Bioaugmentation is an effective treatment method to reduce recalcitrant pollutants from polluted sites. Dimethylformamide (DMF) is a very common toxic organic solvent among the effluents of textile and pharma industries. DMF was degraded by pre-adapted Paracoccus denitrificans SD1 with indigenous mixed cultures in both bioaugmentation and non-bioaugmentation conditions. In free cell condition, augmentation was not much significant due to competition among the bacterial cells and direct exposure of cells to toxic level of DMF. To enhance the degradation of DMF, cells were entrapped in PVA–alginate matrix individually and collectively for bioaugmentation experiments. Bioaugmentation is successful when immobilized P. denitrificans SD1 is introduced higher inoculum volume with indigenous cultures in continuous packed bed reactor system. This treatment has succeeded in removing 91.3% of 3% (v/v) DMF from the industrial effluent. This investigation advocates that bioaugmentation enhances the DMF removal efficiency by about 20% when compared to individual degradation by P. denitrificans SD1.  相似文献

The effect of thermodialysis on the enzymatic kinetic synthesis of the antibiotic cephalexin was investigated. As reference points, two existing models for an immobilised enzyme (Assemblase®) and for the free enzyme were used. For Assemblase®, it is known that diffusion limitation occurs and that therefore considerably more of the undesired side-product phenylglycine is formed.

The enzyme was immobilised on a membrane, and under isothermal conditions (293 K) the course of the reaction resembled that of the Assemblase® enzyme. However, if a temperature gradient was applied across the membrane, with an average temperature of 293 K for the enzyme, than the course of the reaction changed. For large temperature gradients (30° and more), the course of the reaction resembled that of free enzyme. Thermodialysis enhances mass transfer across the membrane and therewith reduces diffusion limitations in the immobilised enzyme on the membrane.

The stability of the immobilised enzyme is such that the reactor can be re-used repeatedly. This, together with the positive effect of the temperature gradient on the course of the reaction, makes thermodialysis an interesting new technique that has potential to be applied on a larger scale if the membrane surface area per volume of reactor can be improved.  相似文献

Nitrobenzene (NB) is an important industrial raw material in organic synthesis. However, successful biological treatment is challenging since NB wastewater is biologically toxic. During the experiment, the performance was examined during the acclimation process of NB in an anaerobic baffled reactor (ABR). The removal efficiencies of NB and chemical oxygen demand were 98% and 90%, respectively. Furthermore, by applying polymerase chain reaction-denaturing gradient gel electrophoresis (DGGE) technology of 16SrDNA, this paper analyzes the structural change of the archaea community in the ABR before and after NB acclimation and identifies the dominant community. The sequence structure analysis of archaea 16S rDNA in DGGE profiles shows that after NB biodegradation, the archaea-dominant community primarily consists of Methanothrix soehngenii from Methanosarcina, Methanosaeta concilii from Methanosaeta, Methanobacterium beijingense 8-2, uncultured Archaeon TA04, and uncultured Methanobacterium sp. isolated from environmental samples, which may be the important functional archaea in an ABR for NB biodegradation. The study of the population structure distribution and the dominant archaea community is helpful for elucidating the mechanisms of the anaerobic biodegradation mechanism of NB.  相似文献

Soil in some parts of the Iowa Army Ammunition Plant in Burlington, Iowa, was contaminated with cyclotetramethyleneter-anitramine, commonly known as high melting explosive (HMX). A laboratory treat-ability study was conducted to find out the ability of the native soil bacteria present in the contaminated site to degrade HMX. The results indicated that the HMX can be removed effectively from soil by native soil bacteria through a co-metabolic process. Molasses, identified as an effective co-substrate, is inexpensive, and this factor makes the treatment system cost-effective. The successful operation of aerobic-anoxic soil slurry reactors in batch mode with HMX-contaminated soil showed that the technology can be scaled up for field demonstration. The HMX concentration in the contaminated soil was decreased by 97% in 4 months of reactor operation. The advantage of the slurry reactor is its simplicity of operation. The method needs only mixing and the addition of molasses as co-substrate.  相似文献

Snapping shrimp (Alpheus heterochaelis) produce a fast, well-focused water jet by rapid closure of their specialised snapper claw. As shown previously, water jets may injure the opponent in interspecific encounters (e.g. with small crabs) although no damage was observed in intraspecific encounters. For conspecific receivers the jet represents a potential hydrodynamic signal and can be analysed with the help of mechanosensory hairs. To gain more insight in the biophysical characteristics of the water jet we visualised and analysed jets of tethered snapping shrimp using standard and high speed video recordings. Water jet width increases with increasing distance from the snapper claw tip, and both width and distance increase with increasing snapper claw size. Water jet distances do not increase with increasing claw cocking duration (building up muscle tension) but medium cocking durations of about 550 ms result in longest distances. Mean water jet velocity is 6.5 m s−1 shortly after claw closure but rapidly decreases subsequently. At the mean distance between snapping conspecifics (9 mm) water jet velocities produced by snapping shrimp with larger snapper claws are significantly higher than those of animals with smaller claws. Interestingly, males with equal snapper claw size as females produce significantly faster water jets. Accepted: 31 March 1999  相似文献

Sialylated galactooligosaccharides (GOS) represent a potential infant formula ingredient, which is believed to contribute with a combination of the beneficial properties of the prebiotic GOS as well as of sialylated human milk oligosaccharides. Sialylated GOS do not exist in natural milk, but can be produced from κ(kappa)-casein glycomacropeptide (CGMP), a sialylated side stream component from cheese-making, by sialidase-catalyzed transsialylation. Using a rationally designed mutant of the sialidase from Trypanosoma rangeli, Tr13, with enhanced transsialylation activity, six different GOS preparations with a varying degree of polymerization (DP) were effectively sialylated with molar yields of 20–30% on the CGMP sialyl in batch reactions. The rate of sialylation of the individual DPs was largely dependent on the DP distribution in each GOS preparation, and Tr13 catalysis did not discriminate against large GOS molecules, providing the novelty point that GOS molecules are sialylated independently of their size by Tr13. Using CGMP, GOS, and Tr13, the production of gram-scale quantities of sialyl-GOS was achieved in 20 L volume reactions. Compared to the benchmark transsialidase from pathogenic Trypanosoma cruzi, the Tr13 was significantly more thermostable. By employing an enzymatic membrane reactor, Tr13 could be recycled and after seven consecutive 1-h reaction cycles, the biocatalytic productivity of the enzyme was increased 7-fold compared to the batch reaction. Assuming that the enzyme may be specific for α-2,3-bound sialyl moieties only, and that only 50% of sialyl linkages in CGMP are α-2,3-linked, the molar yield of sialyl-GOS on the available α-2,3-bound sialyl moieties in CGMP reached 80% in the enzymatic membrane reactor system.  相似文献

Proteolytic digestion of gelatin extracts from Alaska Pollack (Theragra chalcogramma) skin brings about a high angiotensin I converting enzyme (ACE) inhibitory activity. Gelatin extracts were hydrolyzed by serial protease-treatments in the order of Alcalase, pronase E, and collagenase using a three-step recycling membrane reactor. Fragments arising from the third step were composed of peptides ranging from 0.9 to 1.9 kDa and responsible for ACE inhibitory activity. Catalytically active two peptides were separated by the consecutive chromatographic methods including gel filtration, ion-exchange chromatography, and reverse-phase high performance liquid chromatography. The isolated peptides were composed of Gly-Pro-Leu and Gly-Pro-Met and showed IC50 values of 2.6 and 17.13 μM, respectively. These results suggested that Gly-Pro-Leu would be useful as a new antihypertensive agent.  相似文献

Kinetic measurements of the benzyl acetate hydrolysis by pig liver esterase in a two-liquid phase stirred tank reactor were made at a variety of aqueous phase enzyme concentrations and stirrer speeds. All experiments were performed in an inverted liquid-liquid system at a high phase ratio. The results were explained in terms of the aqueous phase bulk reaction model developed from previous Lewis cell studies. An algorithm is presented for the indirect measurement of the substrate mass transfer coefficient and consequently a model was developed to predict reaction rates. While the model describes the kinetics effectively, and could therefore be used to predict reactor behaviour, no difference was observed between kinetic measurements made at a stirrer speed of 750 and 1000 rpm.  相似文献

Archer fishes are skillful hunters of terrestrial prey, firing jets of water that dislodge insects perched on overhead vegetation. In the current investigation, we sought an answer to the question: are distant targets impractical foraging choices? Targets far from the shooter might not be hit with sufficient force to cause them to fall. However, observations from other investigators show that archer fish fire streams of water that travel in a non-ballistic fashion, which is thought to keep on-target forces high, even to targets that are several body lengths distant from the fish. We presented targets at different distances and investigated three aspects of foraging behavior: (i) on-target forces, (ii) shot velocity, (iii) a two-target choice assay to determine if fish would show any preference for downing closer targets or more distant targets. In general, shots from our fish (Toxotes chatareus) showed a mild decrease (less than 15% on average) in on-target forces at our most distant target offered (5.8 body lengths) with respect to the closest target offered (2.3 body lengths). One individual in our investigation showed slightly, but significantly, greater on-target forces as target distance increased. Forces on the furthest targets offered were found to double that of attachment forces for 200 mg insects, even for individuals whose on-target forces showed mild decreases with increases in target distance. High-speed video analysis of jet impact with the target revealed that the shot was traveling in a non-ballistic manner, even to our most distant target offered, corroborating previous suppositions that on-target forces should remain high. Fish were able to accomplish this without large changes to shot velocity, but we did find evidence that the water jets appeared to differ in the timing of their acceleration as target distance increased. Our two-target choice experiment revealed that fish show preference for downing the closer target first, even though impact forces on distant targets only showed mild decreases. Our overall findings (and the findings of others) suggest that archer fish modulate many aspects of their shooting behavior: from target selection to active control over the water jet that allows the fish to deliver reliably forceful impacts to prey over a wide range of distances.  相似文献

Matrix metalloproteinase 9 (MMP-9) plays an important role in cancer invasion and metastasis and has been an attractive target for therapeutic intervention of cancer metastasis. However, considering the high cost and intricacy associated with the expression, isolation and purification of the recombinant enzyme for the screening of drug candidates, alternative methods that explore the recycling of enzymes become desirable. In this study, a new immobilized enzyme reactor (IMER) containing human recombinant MMP-9 enzyme was developed and characterized for the on-line screening of MMP-9 inhibitors. The MMP-9 IMER containing active unit of the enzyme (U = 0.08 μmol/min) on the disk was inserted into a HPLC system connected to a UV–vis detector for on-line chromatographic screening. The resulting conjugated enzyme was shown to be an active and stable catalyst for the hydrolysis of MMP-9 chromogenic thiopeptide substrate Ac-PLG-[2-mercapto-4-methyl-pentanoyl]-LG-OC2H5. The kinetics profile of the enzyme in IMER and free solution were determined and compared. Selected reversible MMP inhibitors, N-isobutyl-N-(4-methoxyphenylsulfonyl)-glycyl hydroxamic acid (NNGH), doxycycline and minocycline were further characterized using the MMP-9 IMER and free enzyme solution assays. Our system demonstrated applicability as a rapid and cost-effective screening tool for inhibitors of the MMP-9 enzyme.  相似文献
