A previous analysis showed that Gammaproteobacteria could be the sole recoverable bacteria from surface-sterilized nodules of three wild species of Hedysarum. In this study we extended the analysis to eight Mediterranean native, uninoculated legumes never previously investigated regarding their root-nodule microsymbionts. The structural organization of the nodules was studied by light and electron microscopy, and their bacterial occupants were assessed by combined cultural and molecular approaches. On examination of 100 field-collected nodules, culturable isolates of rhizobia were hardly ever found, whereas over 24 other bacterial taxa were isolated from nodules. None of these nonrhizobial isolates could nodulate the original host when reinoculated in gnotobiotic culture. Despite the inability to culture rhizobial endosymbionts from within the nodules using standard culture media, a direct 16S rRNA gene PCR analysis revealed that most of these nodules contained rhizobia as the predominant population. The presence of nodular endophytes colocalized with rhizobia was verified by immunofluorescence microscopy of nodule sections using an Enterobacter-specific antibody. Hypotheses to explain the nonculturability of rhizobia are presented, and pertinent literature on legume endophytes is discussed.  相似文献

利用测序及变性高效液相色谱(DHPLC)研究了蒙古羊、哈萨克羊、滩羊和藏绵羊黑素细胞刺激素受体(MSHR)基因单核苷酸多态性(SNP)。结果表明,在扩增片断长度为415bp范围内存在一个T317C突变,DHPLC可检测到该突变并被证明是一种高通量且简便的筛选方法。通过两次DHPLC可确定两个杂合子和一个纯合子,第一次DHPLC可迅速检测出由于形成异源双链而呈肩峰的杂合子(TC),但不能区分两个均呈单峰的纯合子(TT或CC)。第二次DHPLC将未知纯合子与已知序列的纯合子混合后进行,通过判定单峰或肩峰即可推断未知纯合子的基因型。所研究的4个绵羊群体在该突变位点均处于Hardy-Weinberg平衡状态。蒙古羊与哈萨克羊较近的遗传亲缘关系以及滩羊与藏绵羊较近的遗传亲缘关系与线粒体DNA的研究结果一致。  相似文献

The functional activity of the exoglycan complex (EGC) polysaccharides from Rhizobium leguminosarum bv. viciae 250a and its nitrogen-resistant mutant M-71, capable of inducing the formation of nitrogen-fixing nodules on pea roots against a high-nitrogen background (4.8 mM NO3 ), was studied in vegetation tests. For this purpose, the bacterial inoculum washed free of its own exoglycans was supplemented with EGC of the same or another strain grown in the presence of 6 or 20 mM nitrate. The best symbiotic characteristics (nodule number and nitrogenase activity, mass of the roots and aerial parts of plants) were recorded when the inoculum cells and exoglycans were obtained from strain M-71 grown in the presence of 20 mM nitrate. When the plants were inoculated with the cells (grown at 6 mM nitrate) + EGC (obtained at 6 mM nitrate) of this strain, the nodulation characteristics and the effectiveness of symbiosis decreased 1.5- to 2-fold. Partial recovery of the symbiotic potential of strain M-71 was observed when EGC (obtained at 20 mM nitrate) was substituted for its exoglycans (obtained at 6 mM nitrate). In the presence of exoglycans of the parent strain 250a (obtained at 6 or 20 mM nitrate), the mutant formed a substantially lesser number of nodules with a very low nitrogen-fixing activity. In turn, the mutant exoglycans synthesized in medium with either high or low nitrate nitrogen concentration did not recover the fix+ phenotype of strain 250a, capable of forming symbiosis with pea plants only against a low-nitrogen background. In study of the relative content of high-molecular-weight exopolysaccharide components and low-molecular-weight glycans in the exoglycan complex, it was established that, in strain 250a (grown at 6 and 20 mM nitrate), as well as in its mutant M-71 (grown at 6 mM nitrate), exopolysaccharides prevailed, accounting for 72–75% of the sum of both types of glycopolymers, while low-molecular-weight glycans accounted for 25–28%. In contrast, in the EGC of strain M-71 obtained at 20 mM nitrate, which was the most active inducer of the formation of the symbiotrophic system by strain M-71 in the presence of a high mineral nitrogen concentration, low-molecular-weight glycans were the main component, accounting for 61% of total glycopolymers, while the polysaccharide content was 39%. Low-molecular-weight exoglycans are supposed to be involved in maintaining the physiological activity and the symbiotic status of rhizobia under unfavorable environmental conditions.  相似文献

An ethnobotanical study of the palms used by the Huaorani in the Yasuní National Park and Huaorani Ethnic Reserve in Amazonian Ecuador was carried out. In this inventory, 37 palm species were found; all were used by the Huaoranis. One hundred and ninety-one different uses were recorded in eight ethnobotanical categories. Most species (64.9%) were used for house construction and human food. More than half of the species were used for domestic utensils (59.4%) and hunting and fishing implements (54%). A comparison is made between these data and past studies for the other six indigenous communities from Amazonian Ecuador. This paper shows the highest diversity of useful palm species and the highest number of different uses ever recorded for an indigenous group in Amazonian Ecuador. Embryonic axes with cotyledons, shoot-tips of embryonic axes, isolated cotyledons, as well as axillary buds and leaves from 20-year-old trees of Sophora toromiro, were evaluated for their capacity to trigger organogenesis and to regenerate plantlets under in vitro conditions. Embryonic shoot-tips were the only explants capable of regenerating plants. They developed rapidly in vitro in the presence of NAA and BA while in subculture roots were induced at the proximal end in the presence of 0.49 μM IBA within 40–60 days. Development was completed with a subculture phase under non-sterile conditions using a mixture of equal parts of sterilized vermiculite/sand/soil in growth chambers, before final acclimation in the greenhouse. In the presence of NAA, BA and GA3, whole embryonic axes formed multiple shoots that branched when grown in 2.27 or 11.35 μM TDZ in subculture. Similarly, callus was initiated at the embryo axis base, developing into several new shoots in the presence of TDZ. Because of the relatively high shoot induction rate along the embryonic axis, this axis presents a valuable source of new juvenile explants. Growth and rhizogenesis was satisfactory only when organs from seed pods of the year or from the previous season were used. Experiments with isolated cotyledons produced callus only, while axillary buds and leaves did not show any responses in the presence of several growth regulators assayed. Inoculation of seedlings with various strains of rhizobia under in vitro conditions resulted in root outgrowths, but not in nodules that are typical of rhizobia infection. This paper incorporates the indigenous ecological knowledge (IEK) of the Maasai pastoralists and ecological methods to assess effects of grazing and cropping on rangeland biodiversity at macro‐ and micro‐landscape scales in northern Tanzania. The joint surveys with pastoralists identified indicator plant species and their associations with micro‐landscapes and livestock grazing suitability (i.e. for cattle and small ruminant grazing), while traditional calf‐pasture reserves (alalili pl. alalilia) were evaluated for preservation of rangeland biodiversity. The macro‐landscapes comprising the cool high plateau (osupuko pl. isipuki) and montane forest highland (endim) were included in the survey. At micro‐landscape scales, the osupuko was classified into uplands (orkung'u), slopes (andamata) and dry valley bottomlands (ayarata). The micro‐landscapes were assessed in terms of herbaceous plant species and woody species richness and risks of soil erosion. Biodiversity varied at both the macro‐ and micro‐landscape scales and in accordance with the land‐use types. Greater plant species diversity and less erosion risks were found in the pastoral landscapes than in the agro‐pastoral landscapes. The calf‐grazing pastures had greater herbaceous species richness than the non‐calf pastures, which in turn had more woody species. The study concludes that the indigenous systems of landscape classification provides a valuable basis for assessing rangeland biodiversity, which ecologists should incorporate into ecological surveys of the rangelands in East Africa in the future.  相似文献

Data are presented indicating a seasonal mosaic pattern of burning in the savanna of southern Mali. A seasonal mosaic is a landscape that is annually re-created by people, and which contains patches of unburned, early burned, and recently burned vegetation. A survey of over 100 farmers and in-depth interviews demonstrates that rural inhabitants of southern Mali begin an annual burning regime early in the dry season in order to fragment the landscape, with the goal of preventing later fires that can damage natural resources. The process of gradually burning off the driest vegetation creates a seasonal mosaic of habitat patches that increases the potential of the landscape for a variety of dry season land uses, including hunting, gathering of savanna products, and grazing. An analysis of a series of Landsat images shows that the practice of mosaic burning is widespread in the wooded savanna, in which burning usually begins early and large fires are rare. On the basis of recent developments in ecological theory and empirical evidence from similar burning regimes in parts of Australia, it is suggested that seasonal mosaic burning in Mali not only prevents damaging late-season fires but increases biodiversity. It is concluded that discourse on African savanna burning overemphasizes the ecologically detrimental aspects of fire, while neglecting the beneficial ones resulting in misguided policies that pose a threat to human livelihoods and savanna ecosystems.  相似文献
