A purified mitochondrial DNA (mtDNA) probe was used to examine restriction fragment length polymorphisms produced by six restriction enzymes ( Xba I, Eco RV, Ava II, Hinf I, Hae III, Mbo I) in 915 brown trout from western Europe. A total of 20 composite haplotypes were found with one to seven haplotypes in individual populations. Icelandic trout samples from north, south, east, and west coast drainages showed only a single common haplotype in contrast to the high level of polymorphism found in Irish and Scottish populations. The phylogeny of mtDNA haplotypes and the pattern of haplotype distribution suggests that post-glacial colonization of brown trout in NW Europe was more complex than the dual colonization model which has been proposed on the basis of differential LDH-5* allele distribution. One male and one female lake trout, Salvelinus namaycush , were held in a laboratory tank simulating a natural spawning environment. Their behaviour during their reproductive period was videotaped each day for 2 h at dusk. The fish activities were classified into seven types: resting, casual swimming, side-thrusting, chase, chase-nudge, body alignment, shuddering. As the reproductive period progressed the fish became more generally active with a marked increase in interactive behaviours such as pursuit of the female by the male (chasing, chasing-nudging). Around spawning time, shuddering and body alignment activity reached apeak. Each fish was equipped with an internally implanted radiotelemetry package able to detect and transmit continuously over the course of each day signals that reflect production of the electromyograms (EMGs) that accompany fish axial muscle activity (high muscular activity gives low pulse interval times in milliseconds). The lowest EMG pulse intervals corresponded with what, from the video records, appeared to be the time of highest physical activity during the reproductive period–viz. that associated with spawning activity during dusk. Other times during the diel cycles over the reproductive period, except for times of roughly similar duration during early morning (which may also be reflections of spawning) showed considerably lower muscular activity. It is concluded that transmitted EMG records could probably be used as indicators of spawning activity of lake trout in at least some of those places in the field where they cannot normally be located by visual means.  相似文献

The expression of gene(s) governing apomictic reproduction inTripsacum provides the best foundation for comparing the effectiveness of apomictic reproduction in a series of maize-Tripsacum hybrids. Several 38-chromosome, apomictic maize-Tripsacum hybrids are available which possess the gene(s) conferring apomictic reproduction fromTripsacum. Without a base line for comparison, studies directed towards discerning the successful transfer or effectiveness of gene expression in a maize background are hampered. The objectives of this study are to compare the reproductive features found in apomicticTripsacum with those in apomictic maize-Tripsacum hybrids. In addition, this study determined the feasibility of utilizing these maize-Tripsacum hybrid materials to continue an attempt to transfer the genes into a pure maize background. The frequency and occurrence of five unique reproductive features found in apomictic accessions ofTripsacum dactyloides were compared to the reproductive behaviours exhibited in the maize-Tripsacum hybrids. Results indicate the genes controlling apomixis in tetraploidTripsacum are fully functional in maize-Tripsacum hybrids with diploid and triploid maize constitutions. The ability of theTripsacum apomictic genes to retain full expression provides evidence to continue their transfer to a diploid or tetraploid maize background.The use of company names in this publication does not imply endorsement by the USDA-ARS, or the product names or criticism of similar ones not mentioned. Analysis of chorion hardening of eggs of rainbow trout, Oncorhynchus mykiss   总被引:1,自引:1,他引:0  
We estimated changes of chorion hardness of rainbow trout (Oncorhynchus mykiss) egg by the use of three parameters, namely increase of resistance of an egg to rupture by extraneously applied pressure, decrease of solubility of chorion proteins in 8 mol/L urea and a change in the content of γ-glutamyl-ε-lysine crosslink. Unfertilized egg chorions became hardened after egg activation. During chorion hardening, 49, 56 and 65 kDa protein components of the chorion gradually disappeared, high molecular weight intermediates (113,160–170 and higher than 250 kDa) were newly formed and, finally, all components became undetectable by sodium dodecylsulfate-polyacrylamide gel electrophoresis. The content of γ-glutamyl-ε-lysine (γ-Glu-ε-Lys) crosslink in the chorion increased after hardening. Chorion hardening was inhibited by the incorporation of monodansyl-cadaverine, a competitive inhibitor for transglutaminase (TGase), into the chorions. TGase activity was detected in unfertilized eggs and localized in the chorion fraction rather than in the ooplasmic fraction. The findings suggest that chorion hardening depends upon polymerization of the chorion components by TGase-dependent γ-Glu-ε-Lys crosslink formation.  相似文献

As part of the Russian-Finnish research studies on Lake Ladoga, joint expeditions were organized in 1992 and 1993. Water samples were collected for intercalibration of chemical analysis methods and to monitor the chemical quality of the lake water.In August of 1992 water samples were taken from northern Lake Ladoga for intercalibration of Russian and Finnish analysis methods. In August 1993 water samples were collected from 23 sampling stations in all parts of the lake; some of these were also used for intercalibration purposes.The oxygen, colour and CODMn results were at the same level in the intercalibration. In 1993, the Ptot results obtained were acceptable. In Ntot, Fe and Mn analysis there seemed to be systematic and random errors between some results.The Secchi depth ranged from 1.5 m to 3.3 m. The average concentrations for the total phosphorus ranged from 15 µg 1–1 to 29 µg 1–1. The total nitrogen values were from 620 µg 1–1 to 690 µg 1–1. The N:P ratio varied from 24 to 40. The concentration of phosphorus indicated mesotrophic or even eutrophic conditions in the lake. Phosphorus seemed to be the limiting nutrient to bacteria and algae.  相似文献

164 taxa were identified in the net zooplankton of the pelagial of L. Peipsi-Pihkva in 1909–1987, including 3 species of protozoans, 74 species of rotifers, 58 species of cladocerans, 28 species of copepods and 1 mollusc. One rotifer species, Ploesoma peipsiense Mäemets et Kutikova, has been described as new for science here. The zooplankton of L. Peipsi-Pihkva is remarkably rich in species including rarities in Estonia: Limnosida frontosa, Drepanothrix dentata, Bythotrephes longimanus, B. cederstroemi etc. Due to its large surface area, L. Peipsi-Pihkva provides a large scale of biotopes of a diverse trophic state and humic content, which support species with different ecological requirements. Most of the aquatory of the lake has lately been mesotrophic, favouring the coexistence of indicators of oligo- and mesotrophic state and species preferring a higher trophic state. The occurrece of 10 species of the genus Bosmina including B. berolinensis, B. gibbera, B. lilljeborgi, B. thersites and B. crassicornis, sparse in Estonian lakes, is the most noteworthy feature of the zooplankton of L. Peipsi-Pihkva. The coexistence of B. coregoni and B. berolinensis, B. gibbera, B. lilljeborgi etc. which were earlier regarded as subspecies of B. c. coregoni proves that they are different species producing usually no hybrids. The species composition was subjected to certain changes during the years under consideration. Larvae of Dreissena were first found in zooplankton in 1962. The oligo-mesotrophic indicator Holopedium gibberum occurred in the lake in 1909–1964, but was lacking in later samples.  相似文献

Bacterial abundance and production in a vertical profile in Lake Kariba (17dgS), Zimbabwe, were affected by solar irradiance. At the surface, 1.87 × 109 bacteria 1–1 were found and abundance peaked at 10 m (2.5 × 109 bacteria l-1), then decreasing with depth. Bacterial reproduction at the surface(0.145 µg C1–1 h–1) was nearly four times less than the production at 10 m although bacterial numbers were only 26% less. Thus, bacterial production per cell was lower at the surface than deeper down, suggesting that bacterial production is inhibited at the surface.Bacterial production in GF/F filtered lake water in Whirl Pack bags showed an exponential decrease down to 3 m depth. The inhibition was well in accordance with light extinction in the UV region. Phosphatase activity was low in light exposed bags compared to dark, indicating photolysis of extracellular enzymes, or phototransformation of recalcitrant DOM, which substitutes enzyme activity. Hypolimnetic enzyme activity was less affected by solar light than epilimnetic.  相似文献

The late-spring quantitative relationship between epiphyton and macroinvertebrates was analyzed on the basis of units of colonizable plant surface of Typha angustifolia, Phragmites australis and Nuphar lutea (floating leaves) in the shallow euthrophic Lake Loosdrecht (the Netherlands), with a high seston load. The non-predatory chironomid larvae (Glyptotendipes viridis, Endochironomus albipennis, Pentapedilum sordens, Cricotopus sylvestris agg.) dominated among the macroinvertebrate taxa, controlling the diversity and resemblance of macroinvertebrate assemblages. There was a gradient in functional feeding groups among the chironomids from continuous filtering of the seston to prevailing utilization of epiphyton. We found no direct relationship between the total macroinvertebrate abundance and the epiphyton mass on the plants surface. We attribute this to the filter feeding-strategy of the most abundant species, Glyptotendipes viridis, that utilizes seston in the eutrophicated lake.  相似文献

Abstract: The Blue Lagoon in Iceland is a shallow geothermal lake with average temperatures of 37°C, pH 7.5 and about 2.5% salinity. It was formed in 1976 from the effluents of the Svartsengi geothermal power plant and is saturated with silica which constantly precipitates in the lake. It has been colonized by a few types of specialized microorganisms which seem to proliferate in this unusual ecosystem. The average bacterial colony count in the lake was 1.3 × 105 ml−1 on plate count agar made with 50% Blue Lagoon fluid but 2.6 × 106 ml−1 when determined with the MPN method. A total of 99 isolates were purified and characterized by 54 phenotypic tests and then grouped using Numerical Taxonomy. At similarity values of 80%, one major cluster was formed containing 85% of the isolates. Four representative strains from this cluster were further characterized and all shown to be Gram-negative, obligately aerobic, non-motile rods. They were oxidase positive, catalase negative and grew optimally at 45°C and in 3.5% NaCl with doubling time of about 80 min.  相似文献
