Blueberry replant disease (BRD) is an emerging threat to continued blueberry (Vaccinium spp.) production in Georgia and North Carolina. Since high populations of ring nematode Mesocriconema ornatum were found to be associated with commercially grown blueberries in Georgia, we hypothesized that M. ornatum may be responsible for predisposing blueberry to BRD. We therefore tested the pathogenicity of M. ornatum on 10-wk-old Rabbiteye blueberries (Vaccinium virgatum) by inoculating with initial populations (Pi) of 0 (water control), 10, 100, 1,000. and 10,000 mixed stages of M. ornatum/pot under both greenhouse (25 ± 2°C) and field microplot conditions. Nematode soil population densities and reproduction rates were assessed 75, 150, 225, and 255, and 75, 150, 225, and 375 d after inoculation (DAI) in both the greenhouse and field experiments, respectively. Plant growth parameters were recorded in the greenhouse and field microplot experiments at 255 and 375 DAI, respectively. The highest M. ornatum population density occurred with the highest Pi level, at 75 and 150 DAI under both greenhouse (P < 0.01) and field (P < 0.01) conditions. However, M. ornatum rate of reproduction increased significantly in pots receiving the lowest Pi level of 10 nematodes/plant compared with the pots receiving Pi levels of 100, 1,000, and 10,000 nematodes 75 DAI. Plant-parasitic nematode populations were determined in commercial blueberry replant sites in Georgia and North Carolina during the 2010 growing season. Mesocriconema ornatum and Dolichodorus spp. were the predominant plant-parasitic nematodes in Georgia and North Carolina, respectively, with M. ornatum occurring in nearly half the blueberry fields sampled in Georgia. Other nematode genera detected in both states included Tylenchorhynchus spp., Hoplolaimus spp., Hemicycliophora spp., and Xiphinema spp. Paratrichodorus spp. was also found only in Georgia. In Georgia, our results indicate that blueberry is a host for M. ornatum and its relationship to BRD warrants further investigation.  相似文献

A model octapeptide segment derived from vasoactive intestinal peptide (VIP) was utilised to investigate the effect of several conventional cyclisation methods on the α‐helical conformation in short peptide fragments. Three of the classical macrocyclisation techniques (i.e. lactamisation, ring‐closing metathesis and Huisgen cycloaddition) were applied, and the conformations of the resulting cyclic peptides, as well as their linear precursors, were compared by CD analysis. The visibly higher folding propensity of the triazole‐tethered peptide after azide‐alkyne CuAAC macrocyclisation illustrates that the secondary structure of a short peptide fragment can differ significantly depending on the chemical strategy used to covalently cross‐link side chain residues in a ‘helical’ fragment. Forests around the world are subject to risk of high rates of tree growth decline and increased tree mortality from combinations of climate warming and drought, notably in semi‐arid settings. Here, we assess how climate warming has affected tree growth in one of the world's most extensive zones of semi‐arid forests, in Inner Asia, a region where lack of data limits our understanding of how climate change may impact forests. We show that pervasive tree growth declines since 1994 in Inner Asia have been confined to semi‐arid forests, where growing season water stress has been rising due to warming‐induced increases in atmospheric moisture demand. A causal link between increasing drought and declining growth at semi‐arid sites is corroborated by correlation analyses comparing annual climate data to records of tree‐ring widths. These ring‐width records tend to be substantially more sensitive to drought variability at semi‐arid sites than at semi‐humid sites. Fire occurrence and insect/pathogen attacks have increased in tandem with the most recent (2007–2009) documented episode of tree mortality. If warming in Inner Asia continues, further increases in forest stress and tree mortality could be expected, potentially driving the eventual regional loss of current semi‐arid forests.  相似文献

We present prenatal diagnosis and molecular cytogenetic characterization of de novo mosaic r(13). A 32-year-old woman underwent amniocentesis at 18 weeks of gestation because of maternal anxiety. Amniocentesis revealed a karyotype of 46,XY,r(13)[33]/45,XY,-13[19]. aCGH on uncultured amniocytes at repeated amniocentesis detected a 4.22-Mb deletion at 13q34. Interphase FISH on 100 uncultured amniocytes showed the ratio of r(13):-13:idic r(13) as 85%:13%:2%. The cord blood had a karyotype of 46,XY,r(13)[91]/46,XY,idic r(13)[6]/45,XY,-13[3]. The placenta had a karyotype of 46,XY,mar(13)[31]/45,XY,-13[3]. Metaphase FISH confirmed that the marker chromosomes in placenta were derived from chromosome 13. aCGH on cultured placental cells detected a 77.81-Mb deletion at 13q13.3–q34. The fetus postnatally manifested facial dysmorphism. Prenatal diagnosis of r(13) should alert mosaicism for deletion/duplication of r(13) and distal 13q deletion. Fetoplacental chromosomal discrepancy of r(13) may exist in case of mosaic r(13) detected by amniocentesis.  相似文献

We present perinatal findings and molecular cytogenetic characterization of a prenatally detected sacrococcygeal teratoma associated with mosaic r(21). This is the first report of mosaic r(21) presenting with a fetal sacrococcygeal teratoma. We discuss cytogenetic abnormalities associated with fetal sacrococcygeal teratomas.  相似文献

利用树木年轮环境学分析方法,测定了伊春西林铅锌矿附近兴安落叶松年轮中Pb、Cd、Zn、Cu、Mn 5种重金属元素含量的变化.结果表明:在矿区附近的兴安落叶松年轮中,Mn含量最高,Cd含量最低.近地面处年轮中的Cd、Zn、Cu含量显著高于胸高处,而近地面和胸高处的Pb和Mn含量差异不显著.1987-2010年,除Pb含量有轻微上升外,其他4种元素含量均呈下降趋势,其中Cd含量下降趋势最明显,Zn、Cu、Mn含量均先下降后上升.随着兴安落叶松年轮宽度的增加,Pb含量下降,其他4种元素含量均增加.近地面处Pb含量与其他4种元素含量呈显著正相关,而在胸高处的相关性不显著,Cd含量甚至随Pb含量增加而显著下降.兴安落叶松年轮中重金属元素含量变化受铅锌矿产量和采矿活动的影响,可利用年轮中Pb含量重建采矿历史.目前,Pb含量已经对周围环境造成污染,如果继续开采,应加强对尾矿中重金属元素的治理.  相似文献

Over the past 13 years, glyoxal has become the leading alternative to formaldehyde as a histological fixative because of its low inhalation risk, faster reaction rate and selective control over crosslinking. The latter attribute is especially important, because most of the difficulties relating to use of formaldehyde-fixed specimens for immunohistochemistry stem from its aggressive crosslinking behavior. With suitable catalysts or other reaction accelerators, glyoxal forms 2-carbon adducts with nearly all end groups in proteins and carbohydrates, leaving most of them unimpaired for subsequent immunohistochemical demonstration. Only arginine is seriously impaired by the formation of imidazoles, which is the basis for the well known arginine blockade method using glyoxal. A special glyoxal-specific antigen retrieval method using high pH and high temperature effectively reverses the blockade and restores immunoreactivity. Other methods for antigen retrieval are rarely beneficial and in most cases damage the specimen. Special stains work well, except silver methods for Helicobacter pylori. Routine hematoxylin and eosin preparations exhibit clarity and cellular detail rarely seen with formaldehyde.  相似文献

Abstract

Vortex structures, as one of the most important features of cardiac flow, have a crucial impact on the left ventricle function and pathological conditions. These swirling flows are closely related to the presence of turbulence in left ventricle which is investigated in the current study. Using an extended model of the left heart, including a fluid-structure interaction (FSI) model of the mitral valve with a realistic geometry, the effect of using two numerical turbulent models, k-ε and Spalart-Allmaras (SA), on diastolic flow patterns is studied and compared with results from laminar flow model. As a result of the higher dissipation rate in turbulent models (k-ε and SA), vortices are larger and stronger in the laminar flow model. Comparing E/A ratio in the three models (Laminar, k-ε, and SA) with experimental data from healthy subjects, it is concluded that the results from k-ε model are more accurate.  相似文献

In eukaryotes, abnormally circularized chromosomes, known as ‘rings,’ can be mitotically unstable. Some rings derived from a compound X-Y chromosome induce mitotic abnormalities during the embryonic cleavage divisions and early death in Drosophila melanogaster, but the underlying basis is poorly understood. We recently demonstrated that a large region of 359-bp satellite DNA, which normally resides on the X chromosome, prevents sister ring chromatids from segregating properly during these divisions. Cytogenetic comparisons among 3 different X-Y rings with varying levels of lethality showed that all 3 contain similar amounts of 359-bp DNA, but the repetitive sequences surrounding the 359-bp DNA differ in each case. This finding suggests that ring misbehavior results from novel heterochromatin position effects on the 359-bp satellite. The purpose of this view is to explore possible explanations for these effects with regard to heterochromatin formation and replication of repetitive sequences. Also discussed are similarities of this system to a satellite-based hybrid incompatibility and potential influences on genome evolution.  相似文献

Mice lacking the E3 ubiquitin ligase mahogunin ring finger‐1 (MGRN1) have a pleiotropic phenotype that includes spongiform neurodegeneration, embryonic patterning defects, and dark fur due to a defect in pigment‐type switching. The only MGRN1 ubiquitination target identified to date is tumor susceptibility gene 101 (TSG101), a component of the endosomal trafficking machinery. Here, we show that MGRN1 also interacts with but does not ubiquitinate NEDD4, a HECT‐domain ubiquitin ligase involved in endosomal trafficking. Using transgenesis in mice, we demonstrate that pigment‐type switching likely requires MGRN1′s ubiquitin ligase activity but not its ability to bind TSG101 or NEDD4. This indicates that MGRN1‐dependent ubiquitination of an as‐yet unidentified target protein is required for agouti‐mediated melanocortin signaling.  相似文献
