转双价抗虫基因毛白杨无性系85号抗虫性研究   总被引:2,自引:0,他引:2  
以优化的毛白杨无性系85号最适遗传转化体系为基础,通过农杆菌介导法将Bt(CryⅠAc)和API双价抗虫基因导入毛白杨无性系85号基因组.在高浓度卡那霉素的培养基上诱导不定芽和根,初步选择到200株转化植株,PCR检测显示,66株呈阳性反应.Southern杂交和ELISA检测进一步证明,Bt(CryⅠAc)和API双价抗虫基因已整合到毛白杨无性系85号基因组中,并得到了表达.对转化植株用舞毒蛾幼虫进行饲虫试验,结果显示昆虫幼虫的死亡率高达60.0%~77.8%,且存活幼虫的生长发育也受到了明显抑制.研究结果为杨树抗虫育种提供了新的种质资源.  相似文献

Estimates of annual survival are essential for addressing topics in evolutionary and conservation ecology. However, most demographic studies of land birds are based on north temperate species, and few robust estimates of survival based on mark–recapture statistics are available for continental South American birds. We used time‐since‐marking models to estimate apparent survival of adult birds from 7 yr of mist netting data in the Colonche Hills. This site is one of few remaining large tracts of premontane forest in southwestern Ecuador, and an area of high priority for avian conservation. Species with sufficient data for analysis included three hummingbirds (Adelomyia melanogenys, Heliodoxa jacula, Phaethornis baroni), a cotinga (Schiffornis turdinus), and a wren (Henicorhina leucophrys). Our parameter estimates had reduced precision because the number of recaptures was small. Probability of recapture was low in three species , and moderate in two others . Adelomyia and Phaethornis had moderate apparent survival (; probability that a bird neither died nor emigrated from our survey area in a given year). Adults of Adelomyia moved seasonally, and it is possible that permanent emigration from our survey area contributed to low estimates of apparent survival. The other three species had relatively high estimates of adult apparent survival ranging from in Heliodoxa and Schiffornis to a high of in Henicorhina.  相似文献

Environmental conditions are known to affect the reproductive fitness of insects, as do host plants. Although the highly specialized bracts of Himalayan ‘glasshouse’ plants are thought to be an adaptive morphology that allows the plant to reproduce in harsh alpine environments, little information appears to exist concerning the benefit of these bracts for the pollinating insects. In this study, we examine whether the semi‐translucent bracts of Rheum alexandrae, a giant herb endemic to the alpine zone of the Hengduan Mountains in south‐western China, enhance the reproductive fitness of its pollinator. Rheum alexandrae depends mainly on mutualistic seed‐consuming Bradysia flies for pollination. Bracts increased interior temperature on sunny days, acted as a buffer against fluctuating air humidity, greatly decreased the intensities of ultraviolet B/C radiation and provided shelter from strong winds. Bract removal significantly decreased adult oviposition and offspring performance of pollinators during flowering and fruiting, respectively. Our results indicate that the bracts of R. alexandrae enhance the reproductive fitness of its pollinating seed‐consuming mutualist in alpine environments, which may be attributed to the positive effects of the bracts on the interior microenvironment. © 2015 The Linnean Society of London, Botanical Journal of the Linnean Society, 2015, 179 , 349–359.  相似文献

A general field bioassay for detecting biologically active compounds in plants and insects has been developed and tested for efficacy and sensitivity. Methanolic extracts, in sucrose solution, of 20 plant and six caterpillar species were offered to the ponerine ant Paraponera clavata and the feeding preferences observed. The bioassay resulted in the detection of nine plant and three caterpillar species with ant-deterrent extracts, and 11 plant and three caterpillar species with neutral or attractant extracts. All of the plants showing ant-deterrent characteristics which had been chemically investigated in our laboratory, or for which chemical literature was available, contained secondary metabolites of known deterrence. Both naturally occurring and artificial differences in chemical concentrations could be detected using the bioassay. The method provides a means of screening plants and insects for compounds that are insect anti-feedants or that can modify insect behaviour.  相似文献

A technique for the collection of stable hemolymph from larvae of Manduca sexta has been developed. The method avoids the cell clumping and melanization reactions commonly encountered with insect hemolymph by minimizing contact between hemocytes and surfaces which provoke defensive or repair responses. The circulating hemocyte population of second-day, fifth-instar larvae (2dL5) of M. sexta consisted of 4.5 ± 2.5 × 106 cells/ml (n = 15, range 2–7 × 106 cells/ml) and contained five cell types: prohemocytes, plasmatocytes, granulocytes, spherulocytes, and oenocytoids. Two strains of Pseudomonas aeruginosa which differ in pathogenicity (P11-1 and 9027) and Escherichia coli D31 grew well at 26°C in cell-free hemolymph prepared from naive (nonimmunized) 2dL5 M. sexta. When viable cells of any of the three bacteria were injected into M. sexta larvae, changes in both the total hemocyte count (THC) and differential hemocyte count were observed. Viable bacteria were not required to produce these changes since formalin-killed cells of P. aeruginosa 9027 produced a qualitatively and quantitatively similar response. Following injection of bacteria, the THC increased, reaching a maximal level at 1 hr postinjection, and remained elevated for at least 4 hr after injection. While prohemocytes, plasmatocytes, granulocytes, and spherulocytes all increased in number, 80% of the increased cell population at 1 hr postinjection of bacteria were the latter two cell types. Granulocytes and spherulocytes are cells with recognized defensive capabilities. The increased numbers of these cells in circulation soon after injection of bacteria may confer an advantage on M. sexta larvae in dealing with bacterial infections. This could explain in part the unusual resistance of M. sexta to certain bacterial pathogens.  相似文献

The reproductive and diapausing adult females of brachypterous morph and macropterous females with reproductive arrest of non-diapause type, originating from the laboratory cultures of Pyrrhocoris apterus, were studied for their feeding and drinking behaviour, digestive enzyme activities, and carbohydrate and lipid contents. The highest feeding and drinking activities were observed in reproductive brachypters, the lowest in macropters. Macropters also differed from brachypters by lower activities of gut lipase, peptidase and protease, lower concentration of haemolymph sugars, and lower weight of fat body, which probably reflects their low feeding activity. The total content of fat body lipids was also lower in macropters (0.6 mg) than in reproductive and diapausing brachypters (4.6 and 7.5 mg, respectively) on day 14. A very high amount of glycogen was found in the fat body of diapausing brachypters, 363 μg on day 14, as opposed to 15 and 80 μg in macropterous and reproductive brachypterous females, respectively. The obtained data indicate that the most important difference between macropterous and brachypterous females with different types of reproductive arrest consists of an enhanced mobilization of lipids for dispersal in macropters and accumulation of energetic reserves for hibernation in brachypters.  相似文献

This report shows that one of the most important roles of the flower nectar of an autogamous perennialRorippa indica (L.) Hieron is as an attractant for employing some ant species as a defense against herbivorous insects. The plant has flowers from spring to early winter. Its flower nectar is frequently stolen by some ant species (hereafter cited as ants) which also feed on small herbivorous insects on the plant. Internations among the tritrophic levels (R. indica, herbivores, ants) were experimentally examined and the followings became clear. (1) Ants were attracted toR. indica in search of its flower nectar. (2) The gradual secretion of flower nectar seemed to detain ants on the plant. (3)Pieris butterfly lavae were the major herbivores onR. indica and were potentially harmful to the plant. (4) The presence of ants reduced the survival rate ofP. rapae larvae onR. indica. (5) The presence of ants reduced the feeding damage toR. indica. (6) The disadvantage of nectar use by ants seemed to be minimal for the plant since the ants did not disturb the other flower visitors. These facts suggest a mutualistic relationship betweenR. indica and ants. That is, the flower nectar serves as an indirect defense against herbivorous insects.  相似文献

The Australian ‘little ash beetle’ Acanthocnemus nigricans (Coleoptera, Cleroidea, Acanthocnemidae) is attracted by forest fires. A. nigricans has one pair of unique prothoracic sensory organs and it has been speculated that these organs may play a role in fire detection. Each organ consists of a cuticular disc, which is fixed over an air-filled cavity. On the outer surface of the disc, about 90 tiny cuticular sensilla are situated. The poreless outer peg of a sensillum is 3–5 μm long and is surrounded by a cuticular wall. One ciliary sensory cell innervates the peg. As a special feature, the outer dendritic segment is very short already terminating below the cuticle. A massive electron-dense cylindrical rod, which most probably represents the hypertrophied dendritic sheath, extends through the cuticular canal connecting the tip of the outer dendritic segment to the peg. The dendritic inner segment and the soma are fused indistinguishably. Thin, leaflike extensions of glial cells deeply extend into that conjoint and considerably enlarged compartment which also contains large numbers of mitochondria. In summary, the sensilla of the sensory disc of A. nigricans represent a new type of insect sensillum of hitherto unknown function. The possible role of the prothoracic sensory organ in fire detection is discussed.  相似文献

Robots designed to track chemical leaks in hazardous industrial facilities1 or explosive traces in landmine fields2 face the same problem as insects foraging for food or searching for mates3: the olfactory search is constrained by the physics of turbulent transport4. The concentration landscape of wind borne odors is discontinuous and consists of sporadically located patches. A pre-requisite to olfactory search is that intermittent odor patches are detected. Because of its high speed and sensitivity5-6, the olfactory organ of insects provides a unique opportunity for detection. Insect antennae have been used in the past to detect not only sex pheromones7 but also chemicals that are relevant to humans, e.g., volatile compounds emanating from cancer cells8 or toxic and illicit substances9-11. We describe here a protocol for using insect antennae on autonomous robots and present a proof of concept for tracking odor plumes to their source. The global response of olfactory neurons is recorded in situ in the form of electroantennograms (EAGs). Our experimental design, based on a whole insect preparation, allows stable recordings within a working day. In comparison, EAGs on excised antennae have a lifetime of 2 hr. A custom hardware/software interface was developed between the EAG electrodes and a robot. The measurement system resolves individual odor patches up to 10 Hz, which exceeds the time scale of artificial chemical sensors12. The efficiency of EAG sensors for olfactory searches is further demonstrated in driving the robot toward a source of pheromone. By using identical olfactory stimuli and sensors as in real animals, our robotic platform provides a direct means for testing biological hypotheses about olfactory coding and search strategies13. It may also prove beneficial for detecting other odorants of interests by combining EAGs from different insect species in a bioelectronic nose configuration14 or using nanostructured gas sensors that mimic insect antennae15.  相似文献
