Smooth muscle contractility and protein tyrosine phosphorylation 总被引:1,自引:0,他引:1
During the last 5 years several studies have documented an involvement of protein tyrosine kinases (PTKs) in smooth muscle contraction and Ca2+mobilization. Most of these studies have utilized highly selective inhibitors of PTKs, genistein and tyrphostin and have shown that these inhibitors attenuated smooth muscle contraction induced by growth factors - epidermal growth factor (EGF) and platelet derived growth factor (PDGF) and several vasoactive peptides. It has also been demonstrated that inhibitors of protein tyrosine phosphatases (PTPases) such as vanadate and pervanadate mimic growth factors and vasoactive peptides in causing the contraction of smooth muscle. In this brief review, we have summarized some of the recent observations suggesting a possible link between protein tyrosine phosphorylation pathway and smooth muscle contraction. 相似文献

Reciprocal signals between the motor axon and myofiber induce structural and functional differentiation in the developing
neuromuscular junction (NMJ). Elevation of presynaptic acetylcholine (ACh) release on nerve-muscle contact and the correlated
increase in axonal-free calcium are triggered by unidentified membrane molecules. Restriction of axon growth to the developing
NMJ and formation of active zones for ACh release in the presynaptic terminal may be induced by molecules in the synaptic
basal lamina, such as S-laminin, heparin binding growth factors, and agrin. Acetylcholine receptor (AChR) synthesis by muscle
cells may be increased by calcitonin gene-related peptide (CGRP), ascorbic acid, and AChR-inducing activity (ARIA)/heregulin,
which is the best-established regulator. Heparin binding growth factors, proteases, adhesion molecules, and agrin all may
be involved in the induction of AChR redistribution to form postsynaptic-like aggregates. However, the strongest case has
been made for agrin's involvement. “Knockout” experiments have implicated agrin as a primary anterograde signal for postsynaptic
differentiation and muscle-specific kinase (MuSK), as a putative agrin receptor. It is likely that both presynaptic and postsynaptic
differentiation are induced by multiple molecular signals. Future research should reveal the physiological roles of different

Summary A chemically defined culture medium was developed to support the growth of two distinctly different types of normal human
breast epithelial cells (HBEC) derived from reduction mammoplasty. Type I cells expressed luminal epithelial cell markers
and were deficient in gap junctional intercellular communication (GJIC), whereas Type II cells expressed basal epithelial
cell markers and were efficient in GJIC. In this study, we examined and compared the growth factor and hormone requirements
of these two types of cells and a series of cell lines that were obtained by sequential transfection with SV40 DNA (extended
lifespan, nontumorigenic), treatment with 5-bromodeoxyuridine (BrdU)/black light (immortal and weakly tumorigenic), and infection
of a virus carrying the neu oncogene (highly tumorigenic). Growth of Type I cells was inhibited by withdrawing epidermal growth
factor (EGF), hydrocortisone (HC), or insulin (INS) from the culture media, but was enhanced by fetal bovine serum (FBS) supplementation.
Growth of Type II cells was inhibited by withdrawal of EGF, HC, or INS from the media, and was inhibited by FBS supplementation.
Withdrawal of human transferrin (HT) or 17β-estradiol (E2) from the media did not alter the growth of Type I or Type II cells. SV40 transfected Type I cell lines still required EGF,
HC, or INS for optimal growth. However, the highly tumorigenic cell line did not show a growth dependence on EGF, HC, or INS
but did appear to require HT and 3,3′,5-triiodo-D.L. thyronine (T3) for optimal growth. In addition, FBS stimulated the growth of these cell lines. Thus, this study shows that Type I HBEC
are distinctly different from Type II HBEC in growth response to FBS and that neoplastically transformed Type I cells could

采用光合速率和蒸腾速率之比 (Pn/Tr)表示植物水分利用效率 (WUE) ,发现在两种不同水分处理下春小麦拔节期 .WUE日变化中都有上午高于下午值 ,且 8∶0 0~ 1 0∶0 0为明显峰值的特征 ,这种特征与Pn、Tr日变化存在的上、下午值不对称性和反向性 (Pn为上午高于下午 ,Tr相反 )紧密相关 ,水分胁迫处理中 ,Pn、Tr值均有降低 ,但不同品种的WUE反应不一 .研究表明 ,叶水势 ( ψw)、气孔阻力 (Rs)等生理因素和空气相对湿度 (RH)、光照 (Q)及冠层温度 (Tc)等生态因子 ,通过对Tr和Pn的不同影响而同WUE显著相关 ,40 %RH是WUE变化的一个重要阈值 . 相似文献

结合模拟试验,对林下人参的生理活动及生长过程与林内生态因子的关系进行 了观测研究.结果表明,林内光照、水分、温度等因子与林下人参生长的关系极为密切.一 般林内光照在中等条件下,即相对光照在10~35%左右.林分郁闭度约0.6~0.8,土壤含 水量在 35~40%时人参生长最好;林下人参的物候进程及生长与温度的关系密切.林内 温度在5℃左右人参芽胞开始萌动,15℃左右地上茎高生长进入速生期,6月上旬生长停 止.因此,人为调节各种生态因子在适宜的范围内有利于林下人参的生长. 相似文献

应用细胞培养、免疫细胞化学和图像分析方法,探讨了缺氧猪肺动脉内皮细胞条件培养液对肺动脉平滑肌细胞碱性成纤维细胞生长因子(bFGF)表达的影响。本研究表明,肺动脉内皮细胞培养汇合前和汇合后均可表达bFGF,在缺氧时其表达增多。缺氧猪肺动脉内皮细胞条件培养液可刺激猪肺动脉平滑肌细胞表达bFGF,将条件培养液加热100℃处理或加入bFGF抗体后,可使猪肺动脉平滑肌细胞表达bFGF明显减少。这提示缺氧可刺激猪肺动脉内皮细胞表达bFGF,缺氧猪肺动脉内皮细胞条件培养液通过bFGF使肺动脉平滑肌细胞表达bFGF,因此推测缺氧条件下肺动脉内皮细胞bFGF的旁分泌作用可能在肺动脉平滑肌细胞增生中起重要作用,这一过程在缺氧性肺动脉高压时肺动脉结构重建的发生机制中具有重要意义。 相似文献

ERG recordings from German cockroaches showed that the amplitude of light-evoked responses have a circadian rhythmicity in adult males that coincided with the locomotor circadian rhythm. The peak of the response occurred during the subjective night, and the circadian period was less than 24 h under DD condition. In contrast, although the locomotor circadian rhythm was masked by the development of ovaries and pregnancy in females, their visual responses displayed circadian rhythmicity. This inconsistency in expression of locomotor and visual sensitivity circadian rhythms in females implied separate pacemakers for these two overt rhythms. After severing the optic nerves, changes in ERG amplitude of the operated cockroaches still displayed a circadian rhythm under DD condition, demonstrating that the visual sensitive pacemaker was located in the eye and independent from the locomotor pacemaker. 相似文献

Plant hormones play an important role during higher plant embryogenesis. Auxin is central to the development of vascular tissues, formation of lateral and adventitious roots, control of apical dominance, and tropic responses. Auxin response element (AuxRE), present in the promoters of many auxin-induced genes, can confer auxin responsiveness. Using carrot somatic embryo under specific developmental phase, a cDNA expression library was constructed. Several plasmids were recombined containing the tetramer of AuxRE as a bait. After screening by a yeast one-hy-brid system, one positive clone was confirmed and characterized. Electrophoretic mobility shift assay showed that AxRF1 protein expressed in yeast cell could bind AuxRE in vitro. It suggests that AxRF1 participates in regulation of the expression of auxin responsive gene during carrot somatic embryogenesis. 相似文献

Species in which both sex and parthenogenesis co‐occur are extremely valuable for investigating ecological conditions favouring sex. Tychoparthenogenesis is a breeding system characterized by hatching of a small proportion of unfertilized eggs (typically < 10%) from females of sexually reproducing species. With tychoparthenogenesis, both sexual and parthenogenetic reproduction co‐occur within the same population. To identify ecological conditions that may favour this breeding system, I quantified population variation in females’ capacity for tychoparthenogenesis and investigated biotic and abiotic correlates of tychoparthenogenesis. I estimated tychoparthenognetic capacity (proportion of unfertilized eggs hatching) for females from 12 Missouri populations of the mayfly, Stenonema femoratum (Ephemeroptera: Heptageniidae), across three different habitat types – temporary streams, permanent streams and lakes. Tychoparthenogenetic capacity, measured as the population mean hatch success of unfertilized eggs, ranged from 3.8 to 10.7%. Tychoparthenogenetic capacity varied among habitats in 1996, but not in 1997. In 1996, temporary streams showed hatch success of unfertilized eggs twice that of permanent streams and lakes. Tychoparthenogenetic capacity also varied among sampling dates within years. Temporary streams also showed extremely low nymph densities compared to the other two habitats. However, habitats did not differ in adult density. Furthermore, in all populations nymphs showed significantly female‐biased sex ratios. In contrast, adult sex ratios were equal or slightly male biased. Tychoparthenogenetic capacity was negatively correlated with nymph density in 1996, but not in 1997, suggesting possible reproductive assurance in some years. Adult densities also suggested that there may be certain times of year when tychoparthenogenesis may provide benefits of reproductive assurance.
