Colonies of Trioxys pallidus Haliday (Hymenoptera: Aphidiidae) were collected from California walnut orchards during 1985–86 and screened for variability of responses to azinphosmethyl. Variability was found and laboratory selection was initiated with four colonies. All colonies responded to selection; after seven to 12 selections, three colonies were combined (Select colony) and selection was conducted an additional 27 times. The corresponding base colonies were also combined and maintained for comparisons (Base). Concentration/mortality lines obtained for the Select and Base colonies after five and 27 selections indicated seven‐ and 9.2‐fold differences in LC50 values respectively. Survival of the resistant strain varied with bioassay method and decreased in the order: clip cages on treated foliage > treated plastic cups with untreated mesh tops > treated plastic cups with treated mesh tops> treated cups with treated solid lids. The initial bioassay results with treated plastic cups suggested that the selection response was inadequate to allow the resistant strain to survive field rats of azinphosmethyl. When the resistant strain's survival was evaluated with bioassay methods more closely mimicking field conditions (clip cages on field‐treated foliage), we concluded the selected strain could survive field rates of azinphosmethyl.  相似文献

A systematic program of genetic improvement was initiated by assessing the phenotypic variation of Steinernema feltiae strains for two traits assumed to limit efficacy: ultraviolet tolerance and host-finding ability. All of the strains assayed showed both low ultraviolet tolerance and poor host-finding ability, indicating that the likelihood of improving these traits through more extensive population sampling is remote. Limited genetic variation was detected among the strains for tolerance to ultraviolet, suggesting that selective breeding for increased tolerance would be inefficient. By contrast, highly significant phenotypic differences were found with regard to host-finding ability, suggesting that this trait would be responsive to selection. A genetically heterogeneous population was constructed by round-robin mating of 10 strains; it will serve as the foundation population for selective breeding.  相似文献

Polyploidy events (polyploidization) followed by progressive loss of redundant genome components are a major feature of plant evolution, with new evidence suggesting that all flowering plants possess ancestral genome duplications. Furthermore, many of our most important crop plants have undergone additional, relatively recent, genome duplication events. Recent advances in DNA sequencing have made vast amounts of new genomic data available for many plants, including a range of important crop species with highly duplicated genomes. Along with assisting traditional forward genetics approaches to study gene function, this wealth of new sequence data facilitates extensive reverse genetics-based functional analyses. However, plants featuring high levels of genome duplication as a result of recent polyploidization pose additional challenges for reverse genetic analysis. Here we review reverse genetic analysis in such polyploid plants and highlight key challenges.  相似文献

The effects of fosfomycin on penicillin-binding proteins (PBPs) were studied on the methicillin-resistant Staphylococcus aureus strain CIP (Collection de l'Institut Pasteur, Paris, France) 65-25 and on a methicillin-susceptible S. aureus strain CIP 65-6. The combinations of fosfomycin and oxacillin were synergistic, additive or antagonistic, depending on antibiotic concentrations. Fosfomycin induced modifications of the PBP profile of the two strains studied. In particular, it increased the expression of PBP2. This suggested that this protein is inducible; the only PBP not affected by fosfomycin was PBP3.  相似文献

The use of prerecorded data to remotely assess the herd welfare status is a promising approach to reduce the need for costly and time-consuming on-farm welfare assessments. Therefore, the objective of this study was to validate the Herd Status Index, an index developed based on Dairy Herd Improvement data from Canada, to remotely evaluate the welfare status of dairy herds. Herd-level prevalence of five animal-based welfare outcomes, measured once on 2 986 Quebec – Canada dairy herds between 2016 and 2019, were used to generate clusters with different welfare status using the algorithm partitioning around medoids. Dairy Herd Improvement data from 12 months prior to the welfare assessment were extracted and used to calculate the Herd Status Index. A linear model was used to carry out comparisons between clusters. Three stable clusters were found to best describe the data. Cluster two had the best overall welfare status since it had the lowest prevalence of all welfare issues while cluster three had the highest prevalence of most welfare issues, with the exception for the prevalence of neck lesions that was not different than cluster one. Cluster one had an overall intermediate welfare status. The Herd Status Index was higher (i.e., indicating a good welfare status) on cluster two compared to cluster three, but neither cluster three nor two differed to cluster one. In its current format, the Herd Status Index has a weak potential to identify herds with varying prevalence of welfare issues and it requires further improvements before it could be used to accurately assess the welfare status of the herds.  相似文献

Improving traits in wheat has historically been challenging due to its large and polyploid genome, limited genetic diversity and in‐field phenotyping constraints. However, within recent years many of these barriers have been lowered. The availability of a chromosome‐level assembly of the wheat genome now facilitates a step‐change in wheat genetics and provides a common platform for resources, including variation data, gene expression data and genetic markers. The development of sequenced mutant populations and gene‐editing techniques now enables the rapid assessment of gene function in wheat directly. The ability to alter gene function in a targeted manner will unmask the effects of homoeolog redundancy and allow the hidden potential of this polyploid genome to be discovered. New techniques to identify and exploit the genetic diversity within wheat wild relatives now enable wheat breeders to take advantage of these additional sources of variation to address challenges facing food production. Finally, advances in phenomics have unlocked rapid screening of populations for many traits of interest both in greenhouses and in the field. Looking forwards, integrating diverse data types, including genomic, epigenetic and phenomics data, will take advantage of big data approaches including machine learning to understand trait biology in wheat in unprecedented detail.  相似文献

Fragmentation of the Yellowstone River is hypothesized to preclude recruitment of endangered Scaphirhynchus albus (pallid sturgeon) by impeding upstream spawning migrations and access to upstream spawning areas, thereby limiting the length of free‐flowing river required for survival of early life stages. Building on this hypothesis, the reach of the Yellowstone River affected by Intake Diversion Dam (IDD) is targeted for modification. Structures including a rock ramp and by‐pass channel have been proposed as restoration alternatives to facilitate passage. Limited information on migrations and swimming capabilities of pallid sturgeon is available to guide engineering design specifications for the proposed structures. Migration behavior, pathways (channel routes used during migrations), and swimming capabilities of free‐ranging wild adult pallid sturgeon were examined using radiotelemetry, and complemented with hydraulic data obtained along the migration pathways. Migrations of 12–26% of the telemetered pallid sturgeon population persisted to IDD, but upstream passage over the dam was not detected. Observed migration pathways occurred primarily through main channel habitats; however, migrations through side channels up to 3.9 km in length were documented. The majority of pallid sturgeon used depths of 2.2–3.4 m and mean water velocities of 0.89–1.83 m/s while migrating. Results provide inferences on depths, velocities, and habitat heterogeneity of reaches successfully negotiated by pallid sturgeon that may be used to guide designs for structures facilitating passage at IDD. Passage will provide connectivity to potential upstream spawning areas on the Yellowstone River, thereby increasing the likelihood of recruitment for this endangered species.  相似文献

石膏可以改良盐碱土,室内土柱淋洗试验是验证其改良效果的方法之一。本文将石膏计划改良层定为40cm,计算的定量石膏与0~10cm土层混匀后,装填0~40cm土柱于室内进行分次淋洗试验,将收集的滤液测定溶液离子组成并换算成盐分,应用推理计算法分析石膏溶解、转化、渗漏情况,以及石膏与碱性盐及交换性钠镁作用量。结果表明:将石膏与表层0~10cm土层混匀条件下,4次灌水后,0~40cm的土层物理通透性改善,渗透时间变小,pH由9.25降到8.20,碱化度均基本达到10,而且还有残余石膏,应用推理计算法得出,最经济纯石膏理论改良用量为0.05t·hm-2。  相似文献
