Structural characteristics of xyloglucan are constant in the pericarp cell walls of kiwifruit ( Actinidia deliciosa ) throughout fruit enlargement and maturation. Most of the xyloglucan (XG) persists in the cell walls of ripe kiwifruit. XG from the pericarp tissues of 36-h ethylene-treated kiwifruit was extracted as hemicellulose II (HC-II) with 4.28 M KOH containing 0.02% NaBH4 , and purified using iodine precipitation and subsequent anion-exchange chromatography. This purifying protocol increased XG purity from 50 mol% in HC-II fraction to 62 mol% in the purified XG powder. The molar ratio of glucose: xylose: galactose: fucose in the purified XG was 10: 6.9: 2.1: 0.3. Gel permeation chromatography indicated that purified XG had an average molecular-mass of 161 KDa, a value that exceeds the 95 KDa Mr determined for total polymeric sugars. Sugar linkage analysis confirmed the lack of fucose in the kiwifruit XG, but a small amount of arabinoxylan and low Mr glucomannan remained associated with this fraction. 相似文献

目的:研究了蔬菜垃圾好氧处理过程中,纤维素降解菌和半纤维素降解菌(细菌和真菌),纤维素酶活和半纤维素酶活,和有机物降解之间的变化规律。方法:用添加纤维素和半纤维素的牛肉膏蛋白胨培养基和查式培养基,分别培养计数纤维素降解细菌、真菌和半纤维素降解细菌、真菌;马福炉灼烧测有机物含量。结果:好氧处理的初始阶段中,前4d有机物日均降解率5.2%,后3d日均降解率2.2%。结论:半纤维素降解菌的数量比纤维素降解菌的多,半纤维素酶活力,也高于纤维素酶活力;微生物的变化情况为前6d产两种酶的微生物主要有细菌和真菌;从第6d开始真菌快速生长;至第7d真菌纤维素酶和半纤维素酶活力显著升高。 相似文献

亚高山森林林窗可能通过改变冬季雪被格局和生长季水热环境影响林窗内凋落物中半纤维素的分解动态, 但目前对此还缺乏研究。采用凋落物分解袋法, 以亚高山森林5种典型物种岷江冷杉(Abies faxoniana)、红桦(Betula albosinensis)、四川红杉(Larix mastersiana)、方枝柏(Sabina saltuaria)和高山杜鹃(Rhododendron lapponicum)凋落物为研究对象, 研究雪被形成期、雪被覆盖期、雪被融化期和生长季节从林窗中心、林冠林窗、扩展林窗到郁闭林下物种凋落物的半纤维素变化特征。经历一年分解后, 5种凋落物的半纤维素均呈现净累积现象。针、阔叶凋落物半纤维素分别在雪被覆盖期和融化期表现出相对较高的损失率。在雪被覆盖期和融化期, 凋落物半纤维素在林窗中心和林冠林窗具有相对较高的损失率; 而在生长季节, 林窗中心呈现相对较低的凋落物半纤维素累积率。统计分析结果表明凋落物分解过程中半纤维素损失率与环境因子和凋落物质量因子均显著相关。这些结果表明亚高山森林林窗对凋落物分解过程中半纤维素损失率具有显著影响, 分别促进了半纤维素在冬季的损失以及抑制了半纤维素在生长季节的累积, 意味着亚高山森林林窗的形成有利于凋落物半纤维素的降解。 相似文献

Lignocellulose‐derived hydrolyzates typically display a high degree of variation depending on applied biomass source material as well as process conditions. Consequently, this typically results in variable composition such as different sugar concentrations as well as degree and the presence of inhibitors formed during hydrolysis. These key obstacles commonly limit its efficient use as a carbon source for biotechnological conversion. The gram‐negative soil bacterium Pseudomonas putida KT2440 is a promising candidate for a future lignocellulose‐based biotechnology process due to its robustness and versatile metabolism. Recently, P. putida KT2440_xylAB which was able to metabolize the hemicellulose (HC) sugars, xylose and arabinose, was developed and characterized. Building on this, the intent of the study was to evaluate different lignocellulose hydrolyzates as platform substrates for P. putida KT2440 as a model organism for a bio‐based economy. Firstly, hydrolyzates of different origins were evaluated as potential carbon sources by cultivation experiments and determination of cell growth and sugar consumption. Secondly, the content of major toxic substances in cellulose and HC hydrolyzates was determined and their inhibitory effect on bacterial growth was characterized. Thirdly, fed‐batch bioreactor cultivations with hydrolyzate as the carbon source were characterized and a diauxic‐like growth behavior with regard to different sugars was revealed. In this context, a feeding strategy to overcome the diauxic‐like growth behavior preventing accumulation of sugars is proposed and presented. Results obtained in this study represent a first step and proof‐of‐concept toward establishing lignocellulose hydrolyzates as platform substrates for a bio‐based economy. 相似文献

We have compared heterologous expression of two types of carbohydrate binding module (CBM) in tobacco cell walls. These are
the promiscuous CBM29 modules (a tandem CBM29-1-2 and its single derivative CBM29-2), derived from a non-catalytic protein1,
NCP1, of the Piromyces equi cellulase/hemicellulase complex, and the less promiscuous tandem CBM2b-1-2 from the Cellulomonas fimi xylanase 11A. CBM-labelling studies revealed that CBM29-1-2 binds indiscriminately to every tissue of the wild-type tobacco
stem whereas binding of CBM2b-1-2 was restricted to vascular tissue. The promiscuous CBM29-1-2 had much more pronounced effects on transgenic tobacco plants
than the less promiscuous CBM2b-1-2. Reduced stem elongation and prolonged juvenility, resulting in delayed flower development, were observed in transformants
expressing CBM29-1-2 whereas such growth phenotypes were not observed for CBM2b-1-2 plants. Histological examination and electron microscopy revealed layers of collapsed cortical cells in the stems of
CBM29-1-2 plants whereas cellular deformation in the stem cortical cells of CBM2b-1-2 transformants was less severe. Altered cell expansion was also observed in most parts of the CBM29-1-2 stem whereas for
the CBM2b-1-2 stem this was observed in the xylem cells only. The cellulose content of the transgenic plants was not altered. These
results support the hypothesis that CBMs can modify cell wall structure leading to modulation of wall loosening and plant

Aims: To examine the potential use of hemicellulose hydrolysate (HH) for the production of chitosan by Rhizopus oryzae and investigate the influence of contents in HH on mycelia growth and chitosan synthesis. Methods and Results: Compared to xylose medium, HH enhanced mycelia growth, chitosan content and production of R. oryzae by 10·2, 64·5 and 82·1%, respectively. During sulfuric acid hydrolysis of corn straw, sugars (glucose, galactose, etc) and inhibitors (formic acid, acetic acid and furfural) were generated. Acetic acid (2·14 g l?1) and formic acid (0·83 g l?1) were stimulative, while furfural (0·55 g l?1) was inhibitory. Inhibitors, at different concentrations, increased the mycelia growth and chitosan production by 24·5–37·8 and 60·1–207·1%. Conclusions: HH of corn straw is a good source for chitosan production. Inhibitors in HH, at proper concentrations, can enhance chitosan production greatly. Significance and Impact of the Study: This work for the first time reported chitosan production from HH. Chitosan production can be greatly enhanced by cheap chemicals such as inhibitors in HH. 相似文献

The production of xylitol by the yeast Candida guilliermondii was investigated in batch fermentations with aspenwood hemicellulose hydrolysate and compared with results obtained in semi-defined media with a mixture of glucose and xylose. The hemicellulose hydrolysate had to be supplemented by yeast extract and the maximum xylitol yield (0.8 g g–1) and productivity (0.6 g l–1 h–1) were reached by controlling oxygen input. 相似文献

Papaya ( Carica papaya L. cv. Eksotika) fruit softens differentially in relation to position of the tissue. The inner mesocarp tissue is softer, and its firmness decreases more rapidly during ripening than that of the outer mesocarp tissue. As the fruit ripens, pectin solubility and depolymerisation increase. Hemicellulose, too, appears to be depolymerised but, unlike pectins, this apparent degradation of hemicellulose is associated with an increase rather than a decrease in its level. Pectin and hemicellulose depolymerisation began in the inner mesocarp tissue at about the same time as β-galactosidase (EC 3.2,1.23) activity started to increase and tissue firmness began to decrease more rapidly. In contrast, pectin solubilisation in both outer and inner mesocarp tissues occurred steadily throughout ripening at a comparable rate and paralleled closely the increase of polygalacturonase (PG; EC 3.2.1.67) and pectinesterase (EC 3.1.1.11). In general, irrespective of enzyme distribution, tissue softening during ripening was more closely related to changes in β-galactosidase activity than to PG or pectinesterase activity. Papaya, β-galactosidase appears to be an important wall degrading enzyme and may contribute significantly to differential softening, perhaps by complementing the action of polygalacturonase. Polygalacturonase activity increased with increasing depth of the mesocarp tissue, as did softening of the fruit. 相似文献
