为明确朝天椒(品种:BLTY2)叶片饲喂的斜纹夜蛾Spodoptera litura幼虫生长发育受到抑制的关键原因,本研究以人工饲料及牛角椒(品种:FXBX)叶片饲喂的斜纹夜蛾幼虫为对照,通过比较3种食料饲喂后斜纹夜蛾幼虫相关生理生化指标的变化,分析BLTY 2辣椒叶片饲喂所致的斜纹夜蛾幼虫生长发育异常的原因。结果表明,朝天椒辣椒叶片饲喂的斜纹夜蛾幼虫生长发育速率滞后于牛角椒辣椒叶片及人工饲料饲喂的斜纹夜蛾幼虫,且每头幼虫的发育时长均显著延长(P<0.05)。饲喂后第1天、第3天、第4天、第5天、第6天和第7天,饲喂BLTY 2辣椒叶片的斜纹夜蛾幼虫体内保幼激素(JH)含量均显著高于其他2种食料饲喂的斜纹夜蛾幼虫(P<0.05)。饲喂后第3天、第4天、第5天和第7天,饲喂BLTY 2辣椒叶片的斜纹夜蛾幼虫体内蜕皮激素(Ecd)含量均显著高于其他2种食料饲喂的斜纹夜蛾幼虫(P<0.05)。3种食料饲喂后斜纹夜蛾幼虫体内的几丁质酶(CHT)活性变化差异较大,而2种辣椒叶片饲喂后斜纹夜蛾幼虫体内的组织蛋白酶(CTS)活性变化趋势相似,且不同于人工饲料饲喂的斜纹夜蛾幼虫体内的CTS活性变化趋势。由此可见,BLTY 2辣椒主要干扰斜纹夜蛾幼虫体内JH与Ecd的含量抑制其生长发育。本研究结果可为筛选具有抗虫活性的植物次生代谢物质奠定基础。  相似文献

真社会性昆虫是最具代表性的表型可塑性的研究对象之一,其个体之间分工协作的社会性生活方式增强了整个群体的环境适应性和繁殖力。真社会性昆虫虽然具有相同的遗传背景,个体之间却表现出明显的品级分化和个体分工,这是由环境和遗传共同影响的。表观遗传被认为是应对环境条件下重塑基因表达的主要机制,非编码RNA作为一类广泛参与机体生命活动的不编码蛋白的功能性RNA,在真社会性昆虫的品级分化、个体分工等方面起着重要的调控作用。本文从微小RNA、长链非编码RNA、环状RNA、与PIWI蛋白相作用的RNA等非编码RNA,对蜜蜂、蚂蚁及白蚁等真社会性昆虫的非编码RNA调控机制研究进展进行了综述,以加深对真社会性昆虫内在遗传分子基础的理解和认识,也为害虫防治领域提供新的研发视角。  相似文献

旨在探究喀斯特地区退化生态系统植被恢复树种凋落叶分解过程及其对土壤碳排放的激发效应,为选择合适的树种进行植被恢复提供数据支持。以中国林科院热带林业实验中心大青山石山树木园11种适应性强、耐干旱贫瘠的优良石山树种为研究对象,利用13C自然丰度法区分凋落叶和土壤来源CO2并量化土壤激发效应,比较不同生态恢复树种凋落叶分解及其激发效应的差异,探讨凋落物分解及其激发效应与凋落物性状之间的关联。结果表明:(1)11个生态恢复树种凋落叶在碳相关化学性质(水溶性碳、半纤维素和单宁含量等)、养分含量(磷和镁含量等)及化学计量特征(碳磷比和氮磷比)等方面均表现出较高程度变异。(2)不同生态恢复树种凋落叶分解及其诱导的土壤激发效应具有极显著差异(P<0.001);在整个培养实验期间,11个生态恢复树种凋落叶平均分解了35.3%,其中海南椴分解最快,达到50%,而青冈栎分解最慢,仅分解16.5%。(3)总体上看,凋落叶处理的土壤呼吸速率(5.1 mg C kg-1 土壤 d-1)是对照土壤呼吸速率(2.3 mg C kg-1土壤d-1)的2.2倍,凋落叶添加显著促进土壤有机碳分解,平均达到37.6%;其中海南椴、割舌树和任豆凋落叶输入则抑制土壤有机碳分解(抑制程度分别为-13.2%、-6.9%和-22.5%),产生负激发效应。(4)凋落叶分解与非结构性碳(r=0.63,P=0.04)和水溶性碳(r=0.91,P<0.001)呈显著正相关,与叶干物质含量(r=0.64,P=0.03)、纤维素(r=0.62,P=0.04)和锰含量(r=-0.63,P=0.04)呈显著负相关。多元回归分析结果表明,水溶性碳、钾和钙含量相结合可以解释生态恢复树种凋落叶分解变异的98%;然而,凋落叶性状与土壤激发效应强度之间并没有显著相关性。从土壤养分归还角度考虑,喀斯特退化生态系统恢复树种可以选择光皮梾木、海南椴、顶果木和降香黄檀等凋落叶分解较快的树种,以促进土壤养分循环和植被恢复;另一方面,从土壤碳固持角度来看,海南椴、任豆和割舌树等凋落叶输入会抑制土壤有机碳分解,从而有利于提高退化生态系统土壤碳封存能力。  相似文献

The polyphagous beetle Gonioctena quinquepunctata Fabricius (Coleoptera: Chrysomelidae) is a serious leaf pest of the native European bird cherry, Prunus padus L., and the invasive alien black cherry, Prunus serotina Ehrh. (Rosaceae). In the shade, leaf damage is extensive in both species, whereas in full light, it is extensive in P. padus, but very low in P. serotina. We determined the influence of Prunus species and light conditions on differences in performance of both sexes of this folivore. In a laboratory experiment in which larvae were fed with leaves of a single species grown under particular light conditions, we measured larval, pupal, and adult mass, efficiency of conversion of ingested food (ECI), duration of development, total food eaten, and relative growth rate. In the field, we observed differences in beetle mass on shrubs of the two species growing under various light conditions. From the field observations, we hypothesised that leaves of the invasive P. serotina are not an equally good food source as leaves of P. padus for G. quinquepunctata, and the preference of these beetles for shaded shrubs is most favourable for their growth and development. Under laboratory conditions, we found that the beetle growth rate was not affected significantly by Prunus species or light conditions, despite the significant effect of light condition on the structure and chemical composition of Prunus seedlings. The lower ECI value for larvae feeding on sunlit leaves was compensated for by their higher level of consumption. In the field, adult insect mass was higher on P. padus than on P. serotina, and higher on sunlit shrubs of both species than on shaded ones. Under natural conditions, the mass of adult insects is probably also affected by other factors, such as predators and competition among folivores.  相似文献

Summary

The impacts of larvicides used in the control of blackflies (Diptera: Simuliidae) on macroinvertebrates in the stones-in-current biotope were assessed during 8 field trials in the middle Orange River, South Africa. Two Bacillus thuringiensis var. israelensis (B.t.i.) products (VectobacR 12AS and TeknarR HP-D) and the organophosphate temephos (AbateR 200EC) were applied at recommended and high dosages to simulate “operational” and “worst-possible” scenario's respectively. Mortality was evaluated either by direct counting of invertebrates on stones before and after application, or by ranking invertebrates on a 4-point relative abundance scale before and after application. In addition, the re-appearance of benthic invertebrate population densities after temephos application was examined.

At the recommended dosage (1.2 ppm/10 min), B.t.i. significantly reduced blackfly larval numbers (P<0.001) and those of the chironomid Rheotanytarsus fuscus Freeman (P<0.05). At high dosage (20 ppm/10 min), numbers of the filter-feeding mayfly Tricorythus discolor (Burmeister) (P<0.01) and the chironomid Cardiocladius sp. (P<0.05) were also significantly reduced. No Simulium predators were directly affected by B.t.i., but there were indications of food shortage amongst Hydropsychidae and Hirudinea.

Temephos caused significant reductions in the relative abundance of 5 taxa at 0.05 ppm, 3 to 6 taxa at 0.1 ppm, and 9 taxa at 1.0 ppm (P<0.05). “Non-target” organisms which were most affected included the chironomid R. fuscus, the mayflies Baetis glaucus Agnew and Choroterpes elegans Barnard, and the caddisflies Cheumatopsyche thomasseti Ulmer and Amphipsyche scottae Kimmins. The mayfly T. discolor was tolerant of temephos, even at high dosage (1.0 ppm/10 min). In winter, most taxa re-appeared within 19 days, and population densities were back to pre-treatment levels within 35 days.

It is concluded that good reduction of blackfly populations may be obtained with minimal direct impact on the “non-target” fauna, provided recommended dosages of temephos are not exceeded. Overdosing with temephos may result in high mortality of “non-target” organisms, including blackfly predators, and should be avoided.  相似文献

蛋白激酶D(Protein kinase D,PKD)是一种新的丝氨酸/苏氨酸蛋白激酶家族和甘油二酯(Diacylglycerol,DAG)受体,参与细胞内多种生理生化过程。为获得高纯度的PKD1的催化结构域(PKD1-cat)用于晶体学结构的研究,将带有GST标签的PKD1-cat基因克隆到杆状病毒转移载体pFastBac1中,构建了重组质粒。将重组质粒转化到含穿梭载体Bacmid的DH10Bac感受态细胞中,转座后获得了含目的基因GST-PKD1-cat的重组Bacmid。重组Bacmid DNA转染Sf9昆虫细胞后,获得重组杆状病毒并扩毒。将毒种以5 PFU/cell的感染复数感染悬浮培养的T.ni昆虫细胞,SDS-PAGE和Western blotting检测表达产物。结果显示,表达产物在分子量约68 kDa处有一特异条带可与GST单克隆抗体发生反应。经谷胱甘肽琼脂糖凝胶亲和层析纯化和PreScission Protease切除GST标签后,得到了纯度很高的分子量约42 kDa的目的蛋白PKD1-cat。体外PKD激酶活性实验结果显示,随着PKD1-cat浓度的增加,激酶活性增高。这些结果显示截短的重组PKD1-cat有很高的催化活性和纯度,为采用核磁共振或晶体学方法解析PKD1-cat的三维结构奠定了基础。  相似文献

Urbanization is an important driver of the diversity and abundance of tree‐associated insect herbivores, but its consequences for insect herbivory are poorly understood. A likely source of variability among studies is the insufficient consideration of intra‐urban variability in forest cover. With the help of citizen scientists, we investigated the independent and interactive effects of local canopy cover and percentage of impervious surface on insect herbivory in the pedunculate oak (Quercus robur L.) throughout most of its geographic range in Europe. We found that the damage caused by chewing insect herbivores as well as the incidence of leaf‐mining and gall‐inducing herbivores consistently decreased with increasing impervious surface around focal oaks. Herbivory by chewing herbivores increased with increasing forest cover, regardless of impervious surface. In contrast, an increase in local canopy cover buffered the negative effect of impervious surface on leaf miners and strengthened its effect on gall inducers. These results show that—just like in non‐urban areas—plant–herbivore interactions in cities are structured by a complex set of interacting factors. This highlights that local habitat characteristics within cities have the potential to attenuate or modify the effect of impervious surfaces on biotic interactions.  相似文献

Climate change can harm many species by disrupting existing interactions or by favouring new ones. This study analyses the foreseeable consequences of climatic warming in the distribution and dynamics of a Mediterranean pest that causes severe defoliation, the pine processionary caterpillar Thaumetopoea pityocampa, and the effects upon the relict Andalusian Scots pine Pinus sylvestris nevadensis in the Sierra Nevada mountains (southeastern Spain). We correlated a set of regional data of infestation by T. pityocampa upon Scots pine, from a broad ecological gradient, with climatic data for the period 1991–2001, characterized by alternating warm and cold winters. Defoliation intensity shows a significant association with previous warm winters, implying that climatic warming will intensify the interaction between the pest and the Scots pine. The homogeneous structure of the afforested pine woodlands favours the outbreak capacity of the newcomer, promoting this new interaction between a Mediterranean caterpillar pest and a boreal tree at its southern distribution limit.  相似文献

Abstract.  1. Visual surveys for small organisms on complex substrates often yield serious underestimates of true counts. When both visual counts (relative estimates of abundance) and absolute counts can be obtained from the same sample, however, the visual counts can be calibrated such that absolute estimates can be obtained in the future from visual surveys alone.
2. A method is presented for converting quick, timed, visual counts of a sedentary insect on a shrub into absolute estimates of abundance.
3. Analogies were drawn from simple, well-known predation theories to develop a two-parameter non-linear model. Parameter estimates were obtained by both inverse prediction and direct estimation methods; the latter were found to yield more accurate predictions of absolute abundance.
4. The calibration model is mechanistic in its approach, and thus has potential for application in other systems in which all individuals are visible, but able to be missed during timed counts.  相似文献

Abstract.  1. Seed production can be affected by cattle grazing, insect herbivores, and seed predators. Bush vetch, Vicia sepium L. (Fabaceae), possess extra floral nectaries (EFNs), which can attract ants that act as plant defenders.
2. In this field study, four different manipulations were used to investigate interactions between V. sepium , ants, aphids, and seed predators, mainly Bruchus atomarius. The experiment was set up in grazed and non-grazed plots in a semi-natural grassland in south-central Sweden.
3. Grazing negatively affected number of V. sepium buds, flowers, and pods. Seed set, defined as the number of developed seeds per number of ovules in a pod, was higher in grazed plots. Ant abundance, however, was not affected by grazing. Ten different ant species were found on V. sepium and the number of ants was positively correlated with the presence of EFNs. Aphids were found only when ants were excluded from the plants. Pod and seed production of V. sepium plants was not affected by the presence of ants. The seed predator B. atomarius was not affected by the presence of ants. No support was found for the supposition that V. sepium benefited from attracting ants by producing extra floral nectar.
4. A small observational study of V. sepium plants in shrub ( Rosa dumalis ) plots compared with grassland plots was added to the experimental study . Vicia sepium plants were longer and had higher seed-set (seeds/ovules) in shrub plots than in grassland plots.  相似文献
