在光学显微镜下观察了不同产地华中五味子(Schisandra sphenanthera Rehd.et Wils.)叶表皮结构和导管分子特征,应用多元回归方法对不同产地华中五味子的叶表皮特征及导管分子特征与环境因子的关系进行了分析。观察的指标有叶表皮特征(气孔器密度、气孔器指数、气孔器长宽、气孔器面积、气孔极区角质加厚、气孔器类型、表皮细胞垂周壁形状、分泌细胞密度及叶表面角质条纹)和导管分子特征(导管分子长度、直径、长/直径比值及穿孔板类型)。观察结果:上表皮有极少量气孔器和分泌细胞分布。在下表皮,气孔器类型为平列型和侧列型,气孔器指数为15.05%-22.53%,气孔器长59.2—74.2μm,气孔器宽37.9—46.8μm,分泌细胞密度为1.45/mm^2-3.99/mm^2。导管分子以具缘纹孔导管为主,穿孔板包含单穿孔和梯状穿孔板。导管分子长957.8—1270.2μm,导管分子直径85.0—136.7μm。随着产地的不同,华中五味子叶下表皮结构和导管分子的解剖特征有明显的差异。分析结果显示:年均气温与气孔器面积和气孔器长度,年降水量与气孔器宽度以及相对湿度与分泌细胞密度之间关系密切。随着年平均气温的升高,气孔器面积、气孔器长度呈减少的趋势,R^2值分别为0.74、0.71。随着年降水量的增加,气孔器宽呈减小的趋势,R^2值为0.64;导管分子长/直径比值增加,R^2值为0.46。相对湿度与分泌细胞密度呈正相关,R^2为0.63。  相似文献

Vulnerability curves (VCs) generally can be fitted to the Weibull equation; however, a growing number of VCs appear to be recalcitrant, that is, deviate from a Weibull but seem to fit dual Weibull curves. We hypothesize that dual Weibull curves in Hippophae rhamnoides L. are due to different vessel diameter classes, inter‐vessel hydraulic connections or vessels versus fibre tracheids. We used dye staining techniques, hydraulic measurements and quantitative anatomy measurements to test these hypotheses. The fibres contribute 1.3% of the total stem conductivity, which eliminates the hypothesis that fibre tracheids account for the second Weibull curve. Nevertheless, the staining pattern of vessels and fibre tracheids suggested that fibres might function as a hydraulic bridge between adjacent vessels. We also argue that fibre bridges are safer than vessel‐to‐vessel pits and put forward the concept as a new paradigm. Hence, we tentatively propose that the first Weibull curve may be accounted by vessels connected to each other directly by pit fields, while the second Weibull curve is associated with vessels that are connected almost exclusively by fibre bridges. Further research is needed to test the concept of fibre bridge safety in species that have recalcitrant or normal Weibull curves.  相似文献

目的 研究胃癌组织中D2-40、LYVE-1标记的微淋巴管密度(LVD)、血管内皮生长因子受体(VEGFR-3)表达与幽门螺杆菌L型(helicobacter pylori L-form,Hp-L型)感染之间的关系.方法 应用革兰染色和免疫组化SP法检测80例胃癌组织和25例对照组的Hp-L型感染,同时用免疫组化SP法检测上述组织的LVD值和VEGFR-3的表达,分析Hp-L型与LVD以及VEGFR-3表达的关系结果 胃癌组织中革兰染色L型检出阳性率为67.5%;免疫组化Hp-L型抗原表达阳性率为65%,两种方法检测同时阳性的病例50例,占62.5%.胃癌组的Hp-L型阳性率、LVD及VEGFR-3表达阳性率均高于对照组(P<0.01);胃癌组中Hp-L阳性组的LVD值和VEGFR-3表达阳性率高于Hp-L阴性组.LVD与胃癌淋巴结转移具有一定关系.结论 Hp-L型感染与胃癌的发生、发展密切相关,Hp-L型可能是肿瘤淋巴管生成的重要促进因子,影响胃癌的侵袭和转移.  相似文献

In the hemolymph of Melanoplus sanguinipes, a high molecular weight juvenile hormone binding protein (JHBP) was identified by photoaffinity labelling and found to have a Mr of 480,000. The JHBP, purified using native gel electrophoresis followed by electroelution, has an equilibrium dissociation constant for JH III of 2.1 nM and preferentially binds JH III over JH I. Antibody raised against JHBP recognized only the 480,000 band. Under denaturing conditions the native JHBP gave a single band with a Mr 78,000. The antibody against native JHBP recognized only the 78,000 protein in SDS-treated hemolymph samples, indicating that JHBP is a hexamer in this species. The concentration of JHBP fluctuates in both the sexes during nymphal and adult development in parallel with total protein content of hemolymph. © 1995 Wiley-Liss, Inc.  相似文献

Tobacco hornworm larvae, Manduca sexta (L.) (Sphingidae), were administered L-canaline either by parenteral injection or by dietary consumption. The overt toxicity and the alteration of hemolymph amino acids caused by these nonprotein amino acids were evaluated. The LD50 value for parenterally administered canavanine and canaline is 1.0 and 2.5 mg/g fresh body weight, respectively. A dietary concentration of 5.2 mM for canavanine and over 20 mM for canaline represent the respective LC50 values. A large percentage of the larvae reared on diets supplemented with additional arginine, ornithine, or 2,4-diaminobutyric acid in addition to canavanine or canaline were unable to complete larval-pupal ecdysis. These toxic effects were associated with a decreased glutamic acid hemolymph titer and dramatically elevated ornithine. On the other hand, larvae administered canavanine or canaline alone, either by dietary consumption or parenteral injection, experienced less drastic developmental aberrations. These symptoms were in some cases correlated with increased ornithine and glutamic acid titers. Evidence is presented that even a canavanine- and canaline-sensitive insect such as M. sexta has a marked ability to eliminate these protective allelochemicals.  相似文献

An effective procedure for obtaining healthy shoots from nodal segments of Scrophularia yoshimurae is described. Nodal segments cultured on Murashige and Skoog's (MS) basal medium supplemented with 1.0 mg L(-1) benzyladenine (BA) and 0.2 mg L(-1) alpha-naphthaleneacetic acid (NAA) induced multiple shoots in conical flasks that differed in the way they were closed and sealed. Hermitically sealed culture vessels resulted in high hyperhydricity/vitrification. High concentrations of ethylene and CO2 were found to accumulate in these vessels. The hyperhydricity of the shoot cultures could be decreased by progressively ventilating the vessels. Exchange of gases was achieved by removing the Parafilm sealing without affecting sterility. This reduced the hyperhydricity rate and gave a good recovery of vitrified shoots, but resulted in decreased proliferation and a dehydration of proliferating nodal segments and the culture medium. The best number of normal shoots was observed when the parafilm was removed for gaseous exchange after four weeks of culture incubation. The results show that hyperhydricity in shoot cultures of S. yoshimurae could be prevented by sufficient gas exchange during culture.  相似文献

To produce erythropoietin (EPO), Chinese hamster ovary (CHO) cells were first cultured in a medium containing FBS (growth medium) and then in a serum-free medium containing sodium butyrate (production medium). Sodium butyrate increases recombinant protein production, but also induces apoptosis, which reduces cell viability and productivity. In a previous study, we found that silkworm hemolymph (SH), an insect serum, inhibits the apoptosis of insect and mammalian cells. To overcome sodium butyrate-induced apoptosis, we added SH to growth medium. This pretreatment with SH inhibited the sodium butyrate-induced apoptosis of CHO cells and consequently increased their longevity and their ability to produce EPO. As a result, the volumetric productivity of EPO was increased five-fold. SH was found to inhibit cytochrome c release from mitochondria into the cytosol, and prevented the activation of caspase-3 and other subsequent caspase reactions.  相似文献

The precise mechanisms of SDF‐1 (CXCL12) in angiogenesis are not fully elucidated. Recently, we showed that Notch inhibition induces extensive intussusceptive angiogenesis by recruitment of mononuclear cells and it was associated with increased levels of SDF‐1 and CXCR4. In the current study, we demonstrated SDF‐1 expression in liver sinusoidal vessels of Notch1 knockout mice with regenerative hyperplasia by means of intussusception, but we did not detect any SDF‐1 expression in wild‐type mice with normal liver vessel structure. In addition, pharmacological inhibition of SDF‐1/CXCR4 signalling by AMD3100 perturbs intussusceptive vascular growth and abolishes mononuclear cell recruitment in the chicken area vasculosa. In contrast, treatment with recombinant SDF‐1 protein increased microvascular density by 34% through augmentation of pillar number compared to controls. The number of extravasating mononuclear cells was four times higher after SDF‐1 application and two times less after blocking this pathway. Bone marrow‐derived mononuclear cells (BMDC) were recruited to vessels in response to elevated expression of SDF‐1 in endothelial cells. They participated in formation and stabilization of pillars. The current study is the first report to implicate SDF‐1/CXCR4 signalling in intussusceptive angiogenesis and further highlights the stabilizing role of BMDC in the formation of pillars during vascular remodelling.  相似文献
