The oxidative stress response in Bacillus subtilis   总被引:9,自引:0,他引:9  
Abstract Bacillus subtilis undergoes a typical bacterial stress response when exposed to low concentrations (0.1 mM) of hydrogen peroxide. Protection is thereby induced against otherwise lethal, challenge concentrations (10 mM) of this oxidant and a number of proteins are induced including the scavenging enzymes, catalase and alkyl hydroperoxide reductase, and a putative DNA binding and protecting protein. Induced protection against higher concentrations (10–30 mM) of hydrogen peroxide is eliminated in a catalase-deficient mutant. Both RecA and Spo0A influence the basal but not the induced resistance to hydrogen peroxide. A regulatory mutation has been characterized that affects the inducible phenotype and is constitutively resistant to high concentrations of hydrogen peroxide. This mutant constitutively overexpresses the proteins induced by hydrogen peroxide in the wild-type. The resistance of spores to hydrogen peroxide is partly attributable to binding of small acid soluble proteins by the spore DNA and partly to a second step which coincides with the depletion of the NADH pool, which may inhibit the generation of hydroxyl radicals from hydrogen peroxide.  相似文献

Depending on spatial requirements and the distribution of key resources in the environment, social behavior among lizards varies from defense of exclusive territories to the establishment of dominance hierarchies. In captivity or under conditions where dispersal is not possible, dominance hierarchies often emerge in species that are otherwise territorial. This review explores some of the morphological, behavioral, and hormonal determinants of social status in male lizards and how these may lead to differential reproductive function in dominant and subordinate individuals. Emphasis is placed on the importance of population density, local resource dispersion, and the composition and stability of social groups in promoting hierarchical behavior. Results of these studies have ramifications for several aspects of zoo management, including exhibit design, choice of animals to be housed together, provision of resources in space and time, and orientation of enclosures within captive breeding facilities. © 1994 Wiley-Liss, Inc.  相似文献

A relatively drought tolerant cultivar of maize ( Zea mays L. cv. Pioneer 3950) and a drought tolerant line of sorghum ( Sorghum bicolor [L.] Moench cv. ICSV 112) were grown hydroponically for 11 days. Treatments for non-ionic osmotic and salt stresses were started at the 8th day by addition of polyethylene glycol 6000 and NaCl, respectively, at 200 mOsm equivalent concentrations in the presence or absence of 0. 1 μ M abscisic acid. Relative growth rate was depressed by both stress factors, more severely for maize than sorghum. Abscisic acid increased the growth rate and reverted the negative effect of NaCl in maize, while sorghum was only slightly affected. In general, sorghum had higher levels of K+ and lower levels of Na+ and the K+/Na+ ratio was further increased by abscisic acid treatment. From the pressure-volume curves, osmotic potential, the water potential at turgor loss point, bulk elastic modulus and the water saturation deficit at initial turgor loss were estimated. Most significantly, sorghum had a higher elastic modulus than maize and it decreased under osmotic treatment, while in maize it increased under NaCl stress. The results suggest that bulk tissue turgor was not limiting growth under these conditions and underscores the possible implications of changes in the elastic condition of the cell walls in stress responses.  相似文献

The influence of chromium concentration on ethylene production in bean plants ( Phaseolus vulgaris L. cv. Contender) was investigated. A Cr ion-induced inhibition of ethylene synthesis from endogenous 1-aminocyclopropane-1-carboxylic acid (ACC) was observed within both leaf discs floated on 2 m M CrO2−4 or Cr3+ and leaf discs from plants cultured in nutrient solutions containing 10, 20 or 40 μ M CrO2−4. However, Cr ions supplied either to plants with the nutrient solution or to discs with the incubation medium rather increased the conversion of exogenous ACC to ethylene. Primary leaves of plants exposed to CrO2−4-containing nutrient solutions showed a statistically insignificant decrease of ACC-synthase activity. In the trifoliolate leaves of plants exposed to 10 μ M CrO2−4, in which a significant decrease of ethylene production from endogenous ACC was observed, a substantial increase of ACC synthase was found. These results indicate that Cr ion-induced inhibition of ethylene production is not due to a breakdown of membrane integrity, which is necessary for ethylene forming enzyme activity, but caused by metabolic alterations leading to decreased ACC availability. Chromium ions may act by inhibiting ACC synthase activity or by diverting a metabolic step prior to the ACC synthase catalyzed reaction.  相似文献

Ethylene production by primary roots of 72–h-old intact seedlings of Zea mays L. cv. LG11 was studied under ambient and sub-ambient oxygen partial pressures (pO2) using a gas flow-through system linked to a photoacoustic laser detector. Despite precautions to minimize physical perturbation to seedlings while setting-up, ethylene production in air was faster during the first 6h than later, in association with a small temporary swelling of the roots. When roots were switched from air (20–8kPa O2) to 3 or 5kPa O2 after 6h, ethylene production increased within 2—3 h. When, the roots were returned to air 16 h later, ethylene production decreased within 2—3 h. The presence of 10kPa CO2 did not interfere with the effect of 3kPa O2. Transferring roots from air to 12–5kPa did not change ethylene production, while a reduction to 1 kPa O2 induced a small increase. The extra ethylene formed in 3 and 5 kPa O2 was associated with plagiotropism, swelling, root hair production, and after 72 h, increased amounts of intercellular space (aerenchyma) in the root cortex. Root extension was also slowed down, but the pattern of response to oxygen shortage did not always match that of ethylene production. On return to air, subsequent growth patterns became normal within a few hours. In the complete absence of oxygen, no ethylene production was detected, even when anaerobic roots were returned to air after 16 h.  相似文献

The vegetation at various sites within two separate areas (Mechernich and Aachen) of the Eifel Mountains, Germany/Belgium, both characterized by elevated concentrations of heavy metals in their soils, was surveyed in order to investigate the relationships between soil chemical attributes and floristic composition. In both areas, the typical heavy metal communities can form distinct zones, clearly separated from the surrounding heavy metal-sensitive vegetation. However, an intergrading of heavy metal-tolerant and-sensitive vegetation types is not uncommon and such overlaps can occupy large areas. In Mechernich, soil toxicity is primarily determined by the effects of lead, which is best expressed in terms of the Pb/Ca ratio rather than the absolute levels of this metal in the soil. Soils of heavy metal-sensitive vegetation types have a low Pb/Ca ratio, whereas it is considerably higher in areas supporting heavy metal vegetation. Zinc appears to exert little influence on the floristic composition of the investigated vegetation types. In Aachen, zinc is the predominant heavy metal determining vegetation development. Absolute zinc levels of soils do not accurately reflect zinc toxicity. Analogous to the role of the Pb/Ca ratio in the Mechernich area, the Zn/Ca ratio not only separates heavy metal-sensitive and highly tolerant vegetation units, but also gives a good indication of the gradient operating between the two vegetation types. Lead is probably only of local importance in influencing species composition.  相似文献

Gas chromatography-mass spectrometry has been applied to the analysis of plasma linoleic acid and one of its oxidation products, 4-hydroxy-2-nonenal (HNE), in adult patients with the acute respiratory distress syndrome (ARDS). Peak areas of total ion chromatograms showed there to be negative correlations between loss of linoleic acid and formation of HNE (measured by selective ion monitoring) in 7 out 10 patients studied. When HNE was quantitated by selective ion monitoring, with reference to a pure standard of HNE and an internal standard of nonanoic acid, ARDS patients showed significantly increased levels of HNE (0.412 ± 0.023 nmol/ml) compared with normal healthy controls (0.205 ± 0.018 nmol/ml).  相似文献

Abstract: Changes in sympathetic nerve terminals of the heart after varying periods of exposure of rats to 4°C were investigated. Two indices were used for changes in the number of noradrenaline storage vesicles, i.e., vesicular dopamine β-hydroxylase (DBH) activity and noradrenaline storage capacity. The latter was obtained after uptake of [3H]noradrenaline; endogenous content, uptake of exogenous noradrenaline, and degree of saturation of the vesicles were calculated using the specific activity of the [3H]noradrenaline. As a measure of tyrosine hydroxylase activity, whole ventricular noradrenaline, dopamine, and dihydroxyphenylacetic acid content were used. After 4 h of cold exposure there was an increase in vesicular endogenous noradrenaline content, uptake, storage capacity, and DBH activity as well as a large increase in whole ventricular dopamine. After 6 h in the cold, vesicular endogenous noradrenaline content, storage capacity, and DBH activity were decreased. The results suggest that during cold exposure there is an initial increase followed by a decrease in the number of functional vesicles in the nerve terminal, which could explain the fluctuations in the rate of noradrenaline release.  相似文献

The effect of simulated rainfall frequency on the pathogenicity of Pratylenchus zeae and P. brachyurus was studied in four greenhouse experiments. Corn and grain sorghum were watered at different intervals during predetermined cycles to create a gradient of water-stressed plants. Each experiment included nematode and uninoculated treatments. Growth reaction of plants to different frequencies of watering was significant but was not affected by the presence of nematodes. Pratylenchus zeae numbers differed among watering regimens on corn but not on sorghum. Numbers of P. brachyurus did not differ among watering regimens on corn or sorghum. Both lesion nematode species were harmful to corn, but sorghum increased plant growth in response to P. brachyurus. It is concluded that water stress is not the only environmental factor that influences the pathogenicity of these two species on corn and sorghum.  相似文献
