The designed antimicrobial peptide KIGAKIKIGAKIKIGAKI possesses enhanced membrane selectivity for bacterial lipids, such as phosphatidylethanolamine and phosphatidylglycerol. The perturbation of the bilayer by the peptide was first monitored using oriented bilayer samples on glass plates. The alignment of POPE/POPG model membranes with respect to the bilayer normal was severely altered at 4 mol% KIGAKI while the alignment of POPC bilayers was retained. The interaction mechanism between the peptide and POPE/POPG bilayers was investigated by carefully comparing three bilayer MLV samples (POPE bilayers, POPG bilayers, and POPE/POPG 4/1 bilayers). KIGAKI induces the formation of an isotropic phase for POPE/POPG bilayers, but only a slight change in the 31P NMR CSA line shape for both POPE and POPG bilayers, indicating the synergistic roles of POPE and POPG lipids in the disruption of the membrane structure by KIGAKI. 2H NMR powder spectra show no reduction of the lipid chain order for both POPG and POPE/POPG bilayers upon peptide incorporation, supporting the evidence that the peptide acts as a surface peptide. 31P longitudinal relaxation studies confirmed that different dynamic changes occurred upon interaction of the peptide with the three different lipid bilayers, indicating that the strong electrostatic interaction between the cationic peptide KIGAKI and anionic POPG lipids is not the only factor in determining the antimicrobial activity. Furthermore, 31P and 2H NMR powder spectra demonstrated a change in membrane characteristics upon mixing of POPE and POPG lipids. The interaction between different lipids, such as POPE and POPG, in the mixed bilayers may provide the molecular basis for the KIGAKI carpet mechanism in the permeation of the membrane.  相似文献

Allergen‐mediated cross‐linking of the high‐affinity receptor for IgE on mast cells triggers the release of diverse preformed and de novo synthesized immunoregulatory mediators that further the allergic response. A proteomic screen applied to the detection of proteins secreted by the model rat mast cell line, RBL‐2H3 (rat basophilic leukaemia, subline 2H3.1), led to the identification of the cholesterol‐binding glycoprotein, NPC2/RE1 (Niemann–Pick Type C2/epididymal secretory protein 1). Glycosylated NPC2 is secreted early in response to an IgE‐mediated stimulus and co‐localizes with the lysosomal membrane marker, CD63. NPC2 belongs to the ML (MD‐2‐related lipid‐recognition) protein family (155 members), which includes the Toll‐like receptor co‐factors, MD‐1 and MD‐2, and perhaps most interestingly, seven major house dust mite allergens of unknown function (including Der p 2 and Der f 2). Possible role(s) for the protein in the allergic response and future applications of this approach are discussed.  相似文献

用蛙胫前肌小束为材料, 研究了提高胞外钾[K+]O对咖啡因挛缩的作用.[K+]O从2 mmol/L提高到10或25 mmol/L, 由3 mmol/L咖啡因引起的挛缩明显增强.以PKC/PC (PKC和PC分别为在高钾和正常钾条件下的咖啡因挛缩)表示的咖啡因挛缩增强, 依赖[K+]O和高钾作用时间.随着10 mmol/L [K+]O作用时间延长, 直至10 min, 增强逐渐增加.但是, 25 mmol/L [K+]O作用1 min时增强达到最大, 然后下降到对照.PKC/PC变化时程不能用高钾引起的去极化解释, 而与由相似[K+]O引起的胞浆自由钙变化时程相符.提示, 至少在蛙骨骼肌, 高钾引起的咖啡因挛缩增强主要是由胞浆自由钙升高引起的.  相似文献

经SPF鸡胚增殖的虎源高致病性禽流感病毒A/Tiger/Harbin/01/2002(H5N1)株,其对MDCK细胞的TCID50为10-7.36/0.05 mL,经气管接种途径人工感染家猫,对感染致死的家猫和对照组家猫心、肝、脾、肺、肾、脑等脏器进行组织病理学观察和免疫组织化学染色,同时采集咽拭子和各脏器乳剂上清液进行RT-PCR检测,对耐过家猫与对照组家猫进行HI抗体测定。结果表明:剖检感染的死亡家猫以肺脏的损害最为明显,肺叶上有大片暗红色实变灶,呈多病灶融合性肺损伤。组织学光镜观察,病毒对家猫的损害主要见于肺脏,呈融合性炎性病变,浸润的主要为单核细胞,肺泡腔内可见较多量巨噬细胞浸润及少量蛋白样浆液渗出。免疫组织化学染色发现在支气管上皮细胞、少数肺泡上皮细胞和单核细胞胞浆中可见到该病毒的抗原阳性染色颗粒。RT-PCR检测结果在咽拭子以及肺、肾、心、脑组织中均扩出与理论值一致的464bp核酸条带,耐过家猫血清H5N1亚型流感病毒HI抗体效价为1∶32。  相似文献

将水培后盆栽的花生幼苗,置于培养箱42℃高温培养,定时测定幼苗叶光合速率、叶绿素含量和叶绿体Ca2 -ATPase、Mg2 -ATPase的相对活性,并观察幼叶细胞内Ca2 分布的变化。试验结果表明:高温胁迫过程中,光合速率及叶绿素含量都随处理时间的延伸而下降,并呈显著正相关;叶绿体Ca2 -ATPase和Mg2 -ATPase高温胁迫过程中相对活性呈先升后降趋势,Ca2 -ATPase热敏性高于Mg2 -ATPase;高温胁迫过程中,Ca2 具有从胞外转运到胞质内和叶绿体中的趋势,Ca2 能够稳定高温胁迫下叶肉细胞膜和叶绿体的超微结构。  相似文献

Objectives: The relationship of plasma adiponectin levels with various anthropometric and metabolic factors has been surveyed extensively in adults. However, how plasma adiponectin levels are related to various anthropometric indices and cardiovascular risk factors in adolescents is not as vigorously studied. In this study, we investigated this among healthy nondiabetic adolescents. Research Methods and Procedures: Two hundred thirty nondiabetic subjects (125 boys and 105 girls, ~10 to 19 years old) were included. The plasma adiponectin, fasting plasma glucose, insulin, lipids and anthropometric indices including body height, weight, waist circumference, and hip circumference were examined. Body fat mass (FM) and percentage were obtained from DXA scan. The homeostasis model assessment was applied to estimate the degree of insulin resistance. Results: The plasma adiponectin levels were significantly higher in girls (30.79 ± 14.48 μg/mL) than boys (22.87 ± 11.41 μg/mL). The plasma adiponectin levels were negatively related to BMI, FM, FM percentage, waist circumference, waist‐to‐hip ratio, insulin resistance, plasma insulin, triglycerides, and uric acid levels, but positively with high‐density lipoprotein cholesterol (HDL‐C) with the adjustment for age and gender. Using different multivariate linear regression models, only age and HDL‐C were consistently related to the plasma adiponectin levels after adjustment for the other variables. Discussion: The relationship between plasma adiponectin and various anthropometric indices and metabolic factors, especially HDL‐C, previously reported in adults was present in the healthy nondiabetic adolescents. Whether variation of plasma adiponectin levels in healthy nondiabetic adolescents may influence their future coronary artery disease risk warrants further investigation.  相似文献

高原致适应剂新复方党参片预防急性高原反应的效果   总被引:1,自引:0,他引:1  
目的:观察高原致适应剂新复方党参片对急性高原反应(AMS)的预防效果。方法:世居平原者驻守海拔1400m3个月的45名青年男性官兵,随机分为新复方党参片组(30人)和对照组(15人),采用单盲试验方法,于行军前5d开始分别口服新复方党参片和安慰剂片,乘车行军3d,于3700m习服4d,直至进驻高原(海拔5200m)第3天后停药,共服药15d。进驻高原后第1、3、5天,依国家军用标准GJB1098-91急性高原反应的诊断和处理原则,随访记录受试者的AMS症状,然后分度评分,检测受试者的心率(HR)、血氧饱和度(SaO2)。进驻高原后第6天,检测用力肺活量(FVC)、1秒用力呼气量(FEV1.0)、FEV1.0/FVC,一秒率(FEV1%)、最大呼气中期流速(FEF25%~75%)、呼气峰流速(PEF)、最大通气量(MVV)、左右手交叉敲击动作频率总次数(Ttis)、错误次数(Etis)、正确次数(Ctis)、平均时间(Atime)和数字记忆能力试验错误记忆次数总和(Sum)。结果:与对照组比较,进驻高原后第1、3、5d,新复方党参片组AMS症状显著减轻(P0.01);新复方党参片组与对照组的AMS程度分度分布不同(P0.01),新复方党参片组中症状较轻的(基本无反应、轻度反应)占比重较大,而对照组中症状较重的(中度反应、重度反应)占比重较大;新复方党参片组AMS发生率明显降低;与对照组比较,新复方党参片组的FVC、FEV1.0、FEF25%~75%、PEF、MVV升高有统计学意义(P0.05,P0.01),FEV1%差异无统计学意义;与对照组比较,新复方党参片组的Ttis、Ctis增加(P0.05,P0.01),Atime减少(P0.05),Etis和Sum差异无统计学意义。结论:新复方党参片能减轻AMS的程度,减轻AMS的症状,降低AMS发生率;并能显著改善受试者的肺通气功能和手指运动能力。  相似文献

The solubility in water of saturated fatty acids with even carbon numbers from 8 to 18 was measured in the temperature range of 60 to 230°C and at a pressure of 5 or 15 MPa. The pressure had no significant effect on the solubility. The solubility of the fatty acids increased with increasing temperature. At temperatures higher than about 160°C, the logarithm of the solubility in mole fraction was linearly related to the reciprocal of the absolute temperature for each fatty acid, indicating that the water containing solubilized fatty acid molecules formed a regular solution at the higher temperatures. The enthalpy of a solution of the fatty acids in water, which was evaluated from the linear relationship at the given temperatures, increased linearly with the carbon number of the fatty acid.  相似文献

Continuous production of the E7 protein from different types of high risk human papilloma virus (HPV) is required for progression of malignancy. We developed antibodies against HPV type 16 E7 and E2 proteins to evaluate their utility as markers for diagnosis during early stages of cervical cancer. Forty biopsies from uterine cervices were diagnosed as low grade intraepithelial lesion (LSIL), high grade intraepithelial lesion (HSIL), squamous carcinoma (SC), in situ adenocarcinoma (ISA) and invasive adenocarcinoma (AC), all of which were infected with HPV 16. Immunohistochemistry was used to investigate the expressions of E7 and E2 (both from HPV 16) and p16. P16 was expressed in eight of 12 LSILs, in all HSILs, in 16 of 18 SC and in all ACs. E2 was expressed in six of 12 LSILs. E7 was positive in eight of 12 LSILs and in all HSIL and carcinomas. The expressions of E2 and E7 of HPV16 related to p16 expression confirmed the value of the viral oncogenic proteins as complementary to histology and support the carcinogenic model of the uterine cervix, because HPVDNA integration into cellular DNA implies the destruction of the gene encoding E2, which suppresses the expression of the E6 and E7 oncoproteins. E2 from HPV16 could be marker for LSILs, while E7 could be a marker for progression of LSILs to HSILs in patients infected by HPV16, because viral typing has little positive predictive value.  相似文献
