Human umbilical vessels are unique in lacking any innervation; thus endothelial cells may play the major role in local control and regulation of the blood flow. In the present study, we examined ultrathin sections of cultured human umbilical vein endothelial cells and tissue preparations of umbilical vein and artery, immunostained by the post-embedding colloidal gold double-labelling technique. We observed colocalization of atrial natriuretic peptide and neuropeptide Y, as well as colocalization of atrial natriuretic peptide and neuropeptide Y with other vasoactive substances, namely, vasoactive intestinal peptide, substance P, calcitonin gene-related peptide and arginine vasopressin. The functional significance of the colocalization of these vasoactive substances in the human umbilical vessel endothelial cells is discussed.  相似文献

 Storage and remobilization of nitrogen (N) were studied in ash trees (Fraxinus excelsior) under both field and greenhouse conditions. Experiments in the greenhouse providing 15N labelled fertilizer to the trees showed that the major quantity of N remobilized during subsequent spring was from the roots, and only a small amount from the stem. This corresponded with a loss of soluble N (proteins and low-molecular-weight compounds) from both roots and stem. On the two field sites, which differed in water availability, there was a decrease in bark N content during leaf growth, but on the dry site net N export from the bark was sustained throughout the whole vegetation period. Remobilized N was derived from soluble proteins and low-molecular-weight compounds on the moist site, which was demonstrated by the seasonal dynamics of a 56 kDa polypeptide in bark and wood. On the dry site, lower contents of soluble proteins were associated with smaller amounts of N remobilized compared to the moist site. Uptake studies of 15N labelled fertilizer indicated a higher contribution of current uptake to leaf N increment during spring at the dry site compared to the moist site. Differential N availability during the season had a decisive effect on the nitrogen storage dynamics at the two sites. Thus the influence of current N supply on N remobilization and storage as found in the greenhouse-grown plants could be verified under field conditions. Summary Mitochondrial (mt) DNAs of somatic hybrids obtained by electrical and chemical fusion of mesophyll protoplasts of S. brevidens and a dihaploid line of S. tuberosum PDH 40 were analysed by Southern hybridization using the digoxigenin-labelled mtDNA sequences nad5 or orf25. In the Southern analysis of the hybrid mtDNA probed with nad5, most of the 19 hybrids analyzed had an RFLP pattern similar, but not identical, to one of the parents, S. tuberosum, PDH40. Nineteen percent of the hybrids had most of the S. brevidens fragments. Five of the hybrids had an identical RFLP pattern to either one of the parents while another two hybrids had novel RFLP patterns. Similar results were obtained by Southern analysis with orf25. These results clearly show that mtDNA rearrangements had occurred at a high frequency in the somatic hybrids. There were no differences in the frequencies of rearrangements observed between the hybrids regenerated from chemical and electrical fusions.  相似文献

Wetland plants create partly aerobic conditions in the rhizosphere by releasing oxygen to the waterlogged substrate. The present study was conducted to characterise the arrangement of rhizobacteria, especially those active in methane oxidation, in root-associated biofilms of wetland plants. Root cross sections sampled from Typha latifolia L. (broadleafed cattail) and Phragmites australis (Cav.) Trin. ex Steud. (common reed) were scanned using light and electron microscopy. Methane-oxidising bacteria were identified and quantified by immunological labelling of the α-subunit of the methanol dehydrogenase (α-MDH; encoded in mxaF). On roots of both species there was a diverse subset of bacteria arranged in a microbial biofilm around the roots’ exodermis. Similar bacterial densities in the root-associated biofilm were detected in more basal regions and closer to the root tip. Many microbes carried notable internal membrane systems that are characteristic of methanotrophic bacteria. This morpho-anatomical characterisation was confirmed by immunogold labelling with α-MDH antibodies. Quantification of labelled bacteria revealed that 34–43% of the biofilm bacteria were potentially capable of methane turnover. These findings confirm the presence of methane-oxidising bacteria in the root-associated biofilms of the two common macrophytes T. latifolia and P. australis. This implies that the methanotrophs participate essentially in the microbial processes related to oxygen-releasing roots of wetland plants.  相似文献

The present state of worldwide discussions of how to apply LCA in environmental labelling, taking into account the current ISO 14 020 and ISO 14 024 works, is described. There is a consensus to use LCA as a tool for more scientific environmental labelling. The examples presented verify some practical possibilities to realise this approach. As a background to different stages of practical labelling, results from LCA studies are already used in the German “Blue Angel” scheme, e.g. for the definition of the scope in one product category, for the priorisation of specific life cycle phases and criteria, as a basis to establish a scoring system or to emphasise the importance of information on how to use environmentally sound products. Practical examples are presented in detail for hand-drying systems, paper products, milk packages, household equipment, televisions and detergents. Some future perspectives are mentioned.
