Bioarchaeological approaches are well suited for examining past responses to political and environmental changes. In the Andes, we hypothesized that political and environmental changes around AD 1100 resulted in behavioral changes, visible as shifts in paleodiet and paleomobility, among individuals in the San Pedro de Atacama oases and Loa River Valley. To investigate this hypothesis, we generated carbon and oxygen isotope data from cemeteries dating to the early Middle Horizon (Larache, Quitor‐5, Solor‐3), late Middle Horizon (Casa Parroquial, Coyo Oriental, Coyo‐3, Solcor‐Plaza, Solcor‐3, Tchecar), and Late Intermediate Period (Caspana, Quitor‐6 Tardío, Toconce, Yaye‐1, Yaye‐2, Yaye‐3, Yaye‐4). Carbon isotope data demonstrate a greater range of carbon sources during the late Middle Horizon compared with the Late Intermediate Period; while most individuals consumed largely C3 sources, some late Middle Horizon individuals consumed more C4 sources. Oxygen isotope data demonstrate greater diversity in drinking water sources during the late Middle Horizon compared with the Late Intermediate Period. Water samples were analyzed to provide baseline data on oxygen isotope variability within the Atacama Desert, and demonstrated that oxygen isotope values are indistinguishable in the San Pedro and Loa Rivers. However, oxygen isotope values in water sources in the high‐altitude altiplano and coast are distinct from those in the San Pedro and Loa Rivers. In conclusion, instead of utilizing a wider variety of resources after environmental and political changes, individuals exhibited a wider range of paleodietary and paleomobility strategies during the Middle Horizon, a period of environmental and political stability. Stable isotope analysis (SIA) has emerged as an important tool for understanding consumer diets and diet shifts. However, although the general idea behind SIA is clear, the interpretation of data is often fraught with problems because tissue turnover and fractionations are not known. We investigated shifts in stable isotope composition of spiders following a diet shift, using mealworms fed either maize (C4) or wheat (C3) flour. Mealworms had different carbon isotope composition depending on their diet and this difference was reflected in spider body parts. In the experiment, we first fed the spiders on a diet of either maize‐fed or wheat‐fed mealworms and then switched diet at the time of the second molt. Spiders were then sampled repeatedly until the next molt. We sampled both legs and abdomens, as these are presumed to have different turnover of tissue, and also molt remains were sampled when this was relevant. The data indicated that the spider legs had a turnover of about 20 days, whereas the spider abdomens had a turnover of about 8 days. Molt remains had the slowest turnover and reflected the diet at the previous molt, when the exoskeleton was formed. Both these observations indicate that SIA may be successfully used for elucidating diet shifts. More problematic was the fact that fractionation of carbon isotope ratios varied with body parts and diets. When spiders were fed maize‐mealworms then the fractionation was larger for abdomens, but when the spiders were fed wheat‐mealworms then the fractionation was larger for legs. The mechanisms underlying this pattern are unclear and deserve further attention.  相似文献

The unbiased and comprehensive analysis of metabolites in any organism presents a major challenge if proper peak annotation and unambiguous assignment of the biological origin of the peaks are required. Here we provide a comprehensive multi-isotope labelling-based strategy using fully labelled (13) C, (15) N and (34) S plant tissues, in combination with a fractionated metabolite extraction protocol. The extraction procedure allows for the simultaneous extraction of polar, semi-polar and hydrophobic metabolites, as well as for the extraction of proteins and starch. After labelling and extraction, the metabolites and lipids were analysed using a high-resolution mass spectrometer providing accurate MS and all-ion fragmentation data, providing an unambiguous readout for every detectable isotope-labelled peak. The isotope labelling assisted peak annotation process employed can be applied in either an automated database-dependent or a database-independent analysis of the plant polar metabolome and lipidome. As a proof of concept, the developed methods and technologies were applied and validated using Arabidopsis thaliana leaf and root extracts. Along with a large repository of assigned elemental compositions, which is provided, we show, using selected examples, the accuracy and reliability of the developed workflow.  相似文献

Lysyl oxidase (LOX) is implicated in several extracellular matrix related disorders, including fibrosis and cancer. Methods of inhibition of LOX in vivo include antibodies, copper sequestration and toxic small molecules such as β-aminopropionitrile. Here, we propose a novel approach to modulation of LOX activity based on the kinetic isotope effect (KIE). We show that 6,6-d2-lysine is oxidised by LOX at substantially lower rate, with apparent deuterium effect on Vmax/Km as high as 4.35 ± 0.22. Lys is an essential nutrient, so dietary ingestion of D2Lys and its incorporation via normal Lys turnover suggests new approaches to mitigating LOX-associated pathologies.  相似文献

Stable carbon isotope (δ(13)C) series were developed from analysis of sequential radial wood increments from AD 1850 to AD 2009 for four mature primary rainforest trees from the Danum and Imbak areas of Sabah, Malaysia. The aseasonal equatorial climate meant that conventional dendrochronology was not possible as the tree species investigated do not exhibit clear annual rings or dateable growth bands. Chronology was established using radiocarbon dating to model age-growth relationships and date the carbon isotopic series from which the intrinsic water-use efficiency (IWUE) was calculated. The two Eusideroxylon zwageri trees from Imbak yielded ages of their pith/central wood (±1 sigma) of 670 ± 40 and 759 ± 40 years old; the less dense Shorea johorensis and Shorea superba trees at Danum yielded ages of 240 ± 40 and 330 ± 40 years, respectively. All trees studied exhibit an increase in the IWUE since AD 1960. This reflects, in part, a response of the forest to increasing atmospheric carbon dioxide concentration. Unlike studies of some northern European trees, no clear plateau in this response was observed. A change in the IWUE implies an associated modification of the local carbon and/or hydrological cycles. To resolve these uncertainties, a shift in emphasis away from high-resolution studies towards long, well-replicated time series is proposed to develop the environmental data essential for model evaluation. Identification of old (greater than 700 years) ringless trees demonstrates their potential in assessing the impacts of climatic and atmospheric change. It also shows the scientific and applied value of a conservation policy that ensures the survival of primary forest containing particularly old trees (as in Imbak Canyon and Danum).  相似文献

依托FACE技术平台,采用稳定13C同位素技术,通过将小麦(C3作物)种植于长期单作玉米(C4作物)的土壤上,研究了大气CO2浓度升高和不同氮肥水平对土壤排放CO2的δ13C值及根际呼吸的影响.结果表明:种植小麦后土壤排放CO2的δ13C值随作物生长逐渐降低,CO2浓度升高200 μmol·mol-1显著降低了孕穗、抽穗期(施氮量为250 kg·hm-2,HN)与拔节、孕穗期(施氮量为150 kg·hm-2,LN)土壤排放CO2的δ13C值,显著提高了孕穗、抽穗期的根际呼吸比例.拔节至成熟期,根际呼吸占土壤呼吸的比例在高CO2浓度下为24%~48% (HN)和21% ~48% (LN),在正常CO2浓度下为20% ~36% (HN)和19%~32%(LN).不同CO2浓度下土壤排放CO2的δ13C值和根际呼吸对氮肥增加的响应不同,CO2浓度与氮肥用量在拔节期对根际呼吸的交互效应显著.  相似文献

Fundamental to our understanding of the ecology of animal communities in the tropics is knowledge of the effect of seasonal changes in the abundance of food sources in consumer diets. We determined stable‐isotope composition (13C/12C and 15N/14N) in whole blood of 14 resident avian species in a tropical dry forest to quantify the origin of their assimilated protein. We used a probabilistic approach (IsoSource) to estimate the relative contribution of C3 plants, CAM‐C4 plants, C3 insects, and CAM‐C4 insects during the dry and rainy seasons. IsoSource iteratively creates each possible combination of source contribution and produces a distribution of all feasible combinations that adequately predict the observed isotopic signature of the consumer. Granivore–frugivores and granivore–frugivore–insectivores were modeled as predominantly dependent upon plants whereas insectivorous birds were modeled to derive protein almost exclusively from insects. Between these extremes there were several species using mixed diets such as insectivore–frugivores or insectivore–granivores. In most species, virtually all assimilated food was of C3 origin with the exception of Ruddy Ground‐Doves (Columbina talpacoti) in which CAM or C4 plants contributed significantly. Seasonal changes in relative food source contribution were followed in eight species of birds. Of these species, White‐tipped Doves (Leptotila verreauxi), Grayish Saltators (Saltator coerulescens), and Social Flycatchers (Myiozetetes similis) increased their use of insects in the rainy season, in contrast to Great Kiskadees (Pitangus sulphuratus), which decreased their use of insects. Our study suggests that that diverse strategies are used by various avian species to obtain dietary proteins within seasonal habitats.  相似文献

Aim Numerous studies have examined the climatic factors that influence the abundance of C4 species within the grass flora (C4 relative species richness) in various regions throughout the world, but very few have examined the relative abundance of C4 vs. C3 grasses (C4 relative abundance). We sought to determine the climatic factors that influence C4 relative abundance throughout Australia. Location Australia (including Tasmania). Methods We measured C4 relative abundance at 168 locations and measured δ13C (the abundance of 13C relative to 12C) of the bone collagen of 779 kangaroos collected throughout Australia, as bone collagen δ13C was assumed to be proportional to the relative abundance of C4 grasses in the diet. Results Both C4 relative abundance and kangaroo bone collagen δ13C were found to have a strong positive relationship with seasonal water availability, i.e. the distribution of rainfall in the C4 vs. C3 growing seasons (76% and 69% of deviance explained, respectively). There was clear evidence that seasonal water availability was a better predictor of both C4 relative abundance and bone collagen δ13C than other climate variables such as mean annual temperature and January daily minimum temperature. However, seasonal water availability appeared to be a relatively poor predictor of C4 relative species richness, which was most closely related to January daily minimum temperature (90% of deviance explained). Main conclusions Our results highlight the relatively poor relationship between C4 relative abundance and C4 relative species richness, and suggest that these two variables may be related to different climatic factors. They also suggest that caution is required when using C4 relative species richness to infer the relative biomass and productivity of C4 grasses on a global scale.  相似文献

This study is concerned with validating the measurement of the plasma half-life of 11alpha-(2)H cortisol in an attempt to accurately assess the in vivo activity of 11beta-HSD2 in man. Oral administration of 5mg of cortisol-(13)C(4),(2)H(1) to a human subject after repeated ingestions of 130mg/day of glycyrrhetinic acid for 5 days resulted in a decrease in the rate constant of the cortisol-(13)C(4),(2)H(1) to cortisone-(13)C(4) conversion, a direct index reflecting 11beta-HSD2 activity. The reduced 11beta-HSD2 activity led to an increase in the elimination half-life of cortisol-(13)C(4),(2)H(1), indicating that the loss of 11alpha-(2)H is a sensitive in vivo means of assessing 11beta-HSD2 activity. A simultaneous oral administration of 3mg each of [1,2,4,19-(13)C(4),11alpha-(2)H]cortisol (cortisol-(13)C(4),(2)H(1)) and 11alpha-(2)H cortisol to another human subject confirmed the bioequivalency of the two labeled cortisols. The information obtained from the kinetic analysis of the 11beta-HSD2-catalyzed conversion of cortisol-(13)C(4),(2)H(1) to cortisone-(13)C(4) indicated that the elimination half-life of 11alpha-(2)H cortisol was a sensitive index of renal 11beta-HSD2 activity. The use of 11alpha-(2)H cortisol as a tracer appears to offer a significant advance in evaluating human 11beta-HSD2 activity in vivo.  相似文献

Heterotrophic decomposition of organic matter dictates that substrate supply rate, including energy and nutrients, can limit soil microbial activity. In New Zealand, soils are naturally deficient in nitrogen and phosphorus. Fertiliser application is a part of pastoral agriculture, the countrys most widespread land use. We postulated that organic soils under grazed pasture and pristine forest would be at the extremes of substrate quality and supply rate, and thus potential microbial response to food opportunities. Soil microbial responses to the addition of fresh energy (sucrose) were determined by laboratory experiments with root-free samples and intact cores including roots. Responses were quantified by respiration and respired carbon (C) isotope (13C) enrichment measurements. A supra-trace sucrose dose (0.002 mol kg–1 (soil)) caused the forest soils microbial respiration rate to nearly double within 2 h. The peak response took 20 h, and saturation occurred beyond a sucrose dose of 0.05 mol kg–1 (soil). Intact soil cores from the forest had similar respiration rates and responses. For root-free soil samples from the grazed pasture, respiration response to sucrose was nearly immediate, dose dependent, and there was up to a 9-fold increase in the rate. Intact cores from the pasture had much higher respiration rates, but a similar response to sucrose. For both soils, the similarity of sucrose application effects on respiration and relative 13C enrichment of the respired carbon was striking.  相似文献
