Diet studies are fundamental for understanding trophic connections in marine ecosystems. In the southeastern US, the common bottlenose dolphin Tursiops truncatus is the predominant marine mammal in coastal waters, but its role as a top predator has received little attention. Diet studies of piscivorous predators, like bottlenose dolphins, start with assessing prey otoliths recovered from stomachs or feces, but digestive erosion hampers species identification and underestimates fish weight (FW). To compensate, FW is often estimated from the least affected otoliths and scaled to other otoliths, which also introduces bias. The sulcus, an otolith surface feature, has a species‐specific shape of its ostium and caudal extents, which is within the otolith edge for some species. We explored whether the sulcus could improve species identification and estimation of prey size using a case study of four sciaenid species targeted by fisheries and bottlenose dolphins in North Carolina. Methods were assessed first on otoliths from a reference collection (n = 421) and applied to prey otoliths (n = 5,308) recovered from 120 stomachs of dead stranded dolphins. We demonstrated in reference‐collection otoliths that cauda to sulcus length (CL:SL) could discriminate between spotted seatrout (Cynoscion nebulosus) and weakfish (Cynoscion regalis) (classification accuracy = 0.98). This method confirmed for the first time predation of spotted seatrout by bottlenose dolphins in North Carolina. Using predictive models developed from reference‐collection otoliths, we provided evidence that digestion affects otolith length more than sulcus or cauda length, making the latter better predictors. Lastly, we explored scenarios of calculating total consumed biomass across degrees of digestion. A suggested approach was for the least digested otoliths to be scaled to other otoliths iteratively from within the same stomach, month, or season as samples allow. Using the otolith sulcus helped overcome challenges of species identification and fish size estimation, indicating their potential use in other diet studies.  相似文献

Because of the high risk of going unnoticed, cryptic species represent a major challenge to biodiversity assessments, and this is particularly true for taxa that include many such species, for example, bats. Long‐eared bats from the genus Plecotus comprise numerous cryptic species occurring in the Mediterranean Region and present complex phylogenetic relationships and often unclear distributions, particularly at the edge of their known ranges and on islands. Here, we combine Species Distribution Models (SDMs), field surveys and molecular analyses to shed light on the presence of a cryptic long‐eared bat species from North Africa, Plecotus gaisleri, on the islands of the Sicily Channel, providing strong evidence that this species also occurs in Europe, at least on the islands of the Western Mediterranean Sea that act as a crossroad between the Old Continent and Africa. Species Distribution Models built using African records of P. gaisleri and projected to the Sicily Channel Islands showed that all these islands are potentially suitable for the species. Molecular identification of Plecotus captured on Pantelleria, and recent data from Malta and Gozo, confirmed the species' presence on two of the islands in question. Besides confirming that P. gaisleri occurs on Pantelleria, haplotype network reconstructions highlighted moderate structuring between insular and continental populations of this species. Our results remark the role of Italy as a bat diversity hotspot in the Mediterranean and also highlight the need to include P. gaisleri in European faunal checklists and conservation directives, confirming the usefulness of combining different approaches to explore the presence of cryptic species outside their known ranges—a fundamental step to informing conservation.  相似文献

恰玛古(Qamgur, Brassica rapa L.)内生菌的研究主要集中在内生真菌,内生放线菌的研究报道较少。通过研究新疆药食两用植物恰玛古内生放线菌多样性,以期发现产新活性物质的放线菌或新种放线菌,为研究微生物药物奠定基础。从恰玛古根、茎和叶三个部位分离培养获得内生放线菌,对其菌落与个体形态进行观察,并利用序列测定方法进行鉴定,以获取其分类地位。从恰玛古三个部位共分离得到17株内生放线菌,其中12株为革兰氏阳性杆菌,3株为革兰氏阳性球菌,2株为革兰氏阳性丝状菌;17株内生放线菌分属于红球菌属(Rhodococcus)、拟诺卡氏菌属(Nocardiopsis)、链霉菌属(Streptomyces)、短杆菌属(Brevibacterium)、小短杆菌属(Brachybacterium)、两面神菌属(Janibacter)和微杆菌属(Microbacterium)。从新疆药食两用植物恰玛古中分离获得17株内生放线菌以稀有放线菌为主。  相似文献

The purpose of this study was to detail a simple strategy for sexing morphologically-undifferentiated fish using statistical analysis of gene expression patterns characterized by quantitative PCR. This approach is especially relevant for species without known genomic sex markers. The method was developed for early identification of female Siberian sturgeon as part of a genomics study. That study documented activation of the enzyme 17ß-hydroxy-steroid-dehydrogenase (hsd17b1) in future ovarian tissue at 3 months of age, concurrent with a small forkhead box L2 (foxl2) peak and emerging cytochrome P450, family 19, subfamily A (cyp19a1) expression. Major cyp19a1 and foxl2 peaks occurred in presumptive female gonads at 5–6 months. This pattern suggested a genetic relay mediating estrogen production throughout differentiation, possibly to maintain gonadal femininity. Genes involved in stem cell proliferation (lim homeobox 2 (lhx2)) and somatic-germ cell interaction maintenance (iroquois homeobox 5 (irx5) and iroquois homeobox 3 (irx3)) were also expressed during molecular differentiation, at 5–6 months. The roles of lhx2, irx3, and irx5 in fish sex differentiation should be confirmed using other methodologies. These results indicate that estrogens are crucial for ovarian differentiation in basal non-teleost fish, consistent with well-established patterns in teleosts, with hsd17b1 as one of the earliest biomarkers of gonadal development.  相似文献

Natural history collections are repositories of biodiversity and are potentially used by molecular ecologists for comparative taxonomic, phylogenetic, biogeographic and forensic purposes. Specimens in fish collections are preserved using a combination of methods with many fixed in formalin and then preserved in ethanol for long-term storage. Formalin fixation damages DNA, thereby limiting genetic analyses. In this study, the authors compared the DNA barcoding and identification success for frozen and formalin-fixed tissues obtained from specimens in the CSIRO Australian National Fish Collection. They studied 230 samples from fishes (consisting of >160 fish species). An optimized formalin-fixed, paraffin-embedded DNA extraction method resulted in usable DNA from degraded tissues. Four mini barcoding assays of the mitochondrial DNA (mtDNA) were characterized with Sanger and Illumina amplicon sequencing. In the good quality DNA (without exposure to formalin), up to 88% of the specimens were correctly matched at the species level using the cytochrome oxidase subunit 1 (COI) mini barcodes, whereas up to 58% of the specimens exposed to formalin for less than 8 weeks were correctly identified to species. In contrast, 16S primers provided higher amplification success with formalin-exposed tissues, although the COI gene was more successful for identification. Importantly, the authors found that DNA of a certain size and quality can be amplified and sequenced despite exposure to formalin, and Illumina sequencing provided them with greater power of resolution for taxa identification even when there was little DNA present. Overall, within parameter constraints, this study highlights the possibilities of recovering DNA barcodes for identification from formalin-fixed fish specimens, and the authors provide guidelines for when successful identification could be expected.  相似文献

Otolith shape analysis is a powerful method for fish stock identification. We compared the otolith shape of Pagrus pagrus (Linnaeus 1758) along with its distribution in four south-western Atlantic regions where it is commercially fished: Rio de Janeiro, Rio Grande do Sul in southern Brazil, the Argentine-Uruguayan Common Fishing Zone (UA) and the Argentinian Exclusive Fishing Zone (AR). Otolith shapes were compared by Elliptical Fourier and Wavelet coefficients among specimens in a size range with similar otoliths, morphometric parameters and ages. Four potential stocks were identified: one in the AR, a second along the UA which included specimens from southern Brazil with well-marked opaque bands in its otoliths (MRS), the third in southern Brazil with faint or absent opaque bands in its otoliths (FRS) and the fourth along Rio de Janeiro. The difference in the otolith shape among regions followed differences reported using other stock identification techniques. The similarity between otoliths from UA and MRS (ANOVA-like, P > 0.01) can be explained by seasonal short-range migrations. Otoliths shape differences between MRS and FRS (ANOVA-like, P < 0.01) suggest that P. pagrus does not form a homogeneous group in southern Brazil.  相似文献

为了寻找丽江乌头中结构新颖的化合物,丰富丽江乌头的化学成分多样性,本文对丽江乌头(Aconitum forrestii Stapf)根部的化学成分进行研究,采用硅胶柱层析从其乙醇提取物中分离得到14个化合物。通过HR-ESI-MS、1D和2D NMR等波谱技术鉴定了它们的结构,其中化合物1是一个新的乌头碱型二萜生物碱,命名为8-O-methyl-14-O-anisoylchasmanine(1)。其余13个化合物均为已知化合物:14-acetoxy-8-O-methylsachaconitine(2)、14-acetyltalatizamine(3)、talatisamine(4)、chasmanine(5)、ezochasmanine(6)、crassicautine(7)、geniculatine C(8)、cammaconine(9)、vilmoraconitine(10)、aconitramine A(11)、vilmorrianine G(12)、hemsleyaconitine G(13)和heterophylloidine(14)。测试了所有化合物对阿霉素诱导的H9c2心肌细胞损伤的保护活性,结果显示化合物3、10、11和12表现出一定的保护作用。  相似文献

This paper examines the impact of overweight family members on weight outcomes of Italian children aged 6–14 years. We use an original dataset matching the 2012 cross sections of the Italian Multipurpose Household Survey and the Household Budget Survey. Since the identification of within-family peer effects is known to be challenging, we implement our analysis on a partially identified model using inferential procedures recently introduced in the literature and based on standard Bayesian computation methods. We find evidence of a strong, positive effect of both overweight peer children in the family and of overweight adults on children weight outcomes. The impact of overweight peer children in the household is larger than the impact of adults. In particular, the estimated confidence sets associated to the peer children variable is positive with upper bound around one or larger, while the confidence sets for the parameter associated to obese adults often include zero and have upper bound that rarely is larger than one.  相似文献

目的以兰州兴隆山不同区域的土壤微生物为研究对象,分析比较土壤微生物数量与土壤酶活性之间的相关性。方法利用不同方法测定土壤理化性质、微生物数量以及土壤相关酶活特性;采用三区划线法进行土壤微生物的分离与纯化,通过16S rDNA和ITS方法进行优势菌株鉴定。结果兰州兴隆山土壤中微生物菌群数量由多到少依次为细菌、放线菌、真菌。通过分离纯化后,对其中的2株优势菌进行了鉴定,初步推断X2为萎缩芽胞杆菌属(Bacillus atrophaeus)细菌,Z2为栎生青霉属(Penicillium glandicola)真菌。从酶活特性可知,阳面的土壤过氧化氢酶活性比阴面高;随着海拔高度的增加,过氧化氢酶活性呈现增加趋势;阳面的土壤脱氢酶活性总体比阴面高,并且随着海拔梯度的升高,土壤脱氢酶活性也在不断升高。从相关性分析可知,不同海拔土样间微生物数量与酶活性之间表现出明显的相关性。结论兰州兴隆山土壤微生物数量丰富,且细菌数量居多;不同阴、阳面土壤微生物的层次分布以及活性也各有不同。以上研究可为兰州兴隆山土壤生态系统演替等提供参考依据,并为土壤生态环境的治理做铺垫。  相似文献

Bats are a group of mammals well known for forming dynamic social groups. Studies of bat social structures are often based upon the frequency at which bats occupy the same roosts because observing bats directly is not always possible. However, it is not always clear how closely bats occupying the same roost associate with each other, obscuring whether associations result from social relationships or factors such as shared preferences for roosts. Our goal was to determine if bats cohabitating buildings were also found together inside roosts by using anti‐collision technology for PIT tags, which enables simultaneous detection of multiple tags. We PIT‐tagged 293 female little brown myotis (Myotis lucifugus) and installed antennas within two buildings used as maternity roosts in Yellowstone National Park. Antennas were positioned at roost entryways to generate cohabitation networks and along regions of attic ceilings in each building to generate intraroost networks based on proximity of bats to each other. We found that intraroost and cohabitation networks of buildings were significantly correlated, with the same bats tending to be linked in both networks, but that bats cohabitating the same building often roosted apart, leading to differing assessments of social structure. Cohabitation rates implied that bats associate with a greater number of their roost‐mates than was supported by observations within the roost. This caused social networks built upon roost cohabitation rates to be denser, smaller in diameter, and contain nodes with higher average degree centrality. These results show that roost cohabitation does not reflect preference for roost‐mates in little brown myotis, as is often inferred from similar studies, and that social network analyses based on cohabitation may provide misleading results.  相似文献
