Squid giant axons were injected with aequorin or arsenazo III and impaled with a Ca-sensing electrode. The light output of aequorin or the spectrophotometer output when measuring arsenazo was compared with the voltage output of the electrode when the squid axon was depolarized with high-K solutions, when the seawater was made Na-free, or when the axon was tetanized for several minutes. The results from these treatments were that the optical response rose (as much as 50-fold) with all treatments known to increase Ca entry, while the electrode remained unaffected by these treatments. If axons previously subjected to Ca load are treated with electron-transport poisons such as CN, it is known that [Ca]i rises after a time necessary to deplete ATP stores. In such axons one expects a rise of [Ca]i in axoplasm which does not necessarily have to be uniform although the source of such Ca is the mitochondria and these are uniformly distributed in axoplasm. Under conditions of CN application, the optical signals from aequorin or arsenazo and Ca electrode output do rise together when [Ca]i is high, but there is a region of [Ca]i concentration where aequorin light output or arsenazo absorbance rises while electrode output does not. Axons not loaded with Ca but injected with apyrase and vanadate have mitochondria that still retain some Ca and this can be released by CN in a truly uniform manner. The results show that such a release (which is small) can be readily measured with aequorin, but again the Ca electrode is insensitive to such [Ca]i change. 相似文献

- In stream ecosystems, bacterial communities play an important role in nutrient and energy cycling processes as they are among the most numerous and active organisms at the basal trophic level of the stream food web. Bacterial communities in stream biofilms have been shown to correlate well with different catchment land use and therefore provide an opportunity for the development of a novel ecological indicator of stream ecosystem health.
- In this study, a bacterial community index (BCI) model was developed and validated using a national data set of biofilm bacterial community profiles collected from 223 streams across seven geographical regions in New Zealand. The six‐component BCI model was generated using the partial least squares regression method to associate the multivariate bacterial community profile with the macroinvertebrate community index, which is a well‐established indicator of stream health.
- Despite strong regional clustering of the bacterial community profiles, the BCI was indicative of the level of disturbance in the catchment, as shown by significant correlations with a wide range of independent indicators of water quality, macroinvertebrate community data, ecosystem functioning and catchment land‐use data. The BCI was able to explain 35% of the variation in a multi‐metric index incorporating ten common ecological parameters, suggesting that the stream bacterial communities could provide useful information about the ecosystem integrity.
- The BCI provides a novel ecosystem assessment tool, which can be used to complement existing stream health measures in the management of anthropogenic impacts on freshwater streams and rivers.

The study was aimed to investigate a color indicator containing dual curing resin composite luting cement and to plot the color change to the time of solidification of the cement. In addition some physical properties were studied. Specimens were made of a dual-cure resin cement (Maxcem Elite™ Chroma, Kerr, Orange, CA, USA) and polymerized by autopolymerization only, or with light initiated polymerization. A spectrophotometer was used to quantify the color change of the cement as plotted with the curing time. The efficacy of the curing process was studied by measuring water sorption and the ultimate flexural properties of the cement. The results showed that the flexural strength of cement after autopolymerization was 27.3 MPa and after light initiated polymerization 48.1 MPa. Young’s modulus of bending was 2089.3 MPa and 3781.5 MPa respectively for the same cement samples. Water sorption after two weeks for the autopolymerization cement samples was −1.12 wt% and for the light initiated polymerization samples 0.56 wt%. Non-parametric Spearman’s correlation was measured for autopolymerized cement samples between variables for color and solidification load (N), which showed a strong correlation between curing process and color change (p < 0.05). There was a correlation between the color change and degree of monomer conversion of the dual curing resin composite luting cement which contained a color indicator system for polymerization reaction. The study also suggested that autopolymerization only resulted in suboptimal polymerization of the cement. By additional light curing considerably higher flexural properties were obtained. 相似文献

We analyze how measures of adiposity – body mass index (BMI) and waist hip ratio (WHR) – causally influence rates of hospital admission. Conventional analyses of this relationship are susceptible to omitted variable bias from variables that jointly influence both hospital admission and adipose status. We implement a novel quasi-Poisson instrumental variable model in a Mendelian randomization framework, identifying causal effects from random perturbations to germline genetic variation. We estimate the individual and joint effects of BMI, WHR, and WHR adjusted for BMI. We also implement multivariable instrumental variable methods in which the causal effect of one exposure is estimated conditionally on the causal effect of another exposure. Data on 310,471 participants and over 550,000 inpatient admissions in the UK Biobank were used to perform one-sample and two-sample Mendelian randomization analyses. The results supported a causal role of adiposity on hospital admissions, with consistency across all estimates and sensitivity analyses. Point estimates were generally larger than estimates from comparable observational specifications. We observed an attenuation of the BMI effect when adjusting for WHR in the multivariable Mendelian randomization analyses, suggesting that an adverse fat distribution, rather than a higher BMI itself, may drive the relationship between adiposity and risk of hospital admission. 相似文献

Switchgrass is being evaluated as a potential feedstock source for cellulosic biofuels and is being cultivated in several regions of the United States. The recent availability of switchgrass land cover maps derived from the National Agricultural Statistics Service cropland data layer for the conterminous United States provides an opportunity to assess the environmental conditions of switchgrass over large areas and across different geographic locations. The main goal of this study is to develop a data-driven multiple regression switchgrass productivity model and identify the optimal climate and environment conditions for the highly productive switchgrass in the Great Plains (GP). Environmental and climate variables used in the study include elevation, soil organic carbon, available water capacity, climate, and seasonal weather. Satellite-derived growing season averaged Normalized Difference Vegetation Index (GSN) was used as a proxy for switchgrass productivity. Multiple regression analyses indicate that there are strong correlations between site environmental variables and switchgrass productivity (r = 0.95). Sufficient precipitation and suitable temperature during the growing season (i.e., not too hot or too cold) are favorable for switchgrass growth. Elevation and soil characteristics (e.g., soil available water capacity) are also an important factor impacting switchgrass productivity. An anticipated switchgrass biomass productivity map for the entire GP based on site environmental and climate conditions and switchgrass productivity model was generated. Highly productive switchgrass areas are mainly located in the eastern part of the GP. Results from this study can help land managers and biofuel plant investors better understand the general environmental and climate conditions influencing switchgrass growth and make optimal land use decisions regarding switchgrass development in the GP. 相似文献

Regime shifts are abrupt transitions between alternate ecosystem states including desertification in arid regions due to drought or overgrazing. Regime shifts may be preceded by statistical anomalies such as increased autocorrelation, indicating declining resilience and warning of an impending shift. Tests for conditional heteroskedasticity, a type of clustered variance, have proven powerful leading indicators for regime shifts in time series data, but an analogous indicator for spatial data has not been evaluated. A spatial analog for conditional heteroskedasticity might be especially useful in arid environments where spatial interactions are critical in structuring ecosystem pattern and process. We tested the efficacy of a test for spatial heteroskedasticity as a leading indicator of regime shifts with simulated data from spatially extended vegetation models with regular and scale‐free patterning. These models simulate shifts from extensive vegetative cover to bare, desert‐like conditions. The magnitude of spatial heteroskedasticity increased consistently as the modeled systems approached a regime shift from vegetated to desert state. Relative spatial autocorrelation, spatial heteroskedasticity increased earlier and more consistently. We conclude that tests for spatial heteroskedasticity can contribute to the growing toolbox of early warning indicators for regime shifts analyzed with spatially explicit data. 相似文献
