集约种植雷竹林土壤细菌群落结构的演变及其影响因素   总被引:7,自引:1,他引:6  
应用PCR-DGGE技术研究了雷竹林种植过程中土壤细菌群落结构的变化,并运用冗余分析(RDA)手段提取雷竹林土壤细菌群落多样性的干扰控制因子.结果表明:水稻田改种雷竹后,土壤细菌群落的Shannon指数及丰富度指数均显著增加;长期的集约种植过程对雷竹林土壤细菌群落结构产生了影响,主要细菌种群发生了变化,多样性指数表现为先增加、后期大幅降低的趋势.RDA分析表明,土壤pH、总氮、碱解氮以及速效钾这4个变量能够解释高达76.1%的样本总变异,其中土壤pH对细菌群落结构影响最大,但未达到显著水平,表明长期集约种植的雷竹林土壤细菌群落结构的变化是多种环境因子共同作用的结果.  相似文献

以集约和粗放经营的毛竹(Phyllostachys heterocycla ‘Pubescens’)林为研究对象, 探讨了春季毛竹林集约经营后土壤有机碳的变化。结果表明: (1)集约经营后毛竹林0-10和10-20 cm土层土壤总有机碳含量分别下降了7.01%和18.90%, 易氧化碳含量分别下降了31.22%和46.03%, 0-20 cm土层轻组有机质含量下降了19.87%。(2)两种毛竹林的土壤有机碳含量在剖面上整体上均随土层深度的增加而呈下降趋势, 但下降幅度不同。粗放经营的毛竹林土壤易氧化碳的剖面特征与总有机碳相似, 而集约经营的毛竹林存在明显差异。轻组有机质具有表聚性, 主要分布在土壤表层(0-20 cm)。(3)土壤总有机碳、易氧化碳、轻组有机质与土壤养分之间的相关性均达到极显著水平(p < 0.01), 总有机碳与速效磷显著相关(p < 0.05)。(4)集约经营后, 毛竹林0-10 cm土层土壤易氧化碳的碳素有效率和土壤碳库活度分别下降了26.01%和50.52%, 差异显著(p < 0.05); 10-20 cm土层分别下降了35.51%和54.41%。因此, 施加适当配比的有机肥和无机肥, 有利于土壤中各种有机碳的积累, 也可改善土壤的生物化学活性。  相似文献

To elucidate the mode of dissemination of methicillin-resistant Staphylococcus aureus (MRSA) in neonate intensive care units (NICUs), a total of 223 isolates from 3 separate hospitals were investigated between 1994 and 1996 by a DNA fingerprinting technique with pulsed-field gel electrophoresis (PFGE). Exoprotein profiles of some strains were also examined using SDS-polyacrylamide gel-electrophoresis (SDS-PAGE) and the assessment of enzyme/toxin production such as coagulase, enterotoxin and TSST-1. Judging from the strain typing data from PFGE results and the epidemiological data, 2 different types of PFGE patterns (A and B) and their subtypes (A′, A″ and B′) were identified. The A type including A′ and A″ (comprising approximately 95% of the isolates) was markedly dominant. Only 5% of the isolates belonged to type B and subtype B′. On the other hand, MRSA isolated from adult patients admitted to the same hospital showed many different PFGE patterns. The results strongly suggested that some strain(s) with specific PFGE pattern(s) is prevalent in NICUs. Furthermore, isolates which expressed the same PFGE pattern did not always express the same SDS-PAGE pattern. There were some isolates with different abilities to produce coagulase, enterotoxin C and toxic-shock syndrome toxin (TSST)-1, and the abilities had no relation with a particular type of PFGE pattern. Therefore, a combination of PFGE analysis and biochemical analyses of coagulase, enterotoxin C and TSST-1 may provide us with more detailed information for the epidemiological study of MRSA in NICUs.  相似文献

集约用海是一种相对科学、高效、生态的用海方式,但也不可避免的要占用和消耗海洋资源。因此开展集约用海对海洋资源影响的科学评估,将集约用海对海洋资源的影响降到最低就尤为重要。通过分析集约用海工程对海洋资源的影响,明确了集约用海工程对海洋资源影响影响评价的主要内容和思路,从海洋资源的复杂性及其开发利用角度,构建了基于海洋生物资源、海洋空间资源、港口航道资源、滨海旅游资源和其它资源的集约用海对海洋资源影响的综合评价指标体系。其中,从浮游植物、浮游动物、底栖生物和鱼类生物方面反映集约用海工程对海洋生物资源的影响;从滨海湿地、岸线、海域空间利用变化方面反映集约用海工程对海域空间资源的影响;从水动力变化、港口资源利用方面反映集约用海工程影响下港口航道资源的影响;从旅游资源密度反映集约用海工程对滨海旅游资源的影响;从矿产资源、能源资源利用方面反映集约用海工程对其它资源的影响。在此基础上,根据我国海洋资源调查与评估现状,研究并确立了集约用海对海洋资源影响的各评价指标权重、评价标准及评价等级,建立了基于赋值法、综合评价法的集约用海对海洋资源影响的综合评价模型,利用综合评估指数I来反映集约用海工程对海洋资源的影响程度。利用该评价方法评价了曹妃甸区域集约用海对海洋资源的影响,结果表明该区域集约用海对海洋资源造成的影响较大,应严格控制该区域集约用海工程。  相似文献

False discovery rates are routinely controlled by application of the Benjamini–Hochberg step-up procedure to a set of p-values. A method is demonstrated for representing the values so obtained (the BH-FDRs) on a quantile–quantile (Q-Q) plot of the p-values transformed to the negative-logarithmic scale. Recognition of this connection between the BH-FDR and the Q-Q plot facilitates both understanding of the meaning of the BH-FDR and interpretation of the BH-FDR in a particular data set.  相似文献

Golden‐cheeked Warblers (Setophaga chrysoparia) are endangered songbirds that breed exclusively in the Ashe juniper (Juniperus ashei) and oak (Quercus spp.) woodlands of central Texas. Despite being the focus of numerous studies, we still know little about the size of the range‐wide breeding population and how density varies across the spectrum of juniper co‐dominated woodlands. Models that have been tested and shown to be accurate are needed to help develop management and conservation guidelines. We evaluated the accuracy and bias of density estimates from binomial mixture models, the dependent double‐observer method, and distance sampling by comparing them to actual densities determined by intensive territory monitoring on plots in the Balcones Canyonlands Preserve, Austin, Texas. We found that the binomial mixture models consistently overestimated density by 1.1–3.2 times (actual density = 0.07–0.46 males/ha), and the other two models overestimated by 1.1–29.8 times at low density and underestimated by 0.5–0.9 times at high density plots (actual density = 0.01–0.46 males/ha). The magnitude of error for all models was greatest at sites with few or no birds (<0.15 males/ha), with model performance improving as actual density increased. These non‐linear relationships indicate a lack of sensitivity with respect to true changes in density. Until systematic evaluation demonstrates that models such as those we tested provide accurate and unbiased density estimates for a given species over space and time, we recommend additional field tests to validate model‐based estimates. Continued model validation and refinement of point‐count methods are needed until accurate estimates are obtained across the density spectrum for Golden‐cheeked Warblers and other songbird species.  相似文献

The effect of addition of peptone to serum-free and serum supplemented media for the growth of hybridoma cells in various systems was studied. Supplementation of defined medium with either proteose peptone or meat peptone resulted in significant increases in cell number and specific monoclonal antibody production in batch culture system. Other peptones were either inactive or less effective. In continuous culture, using medium supplemented with new born calf serum, the addition of peptone resulted in 125% and 150% increases in cell and antibody concentrations respectively. Similar increase in cell number (128%) was also obtained in spin-filter perfusion culture when medium was supplemented with peptone. By comparison, the substitution of a defined 1xMEM amino acids mixture resulted in only a 50% increase. At higher perfusion rates the cell number maintained in steady state using peptone supplement could be increased to 1.3×107 cells ml–1 while the serum concentration was reduced from 5% to 1% at a perfusion rate of 2.5 volumes per day.  相似文献

Our objective is to define differences in circulating lipoprotein subclasses between intensive versus conventional management of type 1 diabetes during the randomization phase of the Diabetes Control and Complications Trial (DCCT). NMR-determined lipoprotein subclass profiles (NMR-LSPs), which estimate molar subclass concentrations and mean particle diameters, were determined in 1,294 DCCT subjects after a median of 5 years (interquartile range: 4–6 years) of randomization to intensive or conventional diabetes management. In cross-sectional analyses, we compared standard lipids and NMR-LSPs between treatment groups. Standard total, LDL, and HDL cholesterol levels were similar between randomization groups, while triglyceride levels were lower in the intensively treated group. NMR-LSPs showed that intensive therapy was associated with larger LDL diameter (20.7 vs. 20.6 nm, P = 0.01) and lower levels of small LDL (median: 465 vs. 552 nmol/l, P = 0.007), total IDL/LDL (mean: 1,000 vs. 1,053 nmol/l, P = 0.01), and small HDL (mean: 17.3 vs. 18.6 μmol/l, P < 0.0001), the latter accounting for reduced total HDL (mean: 33.8 vs. 34.8 μmol/l, P = 0.01). In conclusion, intensive diabetes therapy was associated with potentially favorable changes in LDL and HDL subclasses in sera. Further research will determine whether these changes contribute to the beneficial effects of intensive diabetes management on vascular complications.  相似文献

A variety of slow release fertilizers, controlled release (availability) fertilizers (CAFs), and stability fertilizers have been developed in response to the serious drawbacks of the conventional fertilizers since the early 1960's. Of these fertilizers, CAFs which are coated with resin are consumed in the largest quantity in the world. Selecting CAFs with higher performance, the author will discuss about: 1) Innovation of agro-technologies for various field crops including new concepts of fertilizer application, 2) high yielding of field crops, 3) enhancing quality and safety of farm products, and 4) controlling the adverse effect of intensive agriculture on the environment.  相似文献

Intensive groundwater extraction causes many environmental problems globally. Reducing groundwater extraction is a primary method for alleviating these problems. However, this reduction may create new pollution issues because of an increase in groundwater vulnerability. A case study was done using the DRAOTIC evaluation method (an improved DRASTIC method for organic pollutant in aquifer vulnerability; soil (S factor) has been updated by organic matter (O factor) in the method) for the Hun River Basin, northeastern China, where groundwater intensive extraction had caused serious environmental and hydrogeological problems. The assessment results show that moderate vulnerability level is the main vulnerability level in the Hun River Basin; high vulnerability level and low vulnerability level categories occupy a smaller area; while very high and very low vulnerability categories occupy the smallest area. By combining the predicted groundwater level distribution and DRAOTIC model, the responses of groundwater vulnerability to different groundwater extraction reductions could be studied. The results show that groundwater vulnerability levels increased as groundwater extraction was reduced; this is because the rising groundwater levels make it easier for pollution coming from the surface to reach the aquifer. The more the reduction in groundwater extraction, the greater the increase in the area with higher vulnerability levels, and the greater the increase in pollution risk.  相似文献
