In an attempt to understand the feasibility of future targeted genome optimization in agronomic crops, we tested the efficiency of homologous recombination-mediated sequence insertion upon induction of a targeted DNA double-strand break at the desired integration site in maize. By the development of an efficient tissue culture protocol, and with the use of an I- Sce I gene optimized for expression in maize, large numbers of precisely engineered maize events were produced in which DNA integration occurred very accurately. In a subset of events examined in detail, no additional deletions and/or insertions of short filler DNA at the integration site were observed. In 30%–40% of the recovered events, no traces of random insertions were observed. This was true for DNA delivery by both Agrobacterium and particle bombardment. These data suggest that targeted double-strand break-induced homologous recombination is a superior method to generate specific desired changes in the maize genome, and suggest targeted genome optimization of agronomic crops to be feasible.  相似文献

Production of folates by yeasts in Tanzanian fermented togwa   总被引:1,自引:0,他引:1  
We have investigated the impact of different yeasts and fermentation time on folate content and composition in a fermented maize-based porridge, called togwa, consumed in rural areas in Tanzania. The yeasts studied, originally isolated from indigenous togwa, belong to Issatchenkia orientalis, Pichia anomala, Saccharomyces cerevisiae, Klyveromyces marxianus and Candida glabrata. The main folate forms found, detected and quantified by HPLC during the fermentations were 5-methyl-tetrahydrofolate (5-CH(3)-H(4)folate) and tetrahydrofolate (H(4)folate). The content of H(4)folate, per unit togwa, remained fairly stable at a low level throughout the experiment for all strains, whereas the 5-CH(3)-H(4)folate concentration was highly dependent on yeast strain as well as on fermentation time. The highest folate concentration was found after 46 h of fermentation with C. glabrata (TY26) (6.91+/-0.14 microg 100 mL(-1)), corresponding to a 23-fold increase compared with unfermented togwa. The cell concentration per se could not predict the togwa folate level, as shown by the much higher specific folate content (g folate CFU(-1)) in the S. cerevisiae strain (TY08) compared with the other species tested. This study provides useful data when trying to maximize folate content in togwa as well as in other yeast-fermented products.  相似文献

Waxy maize was first reported in China in 1909 and is mainly used in food production in Asia. The evidence for strong domestication selection in the Waxy locus of rice and a selective sweep around its genomic region make us to wonder whether there has been similar selection in Waxy in glutinous maize. To address this issue, DNA sequences of Waxy, three flanking genes and an unlinked gene (Adh1) of 30 accessions sampled from Chinese waxy maize accessions, including representative landraces and inbred lines, were determined in this study. Sharp reduction of nucleotide diversity and significant neutrality tests (Tajima’s D and Fu and Li’s F*) were observed in the Waxy locus in Chinese waxy maize but not in nonglutinous maize; comparison with the unlinked gene confirmed that this pattern was different to Waxy. Sequence analysis across a 143 kb genomic segment centered on the Waxy locus revealed patterns consistent with a selective sweep in the upstream region of Waxy. The selective sweep detected based on current limited genomic sequences exceeded over 50 kb, indicating strong selection in this or a bigger region. However, No sweep effect was detected in the repetitive downstream region of Waxy. Phylogenetic analysis indicated that Chinese waxy maize was domesticated from the cultivated flint maize (Zea mays ssp. mays) that was introduced from the new world. At least two independent deletions in exon 7 (30 bp) and 10 (15 bp) were identified in the Chinese accessions respectively. These findings demonstrate a similar pattern of domestication selection in the Waxy genomic region in both glutinous maize and rice, suggesting that this pattern in the rise of glutinous phenotype is likely in other cereal crops.  相似文献

1 Auchenorrhyncha (Planthoppers and Leafhoppers) are not only pests of many crops, but they are also nontarget organisms with respect to Bt‐protein expressing genetically modified plants. As herbivorous arthropods, planthoppers and leafhoppers ingest Cry proteins depending on their feeding behaviour. Consequently, they are directly exposed to these entomotoxic proteins and can also serve as a source of Cry protein exposure to predatory arthropods. Therefore, it is reasonable to use Auchenorrhyncha in the risk assessment of genetically modified crops. 2 During a 2‐year field study, we evaluated four different methods in terms of their feasibility to assess the impacts of plant‐incorporated protectants from Bt‐maize and of insecticide use on this group of arthropods. Visual assessment of plants, sweep netting, yellow traps and custom made sticky traps were utilized in field plots of Bt‐maize MON810, untreated near‐isogenic maize and insecticide‐treated near‐isogenic maize and were compared with respect to their capability to reflect the diversity and abundance of Auchenorrhyncha species. 3 Zyginidia scutellaris (Herrich‐Schäffer) (Cicadomorpha: Cicadellidae) represented more than 94% of all captured individuals in both years. The analysis of Z. scutellaris data showed no consistent differences between Bt‐maize MON810 and the untreated near isogenic hybrid, demonstrating no negative impact of MON810 on this species. Insecticide treatment, on the other hand, was not equivalent to the isogenic maize in terms of Z. scutellaris densities. Based on the collected data and on practical considerations, we recommend the combined use of transect‐wise sweep netting and sticky traps for the sampling of Auchenorrhyncha in maize.  相似文献

The growth and emergence of maize silks has a considerable importance in yield determination under drought conditions. Spatial and temporal patterns of the rates of tissue expansion and of cell division were characterized in silks of plants subjected to different soil water potentials. In all cases, silk development consisted of four phases: (1) cell division and tissue expansion occurred together uniformly all along the silk; (2) cell division progressively ceased from tip to base, while expansion remained spatially uniform including during the phase (3) after the cessation of cell division; and (4) as the silk emerged from the husks, expansion ceased in the emerged portion, probably because of direct evaporative demand, while the relative growth rate progressively decreased in the enclosed part. The rates of tissue expansion and cell division were reduced with water deficit, resulting in delayed silk emergence. The duration of cell division was not affected, and in all cases, the end of cell division in the silk coincided with anther dehiscence. The duration of phase 3, between the end of cell division and the arrest of cell growth in silk apex, considerably increased with water deficit. It corresponded to the anthesis-silking interval used by breeders to characterize the response of cultivars to stress.  相似文献

Standard maize starch was hydrothermally treated by Instantaneous Controlled Pressure Drop (DIC) process at three pressure levels (1, 2 and 3 bar) corresponding to the temperatures of 100, 122 and 135 °C (at 13–27% moisture), respectively. The structural effects of various hydrothermal conditions were examined with differential scanning calorimetry (DSC) and wide-angle X-ray diffraction. In order to understand the changes that occur during DIC treatment, melting endotherms of native maize starch at various moisture contents were determined. The gelatinization temperatures of DIC treated standard maize starch increased with DIC treatment. The transition temperatures (To, Tp) are closely related to the combined effect of pressure and processing time. At approximately 10 min of processing time, To and Tp were 65.7 and 72.3, 68.8 and 73.6 °C, 74.8 and 79.8 °C for pressure levels of 1, 2 and 3 bar, respectively (against 63.1 and 69.6 °C for native starch). DIC treatment narrowed the gelatinization temperature range and decreased gelatinization enthalpy (ΔH), as the severity of processing conditions increased. ΔH decreased from 11.4 J g−1 (native) to 11.0 (1 bar), 9.0 (2 bar) and 1.7 J g−1 (3 bar) for treated maize starch during approximately 10 min. Relative crystallinity of hydrothermally treated starch decreased with increasing DIC conditions. The A-type crystalline pattern was progressively lost (at pressure level 2 bar) and substituted by the Vh-type X-ray diffraction pattern, corresponding to the formation of amylose–lipid complexes. For severe DIC conditions (pressure level of 3 bar), the substitution was completed. Microscopic observations revealed progressive loss of the birefringence of DIC treated starch granules except at low pressure (1 bar), while the integrity of starch granules was preserved for all the conditions. These modifications that reveal important changes in the crystalline organization of the starch granules are related to their functional properties.  相似文献

The maize (Zea mays) spikelet consists of two florets, each of which contains three developmentally synchronized anthers. Morphologically, the anthers in the upper and lower florets proceed through apparently similar developmental programs. To test for global differences in gene expression and to identify genes that are coordinately regulated during maize anther development, RNA samples isolated from upper and lower floret anthers at six developmental stages were hybridized to eDNA rnicroarrays. Approximately 9% of the tested genes exhibited statistically significant differences in expression between anthers in the upper and lower florets. This finding indicates that several basic biological processes are differentially regulated between upper and lower floret anthers, including metabolism, protein synthesis and signal transduction. Genes that are coordinately regulated across anther development were identified v/a cluster analysis.Analysis of these results identified stage-specific, early in development, late in development and bi-phasic expression profiles. Quantitative RT-PCR analysis revealed that four genes whose homologs in other plant species are involved in programmed cell death are up-regulated just prior to the time the tapetum begins to visibly degenerate (i.e., the mid-microspore stage). This finding supports the hypothesis that developmentally normal tapetal degeneration occurs via programmed cell death.  相似文献

The rice BAC-DNA was used as probes and fluorescence in situ hybridization (FISH) was applied to the interphase and metaphase mitotic chromosomes of maize. To optimize the BAC-FISH technique, we respect-ively assayed the effect of several factors, including maize or rice genomic Cot DNA used as blocking reagent of DNA, washing temperatures and FAD concentration in the washing buffer and in the hybrid solution. The results show that Cot DNA of maize genome blocked the repet-itive sequence of the rice BAC-DNA when the Cot value was below 50. Meanwhile, it was necessary to adjust the Cot value according to the different probes and their ratios. Decreasing the concentration of FAD in the hybridization mixtures, adjusting the washing rate after hybridization, and most especially, blocking the rice-specific repetitive sequences of BAC-DNA could improve the positive signals of BAC-FISH.  相似文献

在华北平原灌溉区,采用冬小麦 夏玉米周年轮作田间试验,研究麦季牛场肥水灌溉对冬小麦和夏玉米产量、磷吸收量、磷累计利用率及土壤积累的影响.结果表明:麦季肥水灌溉能显著提高冬小麦和夏玉米产量,冬小麦产量随肥水带入磷的增加先增加后降低,肥水灌溉带入137 kg P2O5·hm-2时冬小麦产量最高,磷的当季利用率较高,分别为7646.4 kg·hm-2和24.8%,肥水灌溉带入过量磷会降低冬小麦产量和磷当季利用率;夏玉米产量和磷素吸收量随冬小麦季肥水灌溉带入磷量增加而增加,后季夏玉米产量增加2222.4~2628.6 kg·hm-2,磷吸收量增加13.9~21.1 kg·hm-2.农民习惯施肥处理夏玉米当季施磷88 kg P2O5·hm-2时,与不施肥处理相比,夏玉米产量增加2235.0 kg·hm-2.随着牛场肥水灌溉年限的推移,作物增产效果逐渐明显,冬小麦 夏玉米轮作体系作物累计磷利用率逐年升高,6季作物收获后,磷累计利用率达40.0%~47.7%.试验条件下,
冬小麦 夏玉米轮作体系进行2次肥水灌溉是较经济安全的灌溉模式.  相似文献
