During drought periods, sugar maple (Acer saccharum) demonstrates hydraulic lift; nocturnal uptake of water by roots from deep soil layers that is released from shallow roots into upper soil layers. Using standard water relations methods and stable hydrogen isotope analysis of both source-water and plant-water, I investigated (1) the magnitude and radial extent of hydraulic lift by mature, relatively open-grown trees, of A. saccharum, (2) the proportion of hydraulically-lifted water (HLW) used by shallow-rooted neighbors growing at different distances from target trees, and (3) the influence that this water source had on stomatal conductance to water vapor (g), water balance and growth of these neighbors. Soil water potentials (s) at –20 and –35 cm showed a distinct diel fluctuation. Soil pits dug beneath three mature trees revealed a distinct hard-pan (e.g. fragipan) layer at a depth of approximately 50 cm. Examination of root distributions obtained from soil cores and soil pits revealed that some larger diameter roots (1.9–3.7 cm) did penetrate the fragipan and were established in the ground water table. The presence of the fragipan indicated that the rewetting of the upper soil layer during the night could not be explained by capillary rise from the shallow water table; it was the trees that were taking up ground water and then redepositing it at night into the upper 35 cm of soil, above the fragipan. The greatest fluctuations in s occurred within 2.5 m of trees and only extended out to approximately 5 m. Application of a two-end-member linear mixing model which used stable hydrogen isotopic data obtained from environmental water sources and xylem-sap demonstrated that all neighbors used some fraction (3–60%) of HLW supplied by sugar maple trees. Plants that used a high proportion of HLW (e.g. rhizomatous or stoloniferous perennials) maintained significantly higher leaf water potentials and g, and showed greater aboveground growth when compared with (i) neighbors that used little or no HLW or (ii) conspecifics found growing at distances greater than about 3 m away from maple trees. Three important conclusions can be drawn from the results of this investigation that have not been demonstrated before: (1) hydraulic lift need not only occur in arid or semi-arid environments where chronic water deficits prevail, but can be important in relatively mesic environments when subjected to periodic soil water deficits, (2) that plants neighboring trees which conduct hydraulic lift can use a significant proportion of this water source, and (3) that the HLW source can effectively ameliorate the influence of drought on the performance and growth of neighboring vegetation. The results are also discussed in terms of their influence on plant nutrient relations (including plant-mycorrhizal associations), the nature of plant-plant interactions and the water balance of individuals, communities and floristic regions.  相似文献

Anterograde tracers, viz. Phaseolus vulgaris leucoagglutinin and fluorescein dextran, were used in conjunction with tyrosine hydroxylase immunohisto-chemistry to study the projections of the A15 dopaminergic cell group towards the median eminence and pituitary in sheep. After injection of the tracers in the retrochiasmatic area, which contains the cell group A15, fibres containing anterograde tracer were observed in the internal zone of the median eminence and in the pars nervosa of the pituitary. Numerous tyrosine hydroxylase immunoreactive fibers were present in the external zone of the median eminence and in the pars intermedia and the pars nervosa of the pituitary, with characteristic patterns of organisation in each area. Most tyrosine hydroxylase-immunoreactive fibres containing fluorescein dextran were located in the pars nervosa, whereas only a few were observed in the internal zone of the median eminence. It was concluded that at least part of the dopaminergic innervation of the pars nervosa originated from the A15 group. These results provide morphological evidence for (1) the role of dopaminergic neurons of the A15 cell group in the seasonal control of prolactin secretion via the release of dopamine in the pars nervosa, and (2) putative physiological interactions between dopamine and the secretion of neurohypophysial hormones in sheep.  相似文献

The distribution of anionic sites on the basal lamina has been examined with highly cationic ferritin. The penetration of ferritins, with a range of charges from anionic to highly cationic, through the basal lamina into the spaces between fat body cells in an insect is correlated with the charge of the tracer. The anionic sites of the basal lamina may therefore affect the composition of the lymph that bathes the fat body cells. There was more cationic ferritin bound to the plasma membrane reticular reticular system than to the lateral plasma membranes, suggesting that there may be regional differences in surface charge.  相似文献

Summary The exchange of protons and deuterons by phosphoglucoisomerase during the single passage conversion of D-[2-13C,1-2H]fructose 6-phosphate in H2O or D-[2-13C]fructose 6-phosphate in D2O to D-[2-13C]glucose 6-phosphate, as coupled with the further generation of 6-phospho-D-[2-13C]gluconate in the presence of excess glucose-6-phosphate dehydrogenase was investigated by 13C NMR spectroscopy of the latter metabolite. In H2O, the intramolecular deuteron transfer from the C1 of D-fructose 6-phosphate to the C2 of D-glucose 6-phosphate amounted to 65%, a value only slightly lower than the 72% intramolecular proton transfer in D2O. Both percentages, especially the latter one, were lower than those previously recorded during the single passage conversion of D-[1-13C,2-2H]glucose 6-phosphate in H2O or D-[1-13C]glucose 6-phosphate in D2O to D-fructose 6-phosphate and then to D-fructose 1,6-bisphosphate. These differences indicate that the sequence of interactions between the hexose esters and the binding sites of phosphoglucoisomerase is not strictly in mirror image during, respectively, the conversion of the aldose phosphate to ketose phosphate and the opposite process.  相似文献

1. To assess the use of stable nitrogen isotopes (δ15N) for reconstructing trophic relationships in planktonic food webs, crustacean zooplankton species and pelagic dissolved and particulate matter were analysed in 14 subarctic lakes in northern Sweden. The lakes are situated along an altitudinal gradient and show a substantial variation in nutrient content and energy mobilization by bacterioplankton and phytoplankton. 2. The δ15N of dissolved and particulate matter was comparatively low, suggesting efficient N recycling and low losses of depleted N from the pelagic zone of these unproductive lakes. 3. Copepods had a systematically higher δ15N than cladocerans, with an average difference of 3.1–4.9‰ within lakes, implying different trophic positions of the two groups. Comparisons of nitrogen pools and energy fluxes suggest that the low cladoceran δ15N was a result of feeding on bacteria. 4. The difference in δ15N between copepods and cladocerans declined with decreasing bacterioplankton production among lakes, due either to increasing trophic isotope fractionation or decreasing relative importance of bacteria in the diet of cladocerans.  相似文献

Lithium was used as a non-radioactive tracer to investigate the root activity of two cereals (wheat and barley), and of two contrasting cultivars of pea (leafy and semi-leafless), both in pure stands and in mixtures. The mixtures included combinations of each cereal with each pea cultivar in single rows, alternative rows and cross-drilled. Total lithium uptake (mg m-2) was higher for wheat than for barley, and higher for semi-leafless pea than for leafy peas. Growing cereals with peas reduced the total lithium uptake by peas, compared with pure stands, especially in alternate-row mixtures. Growing peas with cereals only reduced the total Li uptake by cereals when they were cross-drilled. The Li uptake by wheat, barley and peas generally decreased with soil depth in a similar manner; however, semi-leafless peas absorbed proportionately more Li from close to the soil surface than did leafy peas. Both pea cultivars absorbed more Li at 10–20 cm depth when grown in intimate mixtures with cereals, compared with less intimate mixtures or pure stands. The potential of lithium as a non-radioactive tracer in mixed-cropping studies is briefly discussed.  相似文献

Summary Colloidal ThO2 particles (diameter of 60 Å) were used as electron-opaque markers to trace the intracellular compartments continuous with the bulk interstitial fluid of guinea pig ventricular muscle. Beating and quiescent hearts in a Langendorff preparation were perfused for 30 min with oxygenated Ringer solution containing 1% ThO2. The hearts were immediately fixed by perfusing with glutaraldehyde solution. The colloidal ThO2 particles entered into many of the T tubules and into longitudinallyrunning tubules. No differences in distribution of ThO2 were observed in a heart which was not exposed to ThO2 until after it was fixed. Tracer did not penetrate into the intercalated disk clefts in the guinea pig hearts and one frog heart used for comparison. Tubular profiles filled with ThO2 were not seen in frog heart, an observation which confirms the absence of T tubules in this amphibian. It is concluded that, in mammalian cardiac muscle, the lumens of the longitudinal tubules are continuous with the lumens of the T tubules, forming an extensively interconnected T-L tubular system. Hence, every myofibril has close access to a fluid-filled space which is continuous with the interstitial fluid and which may be of similar cationic composition; such an arrangement should facilitate excitation-contraction coupling.Supported by grants from the American Heart Association and from the Public Health Service (HE-11155, HE-05815 and HE-10384). A confined aquifer in the Malm Karst of the Franconian Alb, South Germany was investigated in order to understand the role of the vadose zone in denitrifiaction processes. The concentrations of chemical tracers Sr2+ and Cl and concentrations of stable isotope 18O were measured in spring water and precipitation during storm events. Based on these measurements a conceptual model for runoff was constructed. The results indicate that pre-event water, already stored in the system at the beginning of the event, flows downslope on vertical and lateral preferential flow paths. Chemical tracers used in a mixing model for hydrograph separation have shown that the pre-event water contribution is up to 30%. Applying this information to a conceptual runoff generation model, the values of 15N and 18O in nitrate could be calculated. Field observations showed the occurence of significant microbial denitrification processes above the soil/bedrock interface before nitrate percolates through to the deeper horizon of the vadose zone. The source of nitrate could be determined and denitrification processes were calculated. Assuming that the nitrate reduction follows a Rayleigh process one could approximate a nitrate input concentration of about 170 mg/l and a residual nitrate concentration of only about 15%. The results of the chemical and isotopic tracers postulate fertilizers as nitrate source with some influence of atmospheric nitrate. The combined application of hydrograph separation and determination of isotope values in 15N and 18O of nitrate lead to an improved understanding of microbial processes (nitrification, denitrification) in dynamic systems.  相似文献

A new method for investigating the binding of ions with polyelectrolytes has been developed. This method, based on Donnan equilibrium and an isotope exchange between the electrolyte and polyelectrolyte, can distinguish territorial from specific binding of ions and can determine fractions of ions bound with the polyion. This method can determine ion binding with polyelectrolytes in a wide range of polyelectrolyte concentrations in multicomponent solutions. The method was tested with radioactive tracers 22Na+, 36Cl- and heparin sodium salt. The influence of the ionic strength on the Na+ binding with heparin was investigated at 310 K. In the limit of zero ionic strength, all Na+ ions are bound to heparin, but only 45% of them are exchangeable. Thus Na+ ions can be bound both territorially and specifically. The fraction of bound ions decreases rapidly with increasing ionic strength. The fraction of the specifically bound ions becomes negligible when the ionic strength exceeds 0.01 M, whereas the fraction of territorially bound ions can be neglected at ionic strengths higher than 0.45 M.  相似文献

Compound-specific hydrogen isotopic compositions (deltaD) of saturated, monounsaturated and polyunsaturated fatty acids have been determined for natural marine macroalgae including two brown algae (Heterokontophyta) and two red algae (Rhodophyta). deltaD values of individual fatty acids from four macroalgae exhibit a wide variation ranging from -189% to +48%. Generally, stearic (18:0), arachidic (20:0) and behenic acids (22:0) are much more enriched in D by up to approximately 180% relative to myristic (14:0), palmitic (16:0), octatetraenoic [18:4(n-3)] and eicosapentaenoic acids [20:5(n-3)]. Other fatty acids such as oleic [18:1(n-9)], lenoleic [18:2(n-6)] and linolenic acids [18:3(n - 3)] fall isotopically between these fatty acids. This wide deltaD variation of fatty acids is probably explained by the hydrogen isotopic fractionation during desaturation being much larger than that during elongation in the network of polyunsaturated fatty acid biosynthesis. A large hydrogen isotopic fractionation during desaturation may cause D-enrichment in the remaining hydrogen of the residual fatty acids, which could be controlled by the relative flux into their desaturates.  相似文献
