Salt Tolerance of Cotton: Some New Advances   总被引:5,自引:0,他引:5  
Referee: Dr. Lin Wu, Department of Environmental Horticulture, University of California, Davis, Davis, CA 95616 Cotton is a dual-purpose crop, widely used for fiber and oil purposes throughout the world. It is placed in the moderately salt-tolerant group of plant species with a salinity threshold level 7.7?dS m?1, its growth and seed yield being severely reduced at high salinity levels and different salts affect the cotton growth to a variable extent. However, inter- and intraspecific variation for cotton salt tolerance in cotton is considerable and thus can be exploited through specific selection and breeding for enhancing salt tolerance of the crop. There are contrasting reports regarding the crop response to salinity at different plant growth stages, but in most of them it is evident that the crop maintains its degree of salt tolerance consistently throughout its entire developmental phases. In the latter case an effective selection for salt tolerance is possible to be made at any growth stage of the crop. The pattern of uptake and accumulation of toxic ions (Na+ and/or Cl?) in tissues of plants subjected to saline conditions appears to be due mostly to the mechanism of partial ion exclusion (exclusion of Na+ and/or Cl?) in cotton. Maintenance of high tissue K/Na and Ca/Na ratios is suggested to be an important selection criterion for salt tolerance in cotton. While judging the appropriate mechanism of ion transport across the membranes in view of existing literature, it was evident that the PM-ATPase responds to increasing supply of Na+ in the growth medium, but the activity of the transport proteins on the plasma membrane alone were insufficient to regulate intracellular Na+ levels. Vacuolar-ATPase is also not responsive to increased external Na+. The inability of V-ATPase to respond to Na+ gave indication of the lack of effective driving force for compartmentalization of Na+ in cotton. However, in view of some latest studies concenrning the role of some antioxidants in salt tolerance of cotton it was suggested that high levels of antioxidants and an active ascorbate-glutathione cycle are associated with salt tolerance in cotton. Genetic studies with cotton in relation to salinity tolerance exhibited that most of growth, yield, and fiber characteristics are genetically based and most being QTL controlled and variable. The high additive component of variation can be exploited for breeding to produce further improvement in the salt tolerance of cotton.  相似文献

The rate of exchange of peptide group NH hydrogens with the hydrogens of aqueous solvent is sensitive to neighboring side chains. To evaluate the effects of protein side chains, all 20 naturally occurring amino acids were studied using dipeptide models. Both inductive and steric blocking effects are apparent. The additivity of nearest-neighbor blocking and inductive effects was tested in oligo-and polypeptides and, suprisingly, confirmed. Reference rates for alanine-containing peptides were determined and effects of temperature considered. These results provide the information necessary to evaluate measured protein NH to ND exchange rates by comparing them with rates to be expected for the same amino acid sequence is unstructured aligo- and polypeptides. Some continental European populations of the land snail Cepaea nemoralis have mitochondrial haplotypes that differ by up to 20% at the 16S rRNA locus. I mapped the distribution of different lineages in populations from 36 different sites in Britain and Ireland. In 93% of individuals, one of two mitochondrial lineages was found, A or. N , which differ from each other by about 6% using a 16S rRNA fragment {approximately 300 base pairs). The distribution of these two types is very striking–one is confined to Wales, West and central England, and Scotland, while the other is found mainly in East and central England. The two types meet in a transition zone. The most likely explanation for the distribution is that it reflects two routes of colonization after the last ice age. Cepaea dispersal is leptokurtic, and only limited gene flow occurs between established populations, so that the original pattern could have been retained since the post-glacial colonization. However, many environmental gradients are orientated East-West, so alternative selective explanations are possible. A distinct mitochondrial lineage, as well as fossil evidence, suggests that Ireland was colonized separately from Britain. The implications of these distributions for the origins of the puzzling geographical patterns of shell types known as 'area effects' is discussed.  相似文献

Because of the recognized inaccuracy and unreliability of currently available methods for the quantification of histamine in biological fluids, a method for quantification of urinary histamine by stable isotope dilution assay with negative ion chemical ionization mass spectrometry has been developed. Following the addition of [2H4]histamine to 1 ml of urine, histamine is extracted into butanol, back-extracted into HCl, derivatized to the pentafluorobenzyl derivative (CH2C6F5)3-histamine, extracted into methylene chloride, and then quantified with negative ion chemical ionization mass spectrometry by selected ion monitoring of the ratio of ions mz430434. Twenty samples can be assayed in 2 days. Precision of the assay is ±2.7% and the accuracy is 97.6%. Lower limits of sensitivity are approximately 100–500 fg injected on-column. This assay provides a very sensitive, accurate, and efficient method for the quantification of histamine in human urine.  相似文献

后生效应是近几年才定义清楚的一类现象,指所有那些DNA序列没有改变的基因表达后体细胞的遗传表型发生改变的现象。它对生物体的性状发育如性别决定、着丝粒的功能决定、果蝇复眼发育、朊病毒致病性的获得及癌的发生等等方面有着重要的影响。多方面的研究表明,导致后生效应的可能原因主要有四个方面:DNA的甲基化、组蛋白的乙酰化、染色质结构背景的改变和蛋白质构型的变化。 Abstract:Epigenetic effects,which were defined clearly in recent years,are most often used to denote all those somatically heritable changes in gene expression that do not involve changes in DNA sequence.They affect the development of organismal characters such as sex determination,the functional determination of centromeres,the development of Drasophila's eys,the prion pathogenicity and cancer formation greatly.Studies on many aspects show that there are four main reasons which cause the epigenetic effects;the methylation of DNA,the acetylation of histones,the change of chromatin structure and the alteration of protein configuration.  相似文献

Ligand-gated ion channels contain a conserved leucine at position 9′ (L9′) in the M2 transmembrane domain. We used multiple substitutions at this position in the γ subunit of the mouse acetylcholine receptor (AChR) (γL9′) to examine the role of residue polarity at this position in the gating process at both the macroscopic and single-channel levels. The midpoint of the macroscopic dose-response relationship (EC50) and the channel closing rate constant, α, decreased as the polarity of the residue at that position increased, suggesting a stabilization of the open state of the channel. Both parameters showed similar dependencies on the polarity of the substituted residue. These data support the notion that during AChR gating, the amino acid at the 9′ position moves into a polar environment, and that interactions between this residue and the polar environment determine the stability of the open state. Since this residue is conserved in all other members of the ligand-gated ion channel family, we suggest that a similar mechanism applies to the other members of the family. The main concern regarding methylmercury neurotoxicity relates to adverse effects on the brain during development. Many environmental chemicals may act as developmental neurotoxicants, but solid documentation from epidemiological studies exists only on methylmercury, lead, and polychlorinated biphenyls (PCBs). Neurobehavioral tests may reveal subtle dysfunctions, but the tests chosen must be valid and appropriate for the setting. In a prospective study in the Faroe Islands, the main neuropsychological functions affected by prenatal methylmercury exposure were attention, language and memory. Deficits in visuospatial function were mainly related to postnatal exposures. These associations were stable after adjustment for confounders and exclusion of the children with the highest exposures to methylmercury and PCBs. Tests with good psychometric properties were more likely to show an association with mercury exposure. Greater sensitivity was also seen with tests administered by specialized academic staff rather than a trained technician. Despite highly significant effects on nervous system function, the deficits were subtle, and mercury exposure explained only a small part of the variation. Available evidence suggests that neurotoxicity may have severe implications on public health, but current methods are not amenable to application as sentinels of adverse health effects in environmental health surveillance.  相似文献

Abstract Possible effects of increased atmospheric concentrations of CO2 on forest ecosystems are discussed and as an example a simulation case study using a set of mixed-age and mixed-species forest stand models is presented. The responses of the models to a simple scenario (uniform growth increase of all trees as a response to CO2 enrichment) include increases in biomass that are considerably less than the increases in growth rate of the trees. These simulations and more general discussion of the possible effects of increased photosynthetic production identify the problem of scaling-up small time-scale and space-scale measurements of plant responses to CO2 enrichment to the ecosystem level.  相似文献

Brugada syndrome (BrS) is a life-threatening cardiac rhythm disorder characterized by persistent STsegment elevation in leads V1–V3 and right bundle branch block on electrocardiograms (ECG), and by syncope and sudden death from ventricular tachycardia (VT) and ventricular fibrillation (VF). BrS is responsible for nearly 4% of sudden cardiac deaths and considered to be the most common cause of natural death in males younger than 50 years in some Asian countries. Since the first diseasecausing gene for BrS (the cardiac sodium channel gene SCN5A) was identified in 1998, extensive investigations on both clinical and basic aspects of BrS have occurred rapidly. SCN5A mutations remain the most common cause of BrS; nearly 300 SCN5A mutations have been identified and are responsible for 20%–30% of BrS cases. Commercial genetic testing is available for SCN5A. Recently, seven other disease-causing genes for BrS have been identified and include GPD1L (BrS2), CACNA1C (Cav1.2, BrS3), CACNB2 (Cavβ2, BrS4), SCN1B (Navβ1, BrS5), KCNE3 (MiRP2, BrS6), SCN3B (Navβ3, BrS7), and HCN4 (BrS8). This article will briefly review the progress made over the past decade in our understanding of the clinical, genetic and molecular aspects of BrS.  相似文献

The Cry9Aa entomocidal toxin from Bacillus thuringiensis ssp. galleriae (Btg) and an epiphytic Pseudomonas sp. derivative carrying the cloned cry9Aa gene from Btg are active against the pine processionary moth Thaumetopoea pityocampa and the laboratory model species Galleria mellonella. A laboratory study was conducted to investigate the side effects of the Cry9Aa toxin and the engineered bacterium on the post-embryonic development of Exorista larvarum, a larval parasitoid of forest lepidopterous defoliators, cultured in the factitious host G. mellonella. In a first experiment, the purified toxin and the commercial Bt preparation Foray 48B induced a mortality of G. mellonella sixth-instar larvae significantly higher than that of the distilled water control. In parallel, the development of E. larvarum in this host was assessed, but no significant difference was found for any of the parasitoid parameters examined (i.e., eggs oviposited, percentage of puparia and adults and puparial weights). In subsequent experiments, cry9Aa-Pseudomonas suspension significantly increased the mortality of sixth instar G. mellonella larvae compared to untransformed Pseudomonas sp. suspension and distilled water. As to the parasitoid parameters, the cry9Aa-Pseudomonas did not significantly affect the number of oviposited eggs, percentage of puparia and puparial weights. It can be concluded that the post-embryonic development of E. larvarum was not affected by host treatment with either Cry9Aa toxin or cry9Aa-Pseudomonas under the laboratory conditions tested. Although direct effects on parasitoid performance have not been shown, indirect effects could still occur and need to be considered in future studies concerning the effects of genetically modified Bt-derivatives.  相似文献
