The inhibitory effect of a series of analogs of CPTA, 2-(4-chlorophenylthio)-triethylamine-HCl, and ammonia derivatives on carotenoid biosynthesis in Phycomyces blakesleeanus mutants was studied. The types of inhibition exhibited allowed no firm conclusions about the biosynthetic route to β-carotene from either β-zeacarotene or lycopene. However, the evidence suggests at present that both pathways are operative. It was found that a slight change in structure of inhibitor resulted in a different type of action. Conclusions based on a single inhibitor could be cited as “evidence” for a certain pathway. 相似文献

Abstract Several approaches have been followed to increase the nitrogenase level in filaments of Anabaena ATCC33047. In a nitrogen-free medium lacking added molybdate and supplemented with 10 mM tungstate, growth was impaired as a result of decreased nitrogenase activity level. Under these conditions, the filaments exhibited nitrogen starvation symptoms and a high heterocyst frequency, with heterocysts being up to 28% of the total number of cells in the filaments, while a regular pattern of heterocyst distribution was maintained. Normal nitrogenase level and nitrogen status were recovered upon molybdate addition, with resumption of growth and decrease of the heterocyst frequency with time until reaching a value of about 10%. The yield of ammonium photoproduction from N2 by filaments displaying different heterocyst frequencies and treated with l -methionine- d,l -sulfoximine (MSX) was determined. Maximal rates were obtained with filaments containing 16% of the cells differentiated as heterocysts. Results indicate that appropriate manipulation of the heterocyst frequency leads to an improvement in the efficiency of conversion of light energy into chemical energy through photoreduction of N2 to ammonium. 相似文献

Two of nine sulfate reducing bacteria tested,Desulfobulbus propionicus andDesulfovibrio desulfuricans (strain Essex 6), were able to grow with nitrate as terminal electron acceptor, which was reduced to ammonia.
Desulfovibrio desulfuricans was grown in chemostat culture with hydrogen plus limiting concentrations of nitrate, nitrite or sulfate as sole energy source. Growth yields up to 13.1, 8.8 or 9.7 g cell dry mass were obtained per mol nitrate, nitrite or sulfate reduced, respectively. The apparent half saturation constants (K
s) were below the detection limits of 200, 3 or 100 mol/l for nitrate, nitrite of sulfate, respectively. The maximum growth rates {ie63-1} raised from 0.124 h-1 with sulfate and 0.150 h-1 with nitrate to 0.193 h-1 with nitrite as electron acceptor. Regardless of the electron acceptor in the culture medium, cell extracts exhibited absorption maxima corresponding to cytochromec and desulfoviridin. Nitrate reductase was found to be inducible by nitrate or nitrite, whereas nitrite reductase was synthesized constitutively. The activities of nitrate and nitrite reductases with hydrogen as electron donor were 0.2 and 0.3 mol/min·mg protein, respectively. If limiting amounts of hydrogen were added to culture bottles with nitrate as electron acceptor, part of the nitrate was only reduced to the level of nitrite. In media containing nitrate plus sulfate or nitrite plus sulfate, sulfate reduction was suppressed.The results demonstrate that the ammonification of nitrate or nitrite can function as sole energy conserving process in some sulfate-reducing bacteria. 相似文献

Mutant 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductases of the halophilic archaeon Haloferax volcanii were constructed to test the proposed mechanism that phosphorylation downregulates the activity of higher eukarya HMG-CoA reductases via charge-charge interaction with the active site histidine. To first verify the sequence-based inference that His 398 is the catalytic histidine of the H. volcanii enzyme, enzyme H398Q was constructed, purified, and assayed for catalysis of three reactions: [1] reductive deacylation of HMG-CoA, [2] reduction of mevaldehyde, and [3] oxidative acylation of mevaldehyde. Enzyme H398Q had low activity for catalysis of reaction [1] or [3], but readily catalyzed mevaldehyde reduction. By analogy to hamster HMG-CoA reductase, we conclude that His 398 is the active site histidine. Mutant forms of the 403-residue H. volcanii enzyme were constructed to model phosphorylation and infer whether attenuated activity involved interaction with His 398. Chimeric H. volcanii-hamster enzymes constructed in an effort to create an active, phosphorylatable chimeric enzyme were inactive or not phosphorylated. We therefore added Asp at position 404 to mimic the introduction of negative charge that would accompany phosphorylation. Enzyme 404D/H398Q was inactive for reaction [1] or [3], but catalyzed reaction [2] at 35% the wild-type rate. These observations are consistent with the model that attenuation of catalytic activity results from an ionic interaction between the imidazolium cation of His 398 and the carboxylate anion of Asp 404. 相似文献

Two experimental high rate algal ponds (HRAPs) (1.5m2, 570 L per unit), each with a secondaryclarifier for algal biomass separation (0.025 m2,without recirculation), were fed with urban wastewaterfor a one-year period (June 1993 to July 1994). TheHRAPs were installed on the roof of the Department ofHydraulic, Coastal and Environmental Engineering ofthe Technical University of Catalonia, Barcelona,Spain (lat. 41° 24 42 N; long. 2° 742 E). Nitrogen removal efficiency and changes intotal nitrogen, total organic nitrogen,NH4
+-N, and oxidized nitrogen underdifferent hydraulic retention times (HRTs) werecompared. HRAP A was always operated at a higherHRT than HRAP B. Both HRAPs were subjected to thesame environmental conditions of solar radiation, airtemperature and influent water quality. Grab samplesof influent, effluent of the HRAP (mixed liquor) andfinal effluent from the clarifiers were taken once aweek. The annual average nitrogen removal was 73% forHRAP A, and 57% for HRAP B. Higher removal in HRAP Awas due to a lower inorganic nitrogen concentration inits effluent. Significant differences (p> 0.05) inthe relative proportions of nitrogen forms between thetwo HRAPs were observed only in autumn and winter.This was mainly because HRAP B did not achieve a highlevel of NH4
+-N removal by stripping andalgal uptake, as observed in HRAP A. NH4
+-Nstripping was the most important mechanism fornitrogen removal (mean efficiency of 47% and 32% inHRAP A and B, respectively) followed by algal uptake,and subsequent algal separation in the clarifiers(mean efficiency of 26% and 25% in HRAP A and Brespectively). The conclusion of this study is thatHRT determines both the nitrogen removal efficiencyand the distribution of nitrogen forms in the effluentof a HRAP. The nitrogen removal level can becontrolled through suitable HRT operating strategies.By operating at a HRT of 4 days in spring and summer,and 10 days in autumn and winter, nitrogenconcentration in the effluent of a HRAP system can bereduced to less than 15 mg L-1 N. 相似文献

采用通气法测定了北方冬小麦/夏玉米轮作体系田间土壤的原位氨挥发。结果表明,与冬小麦施用基肥相比,夏玉米追肥后土壤的氨挥发速率很快升高,但军发高峰期持续时间较短,最大氨挥发速率亦低于冬小麦,冬小麦拨节期追肥,氨挥发速率低且呈波动变化,未出现高峰值,从整个生长季节来看,冬小麦不施氮和每公顷施氮120、240、360kg时的累计挥发量分别为4.4、6.9、13.0、38.4kgN/hm^2,夏玉米为8.4、15.1、20.0、26.1kgN/hm^2。按我国北方冬小麦/夏玉米播种面积1864.4万hm^2计,每年由氨挥发向大气排放的氨素达23.8-120.2万t,其中17.2-96.4万t来自氮肥,相当于氮肥投入的2.1%-9.5%。 相似文献
