Monitoring of three-dimensional floodplain vegetation structure is essential for ecological studies, as well as for hydrodynamic
modelling of rivers. Height and density of submerged vegetation and density of emergent vegetation are the key characteristics
from which roughness parameters in hydraulic models are derived. Airborne laser scanning is a technique with broad applications
in vegetation structure mapping, which therefore may be a promising tool in monitoring floodplain vegetation for river management
applications. This paper first provides an introduction to the laser scanning technique, and reviews previous studies on the
extraction of vegetation height and density of forests, low vegetation and meadows or unvegetated areas. Reliable predictions
using laser scan data have been reported for forest height (R2=0.64–0.98), parameters related to forest density, such as stem number, stem diameter, biomass, timber volume or basal area
(R2=0.42–0.93), and herbaceous vegetation height (summer condition; R2=0.75–0.89). No empirical relations have been reported on density of herbaceous vegetation. Laser data of meadows and unvegetated
areas show too much noise to predict vegetation structure correctly. In a case study for the lower Rhine river, the potential
of laser scan mapping of vegetation structure was further explored for winter conditions. Three laser-derived metrics that
are often reported in the literature have been applied to characterize local vertical distributions of laser reflections.
The laser data clearly show the large structural differences both between and within vegetation units that currently are the
basis of floodplain vegetation and roughness mapping. The results indicate that airborne laser scanning is a promising technique
for extraction of 3D-structure of floodplain vegetation in winter, except for meadows and unvegetated areas. 相似文献

本文旨在建立基于高效体积排阻色谱(high-performance size-exclusion chromatography,HPSEC)偶联多角度激光散射仪(multi-angle laser light scattering,MALLS)的猪圆环病毒2型(porcine circovirus type 2,PCV2)疫苗抗原检测方法。以纯化的PCV2灭活病毒及病毒样颗粒(virus-like particles,VLP)为参照,对4家生产企业的2种PCV2灭活病毒疫苗(a、b)及VLP疫苗(c、d)破乳后进行HPSEC-MALLS检测及分子量分析;结合PCV2抗原检测卡、Western blotting和透射电子显微镜(transmission electron microscope,TEM),鉴定了特征色谱峰;考察了方法的重复性和检测线性。结果表明,两家企业生产的PCV2灭活病毒疫苗破乳液水相经HPSEC分离,在保留时间约13.3 min处出现抗原特征峰;MALLS计算该色谱峰分子量分别为2.61×106(±4.34%) Da和2.40×106(±2.51%) Da。两种VLP疫苗也在13.3 min处出现抗原特征峰,分子量分别为2.09×106(±2.94%) Da和2.88×106(±11.85%) Da,接近PCV2的理论分子量;同时在保留时间约11.4 min处也出现色谱峰,经检测分子量为4.37×106(±0.42%) Da,TEM表征显示为VLP二聚体。取疫苗d和PCV2 VLP纯品进行重复检测,抗原色谱峰面积的RSD(n=3)均小于1.5%,重复性好;将PCV2 VLP纯品梯度稀释检测,VLP及其多聚体的色谱峰面积与浓度均呈良好的线性关系,R2分别为0.999及0.997,能够满足定量及多聚体含量分析。该方法有望成为一种准确、高效的PCV2疫苗的体外评价方法,用于质量评价与提升。 相似文献

目的:探讨A型肉毒毒素联合CO_2点阵激光治疗眼周皱纹的近期临床疗效。方法:选择2013年6月到2014年6月我院接收眼部皱纹改善手术的患者90例,随机分为激光组、肉毒毒素组及联合治疗组,各30例,分别给予CO_2点阵激光治疗、A型肉毒毒素注射和A型肉毒毒素注射联合CO_2点阵激光治疗。所有患者在治疗后7 d、1月及3月进行疗效随访评价,比较三组不良反应情况。结果:三组患者眼周皱纹均有所改善,治疗1个月时效果最为明显。激光组的静态皱纹治疗效果明显,对动态皱纹治疗效果不明显;肉毒毒素组对动态皱纹治疗效果明显,对静态皱纹治疗效果不明显;联合治疗组对静态皱纹和动态皱纹均有明显的改善,其满意度评价总分数明显高于其他两组。三组患者对CO_2点阵激光和注射A型肉毒毒素所引起疼痛均能耐受,安全性好。结论:A型肉毒毒素联合CO_2点阵激光对静态皱纹和动态皱纹均有明显的改善作用,且不良反应轻,安全性好。 相似文献

Crop wild relatives (CWR) provide an important source of allelic diversity for any given crop plant species for counteracting the erosion of genetic diversity caused by domestication and elite breeding bottlenecks. Hordeum bulbosum L. is representing the secondary gene pool of the genus Hordeum. It has been used as a source of genetic introgressions for improving elite barley germplasm (Hordeum vulgare L.). However, genetic introgressions from H. bulbosum have yet not been broadly applied, due to a lack of suitable molecular tools for locating, characterizing, and decreasing by recombination and marker‐assisted backcrossing the size of introgressed segments. We applied next‐generation sequencing (NGS) based strategies for unlocking genetic diversity of three diploid introgression lines of cultivated barley containing chromosomal segments of its close relative H. bulbosum. Firstly, exome capture‐based (re)‐sequencing revealed large numbers of single nucleotide polymorphisms (SNPs) enabling the precise allocation of H. bulbosum introgressions. This SNP resource was further exploited by designing a custom multiplex SNP genotyping assay. Secondly, two‐enzyme‐based genotyping‐by‐sequencing (GBS) was employed to allocate the introgressed H. bulbosum segments and to genotype a mapping population. Both methods provided fast and reliable detection and mapping of the introgressed segments and enabled the identification of recombinant plants. Thus, the utilization of H. bulbosum as a resource of natural genetic diversity in barley crop improvement will be greatly facilitated by these tools in the future. 相似文献

The dipeptidyl peptidases (DPP) 8 and 9 belong to the DPP4 activity and/or structure homologues (DASH). Recently, a DPP9-like protein was purified from bovine testes. The aim of the present study was to prove its identity and to investigate the characteristics of this natural enzyme. We report the identification and N-terminal sequence analysis by MALDI-TOF/TOF MS, of the purified bovine enzyme as DPP9. The tryptic peptides after in-gel digestion covered 41% and 38% of the short and full-length variants of bovine DPP9, respectively. Using Asp-N digestion combined with a very recently described mass spectrometric method using DITC glass beads, the N-terminal peptide (XTGALTSERG) was isolated. It corresponds to the N-terminus of the short form of bovine DPP9. There was no evidence for glycosylation of purified bovine DPP9. The purified DPP9 was activated and stabilized by DTT. Bovine DPP9 lost its activity almost completely after alkylation with N-ethylmaleimide. Also alkylation with iodoacetamide inhibited DPP9, albeit only 70%. Other properties of bovine DPP9 are reported, including functional stability and sensitivity towards metal ions. Our results indicate that the short form of DPP9 can be isolated from bovine testes and that it behaves as a stable enzyme suitable for further functional and biochemical characterization as well as for inhibitor screening and characterization. 相似文献

The close association of the heme enzyme myeloperoxidase to phosphatidylserine epitopes on the surface of non-vital polymorphonuclear leukocytes (PMNs) and other apoptotic cells at inflammatory sites favours modifications of this phospholipid by myeloperoxidase products. As detected by matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry, ammonium ions inhibit in a concentration-dependent manner the hypochlorous acid-mediated formation of aldehyde and nitrile products from 1,2-dipalmitoyl-sn-glycero-3-phosphoserine (DPPS). Concomitantly, the formation of monochloramine (NH2Cl) raises with increasing NH4+ concentrations. A transchlorination from monochlorinated O-phospho-l-serine to NH4+ with the formation of NH2Cl occurs only when extraordinary high NH4+ concentrations are applied. Due to the low rate of 0.044 M− 1 s− 1 for this process, a transhalogenation reaction from transient chlorinated intermediates of the serine moiety to NH4+ can be ruled out as an important process contributing to the HOCl-mediated formation of NH2Cl. A significant formation of NH2Cl by myeloperoxidase interacting with DPPS in the presence of ammonium ions takes only place at acidic pH values around 5, a scenario that may occur in phagosomes of macrophages after the uptake of apoptotic PMNs. 相似文献
