO2 uptake of maize leaves, leaf pigment composition and chlorophyll fluorescence was more pronounced in the plants supplied with Fe3+ chelates of polymers bearing 8QOH groups attached at 5-position, compared to the other tested Fe3+complexes. The importance of the stability of Fe3+ complexes, structure of the chelating agent and the necessity of effective ligand exchange between synthetic chelators and free phytosiderophore in iron uptake by strategy II plants was discussed. Tomatoes grown in traditional mediums such as peat moss are usually supplied with essential nutrients by preplant fertilization and in the later growth stages by top dressing application to the root zone. According to the data from literature the efficient method of nutrient supply during the intensive stage of growth may be estimated as by foliar spraying. In the greenhouse experiment conducted in 1998–1999 a preplant fertilization by multicomponential fertilizer MIS-4 was applied to peat moss growing medium in full (4 kg per m3) and half (2 kg per m3) of recommended rate. Supplemental fertilization during the growing period was provided to the root zone or by using liquid multicomponential fertilizers Ekolist S and Mikrosol U in 5 sprays conducted at two week intervals. Results of the study proved that advantageous effect of foliar tomato nutrion grown in eat substrate was observed only in treatments with restricted supply of fertilizers to the growing medium. The maximum fruit yield was received from plots provided by the reduced preplant MIS-4 dose to 50 % of recommended rate combined with foliar sprays by Ekolist S. Such system of fertilization enhanced the marketable yield of fruit by 9.8 % and early yield by 11.3 % as compared to that obtained with preplant and top dressing soil application. Foliar nutrion did not change dry matter and vitamin C content and increased total and reducing sugars accumulation in tomato fruits.  相似文献
