The B800–850 antenna complex of Rhodopseudomonas sphaeroides was studied by comparing the spectral properties of several different types of complexes, isolated from chromatophores by means of the detergents lithium dodecyl sulfate (LDS) or lauryl dimethylamine N-oxide (LDAO). Fluorescence polarization spectra of the BChl 800 emission at 4 K indicated that rapid energy transfer between at least two BChl 800 molecules occurs with a rate constant of energy transfer kET > 3 · 1012 s?1. The maximal dipole-dipole distance between the two BChl 800 molecules was calculated to be 18–19 Å. The porphyrin rings of the BChl 800 molecules are oriented parallel to each other, while their Qy transition moments are mutually perpendicular. The energy-transfer efficiency from carotenoid to bacteriochlorophyll measured in different complexes showed that two functionally different carotenoids are present associated with, respectively, BChl 800 and BChl 850. Fluorescence polarization and linear dichroism spectra revealed that these carotenoids have different absorption spectra and a different orientation with respect to the membrane. The carotenoid associated with BChl 800 absorbs some nanometers more to the red and its orientation is approximately parallel to the membrane, while the carotenoid associated with BChl 850 is oriented more or less perpendicular to the membrane. The fluorescence polarization of BChl 850 was the same for the different complexes. This indicates that the observed polarization of the fluorescence is determined by the smallest complex obtained which contains 8–10 BChl 850 molecules. The B800–850 complex isolated with LDAO thus must consist of a highly ordered array of smaller structures. On basis of these results a minimal model is proposed for the basic unit consisting of four BChl 850 and two BChl 800 and three carotenoid molecules.  相似文献

Aim This paper examines eight fossil pollen datasets from Romania with the aim of exploring regional and elevational patterns in site similarity throughout the Holocene. In particular, we aim to determine whether there are clear intervals of homogenization/differentiation and to ascertain the potential driving factors. Location Romania. Methods Qualitative (pollen diagrams) and numerical methods including principal components analysis and Bray–Curtis similarity analyses were used. Results We found strong variability in the past vegetation dynamics during the Holocene. Bray–Curtis similarity analyses show large fluctuations in vegetation similarity and distinct periods of homogenization and differentiation throughout the Holocene. The magnitude and length of these periods appear quite variable in time, but the significant ones can be delimited as follows: (1) differentiation between 11,250 and 11,000 cal. yr bp , 10,000 and 9750 cal. yr bp , 6000 and 5750 cal. yr bp , 2500 and 2250 cal. yr bp , and especially over the last 200 years; and (2) homogenization between 9750 and 9500 cal. yr bp , and 2750 and 2500 cal. yr bp , with more stable periods between 9000 and 7750 cal. yr bp , 4750 and 3500 cal. yr bp , and 2000 and 1000 cal. yr bp . Main conclusions First, periods of biotic homogenization that occurred before significant anthropogenic impact on vegetation demonstrate that not all homogenization is a product of anthropogenic change: it can also be driven by natural causes. In fact, recent human impact (over the last 200 years) appears to have resulted in increased regional differentiation and not in homogenization – a result that contradicts most studies based on more modern, short‐term records. Second, both abiotic (climate and disturbance) and biotic factors are likely drivers of intervals of differentiation and homogenization. We suggest that differentiation may be triggered primarily by climate changes and disturbances (mostly natural pre‐2500 cal. yr bp and human‐induced thereafter), whereas homogenization may be driven predominantly by biotic interactions (e.g. immigration and interspecific competition). Third, this long‐term study raises awareness that assessments of pattern in vegetation homogenization/differentiation may depend on the specific time period and length of investigation. Long‐term investigations through multiple generations are likely to yield particularly useful information on the mechanisms and effects of biotic homogenization.  相似文献

Aim We present a continental‐scale analysis that explores the processes controlling woody community structure in tropical savannas. We analyse how biotic and abiotic factors interact to promote and modify tree cover, examine alternative ecological hypotheses and quantify disturbance effects using satellite estimates of tree cover. Location African savannas. Methods Tree cover is represented as a resource‐driven potential cover related to rainfall and soil characteristics perturbed by natural and human factors such as fire, cattle grazing, human population and cultivation. Within this framework our approach combines semi‐empirical modelling and information theory to identify the best models. Results Woody community structure across African savannas is best represented by a sigmoidal response of tree cover to mean annual precipitation (MAP), with a dependency on soil texture, which is modified by the separate effects of fire, domestic livestock, human population density and cultivation intensity. This model explains c. 66% of the variance in tree cover and appears consistent across the savanna regions of Africa. Main conclusions The analysis provides a new understanding of the importance and interaction of environmental and disturbance factors that create the broad spatial patterns of tree cover observed in African savannas. Woody cover increases with rainfall, but is modified by disturbances. These ‘perturbation’ effects depend on MAP regimes: in arid savannas (MAP < 400 mm) they are generally small (< 1% decrease in cover), while in semi‐arid and mesic savannas (400–1600 mm), perturbations result in an average 2% (400 mm) to 23% (1600 mm) decrease in cover; fire frequency and human population have more influence than cattle, and cultivation appears, on average, to lead to small increases in woody cover. Wet savannas (1600–2200 mm) are controlled by perturbations that inhibit canopy closure and reduce tree cover by, on average, 24–34%. Full understanding of the processes determining savanna structure requires consideration of resource limitation and disturbance dynamics.  相似文献

Forest harvesting and wildfire were widespread in the upper Great Lakes region of North America during the early 20th century. We examined how long this legacy of disturbance constrains forest carbon (C) storage rates by quantifying C pools and fluxes after harvest and fire in a mixed deciduous forest chronosequence in northern lower Michigan, USA. Study plots ranged in age from 6 to 68 years and were created following experimental clear‐cut harvesting and fire disturbance. Annual C storage was estimated biometrically from measurements of wood, leaf, fine root, and woody debris mass, mass losses to herbivory, soil C content, and soil respiration. Maximum annual C storage in stands that were disturbed by harvest and fire twice was 26% less than a reference stand receiving the same disturbance only once. The mechanism for this reduction in annual C storage was a long‐lasting decrease in site quality that endured over the 62‐year timeframe examined. However, during regrowth the harvested and burned forest rapidly became a net C sink, storing 0.53 Mg C ha−1 yr−1 after 6 years. Maximum net ecosystem production (1.35 Mg C ha−1 yr−1) and annual C increment (0.95 Mg C ha−1 yr−1) were recorded in the 24‐ and 50‐year‐old stands, respectively. Net primary production averaged 5.19 Mg C ha−1 yr−1 in experimental stands, increasing by < 10% from 6 to 50 years. Soil heterotrophic respiration was more variable across stand ages, ranging from 3.85 Mg C ha−1 yr−1 in the 6‐year‐old stand to 4.56 Mg C ha−1 yr−1 in the 68‐year‐old stand. These results suggest that harvesting and fire disturbances broadly distributed across the region decades ago caused changes in site quality and successional status that continue to limit forest C storage rates.  相似文献

Cyclo[EKTOVNOGN] (AFPep), a cyclic 9‐amino acid peptide derived from the active site of alpha‐fetoprotein, has been shown to prevent carcinogen‐induced mammary cancer in rats and inhibit the growth of ER+ human breast cancer xenografts in mice. Recently, studies using replica exchange molecular dynamics predicted that the TOVN region of AFPep might form a dynamically stable putative Type I beta‐turn, and thus be biologically active without additional amino acids. The studies presented in this paper were performed to determine whether TOVN and other small analogs of AFPep would inhibit estrogen‐stimulated cancer growth and exhibit a broad effective‐dose range. These peptides contained nine or fewer amino acids, and were designed to bracket or include the putative pharmacophoric region (TOVN) of AFPep. Biological activities of these peptides were evaluated using an immature mouse uterine growth inhibition assay, a T47D breast cancer cell proliferation assay, and an MCF‐7 breast cancer xenograft assay. TOVN had very weak antiestrogenic activity in comparison to AFPep's activity, whereas TOVNO had antiestrogenic and anticancer activities similar to AFPep. OVNO, which does not form a putative Type I beta‐turn, had virtually no antiestrogenic and anticancer activities. A putative proteolytic cleavage product of AFPep, TOVNOGNEK, significantly inhibited E2‐stimulated growth in vivo and in vitro over a wider dose range than AFPep or TOVNO. We conclude that TOVNO has anticancer potential, that TOVNOGNEK is as effective as AFPep in suppressing growth of human breast cancer cells, and that it does so over a broader effective‐dose range. Functional groups (FG) are an useful generalization to investigate environmental change effects on biotic communities. Assigning species to FGs is a contextual task and carries an arbitrary element, regardless of whether the grouping is obtained a priori or by sophisticated numerical methods. Using two grassland community case studies, we show that even simple FG allocation based on growth form, architecture and longevity (plants and mosses), or foraging characteristics (above-ground invertebrates) can be useful to increase our understanding of community processes. For example, the sensitivity of organisms to climate change increases with trophic rank and is higher in disturbed than in undisturbed communities. Complexity of interaction webs (in terms of web connectance), however, is larger in undisturbed than in disturbed communities. A significant and important relationship is likely to exist between anthropogenic disturbances, community complexity and the ecosystem effects of climate change. Trophic interactions may be disrupted much easier by climate changes in disturbed than in undisturbed communities where complexity may be buffering these effects.  相似文献
