Antifeedant and toxic activity of Trichilia americana extract against the larvae of Spodoptera litura 总被引:3,自引:0,他引:3
The biological activity of a crude methanolic extract of Trichilia americana (Sesse and Mocino) Pennington (Meliaceae) was assessed using the Asian armyworm, Spodoptera litura (Fabr.) (Lepidoptera: Noctuidae). The extract exhibited strong antifeedant activity in a choice leaf disc bioassay with 0.18 g cm–2 extract deterring feeding by 50%. In nutritional assays, the crude extract reduced growth, consumption and the utilisation of ingested and digested food in a dose-dependent manner when fed to larvae, suggesting both antifeedant and toxic activities. When relative growth rates were plotted against relative consumption rates, the growth efficiency of the S. litura fed on diet containing T. americana crude extract was significantly less than that of control larvae. This result further indicates that the extract acts as both an antifeedant and chronic toxin. Toxicity is only seen following ingestion and was not observed following topical application or injection into the hemocoel. Larvae reared initially on extract-containing diet then transferred to control diet showed nutritional indices comparable to those of larvae fed continuously on control diet. This suggests that the extract is not permanently damaging the insect's digestive tract. The mode-of-action of the extract as a chronic toxin remains unknown. 相似文献

Avoidance thresholds and 96-h LC50 values were determined for golden shiners, Noiemigonus crysoleucas , for five individual elements: chromium, Cr; copper, Cu; cadmium, Cd; arsenic, As; selenium, Se. The avoidance concentrations were 73, 26 and 28 μgl-1 for Cr-VI, Cu and As-III, respectively. Cadmium and Se were not avoided at experimental concentrations up to 68 and 3489 μg1-1 , respectively. Acute flow-through 96-h LC50 values were Cr-VI 55·0, Cu 84·6, and As-III 12·5 mg 1-1 , which were more than two orders of magnitude above avoidance concentrations. The acute flow-through 96-h LC50 values for Cd and Se were 2·8 and 11·2 mg 1-1 , respectively. These concentrations are 31 and 2·2 times the highest concentration employed in the avoidance tests, neither of which were avoided by the test organisms. Thus, simple toxicity tests do not identify the environmental hazard of some elements, and the most toxic elements may not elicit a behavioural response. When used in concert with tests of organism function, more realistic indicators of environmental hazard or safety may be determined. 相似文献

Desulfovibrio vulgaris Hildenborough is a well-studied sulfate reducer that can reduce heavy metals and radionuclides [e.g., Cr(VI) and U(VI)].
Cultures grown in a defined medium had a lag period of approximately 30 h when exposed to 0.05 mM Cr(VI). Substrate analyses
revealed that although Cr(VI) was reduced within the first 5 h, growth was not observed for an additional 20 h. The growth
lag could be explained by a decline in cell viability; however, during this time small amounts of lactate were still utilized
without sulfate reduction or acetate formation. Approximately 40 h after Cr exposure (0.05 mM), sulfate reduction occurred
concurrently with the accumulation of acetate. Similar amounts of hydrogen were produced by Cr-exposed cells compared to control
cells, and lactate was not converted to glycogen during non-growth conditions. D. vulgaris cells treated with a reducing agent and then exposed to Cr(VI) still experienced a growth lag, but the addition of ascorbate
at the time of Cr(VI) addition prevented the lag period. In addition, cells grown on pyruvate displayed more tolerance to
Cr(VI) compared to lactate-grown cells. These results indicated that D. vulgaris utilized lactate during Cr(VI) exposure without the reduction of sulfate or production of acetate, and that ascorbate and
pyruvate could protect D. vulgaris cells from Cr(VI)/Cr(III) toxicity.

We have recently shown that MEHP induces spermatogenic cell apoptosis in guinea pigs at prepubertal stage in vitro. To evaluate the effects of MEHP on the testicular tissues of guinea pigs in vivo, we conducted this research work. Five weeks old male guinea pigs were used in this experiment. They received a single oral dose of 2000 mg/ml of MEHP in corn oil by gavage at a volume equal to 4 ml/kg. Control group received a similar volume of corn oil vehicle. Vehicle- and MEHP-treated guinea pigs were sacrificed at the interval of 3, 6, and 9 h, and the testicular tissues were processed for histopathological studies. Distinct histopathological changes were recognized in testes. Detachment and displacement of spermatogenic cells, thin seminiferous epithelia, vacuolization of Sertoli cells were prominent at 6 h after MEHP treatment. The lumina of the efferent ductules were frequently occupied with sloughed seminiferous epithelia from 6 to 9 h after MEHP treatment. Apoptotic spermatogenic cells appeared at 3 h in the control group. The incidence of apoptotic spermatogenic cells significantly increased (*p<0.05) from 3 to 9 h, and the maximal increase of apoptotic spermatogenic cells were observed at 9 h after MEHP treatment. Time-dependent increases of apoptotic spermatogenic cells was recognized throughout the experimental period. It may be suggested here that MEHP also induces spermatogenic cell apoptosis in guinea pigs in vivo and guinea pigs may be considered as a useful animal model for sensitivity test of the reproductive toxicity to some phthalate esters at their earlier stage in vivo. 相似文献

The effects of cadmium (Cd) uptake on ultrastructure and lipid composition of chloroplasts were investigated in 28-day-old tomato plants (Lycopersicon esculentum var. Ibiza F1) grown for 10 days in the presence of various concentrations of CdCl2. Different growth parameters, lipid and fatty acid composition, lipid peroxidation, and lipoxygenase activity were measured in the leaves in order to assess the involvement of this metal in the generation of oxidative stress. We first observed that the accumulation of Cd increased with external metal concentration, and was considerably higher in roots than in leaves. Cadmium induced a significant inhibition of growth in both plant organs, as well as a reduction in the chlorophyll and carotenoid contents in the leaves. Ultrastructural investigations revealed that cadmium induced disorganization in leaf structure, essentially marked by a lowered mesophyll cell size, reduced intercellular spaces, as well as severe alterations in chloroplast fine structure, which exhibits disturbed shape and dilation of thylakoid membranes. High cadmium concentrations also affect the main lipid classes, leading to strong changes in their composition and fatty acid content. Thus, the exposure of tomato plants to cadmium caused a concentration-related decrease in the fatty acid content and a shift in the composition of fatty acids, resulting in a lower degree of fatty acid unsaturation in chloroplast membranes. The level of lipid peroxides and the activity of lipoxygenase were also significantly enhanced at high Cd concentrations. These biochemical and ultrastructural changes suggest that cadmium, through its effects on membrane structure and composition, induces premature senescence of leaves. 相似文献

Forty pregnant long-tailed macaques were treated daily for 30 d with 0, 25, 150 or 300 μg selenium as L-selenomethionine/kg
body weight. Erythrocyte and plasma selenium and glutathione peroxidase specific activities, hair and fecal selenium, and
urinary selenium excretion were increased by and were linearly related to L-selenomethionine dose. Hair selenium was most
sensitive to L-selenomethionine dose, with an 84-fold increase in the 300 μg selenium/(kg-d) group relative to controls (r=0.917). Daily urinary selenium excretion (80-fold,r=0.958), plasma selenium (22-fold,r=0.885), erythrocyte selenium (24-fold,r=0.920), and fecal selenium (18-fold,r=0.911) also responded strongly to L-selenomethionine. Erythrocyte and plasma glutathione peroxidase specific activities increased
154% and 69% over controls, respectively. Toxicity was associated with erythrocyte selenium >2.3 μg/mL, plasma selenium >2.8
μg/mL, and hair selenium >27 μg/g. Plasma, erythrocyte, and hair selenium concentrations may be useful for monitoring and

龙柏精油对小菜蛾Plutella xylostella(L.)的室内生物活性测定结果表明,熏蒸作用是龙柏精油对小菜蛾的主要作用方式,熏蒸实验中当龙柏精油浓度为5.00 mg/L2、.50 mg/L时,处理后48 h小菜蛾2龄幼虫的校正死亡率分别为92.33%、80.29%。龙柏精油对小菜蛾具有较强的非选择性拒食作用和产卵忌避作用,但由于精油的挥发性效果不持久。当龙柏精油浓度为5.00 mg/L和2.50 mg/L时,24 h小菜蛾的产卵忌避率分别为80.45%和77.42%;而48 h则降为72.30%和61.79%。当龙柏精油浓度为5.00,2.50和1.25 mg/L时,24 h的非选择性拒食率分别为86.70,74.36和70.93%,48 h的非选择性拒食率分别降为82.36,71.40和60.40%。龙柏精油对小菜蛾具有一定的触杀活性,当龙柏精油浓度为5.00 mg/L时效果最好,24 h和48 h的校正死亡率分别为60.00%和76.67%。 相似文献

The mechanism of resistance toB toxicity in barley and wheat was studied in a solution culture experiment using several cultivars displaying a large range of sensitivity to excessB supply. Plants were cultured for 35 d atB concentrations ranging from normal to excessive (15 to 5000 M, respectively) then examined for dry matter production and theB distribution between roots and shoots.In both species, increasedB supply was accompanied by increased tissueB concentrations, development ofB toxicity symptoms and depressed growth. At each level ofB supply, however, resistant cultivars accumulated considerably lessB than did sensitive cultivars, in both roots and shoots. Even at the lowestB supply, at which noB toxicity symptoms developed and growth was not affected, resistant cultivars maintained relatively low tissueB concentrations. No cultivar displayed an ability to tolerate high tissueB concentrations.These results indicate that sensitivity toB toxicity in barley and wheat is governed by the ability of cultivars to excludeB. If theB concentrations of tissues is used to indicate resistance toB toxicity, then cultivars have the same ranking whether cultured at a normal or excessB supply. 相似文献
