Expression of C4-like photosynthesis in several species of Flaveria   总被引:4,自引:2,他引:2  
Abstract Photosynthetic metabolism was investigated in leaves of five species of Flaveria (Asteraceac), all previously considered to be C4 plants. Leaves were exposed to 14CO2 for different intervals up to 16s. Extrapolation of 14C-product curves to zero time indicated that only F. trinervia and F.bidentis assimilated atmospheric CO2 exclusively through phosphoenolpyruvate carboxylase. The proportion of direct fixation of 14CO2 by ribulose-I, 5-bisphosphate carboxylase/oxygenase (Rubisco) ranged from 5 to 10% in leaves of F. australasica. F. palmeri and F. vaginata. Protoplasts of leaf mesophyll and bundle sheath cells were utilized to examine the intercellular compartmentation of principal photosynthetic enzymes. Leaves of F. australasica, F. palmeri and F. vaginata contained 5 to 7% of the leaf's Rubisco activity in the mesophyll cells, while leaves of F. trinervia and F. bidentis contained at most 0.2 to 0.8% of such activity in their mesophyll cells. Thus, F. trinervia and F. bidentis have the complete C4 syndrome, while F. australasica, F. palmeri and F. vaginata are less advanced, C4-like species.  相似文献

Cholesterylphosphoryldimethylethanolamine is a zwitterionic compound which is a good bilayer stabilizer. As has been found with many other compounds having these properties, cholesterylphosphoryldimethylethanolamine is found to be a potent inhibitor of protein kinase C in both vesicle and micelle assay systems. The kinetics of the inhibition in Triton X-100 micelles was non-competitive with respect to ATP, histone, diolein, phorbol ester and Ca2+. It has a Ki of about 30 m. The inhibition kinetics as a function of phosphatidylserine concentration is more complex but suggestive of competitive inhibition. Cholesterylphosphoryldimethylethanolamine does not prevent the partitioning of protein kinase C into the membrane. This inhibitor lowers the Ca2+-phosphatidylserine-independent phosphorylation of protamine sulfate by protein kinase C and directly affects the catalytic segment of the enzyme generated by tryptic hydrolysis. Thus, this zwitterionic bilayer stabilizing inhibitor of protein kinase C both competes with the binding of phosphatidylserine as well as affects the active site of protein kinase C.Abbreviation CPD cholesterylphosphoryldimethylethanolamine  相似文献

A monospecific inhibitory antibody directed to phospholipase C (phosphoinositidase C) blocked the antiviral effect of human interferons alpha and beta when tested on human quiescent fibroblasts challenged with the vesicular stomatitis virus. This action was due to specific inhibition of polyphosphoinositide hydrolysis because (a) the F(ab)2 fragment of the antibody molecule was also inhibitory; (b) excess antibodies directed to phospholipase A2 and to a phosphatidylcholine-preferring phospholipase C did not have any inhibitory effect, and (c) the combination of 12-O-tetradecanoylphorbol-acetate and calcium ionophore A23187 had an interferon-like antiviral effect which was not influenced by the inhibitory anti-phospholipase C antibodies. To avoid an interferon-like effect due to induction of interferon by second messengers, Vero cells, which lack interferon biosynthesis, were also used. Liposomes containing inositol 1,4,5-triphosphate and 1-oleoyl-2-acetyl-rac-glycerol protected Vero cells against the infection with the vesicular stomatitis virus. These results taken together show that phosphoinositide-derived second messengers are involved in triggering the antiviral effect of interferons alpha and beta.  相似文献

Summary Pre-implantation-stage embryos from rats, mice, and guinea-pigs were transferred to a non-uterine site — the anterior chamber of the eye — of female recipients. All 9 combinations of transfers were performed: 3 allogeneic (intraspecies) transfers as controls, and 6 xenogeneic (interspecies) transfers. Implantation, as judged by extravasation from blood vessels of the iris or ciliary body occurred with success rates of 90.4% per transfer in the control rat group, 76.9% in the control mouse group, and 81.8% in the control guinea-pig group. Significantly reduced implantation rates occurred in the rat to guinea-pig (0%), mouse to rat (46.9%), mouse to guinea-pig (6.7%), and guinea-pig to rat (0%) groups compared to controls. Reductions, although not significant, also occurred in the other 2 groups: rat to mouse (77.8%), and guinea-pig to mouse (44.4%). These results together with some ultrastructural and lightmicroscopical observations suggest a degree of species specificity involved in the vascular response to the implanting embryo. We propose that the peri-implantation embryo produces a signal(s) which is to some extent species specific and which in the normal allogeneic situation is responsible for the early vascular effects seen at implantation in most eutherian mammals.  相似文献

Summary In-situ hybridization experiments have been performed using isoactin ( and )-specific riboprobes in various tissues of the rat and mouse. Distribution of the grains of actin mRNAs for both and types was similar throughout sections of the rat testis. Although both mRNAs were evenly distributed in the seminiferous tubule, extremely heavy labeling was observed in about 10% of the seminiferous tubules that could be identified as stage XII of spermatogenesis. At high magnification, grains of the mRNA were found in the cytoplasm of elongating spermatids and in the Sertoli cell cytoplasm at the adluminal side. Much higher density of the grains of mRNA was observed in the neck region of the spermatids at stage XII. Thus, the dense distribution of cytoskeletal actin mRNAs is stage-specific in the tubule during spermatogenesis in the rat. The high expression of both and actin mRNAs was also observed in the epithelial cells of the intestinal crypts.  相似文献

The intracellular pH of the halotolerant green algae Dunaliella tertiolecta, was determined by the distribution of 5,5-dimethyl-2(14C)-oxalolidine-2,5-dione (DMO) between the cell and the surrounding medium. 5,5-dimethyl-2(14C)oxalolidine-2,4-dione was not metabolized by the algal cells. The intracellular pH of Dunaliella tertiolecta was 6.8 in the dark and 7.4 in the light. During a salt stress, after two hours, the intracellular pH was increased by 0.2 pH units in both light and dark. The salt stressed cells maintained a constant pH of about 7.5 over the pH range of 6.5 to 8.5. Because of the relatively low permeability coefficient of the plasma membrane for DMO, this technique does not permit rapid pH determinations during the induction period after a salt stress. The magnitude of the salt induced pH changes measured 2 h after the salt stress implies a minor importance of this alkalization in this time range, but does not exclude a larger importance of pH changes for osmoregulation during the induction period.Abbreviations Chl chlorophyll - DMO 5,5-dimethyl-2(14C)oxalolidine-2,4-dione - PCV packed cell volume - SDS sodium dodecyl sulfate  相似文献

In laboratory experiments treatment of sugar-beet plants with aldicarb stimulated the mobility of Aphis fabae and two clones of Myzus persicae which were susceptible (S) and resistant (R) to carbamate-based insecticides, respectively. On the other hand, the number of aphids probing and the total number of probes made was reduced, and hence the transmission of beet mosaic virus (BMV) was restricted. In outdoor experiments the spread of BMV from aldicarb-treated plants by naturally infesting aphids was also restricted. The number of infected plants decreased with increasing distance from the sources of infection.
Résumé Des plantes de betterave traitées au laboratoire avec de l'aldicarbe ont stimulé la mobilité d'Aphis fabae et de deux clones de Myzus persicae, l'un sensible et l'autre résistant à des insecticides contenant des carbamates. Par ailleurs, le nombre de pucerons en train de sonder les feuilles ainsi que le nombre total de sondages ont été réduits et ainsi la transmission du virus de la mosaïque de la betterave (BMV) a été limitée. Dans des expériences à l'extérieur, la vitesse de propagation de BMV par des pucerons sur des plantes traitées à l'aldicarbe a été aussi plus limitée. Le nombre de plantes contaminées diminuait avec la distance de la source de contamination.

Earlier studies have indicated that interior (physical and/or chemical) properties of a plant may be responsible for feeding-site selection by the greenhouse whitefly, Trialeurodes vaporariorum (Westw.). In order to study the process of feeding-site selection further, stylet-penetration activities and the pathway followed by the stylets in host-plant tissue were investigated using a DC electrical recording method and transmission electron microscopy (TEM). Penetrating whiteflies attached to a gold wire were included in an electrical circuit to record electrical penetration graphs (EPGs). Seven EPG patterns have been distinguished, five of which could be correlated with components of the stylet-penetration process: 1) one with penetration of the leaf surface, 2) one with intercellular penetration and salivary-sheath secretion, 3) one with sieve element penetration and ingestion, 4) one with short penetration of a cell, and 5) one with xylem penetration. The stylet pathway is almost completely intercellular before the phloem is reached and in contrast to aphids, brief symplast punctures are very rare. In general, it takes T. vaporariorum more than half an hour from the start of a penetration to reach a sieve element. Rejection of feeding sites occurs within a few minutes of penetration by adult whiteflies, a time span in which stylets are presumed to penetrate just beyond the epidermis. Properties of the apoplast close to the leaf surface seem therefore to play a major role in feeding-site selection.
Résumé De précédentes études ont montré que les propriétés internes (physiques et/ou chimiques) d'une plante peuvent induire la sélection du site de nutrition de la mouche blanche de serres, Trialeurodes vaporariorum (Westw.). Afin d'étudier plus avant le processus de sélection du site de nutrition, les activités de piqûre et le chemin suivi par le stylet dans les tissus de la plante-hôte ont été étudiés par une méthode d'enregistrement électrique en courant continu ainsi que par microscopie à transmission d'électrons (TEM). Des aleurodes en activité de piqûre attachées à un fil d'or ont été incluses dans un circuit électrique pour enregistrer des graphes de pénétration électriques (EPG). Sept motifs d'EPG ont été distingués, dont cinq peuvent être corrélés aux composantes du processus de pénétration du stylet: 1) un avec pénétration de la surface foliaire, 2) un avec pénétration interecellulaire et sécrétion d'une gaine de salive, 3) un avec pénétration du phloème, 4) un avec courte pénétration d'une cellule, et 5) un avec pénétration du xylème. Le parcours du stylet est presque entièrement intercellulaire avant que le phloème soit atteint. Contrairement aux pucerons, les ponctions brèves dans le symplasme sont rares. Généralement, T. vaporariorum met plus d'une demi-heure, à partir du début d'une piqûre, pour atteindre un vaisseau. Le rejet des sites de nutrition par les aleurodes adultes se passe quelques minutes après la pénétration du stylet; pendant ce laps de temps, le stylet pénétrerait juste sous l'épiderme. Le rôle des propriétés de l'apoplaste près de la surface foliaire semble donc majeur dans la sélection des sites de nutrition.

Inhibition of protein kinase C (PKC) by calmodulin is investigated and we describe the localization of inhibitory sequences within the calmodulin molecule. We present evidence that calmodulin inhibits PKC through an inhibition of the activation of PKC associated with lipid membranes: Binding of PKC to lipid vesicles is not affected, but activation is abolished. The potent calmodulin antagonist R24571 (calmidazol) did not affect the inhibition of PKC by calmodulin at concentrations up to 10–5 M. Two tryptic fragments of calmodulin were isolated which inhibited PKC. They were only slightly less potent than intact calmodulin with an IC50 of 6 µ M compared to 1 µ M of intact calmodulin. They were identified as Ser38-Arg74 and His107-Lys148. Each of the inhibiting fragments contains an intact Ca2+-binding domain with complete helix-loop-helix structure (EF hand). Other calmodulin peptides showed only weak inhibitory activity. Both fragments did not stimulate cAMP phosphodiesterase even at concentrations 100-fold higher than the calmodulin concentration needed for maximal stimulation. None of the fragments acted as a calmodulin antagonist.  相似文献
