Unidirectional flux rates of saturated fatty acids, saturated alcohols, and bile acids were measured in an intact rat diaphragm preparation. The logarithm of the permeability coefficients for fatty acids containing from five to ten carbon atoms was a linear function of the number of carbon atoms in the fatty acid chain. Incremental free energies of solution were +336 cal · mol−1 for the addition of a hydroxyl group and −258 cal · mol−1 for the addition of a methylene group. These incremental free energies were similar to those obtained by other investigators in other animal tissues, and our data suggest a structural similarity between membranes in different tissues and in different species. The muscle membrane exhibited anomalously high permeabilities for fatty acids containing less than five carbon atoms. Since muscle lacks tight junctions, this result suggests that small non-electrolytes traverse polar regions or aqueous pores within the cellular membrane.  相似文献

A study on nutrient regeneration processes and a measure of their fluxes at the sediment-water interface was carried out in two different stations of a shallow lagoon of the Po delta river (Italy). A few parameters on the solid fraction (grain-size, porosity, C, N) and pore water profiles of o-P, NH3, NO inf3 sup– , SiO2, Tot-CO2, SO inf4 sup2– , Fe, Mn, Ca, Mg, pH, Eh were determined. At both stations the results were typical for fine sediments rich in organic matter. The ratio of variations of sulphate (SO inf4 sup2– ) to total carbonate demonstrates the main role sulphate reduction plays on the organic matter decay. The use of the ratios of variations of sulphate (SO inf4 sup2– ) to ammonia (NH3) and of sulphate (SO inf4 sup2– ) to phosphate (o-P) in pore waters enabled us to calculate the C/N/P of the decomposing organic matter. Obtained C/N/P indicated an enrichment of N and P with regard to C/N/P ratios of the solid fraction, due to the selective stripping of N and P during organic matter mineralization. This phenomenon decreases with depth, where organic matter becomes more refractory. Calculations on saturation degrees have shown the possibility of authigenic calcite, apatite and rhodochrosite precipitation in sediments. Nutrient fluxes were estimated for SiO2, NH3 and o-P by means of benthic chambers and modelling the pore water profiles. The model used for the calculation of fluxes allowed us to account for the bioturbation-irrigation influence near the interface, by means of a cumulative diffusion coefficient. Directly measured fluxes proved to be always significantly greater than the theoretical ones. These differences seem to be due to surface processes which do not affect pore water concentrations (degradation of fresh materials at the interface; micro-bioturbation by small gasteropoda such as Hydrobia ventrosa) and/or to the different concept of the two methods in time and space. Number, size and biomass of macrobenthic species living in the sediment underneath the benthic chambers were determined. The comparison between data on macrobenthic populations and flux values showed a good relationship between the number of organisms and benthic fluxes within each station. However, flux variations between stations are to be attributed mainly to the different arrangement of the tubes of the polychaetes Polydora ciliata in the sediment.  相似文献

A mathematical treatment is given for 1/f noise observed in the ion transport through membranes. It is shown that this noise can be generated by current or voltage fluctuations which occur after step changes of the membrane permeability. Due to diffusion polarization in the unstirred solution layers near the membrane these fluctuations exhibit a 1/t time course which produces noise with a 1/f frequency dependence. The spectral density of 1/f noise is calculated for porous membranes with random switches between a finite and zero pore permeability. A wide frequency range and a magnitude of 1/f noise are obtained which are compatible with experimental data of 1/f noise reported for nerve membranes.Supported by Deutsche Forschungsgemeinschaft, Sonderforschungsbereich 38 Membranforschung.  相似文献

Summary Ultrastructural observation of the sensory pore of several species of Natantia reveals a twofold organ. A main sensory pore (M.S.P.) comprises a layer of supporting cells which encapsulate the terminal region of sensory cell bodies. These sensory cells include two ciliary processes dividing into a flat sub-cuticular cavity. The cuticle opposite is thin and perforated with crater-like paired micropores. Next to the main sensory pore, a second organ, the lateral sensory pore (L.S.P.), is smaller and more difficult to observe. A complex-shaped cavity underlies a contorted epicuticular invagination. Ciliary outer segments, belonging to a bundle of sensory cells, branch out in this cavity.M.S.P. and L.S.P. appear to be chemoreceptors.I thank Mme Colette Besse for her technical assistance and Mr. A. Mitochondria from different organisms can undergo a sudden process of inner membrane unselective leakiness to molecules known as the mitochondrial permeability transition (MPT). This process has been studied for nearly four decades and several proteins have been claimed to constitute, or at least regulate the usually inactive pore responsible for this transition. However, no protein candidate proposed as the actual pore-forming unit has passed rigorous gain- or loss-of-function genetic tests. Here we review evidence for -and against- putative channel-forming components of the MPT pore. We conclude that the structure of the MPT pore still remains largely undefined and suggest that future studies should follow established technical considerations to unambiguously consolidate the channel forming constituent(s) of the MPT pore.  相似文献

Human erythrocytes were exposed to oxidative stress by iodate and periodate. Oxidation causes a time- and concentration-dependent increase in membrane permeability for hydrophilic molecules and ions. The induced leak discriminates nonelectrolytes on the basis of molecular size and exhibits a very low activation energy (Ea = 1–4 kcal · mol?1). These results are reconcilable with the formation of aqeous pores. The pore size was approximated to be between 0.45 and 0.6 nm. This increase in permeability is reversible upon treatment with dithioerythritol. Blocking of membrane thiol groups with N-ethylmaleimide protects the membranes against leak formation. The oxidation causes dithioerythritol-reversible modification of membrane proteins as indicated by the gel electrophoretic behavior. These modifications can also be suppressed by blocking the membrane thiol groups with N-ethylmaleimide. About half of the membrane methionine is oxidized to acid hydrolysis-stable derivatives. A fast saturating increase in diene conjugation was observed in whole cells but not in isolated membranes, with only minor degradation of fatty acid chains. The oxidation of cell membrane lipids as well as oxidation of cell surface carbohydrates are not involved in leak formation. Taken together with earlier data (Deuticke, B., Poster, B., Lütkemeier, P., and Haest, C.W.M. (1983) Biochim. Biophys. Acta 731, 196–210), these findings indicate that formation of disulfide bonds by different oxidative mechanisms results in leaks with similar properties.  相似文献

【背景】雷可肽(Lexapeptide)为首例V型羊毛硫肽家族化合物,具有较好的抗革兰氏阳性菌活性,对耐甲氧西林金黄色葡萄球菌(Methicillin-Resistant Staphylococcus aureus,MRSA)和表皮葡萄球菌(Methicillin-Resistant Staphylococcus epidermidis,MRSE)的抑制作用强于广泛应用的食品防腐剂乳酸链球菌素,其对pH和高温的稳定性也优于乳酸链球菌素,具有较好的应用前景。由于抑菌机制不明确,限制了雷可肽的开发应用。【目的】探究雷可肽抑菌作用特征以及作用机制,为雷可肽开发应用奠定基础。【方法】通过菌落计数法与Mg2+试验表征雷可肽抑菌动力学曲线;采用流式细胞仪和透射电子显微镜研究雷可肽在靶细胞表面的成孔性;利用高效液相色谱与基质辅助激光解吸电离的时间飞行质谱分析雷可肽处理对革兰氏阳性菌肽聚糖前体积累的影响。【结果】雷可肽在抑菌动力学上与乳酸链球菌素没有显著差别,但在更宽的Mg2+浓度范围内仍可保持抑菌活性。雷可肽处理后的细胞具有透过荧光染料的能力,生物型透射电镜观察到细胞发生破损。此外,在雷可肽作用后的细胞中检测到肽聚糖合成的前体尿嘧啶核苷二磷酸-N-乙酰胞壁酸五肽。【结论】雷可肽能够通过抑制细胞壁肽聚糖生物合成并造成细胞损伤进而获得通透性,以此来抑制革兰氏阳性菌生长。  相似文献

A key factor influencing the quality of long oligonucleotides is the choice of controlled pore glass (CPG) which is used as a solid support during oligonucleotide synthesis. We studied the influence of CPG pore size on the quality of 75-mer oligonucleotides. Using electrophoresis and HPLC, we demonstrated failure modes that can occur at certain oligo lengths with 1000A pore size, and compared yield and purity of 75-mer oligos using 1000A and larger pore size CPG. We showed that oligonucleotides with much better quality are obtained using CPG with pore sizes of 1400A and larger. We also identified the key characteristics for CPG selection that lead to the best CPG performance.  相似文献

Annual production of crop residues has reached nearly 4 billion metric tons globally. Retention of this large amount of residues on agricultural land can be beneficial to soil C sequestration. Such potential impacts, however, may be offset if residue retention substantially increases soil emissions of N2O, a potent greenhouse gas and ozone depletion substance. Residue effects on soil N2O emissions have gained considerable attention since early 1990s; yet, it is still a great challenge to predict the magnitude and direction of soil N2O emissions following residue amendment. Here, we used a meta‐analysis to assess residue impacts on soil N2O emissions in relation to soil and residue attributes, i.e., soil pH, soil texture, soil water content, residue C and N input, and residue C : N ratio. Residue effects were negatively associated with C : N ratios, but generally residue amendment could not reduce soil N2O emissions, even for C : N ratios well above ca. 30, the threshold for net N immobilization. Residue effects were also comparable to, if not greater than, those of synthetic N fertilizers. In addition, residue effects on soil N2O emissions were positively related to the amounts of residue C input as well as residue effects on soil CO2 respiration. Furthermore, most significant and stimulatory effects occurred at 60–90% soil water‐filled pore space and soil pH 7.1–7.8. Stimulatory effects were also present for all soil textures except sand or clay content ≤10%. However, inhibitory effects were found for soils with >90% water‐filled pore space. Altogether, our meta‐analysis suggests that crop residues played roles beyond N supply for N2O production. Perhaps, by stimulating microbial respiration, crop residues enhanced oxygen depletion and therefore promoted anaerobic conditions for denitrification and N2O production. Our meta‐analysis highlights the necessity to connect the quantity and quality of crop residues with soil properties for predicting soil N2O emissions.  相似文献
