To discover new inhibitors on tissue factor procoagulant activity, 20 pentacyclic triterpenes were semi-synthetized through microbial transformation and assayed on the model of human THP-1 cells stimulated by lipopolysaccharide. In the biotransformation two types of reactions were observed, regio-selective hydroxylation and glycosylation. The bioassay results showed that most of tested compounds were significant effective on this model and two of the biotransformation products 23-hydroxy-28-O-β-d-glucopyranosyl betulinic acid (3d) and 28-O-β-d-glucopyranosyl oleanic acid (1a) exhibited most potential activities with the IC50 values of 0.028, 0.035 nM respectively. The preliminary structure and activity relationship analysis revealed that the aglycones with single free hydroxyl group on the skeleton (1, 1j) were less effective than that with more free hydroxyl groups (1d, 1f, 2), mono-glycosylation can significantly enhance their inhibitory effects. Our findings also provide some potential leading compounds for tissue factor-related diseases, such as cancer and cardiovascular diseases.  相似文献

从万寿菊(Tagetes ereata L.)花中分得8个已知化合物,分别为丁香酸(Syringic acid,1),3,4二羟基苯甲酸(3,4-dihydroxybenzonic acid,2),β-香树脂醇(β-amyrin,3),槲皮万寿菊素(Quercetagetin,4),Quercetagetin 7-methyl(5),β-谷甾醇(β-sitosterol,6),豆甾醇(stigmasterol,7),正三十四烷(Tetratriacontane,8)。应用光谱法鉴定了所有化合物的结构。  相似文献

从镰叶西番莲(Passiflora wilsonii)的腾茎中分离到6个化合物,经理化性质和波谱分析,分别确定为:glut-5-en-3β-ol(I)、maslinic acid(Ⅱ),friedelin(Ⅲ),ergosterol epidioxide(Ⅴ)和β-谷甾醇(Ⅳ)。以上化合物均为首次从镰叶西番莲的腾茎中分离得到。  相似文献

The oleanane triterpenes arjunic acid, arjungenin and their glucosides, arjunetin and arjunglucoside II, were isolated from the bark of Terminalia arjuna. Arjungenin and its glucoside exhibited a moderate free radical scavenging activity while all the compounds showed no effect on the superoxide release from PMN cells. Further arjungenin also exhibited greater inhibitory action on the hypochlorous acid production from human neutrophils.  相似文献

The translocation of several plant sterols and a triterpene was studied in geranium and sunflower plants. Upward translocation of sitosterol-[14C] and β-amyrin-[14C] was shown within 48 hr to the upper parts of a geranium plant sectioned previously above the roots. Downward translocation of sitosterol-[14C] from the leaf of application was evident in intact plants after 48 hr. In addition to free sitosterol-[14C] considerable amounts of sitosteryl-[14C] glycoside and traces of sitosteryl-[14C] ester were found in most parts examined. Very slow downward translocation of cholesterol-[14C] but not of desmosterol-[14C], sitosteryl-[14C] palmitate or β-amyrin-[14C] was shown in geranium. In sunflower no downward translocation of cholesterol-[3H], sitosteryl-[3H] acetate or palmitate could be detected. In geranium, sitosteryl-[14C] glycoside translocated downward from the leaf of application to all other plant parts, except other leaves, and was found in these parts after 10 days as the unchanged glycoside, free sterol and steryl ester. The effect of drying the plant parts on the recovery of radioactive steroidal material is discussed. Traces of a water soluble, dialyzable form of sterol-[14C] were also detected in dried geranium roots after treatment with strong acid or alkali.  相似文献

From the resins of Dipterocarpus hispidus, Dipterocarpus zeylanicus and Doona macrophylla, asiatic (2α,3β,23α-trihydroxyurs-12-en-28-oic) and 2α,3β-dihydroxyurs-12-en-28-oic acids have been isolated. The resin of Doona macrophylla contains ursolic acid and that of Doona congestiflora asiatic acid, 20β-hydroxy-3-oxo dammar-23-ene (Dipterocarpol) and a dihydroxyolean-12-en-28-oic acid. The bark of Dipterocarpus hispidus contains betulinic acid, dipterocarpol, and 3β,20β-dihydroxydammar-23-ene (dammarenediol 20S) whilst the timber contained dipterocarpol and asiatic acid.  相似文献
