Production of transgenic alfalfa plants by Agrobacterium-mediated transformation requires Agrobacterium infection and regeneration from tissue culture. Variation in alfalfa (Medicago sativa L.) germplasm for resistance to oncogenic and disarmed strains of A. tumefaciens (Smith & Townsend) Conn was tested in plant populations representing the nine distinct sources of alfalfa germplasm introduced into North America and used to develop modern varieties. For each of the virulent strains there was a positive correlation (p=0.05) of resistance to tumorigenesis with the trait for fall dormancy. There was also a significant correlation between plants selected for ineffective nodulation and resistance to tumorigenesis suggesting that the genetic loci required for successful symbiosis are also involved in tumorigenesis. Tissue explants of seedlings from the nine diversity groups were tested for transformation by three disarmed strains containing a plasmid with the scorable marker -glucuronidase. The strong correlation between dormancy and resistance to oncogenic strains was not observed with disarmed strains. However, there was a strong germplasm-strain interaction or transformation and embryogenesis in a highly embryogenic genotype. Thus, transformation at the whole plant level is germplasm dependent while in tissue culture the bacterial strain used is the critical variable for successful transformation.Abbreviations pTi tumor-inducing plasmid - GUS -glucuronidase  相似文献

We studied the inheritance of survival ability in host-associated populations of the tephritid fly, Eurosta solidaginis, to test predictions of sympatric speciation models. Eurosta solidaginis induces galls on two species of goldenrod, Solidago altissima and S. gigantea. The host-associated populations have been hypothesized to be host races that originated in sympatry (Craig et al. 1993). We found evidence for disruptive selection for host use, which is a critical assumption of sympatric speciation models. Each host race had higher survival rates on their host plant than on the alternative host. F1 and backcross hybrids also had lower survival rates than the pure host-race flies on their host plant. Since assortative mating occurs due to host-plant preference (Craig et al. 1993) this would select for divergence in host preference. Low hybrid survival could have been due to strong genetic incompatibilities of the populations or due to host adaptation by each population. Strong genetic incompatibilities would result in poor survival on all host plants, while host adaptation could result in low overall survival with high hybrid survival on some host plants with particularly “benign” environments. High survival of F1, F2, and backcross hybrids on some plant genotypes in some years supported the host adaptation hypothesis. F1 flies mated and oviposited normally and produced viable F2 and backcross hybrids indicating gene flow is possible between the host races. A few flies developed and emerged on the alternative host plant. This demonstrates that genes necessary to utilize the alternative host exist in both host races. This could have facilitated the origin of one of the populations via a host shift from the ancestral host. The inheritance of survival ability appears to be an autosomal trait. We did not find evidence that survival ability was maternally influenced or sex linked.  相似文献

We examined whether larvae of the gall midge Rabdophaga rigidae (Diptera: Cecidomyiidae) can modify the seasonal dynamics of the density of a leaf beetle, Plagiodera versicolora (Coleoptera: Chrysomelidae), by modifying the leaf flushing phenology of its host willow species, Salix serissaefolia and Salix eriocarpa (Salicaceae). To test this, we conducted field observations and a laboratory experiment. The field observations demonstrated that the leaf flushing phenology of the willows and the seasonal dynamics of the beetle density differed between shoots with stem galls and shoots without them. On galled shoots of both willow species, secondary shoot growth and secondary leaf production were promoted; consequently, leaf production showed a bimodal pattern and leaf production periods were 1 to 2 months longer than on non‐galled shoots. The adult beetle density on galled shoots was thus enhanced late in the season, and was found to change seasonally, synchronizing with the production of new leaves on the host willow species. From the results of our laboratory experiment, we attributed this synchrony between adult beetle density and willow leaf flush to beetles’ preference to eat new leaves rather than old. Indeed, beetles consumed five times more of the young leaves when they were fed both young and old leaves. These results indicate that stem galls indirectly enhance the adult beetle density by enhancing food quality and quantity late in the beetle‐feeding season. We therefore conclude that midge galls widen the phenological window for leaf beetles by extending the willows’ leaf flush periods.  相似文献

We reared six idiobiont braconids, Bracon asphondyliae, B. sunosei, B. tamabae, Simplicibracon curticaudis, Testudobracon longicaudis and T. pleuralis from 22 identified species and 11 unidentified segregates of Asphondyliini (Diptera: Cecidomyiidae) in Japan. A total of 22 cecidomyiid species and segregates were newly recorded as hosts of the braconids. Analysis of cytochrome oxidase subunit I (COI) did not show any evidence of host races among the braconids. Bracon sunosei, which was synonymized with B. asphondyliae, is restored to a valid species. The host range of the braconid species seemed to be related to the lineage of host genera within Asphondyliini.  相似文献

Saddle gall midge Haplodiplosis marginata (Diptera: Cecidomyiidae) is a pest of cereals across Europe. The occasional nature of this pest has resulted in limited and sporadic research activity. There remain important gaps in knowledge due either to a genuine lack of research or to previous research being difficult to access. These knowledge gaps make the development of effective control options difficult. Here, we review the existing literature in an attempt to consolidate the information on H. marginata from research which spans several decades and encompasses many different countries. The current distribution and pest status of this insect are updated, along with the methods of cultural and chemical control available to growers. The biology and life history of the insect are described in detail and the ecological processes governing them are discussed. A forecasting model is presented which allows the emergence of this pest in the UK to be predicted from degree day data, and the potential application of this model in management decisions is discussed. Finally, the areas in most need of further research are identified, along with suggestions of how this information can be used to help develop effective and sustainable management solutions for this pest.  相似文献

Summary S-(2-aminoethyl-)L-cysteine and L-canavanine were less toxic for octopine-type crown gall tissues that contained lysopine dehydrogenase than for other crown gall or habituated tissues. These analogs are substrates for lysopine dehydrogenase in vitro and in vivo. Summary In vitro growth rates of transformed (crown gall) and nontransformed cultures ofVinca rosea L. were greater at 32°C than at 25°C. The growth of transformed cells was significantly inhibited by kanamycin, neomycin, and chloramphenicol but not by cycloheximide. Nontransformed cells were inhibited by all four antibiotics., The relative growth rates of transformed cultures induced by four different strains, ofAgrobacterium tumefaciens did not correspond to the relative rates of tumor weight increase observed in vivo nor with the relative weights of tumor tissue in, plants 8 weeks after inoculation with the corresponding bacterial strains.  相似文献

SYNOPSIS. Zschokkella floridanae sp. n. (Myxosporidea) is described from the gall bladder of the goldspotted killifish, Floridichthys carpio, collected in south Florida. The fresh spore is circular to ovoid in front view and irregularly oblong in sutural view. It averages 14.5 in length × 13.3 μm in width.
