Summary The RNA structures synthesized in vitro by a crude enzyme complex from tobacco mosaic virus (TMV)-infected leaves have been analyzed; the major viral-specific products were similar to TMV-replicative form (RF) and-replicative intermediate (RI) in electrophoretic behavior and ribonuclease sensitivity. Synthesis of these RF-like and RI-like structures neither required nor responded to added viral RNA, but did require all four ribonucleotide triphosphates. Enriched radiolabeled RF-like and RI-like RNA fractions were isolated from non-denaturing agarose gels by electroelution and hybridized to a collection of TMV sequences cloned into bacteriophage M13. Enriched RF-RNA hybridized to sequences of both plus and minus polarity, while enriched RI-RNA hybridized only to inserts of minus polarity, indicating only plus strand synthesis in this fraction. Most of the label incorporated into the plus strand of the enriched RF-RNA was found near the 3-end of this strand, while most of the label incorporated into enriched RI-RNA was found several hundred bases from the 5-end of the plus strand.Paper presented at the first International Congress of Plant Molecular Biology (Savannah, GA, 1985). 相似文献

Summary Plant cells are sensitive to the antibiotic bleomycin, a DNA damaging glycopeptide. A bleomycin resistance determinant, located on transposon Tn5 and functional in bacteria, has been cloned in a plant expression vector and introduced into Nicotiana plumbaginifolia using Agrobacterium tumefaciens. ABSTRACT. Eggs and neonate larvae of chrysomelid beetles (sub-tribes Chrysomelina and Phyllodectina) were investigated for the presence of defensive substances.
The two isoxazolinone glucosides (compounds 1 and 2), characteristic of the adult defence secretion, were detected in the eggs of all studied species. Compound 2, containing a nitropropionate, is always present in concentrations (above 10-2 M), which are highly deterrent to the ant Myrmica rubra. This compound is not at all or only slightly toxic to ants at 10-2 M. Compound 1, devoid of nitropropionate, is a minor constituent, and is neither deterrent nor toxic to ants.
The five Chrysomela species studied and Phratora vitellinae also sequester salicin in their eggs in amounts highly deterrent and toxic to ants. A single Chrysomela egg often contains enough salicin to kill an ant. While the isoxazolinones are discarded with the egg shells, salicin is used by neonate larvae as a precursor for the production of salicylaldehyde in the thoracic defence glands, already functional at hatching. No salicin could be detected in the eggs of those species whose larvae produce cyclopentanoid monoterpenes, even if they feed on Salicaceae. No larva of any species seems to be able to produce detectable amounts of monoterpenes at birth. A very early defence, possible only in those species using salicin as the precursor for their defensive secretion, could be highly advantageous in protecting the clustered larvae during the long process of hatching and in avoiding cannibalism between siblings.
Only trace amounts of oleic acid were found in the eggs of Gastrophysa viridula , in contrast to previous reports on its presence in large quantities in the American G. cyanea. 相似文献

The gibberellin (GA) 2-hydroxylases in mature and immature seeds of Pisum sativum have been partially purified and characterised. The enzymes are unstable when stored below pH 7.0 or in the absence of a thiol reagent. The optimum assay pH is between 7.4 and 7.8 and activity is dependent upon the presence of -ketoglutarate, Fe2+ and ascorbate. The 2-hydroxylase activities for GA1, GA4, GA9 and GA20 are chromatographically inseparable and correspond to a protein of Mr 44000. The rate of GA 2-hydroxylation varies according to substrate and some evidence indicates that the 2-hydroxylase activities for GA1 and GA4 and for GA9 and GA20 may reside in different proteins. During pea seed maturation, the specific activity of the enzyme(s) increases dramatically and reaches a maximum at a time when endogenous GA9, GA20, GA29 and GA51 are also at their greatest concentration. This correlation is not the result of substrate induction of enzyme activity. Since the GA 2-hydroxylases operate at maximal rate at low substrate concentrations they are incapable of rapidly 2-hydroxylating excessive quantities of (exogenously applied) GA1 or GA20. On the basis of the kinetic parameters of the GA 2-hydroxylase activities, a generalised model is discussed for the control of the steady-state levels of bioactive hormone under normal physiological conditions.Abbreviations DTE

The rise time, of Signal IIf and the decay time of P-680+ have been measured kinetically as a function of pH by using EPR. The Photosystem II-enriched preparations which were used as samples were derived from spinach chloroplasts, and they evolved oxygen before Tris washing. The onset kinetics of Signal IIf are in agreement, within experimental error, with the fast component of the decay of an EPR signal attributable to P-680+. The signal IIf rise kinetics also show good agreement with published values of the pH dependence of the decay of P-680+ measured optically (Conjeaud, H. and Mathis, P. (1980) Biochim. Biophys. Acta 590, 353–359). These results are consistent with a model where the species Z (or D1) responsible for Signal IIf is the immediate electron donor to P-680+ in tris-washed Photosystem II fragments. 相似文献

Abstract: The concentration of γ-aminobutyrate (GABA) and the activity of glutamate decarboxylase and GABA-transaminase were measured in extracts of mouse brain before the onset and during the course of generalized seizures induced by systemic administration of homocysteine thiolactone. The results indicate that whole brain GABA metabolism is unaffected by subconvulsive and convulsive doses of homocysteine at all stages of the generalized seizure. Electroencephalographic monitoring of rat brain electrical activity via hippocampal electrode implantation allowed the course of homocysteine-induced seizures to be followed and afforded a means of quantifying such seizures. 相似文献

The effects of ultraviolet-B radiation on plant competition in terrestrial ecosystems 总被引:3,自引:0,他引:3
Evidence regarding the interaction of ultraviolet-B (UV-B, 280-320 nm) radiation and plant competition in terrestrial ecosystems is examined. The competitive interactions of some species pairs were affected even by ambient solar UV-B radiation (as exists without ozone depletion), when compared to control pairs grown without UV-B. Also, the total shoot biomass of these species pairs was depressed under ambient UV-B. Relatively large increases in UV-B radiation (approximating a 40% ozone layer reduction when weighted with the generalized plant action spectrum) altered the competitive interactions of some species pairs grown in pots under field conditions, but did not affect the total shoot biomass production of those pairs. Recent field experiments have examined the competitive interactions of wheat ( Triticum aestivum L. cv. Bannock) and wild oat ( Avena fatua L.) under a simulated increased UV-B regime resulting from a 16% ozone layer reduction when weighted with the generalized plant action spectrum. This increase in UV-B altered the competitive interactions of these two species without affecting the total shoot biomass production of the species pair. The manner in which increased UV-B affected the relative competitive abilities of the two species was highly dependent upon the environmental conditions during the early life stages of the plants. The implications of these results for both agricultural and natural plant communities are discussed. 相似文献

In addition to known derivatives, four new sesquiterpene-coumarin ethers were isolated from the roots of Achillea ochroleuca and Artemisia tripartita and identified by 1H and 13C NMR spectroscopy, including lanthanide induced shifts. The new compounds are isofraxidin derived ethers which differ from the previously described derivatives by ring cleavage and methyl migration within the terpenoid unit. The chemosystematic importance of sesquiterpene-coumarin ether accumulation within the two genera is briefly discussed. 相似文献

Studies using [3H]chlorogenic acid and [3H]rutin demonstrated that the kinetics of uptake of these plant phenolics into the haemolymph of 5th-instar Heliothis zea (Boddie) following actue oral administration is a first-order process. The total quantity of either phenolic present in the haemolymph within 1 hr amounts to 5% or less of the total ingested dose. Based on TLC analyses, 80% or more of the radioactivity in the haemolymph occurs as the parent phenolic. Retention of [3H]-chlorogenic acid or [3H]-rutin in H. zea following chronic feeding from 1st to 3rd-instar larvae is also linearly related to dietary dose. Chlorogenic acid and rutin are both equitoxic and equivalent in bioavailability to H. zea.Loss of [3H]-rutin from the haemolymph of 5th-instar larvae following injection is biphasic. One half of the injected dose is excreted in the frass in the first 6 hr after injection; the other half is thereafter eliminated at 1/20th of the initial rate. Analyses of extracts of frass by thin-layer chromatography indicate that after either chronic or acute feeding 90% of the ingested phenolic is excreted unchanged. Possible sites and modes of action of phenolics in insects are discussed in light of these findings. 相似文献

Summary Plants have been regenerated from nodular, green callus derived from cotyledon, petiole and leaf lamina explants ofG. argyrea, a perennial relative of the soybean (G. max). The degree of response obtained was governed primarily by the genotype used, accession G1626 proving the most responsive.
Shoots were also recovered from about 6.0% of cotyledon protoplasts of this genotype. The implications of these results are
