The protochlorophyll(ide) forms and plastid ultrastructure were investigated in hypocotyls of dark-grown seedlings of kidney bean ( Phaseolus vulgaris L. cv. Brede zonder draad). By deconvolution of the fluorescence emission spectra into Gaussian components three protochlorophyll(ide) forms were found with maxima at 633, 642 and 657 nm, respectively. The ratio of protochlorophyll(ide) emitting at 657 nm to protochlorophyll(ide) emitting at 633 nm decreased downwards the hypocotyl. The gradient was established already after 4 days in dark-grown Phaseolus and was also seen in hypocotyls of 7-day-old dark-grown plants of 8 other species. Ultrastructural observations revealed a plastid developmental sequence along the hypocotyl. Plastids in the upper parts of the hypocotyl contained prolamellar bodies typical of etiolated leaves while those in the lower parts contained only stroma lamellae. Immunological detection of NADPH-protochlorophyllide oxidoreductase (EC 1.3.1.33) on nitrocellulose membranes after sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDSPAGE) indicated the occurrence of the enzyme in upper, middle and lower sections of hypocotyls and in the root tips. 相似文献

Circadian clocks are endogenous auto-regulatory mechanisms that allow organisms, from bacteria to humans, to advantageously time a wide range of activities within 24 h environmental cycles. Here we report the identification and characterization of an MYB-related gene, designated Circadian 1 (CIR1), that is involved in circadian regulation in Arabidopsis. Expression of CIR1 is transiently induced by light and oscillates with a circadian rhythm. The rhythmic expression of CIR1 is controlled by the central oscillator. Constitutive expression of CIR1 resulted in a shorter period length for the rhythms of four central oscillator components, and much lower amplitude for the rhythms of central oscillator components CCA1 and LHY. Furthermore, CIR1 over-expression severely affected the circadian rhythms of its own RNA and those of the slave oscillator EPR1 and effector genes Lhcb and CAT3. Plants that constitutively expressed CIR1 displayed delayed flowering, longer hypocotyls and reduced seed germination in the dark. These results suggest that CIR1 is possibly part of a regulatory feedback loop that controls a subset of the circadian outputs and modulates the central oscillator. 相似文献

Hypocotyl-derived callus from the Helianthus annuus L. inbred line SS415B regenerated significantly more plants if the seedlings were grown in the light. The difference between light- and dark-grown seedlings was not correlated with differences in seedling ethylene production, but seemed to be due to a difference in sensitivity to ethylene at a specific time during seedling growth. Treating 3-day-old dark-grown seedlings with 10 μ M aminoethoxyvinylglycine (AVG) effectively inhibited ethylene production for at least 7 days. Hypocotyl callus derived from AVG-treated seedlings gave the same amount of regeneration as callus from light-grown seedlings. Promotion of regeneration by AVG was not seen unless the 3-day-old seedlings were grown for 4 additional days prior to culturing hypocotyl explants. The effects of AVG could be reversed by treatment with 1-aminocyclopropane-1-carboxylic acid (ACC) during these 4 days. After the 4 days, ACC was no longer effective. 相似文献

Various explants of salad burnet (Poterium sanguisorba L.=Sanguisorba minor Scop.) were cultured on semi-solid MS or B5 media containing factorial combinations of various plant growth regulators. Hypocotyl
and petiole explants, after initial callus stage, regenerated prolific adventitious shoots via organogenesis, when placed
on Murashige and Skoog basal medium containing 1–2 μM α-naphthaleneacetic acid and 4-20 μM thidiazuron. Any other growth regulator
combination tested failed to respond. The addition of the biocide, Plant Preservative Mixture, was effective in controlling
contamination and did not impair regeneration. Elongated shoots at 2–4 cm were transferred to rooting medium containing semi-solid
Murashige and Skoog plus 1 μM α-naphthaleneacetic and rooted plants were transferred to the glasshouse. This is the first
report on in vitro plant regeneration within the genusPoterium.

The activities of two mono- and two di-fluorogibberellins are compared with the equivalent unfluorinated compounds in the barley half-seed, lettuce hypocotyl and Tanginbozu dwarf rice bioassays. Interactions between the substituted and unsubstituted gibberellins are also examined. It is concluded that the effects of fluorination are dependent upon the site and degree of substitution and vary with the type of bioassay tissue involved. In the interaction studies the fluorogibberellins generally proved inhibitory, and competitive inhibition was indicated. 相似文献
