Six features are covered. (1) The high endemism, which is not discussed in detail, is all-pervasive, and has resulted from the isolation of Madagascar from Africa some 125 million years ago and their present separation by 430 km. (2) The great richness in plant species (especially relative to Africa), seen particularly in the families of woody species in the wetter vegetation-types, involves both sympatry and allopatry within genera, and is explicable in terms of much less extreme drying out than in Africa during the Pleistocene and effective ‘species-pumping’ rather than mass extinctions during that period. (3) The abundance and species-richness of palms, pandans, tree-ferns, bamboos, and certain families of dicot trees (notably Lauraceae, Monimiaceae, Myrsinaceae and Myristicaceae) in the lowland rain forests also appears to be a result of both past and present wetness of the climate, while it is hypothesized that the low stature of most lowland rain forests, paucity of large-girth trees, and small size and sparsity of broad-leaved herbs, are a result of most rain forest soils being old and relatively nutrient-poor. (4) Within the dry evergreen forest region where rainfall is moderate (900–1600 mm yr−1) a sub-set of trees with fire-resistant bark seems to have evolved at sites prone to frequent ground fires, some perhaps spreading out of adjacent palm savanna on seasonally flooded sites. (5) Both the evolution of thicket rather than grassy woodland in the driest areas (300–600 mm yr−1), and the abundance of evergreen trees and shrubs on ordinary soils – not confined to run-on sites – are explicable in terms of there being a finite chance of rain throughout the year rather than one short wet season, coupled with relatively high values for air humidity throughout the year. The same factors probably explain the abundance and variety of succulents in the thicket; they are found throughout and not just on rocks. (6) Concerning physical defence against herbivores, the rain forests, dry evergreen forests and deciduous forests all show a complete lack of plants with physiognomic features plausibly related to browsing by extinct giant birds (a strong contrast with New Zealand), but in the semi-deciduous thicket there are many tiny-leaved, mostly non-spiny shrubs and small trees, whose dense branching and impenetrability have plausibly evolved as a defence against browsing by elephant birds. The Didiereaceae of the thicket are spiny (unlike members of the same family in Africa), and are giant analogues of the ‘ocotillo’ (Fouquieria splendens) in western North America rather than of Cactaceae; their spines appear to be protecting the leaves more than the stems against arboreal primates, spine length paralleling leaf length.  相似文献

We describe here the oncostatin M (OSM)-dependent inhibition of in vivo tumour formation after intracerebral inoculation of glioblastoma cells in mice. We generated human glioblastoma cells transfected with the OSM gene under the control of a tetracycline-response promoter. Upon removal of tetracycline from the medium, cells exhibited a differentiated cell morphology, while proliferation was significantly inhibited. After implantation of these cells into nude mice brains, large tumours developed in animals lacking OSM expression, whereas no tumour formation was observed in mice with induced OSM expression. Our results suggest that OSM exerts pronounced antitumorigenic effects on glioblastoma cells in vivo and provide arguments for a therapeutic application of OSM in humans.  相似文献

Glutathione S-transferase P1 (GSTP1) is a ubiquitous expressed protein which plays an important role in the detoxification and xenobiotics metabolism. Previous studies showed that GSTP1 was upregulated by the LPS stimulation in RAW264.7 macrophage-like cells and GSTP1 overexpression downregulated lipopolysaccharide (LPS) induced inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression. Here we show that GSTP1 physically associates with the oxygenase domain of iNOS by the G-site domain and decreases the protein level of iNOS dimer. Both overexpression and RNA interference (RNAi) experiments indicate that GSTP1 downregulates iNOS protein level and increases S-nitrosylation and ubiquitination of iNOS. The Y7F mutant type of GSTP1 physically associates with iNOS, but shows no effect on iNOS protein content, iNOS S-nitrosylation, and changes in iNOS from dimer to monomer, suggesting the importance of enzyme activity of GSTP1 in regulating iNOS S-nitrosylation and stability. GSTM1, another member of GSTs shows no significant effect on regulation of iNOS. In conclusion, our study reveals the novel role of GSTP1 in regulation of iNOS by affecting S-nitrosylation, dimerization, and stability, which provides a new insight for analyzing the regulation of iNOS and the anti-inflammatory effects of GSTP1.  相似文献

An inducible expression vector, pSH19, which harbors regulatory expression system PnitA-NitR, for streptomycetes was constructed previously. Here, we have modified pSH19 to obtain shuttle vectors for Streptomyces-E. coli by introducing the replication origin of a plasmid for E. coli (ColE1) and an antibiotic-resistant gene. Six inducible shuttle vectors, pESH19cF, pESH19cR, pESH19kF, pESH19kR, pESH19aF, and pESH19aR, for Streptomyces-E. coli, were successfully developed. The stability of these vectors was examined in five different E. coli strains and Streptomyces lividans TK24. The stability test showed that the pSH19-derived shuttle vectors were stable in E. coli Stbl2 and S. lividans TK24. Heterologous expression experiments involving each of the catechol 2,3-dioxygenase, nitrilase, and N-substituted formamide deformylase genes as a reporter gene showed that pESH19cF, pESH19kF, and pESH19aF possess inducible expression ability in S. lividans TK24. Thus, these vectors were found to be useful expression tools for experiments on both Gram-negative and Gram-positive bacterial genes.  相似文献

Summary Studies on rapidly inducible resistance in trees against insect herbivores show substantial variation in the strength of responses. Here we report the results of a study which examined causes of this variation. We bioassayed the quality of leaves of two developmental phases (young vs. mature) of the mountain birch Betula pubescens ssp. tortuosa by measuring the growth of two instars of Epirrita autumnata larvae. We used only short shoot leaves from trees of a natural stand, uniform in size and age. Damage was caused by larvae and artificial tearing of leaf lamina, varying the scale and time. We separated seasonal changes in plants from instar-dependent effects of the animals by testing experimental larvae in two subsequent growth trials. We found that only larval-made damage induced responses in leaves that made the leaves significantly poorer quality for the test larvae. Artificial damage induced only weak responses, and artificial canopy-wide damage even caused slight improvement of leaf quality. Cumulative leaf damage did not strengthen birch responses. Leaves that were in the expansion phase responded to damage while fully-expanded, mature leaves showed no response. The pattern of responses indicated that there might be physiological constraints: small-scale damage induced resistance against the larvae but largescale damage did not. Prevalent weather conditions might have modified these responses. Larvae of two instars and sexes, of low- and high-density populations responded to leaf damage similarly. However, prior experience of larvae with the host plant may have affected subsequent larval performance. Variation in rapidly inducible responses in birches was caused by plant characters rather than by test animals.  相似文献

The oxygenase domain of the inducible nitric oxide synthase, Δ65 iNOSox is a dimer that binds heme, L-Arginine (L-Arg), and tetrahydrobiopterin (H4B) and is the site for NO synthesis. The role of H4B in iNOS structure-function is complex and its exact structural role is presently unknown. The present paper provides a simple mechanistic account of interaction of the cofactor tetrahydrobiopterin (H4B) with the bacterially expressed Δ65 iNOSox protein. Transverse urea gradient gel electrophoresis studies indicated the presence of different conformers in the cofactor-incubated and cofactor-free Δ65 iNOSox protein. Dynamic Light Scattering (DLS) studies of cofactor-incubated and cofactor-free Δ65 iNOSox protein also showed two distinct populations of two different diameter ranges. Cofactor tetrahydrobiopterin (H4B) shifted one population, with higher diameter, to the lower diameter ranges indicating conformational changes. The additional role played by the cofactor is to elevate the heme retaining capacity even in presence of denaturing stress. Together, these findings confirm that the H4B is essential in modulating the iNOS heme environment and the protein environment in the dimeric iNOS oxygenase domain. Phenotypic plasticity is central to the persistence of populations and a key element in the evolution of species and ecological interactions, but its mechanistic basis is poorly understood. This article examines the hypothesis that epigenetic variation caused by changes in DNA methylation are related to phenotypic plasticity in a heterophyllous tree producing two contrasting leaf types. The relationship between mammalian browsing and the production of prickly leaves was studied in a population of Ilex aquifolium (Aquifoliaceae). DNA methylation profiles of contiguous prickly and nonprickly leaves on heterophyllous branchlets were compared using a methylation‐sensitive amplified polymorphism (MSAP) method. Browsing and the production of prickly leaves were correlated across trees. Within heterophyllous branchlets, pairs of contiguous prickly and nonprickly leaves differed in genome‐wide DNA methylation. The mean per‐marker probability of methylation declined significantly from nonprickly to prickly leaves. Methylation differences between leaf types did not occur randomly across the genome, but affected predominantly certain specific markers. The results of this study, although correlative in nature, support the emerging three‐way link between herbivory, phenotypic plasticity and epigenetic changes in plants, and also contribute to the crystallization of the consensus that epigenetic variation can complement genetic variation as a source of phenotypic variation in natural plant populations. © 2012 The Linnean Society of London  相似文献

Four natural populations of the grasshopper Eyprepocnemis plorans in the Mallorca island were analysed for several years revealing the recent invasion of the B1 chromosome from the south‐west part of the island (Palma region) towards the north and to the east. In only 10 years, the mean number of Bs in the northern population at Pollença increased from 0.053 to 0.692. Therefore, B chromosome invasion seems to be very rapid and has recently arrived to the north of the island. The south‐west (close to Palma) is the most likely point at which B invasion started in the Mallorca Island. Finally, the number of B chromosomes was significantly associated to an increase in chiasma frequency (and thus recombination) in A chromosomes.  相似文献
