164 taxa were identified in the net zooplankton of the pelagial of L. Peipsi-Pihkva in 1909–1987, including 3 species of protozoans, 74 species of rotifers, 58 species of cladocerans, 28 species of copepods and 1 mollusc. One rotifer species, Ploesoma peipsiense Mäemets et Kutikova, has been described as new for science here. The zooplankton of L. Peipsi-Pihkva is remarkably rich in species including rarities in Estonia: Limnosida frontosa, Drepanothrix dentata, Bythotrephes longimanus, B. cederstroemi etc. Due to its large surface area, L. Peipsi-Pihkva provides a large scale of biotopes of a diverse trophic state and humic content, which support species with different ecological requirements. Most of the aquatory of the lake has lately been mesotrophic, favouring the coexistence of indicators of oligo- and mesotrophic state and species preferring a higher trophic state. The occurrece of 10 species of the genus Bosmina including B. berolinensis, B. gibbera, B. lilljeborgi, B. thersites and B. crassicornis, sparse in Estonian lakes, is the most noteworthy feature of the zooplankton of L. Peipsi-Pihkva. The coexistence of B. coregoni and B. berolinensis, B. gibbera, B. lilljeborgi etc. which were earlier regarded as subspecies of B. c. coregoni proves that they are different species producing usually no hybrids. The species composition was subjected to certain changes during the years under consideration. Larvae of Dreissena were first found in zooplankton in 1962. The oligo-mesotrophic indicator Holopedium gibberum occurred in the lake in 1909–1964, but was lacking in later samples. 相似文献

Recent investigations concentrate on the correlation between the myocardial expression of the inducible 70-kDa heat shock protein (HSP70i) by different stress conditions and its possible protective effects. Only few studies have focused on the involvement of small heat shock proteins in this process. We analyzed the location of the small heat shock protein HSP25 in isolated cardiomyocytes as well as its location and induction in isolated perfused hearts of rats. By immunofluorescence microscopy HSP25 was found to colocalize with actin in the I-band of myofibrils in cardiomyocytes of isolated perfused hearts as well as in isolated neonatal and adult cardiomyocytes. Hyperthermic perfusion of isolated hearts for 45 min resulted in modulation of different parameters of heart function and in induction of HSP25 and HSP70i. Temperatures higher than 43°C (44–46°C) were lethal with respect to the contractile function of the hearts. Compared to control hearts perfused at 37°C, significant increases during hyperthermic perfusion at 42°C and 43°C were obtained for heart rate, contraction velocity and relaxation velocity. In response to hyperthermia at 43°C and after subsequent normothermic perfusion for 135 min at 37°C, left ventricular pressure, contraction velocity and relaxation velocity remained significantly elevated. However, heart rate returned to control values immediately after the period of heat treatment. HSP25 is constitutively expressed even in normothermic perfused hearts as shown by Western blotting. Hyperthermia increased the content of HSP25 only in the left ventricular tissue. In contrast, HSP70i was strongly induced in all analyzed parts of the myocardium (left ventricle, right ventricle, septum). Our findings suggest a differential regulation of HSP25 and HSP70i expression in response to hyperthermia in isolated perfused hearts. The constitutively expressed HSP25 seems to be located adjacent to the myofibrils which implies a specific role of this protein even under unstressed conditions for the contractile function of the myocardium. 相似文献

Intracellular free Ca2+ concentration has been shown to be elevated in platelets of patients with essential hypertension. This study was designed to characterize Ca2+-pump activity of the platelet membranes (surface and intracellular) in these patients. A double-blind study was carried out. Untreated and treated (on R-blockers) essential hypertensives were studied in comparison with normotensive control subjects. First degree blood relatives of essential hypertensives were also studied. The Ca2+-activation kinetics of the enzyme showed a significant decrease in the Vmax. (for the plasma- and intracellular membranes) and Km (for the intracellular membranes) in the essential hypertensive patients. Increased platelet membrane cholesterol content was observed in these patients. Lowered Ca2+-efflux by Ca2+-ATPase may lead to elevated intracellular free Ca2+-levels in platelets of essential hypertensives. A lowered Ca2+-ATPase activity may emerge as a marker for essential hypertension. 相似文献

Overexpression of the MDR protein, or p-glycoprotein (p-GP), in cells leads to decreased initial rates of accumulation and altered intracellular retention of chemotherapeutic drugs and a variety of other compounds. Thus, increased expression of the protein is related to increased drug resistance. Since several homologues of the MDR protein (CRP, ltpGPA, PDR5, sapABCDF) are also involved in conferring drug resistance phenomena in microorganisms, elucidating the function of the MDR protein at a molecular level will have important general applications. Although MDR protein function has been studied for nearly 20 years, interpretation of most data is complicated by the drug-selection conditions used to create model MDR cell lines. Precisely what level of resistance to particular drugs is conferred by a given amount of MDR protein, as well as a variety of other critical issues, are not yet resolved. Data from a number of laboratories has been gathered in support of at least four different models for the MDR protein. One model is that the protein uses the energy released from ATP hydrolysis to directly translocate drugs out of cells in some fashion. Another is that MDR protein overexpression perturbs electrical membrane potential () and/or intracellular pH (pHi) and therebyindirectly alters translocation and intracellular retention of hydrophobic drugs that are cationic, weakly basic, and/or that react with intracellular targets in a pHi, or -dependent manner. A third model proposes that the protein alternates between drug pump and Cl– channel (or channel regulator) conformations, implying that both direct and indirect mechanisms of altered drug translocation may be catalyzed by MDR protein. A fourth is that the protein acts as an ATP channel. Our recent work has tested predictions of these models via kinetic analysis of drug transport and single-cell photometry analysis of pHi, , and volume regulation in novel MDR and CFTR transfectants that have not been exposed to chemotherapeutic drugs prior to analysis. This paper reviews these data and previous work from other laboratories, as well as relevant transport physiology concepts, and summarizes how they either support or contradict the different models for MDR protein function. 相似文献

The effects of lead on Ca2+ homeostasis in nerve terminals was studied. Incubation with leadin vitro stimulated the activity of calmodulin and the maximum effect was observed at 30 M lead, higher concentrations had an inhibitory effect.In vivo exposure to lead increased the activity of calmodulin by 45%. Lead had an inhibitory effect on Ca2+ ATPase activity in both calmodulin-rich and calmodulin-depleted synaptic plasma membranes, the IC50 values for inhibition being 13.34 and 16.69 M respectively. Exogenous addition of calmodulin (5 g) and glutathione (1 mM) to calmodulin rich synaptic plasma membranes reversed the inhibition by IC50 concentration of lead.In vivo exposure of lead also significantly reduced the Ca2+ ATPase activity, resulting in an increase in intrasynaptosomal calcium. Concomitant with the increase in intrasynaptosomal calcium, lipid peroxidation values also increased significantly in lead-treated animals. In addition lead also had an inhibitory effect on depolarization induced Ca2+ uptake and the inhibition was found to be a competitive one. The results sugest that lead exerts its toxic effects by modifications of the intracellular calcium messenger system which would have serious consequences on neuronal functioning. 相似文献

Nutrient additions by waterfowl to lakes and reservoirs: predicting their effects on productivity and water quality 总被引:3,自引:3,他引:0
Lakes and reservoirs provide water for human needs and habitat for aquatic birds. Managers of such waters may ask whether nutrients added by waterfowl degrade water quality. For lakes and reservoirs where primary productivity is limited by phosphorus (P), we developed a procedure that integrates annual P loads from waterfowl and other external sources, applies a nutrient load-response model, and determines whether waterfowl that used the lake or reservoir degraded water quality. Annual P loading by waterfowl can be derived from a figure in this report, using the days per year that each kind spent on any lake or reservoir. In our example, over 6500 Canada geese (Branta canadensis) and 4200 ducks (mostly mallards, Anas platyrhynchos) added 4462 kg of carbon (C), 280 kg of nitrogen (N), and 88 kg of P y–1 to Wintergreen Lake in southwestern Michigan, mostly during their migration. These amounts were 69% of all C, 27% of all N, and 70% of all P that entered the lake from external sources. Loads from all external sources totaled 840 mg P m–2 y–1. Application of a nutrient load-response model to this concentration, the hydraulic load (0.25 m y–1), and the water residence time (9.7 y) of Wintergreen Lake yielded an average annual concentration of total P in the lake of 818 mg m–3 that classified the lake as hypertrophic. This trophic classification agreed with independent measures of primary productivity, chlorophyll-a, total P, total N, and Secchi disk transparency made in Wintergreen Lake. Summary The cytoplasmic content and the distribution of intramembrane particles (IMPs) of the plasma membrane of isolated sperm cells of perennial ryegrass (Lolium perenne L.) have been characterized using flow cytometry, transmission electron microscopy, confocal scanning laser microscopy and freeze-fracture studies. The isolated haploid sperm cells contain a variety of cell organelles with the exception of microtubules. Proplastids and plastids with starch were observed, although only rarely. Vacuoles containing remnants of organelles and stacked lamellae of endoplasmic reticulum with cytoplasmic inclusions were observed frequently, indicating that autophagy takes place. The number of mitochondria varies from 11 to 26 with an average of 17. Generally, the nucleus has a lobed shape and displays various interphasic stages of chromatin condensation. The analysis of the number of mitochondria and the nuclear state did not show evidence of sperm cell dimorphism. The cytological variability observed, could be explained by differences in developmental stages already present in vivo at the moment of isolation. No correlation between the number of mitochondria and the nuclear cross-sectioned area and/or the condensation state of the chromatin could be found. The density of intramembrane particles of the plasma membrane on the exoplasmic fracture face is more than twice that on the protoplasmic fracture face. That is the opposite of what was found for sporophytic cells of perennial ryegrass. These results are discussed in relation to the potential use of these cells for biotechnology and developmental studies. 相似文献

The effects of pulsed microwaves (2.45 GHz, 10 μs, 100 pps, SAR: 81.5 kW/kg peak, 81.5 W/kg average) on membrane input resistance and action potential (AP) interval statistics were studied in spontaneously active ganglion neurons of land snails (Helix aspersa), at strictly constant temperature (20.8±.07°C worst case). Statistical comparison with sham-irradiated neurons revealed a significant increase in the mean input resistance of neurons exposed to pulsed microwaves (P ? .05 ). Pulsed microwaves had no visible effect on mean AP firing rate; this observation was confirmed by analysis of interspike intervals (ISIs). Using an integrator model for spontaneously active neurons, we found the net input current to be more variable in neurons exposed to pulsed microwaves. The mean input current was not affected. The standard deviation of ISIs and the autocorrelation of the input current were marginally affected, but these changes were not consistent across neurons. Although the observed effects were less obvious than those reported in other studies, they represent evidence of a direct interaction between neurons and pulsed microwaves, in the absence of macroscopic temperature changes. A survey of base-rich wetlands in The Netherlands is presented. The main area of their occurrence is the low-lying Holocene
part of the country, until some thousand years ago a large and coherent wetland landscape: the Holland wetland. The development
of various parts of the Holland wetland into marshes, fens and bogs can be understood from hydrological relations in mire
basins, as recognized in the distinction of primary, secondary and tertiary mire basin stages. Presently, the remnants of
the Holland wetland are separate base-rich wetlands. The succession of their vegetation reflects various abiotic conditions
and human influences. Three main developmental periods are distinguished as regards these factors. The first, geological period
of mire development is seen as a post-glacial relaxation, with the inertia due to the considerable mass of wetland as a stabilizing
factor. Biological “grazing” influences, as an aspect of utilization by humans, converted base-rich wetlands to whole new
types in the second, historical period. Presently, mass and harvesting have decreased in importance, and actual successions
in terrestrializing turbaries seem to reflect rapidly changing environmental conditions. Human control could well become the
most important factor in the future development of wetland nature. The present value of open fen vegetation strongly depends
on the continuation of the historical harvesting. The development of wooded fen may help to increase the mass of wetland in
the future. Best results in terms of biodiversity are expected when their base state is maintained through water management.
The vegetation and hydrology of floating fens in terrestrializing turbaries is treated in some more detail. Various lines
and phases in the succession are distinguished. Open fen vegetation at base-rich, yet nutrient-poor sites is very rich in
species threatened elsewhere. The fast acidification of certain such fens is attributed to hydrological and management factors.
This acidification is illustrated in the profile of a floating raft sample. At the scale of these small fens, the elemental
structure comprising base-rich fen, transitional fen and bog vegetation, is not as stable as it was in the large Holland wetland.
A critical role seems to be played by the supply of bases with the water influx. The changing base state is supposed to change
the nutrient cycling to such an extent that it would be correct to call this trophic excitation of the ecosystem, rather than
just eutrophication. Eutrophication indicates a quantitative reaction to an increased nutrient supply, the internal system
being unaltered. The drainage of fens, resulting in an increased productivity of the vegetation, provides another example
of excitation, to the effect that the functional system is dramatically changed internally. 相似文献
