Regulators of G-protein signaling (RGS) 9-2 is a striatal enriched protein that controls G protein coupled receptor signaling duration by accelerating Galpha subunit guanosine triphosphate hydrolysis. We have previously demonstrated that mice lacking the RGS9 gene show enhanced morphine analgesia and delayed development of tolerance. Here we extend these studies to understand the mechanism via which RGS9-2 modulates opiate actions. Our data suggest that RGS9-2 prevents several events triggered by mu-opioid receptor (MOR) activation. In transiently transfected PC12 cells, RGS9-2 delays agonist induced internalization of epitope HA-tagged mu-opioid receptor. This action of RGS9-2 requires localization of the protein near the cell membrane. Co-immunoprecipitation studies reveal that RGS9-2 interacts with HA-tagged mu-opioid receptor, and that this interaction is enhanced by morphine treatment. In addition, morphine promotes the association of RGS9-2 with another essential component of MOR desensitization, beta-arrestin-2. We also show that over-expression of RGS9-2 prevents opiate-induced extracellular signal-regulated kinase phosphorylation. Our data indicate that RGS9-2 plays an essential role in opiate actions, by negatively modulating MOR downstream signaling as well as the rate of MOR endocytosis. 相似文献

Studied contents of macro-, microelements and long-lived radionuclides in the following medicinal plants belonging to the wetland community of Siberian region, Russia: Sphagnum fuscum (Schimp.) Klinggr., Sphagnum balticum (Russ.) Russ. ex С. Jens., Polytrichum strictum Brid., Pinus sylvestris L., Pinus sibirica Du Tour, Betula pubescens Ehrh., Andromeda polifolia L., Ledum palustre L., Oxycoccus palustris Pers., Rubus chamaemorus L., Vaccinium uliginosum L., Comarum palustre L., Chamaedaphne calyculata (L.) Moench. Samples were collected in the sphagnum wetlands of Tomsk region and Khanty-Mansi Autonomous Okrug. For the purpose of research, we used roots, shoots and bark of the plants in the Pinophyta and Magnoliophyta species and turf of the Bryophyta species. The content of macro-, microelements in the plants was determined by an atomic emission spectrometer with inductively coupled plasma “iCAP 6300 Duo” of the “Thermo Scientific” company. The specific activities of the long-lived radionuclides Th-232, K-40, Ra-226, Cs-137 were measured by a gamma-ray spectrometric complex with a high purity germanium semiconductor detector by “ORTEC” (AMETEK) and a digital analyser “ORTEC DSPEC LF”. Nectar robbers access floral nectar in illegitimate flower visits without, in general, performing a pollination service. Nevertheless, their effect on fruit set can be indirectly positive if the nectar removal causes an incremental increase in the frequency of legitimate flower visits of effective pollinators, especially in obligate outcrossers. We studied pollination and the effect of nectar robbers on the reproductive fitness of Jacaranda rugosa , an endemic shrub of the National Park of Catimbau, in the Caatinga of Pernambuco, Brazil. Xenogamous J . rugosa flowers continuously produced nectar during the day at a rate of 1 μl·h−1 . Female and male Euglossa melanotricha were the main pollinators. Early morning flower visits substantially contributed to fruit set because stigmas with open lobes were almost absent in the afternoon. Ninety-nine per cent of the flowers showed damage caused by nectar robbers. Artificial addition of sugar water prolonged the duration of flower visits of legitimate flower visitors. Removal of nectar, simulating the impact of nectar robbers, resulted in shorter flower visits of euglossine bees. While flower visits of nectar-robbing carpenter bees ( Xylocopa frontalis , X . grisescens, X . ordinaria) produced only a longitudinal slit in the corolla tube in the region of the nectar chamber, worker bees of Trigona spinipes damaged the gynoecium in 92% of the flowers. This explains the outstandingly low fruit set (1.5%) of J. rugosa in the National Park of Catimbau. 相似文献

Aims: The aim of the study was to evaluate the in vitro antibacterial activity of glucosinolates and their enzymatic hydrolysis product against bacteria isolated from the human intestinal tract.
Methods and results: Using a disc diffusion bioassay, different doses of intact glucosinolates and their corresponding hydrolysis products were tested. There were clear structure–activity and concentration differences with respect to the in vitro growth inhibition effects as well as differences in the sensitivities of the individual bacteria. The most effective glucosinolate hydrolysis products were the isothiocyanates; sulforaphane and benzyl isothiocyanate were the best inhibitors of growth. Indole-3-carbinol had some inhibitory effects against the Gram-positive bacteria but had no effect, even at the highest dose, against the Gram-negative bacteria. Indole-3-acetonitrile had some inhibitory activity against the Gram-negative bacteria. Glucosinolates, nitriles and amines were ineffective at all the doses used.
Conclusions: Glucosinolate hydrolysis products and specifically the isothiocyanates SFN and BITC have significant antimicrobial activity against Gram-positive and Gram-negative bacteria, and might be useful in controlling human pathogens through the diet.
Significance and Impact of the Study: This the first major in vitro study demonstrating the potential of these natural dietary chemicals as an alternative to, or in combination with, current antibiotic-based therapies for treating infectious diseases. 相似文献

Cardiac contractile dysfunction is frequently reported in human patients and experimental animals with type-1 diabetes mellitus. The aim of this study was to investigate the voltage-dependence of contraction in ventricular myocytes from the streptozotocin (STZ)-induced diabetic rat. STZ-induced diabetes was characterised by hyperglycaemia and hypoinsulinaemia. Other characteristics included reduced body and heart weight and raised blood osmolarity. Isolated ventricular myocytes were patched in whole cell, voltage-clamp mode after correcting for membrane capacitance and series resistance. Moreover, the amplitude of contraction was significantly reduced (p < 0.05) in myocytes from STZ-treated rats at all test potentials between –20 mV and +30 mV. However, in electrically field-stimulated (1 Hz) myocytes, the amplitude of contraction was not altered by STZ-treatment. It is suggested that in field-stimulated myocytes taken from STZ-induced diabetic hearts, prolonged action potential duration may promote increased Ca2+ influx via the sodium-calcium exchanger (NCX), which may compensate for a reduction in Ca2+ trigger through L-type-Ca2+-channels and lead to normalised contraction. 城市化水平阈值是指在水资源硬约束下,在确保经济发展达到一定速度与规模、生态环境建设得到基本保障、并具备足以支撑城市接纳一定数量农民进城转为非农人口的经济能力时,能达到的“农转非”人口占总人口的最大比例。以干旱区河西走廊为例,分析了在水资源约束下城市经济发展总量及其对应的城市化阈值。采用阈值模型计算表明,未来30a在不跨区调水的前提下,河西走廊总需水量的上限阈值为7.81×109m3,其中生态需水、生产需水和生活需水比例调整为13.2:83.5:3.3,总需水量对应的国内生产总值为1.81×1011元,历年平均增长速度最快可达到6.91%,第一、二、三产业结构调整为18.99:47.20:33.81。在总需水量阈值、经济总量、需水结构与产业结构调整优化和经济增长速度不低于7%、农业需水按0.61%的速度退水9.12×108m3、生态需水比例不低于13%等多重条件约束下,求得河西走廊未来30a能够达到的总人口为5.82×106人,城镇非农业人口可达到2.05×106人,城市化水平可达到35.14%,这一水平仅相当于中国2001年的平均城市化水平。在跨区调水1.31×109m3的条件下,城市化水平可望达到47.17%。进而采用总人口人均拥有的GDP、非农业人口人均拥有的第二、三产业增加值、农业人口人均拥有的第一产业增加值3项经济指标对水资源约束下经济发展总量对城市化水平的保障程度进行了验证分析。由于各城市的缺水状况、取水条件、发展性质、发展阶段与经济发展实力与前景等各不相同,致使不同城市之间对应的总需水量、经济总量、经济增长速度和城市化水平等均表现出较大的差异性。 相似文献

To prevent diabetic complications derived from enhanced glucose flux via the polyol pathway the development of aldose reductase inhibitors (ARIs) has been established as a promising therapeutic concept. Here, we study the binding process of inhibitors to aldose reductase (ALR2) with respect to changes of the protonation inventory upon complex formation. Knowledge of such processes is a prerequisite to factorize the binding free energy into enthalpic and entropic contributions on an absolute scale. Our isothermal titration calorimetry (ITC) measurements suggest a proton uptake upon complex formation with carboxylate-type inhibitors. As the protonation event will contribute strongly to the enthalpic signal recorded during ITC experiments, knowledge about the proton-accepting and releasing functional groups of the system is of utmost importance. However, this is intricate to retrieve, if, as in the present case, both, binding site and ligand possess several titratable groups. Here, we present pKa calculations complemented by mutagenesis and thermodynamic measurements suggesting a tyrosine residue located in the catalytic site (Tyr48) as a likely candidate to act as proton acceptor upon inhibitor binding, as it occurs deprotonated to a remarkable extent if only the cofactor NADP+ is bound. We furthermore provide evidence that the protonation state and binding thermodynamics depend strongly on the oxidation state of the cofactor;s nicotinamide moiety. Binding thermodynamics of IDD 388, IDD 393, tolrestat, sorbinil, and fidarestat are discussed in the context of substituent effects. 相似文献

The functional state of the Photosystem (PS) II complex in Arabidopsis psbR T-DNA insertion mutant was studied. The ΔPsbR thylakoids showed about 34% less oxygen evolution than WT, which correlates with the amounts of PSII estimated from YDox radical EPR signal. The increased time constant of the slow phase of flash fluorescence (FF)-relaxation and upshift in the peak position of the main TL-bands, both in the presence and in the absence of DCMU, confirmed that the S2QA− and S2QB− charge recombinations were stabilized in ΔPsbR thylakoids. Furthermore, the higher amount of dark oxidized Cyt-b559 and the increased proportion of fluorescence, which did not decay during the 100s time span of the measurement thus indicating higher amount of YD+QA− recombination, pointed to the donor side modifications in ΔPsbR. EPR measurements revealed that S1-to-S2-transition and S2-state multiline signal were not affected by mutation. The fast phase of the FF-relaxation in the absence of DCMU was significantly slowed down with concomitant decrease in the relative amplitude of this phase, indicating a modification in QA to QB electron transfer in ΔPsbR thylakoids. It is concluded that the lack of the PsbR protein modifies both the donor and the acceptor side of the PSII complex. 相似文献
