Polymerase chain reaction (PCR) has been used extensively in the construction of linkage maps for many cultivated crops including soybean, [Glycine max (L.) Merr]. In this study, four sets of oligonucleotide primer pairs of known genes (pearl millet Adh 1, nodule specific proline-rich protein, Drosophila homeobox, heat shock protein), several different combinations from kits A, D, E, and J of arbitrary primers and five primer pairs of soybean simple sequence repeats of varying length (Satt 9, Satt 20, Satt 42, Satt 64, and Satt 30) were utilized in PCR to identify molecular markers which were then used to construct a genetic linkage map. DNA for the PCR reactions was isolated from 65 recombinant inbred soybean lines resulting from crossing PI 290,136 and BARC-2 (Rj 4 ), followed by self-pollination for seven generations without selection. Mapmaker 3.0, a computer package, was used for construction of the linkage map. A total of 43 polymorphic markers were identified; 30 markers were linked and distributed among 5 linkage groups while 13 markers were unlinked. Arbitrary primers revealed more polymorphisms than specific primers. A combination of arbitrary primers A5 and A18 revealed the maximum number of polymorphic bands. Five observed linkage groups can be expanded in future soybean research by using additional markers.  相似文献

Urbanization is currently one of the most pervasive threats to biodiversity worldwide, yet traits permitting birds to exploit urban environments are not fully understood. I used bird traits related to diet (dietary item and foraging stratum), accounting for latitude, body size, development mode and phylogeny, to compare diet breadths of urban exploiters and urban avoiders, using a global dataset (463 bird species). Urban exploiters (urban species) were larger, consumed more vertebrates and carrion, and fed more frequently on the ground or aerially, and also had broader diets than urban avoiders (non-urban species). In contrast, urban species had narrower foraging strata ranges than non-urban species. These results not only support the hypothesis that urban species share dietary traits allowing them to cope with urban environments but also highlight the importance of considering multiple dietary traits to properly quantify species niches when assessing a species’ response to environmental change.  相似文献

Prefoldin is a jellyfish-shaped hexameric chaperone that captures a protein-folding intermediate and transfers it to the group II chaperonin for correct folding. In this work, we characterized the organic solvent tolerance of Escherichia coli cells that overexpress prefoldin and group II chaperonin from a hyperthermophilic archeaum, Pyrococcus horikoshii OT3. The colony-forming efficiency of E. coli cells overexpressing prefoldin increased by 1,000-fold and decreased the accumulation of intracellular organic solvent. The effect was impaired by deletions of the region responsible for the chaperone function of prefoldin. Therefore, we concluded that prefoldin endows E. coli cells by preventing accumulation of intracellular organic solvent through its molecular chaperone activity.  相似文献

Helicobacter pylori is a cause of chronic gastritis and leads to development of atrophy in some cases. There is evidence that the heat shock protein 60 (HSP60) of H. pylori is involved in induction of chronic inflammation. Seroprevalence of IgG antibodies to H. pylori HSP60 in an adult cohort from Saaremaa, Estonia (68 persons, median age 57 years), with a high prevalence of antibodies to cell surface proteins of H. pylori (92%) and a well characterized dynamics of chronic gastritis in an 18-year follow-up study, was tested using purified H. pylori HSP60 at a concentration of 1 microg ml(-1) with ELISA. The state of the gastric mucosa and the presence of H. pylori in histological sections in the samples of 1979 and 1997 were assessed in accordance with the Sydney system. Seropositivity for H. pylori HSP60 was 65%. Immunological response to H. pylori HSP60 is associated with the morphological presence of H. pylori in the antrum and corpus (P=0.01) and is strongly correlated with the grade of chronic inflammation, particularly in the antrum mucosa (r=0.34; P=0.003; OR=5.97 (95% CI 1.21-29.3)), but is not associated with development of atrophy during 18 years of follow-up, or with the activity of gastritis. This finding supports the evidence that immunological response to H. pylori HSP60 may play a role in triggering of the inflammatory process in the gastric mucosa.  相似文献

Abstract. 1. In central Japan, Drosophila curviceps Okada and Kurokawa was collected in spring and autumn but not in summer at lowlands (alt. 500–1200 m), while it was collected only in summer at highlands (1500–2000 m). Experiments on its thermal tolerance suggested that summer heat at the lowlands and winter low temperatures at the highlands were adverse to this species. It is considered that this species escapes from these extreme temperatures by undergoing seasonal migration between the lowlands and the highlands. This species had no photo-periodic diapause and bred at both lowlands and highlands.
2. D.immigrans Sturtevant was less cold-hardy but more heat-tolerant than D.curviceps. It is considered that this species is unable to overwinter outdoors at least in the study areas (i.e. alt. 500m or higher in central Japan) and its populations in these areas originate with migrants from warmer areas.
3. D.albomicans Duda, a subtropical species, was less cold-hardy but more heat-tolerant than the above two species.
4. Climatic adaptations and distributions of these species are discussed with reference to their thermal tolerance.  相似文献

Exposure of growing rabbits to heat stress during summer adversely affects their performance leading to major production losses. A total number of 48 rabbits, unsexed V-line weaned rabbits, were randomly divided into four experimental groups, temperature ranged from high at 32°C to low at 23°C. Animals of the 2nd, 3rd and 4th group were individually orally given 200, 400 or 800 mg royal jelly (RJ)/kg BW once a week, respectively, to evaluate RJ ability to reduce physiological strain resulted from heat stress. Weekly BW gain increased by 10.4, 11.8 and 10.8%, and feed conversion ratio was significantly improved by 20, 24 and 18% with RJ treatments. Serum total protein, albumin and globulin increased, whereas serum total lipids, cholesterol and triglycerides decreased with RJ treatments. Creatinine was reduced by 21, 30 and 18% and uric acid by 14, 25 and 18% compared with the heat stressed control with the three doses of RJ. Glucose level increased significantly to reach 116, 125, and 120% of heat stressed control. Calcium, phosphorus and alkaline phosphatase increased significantly with RJ treatments indicating the occurrence of active bone deposition. Thyroid hormone levels increased significantly to reach 108, 111, and 112% of heat stressed control rabbits with the three doses of RJ, counteracting the hypothyroid state resulted from heat stress. It can be concluded that RJ administration to heat stressed growing rabbits can reduce physiological strain resulted from heat stress.  相似文献

利用东北地区48个农气站1961-2010年气象资料和近20多年玉米生育期资料,建立生育阶段热量指数和冷害指数,分析气候变暖对东北玉米4个生育阶段热量及低温冷害分布格局的影响。结果表明:热量指数总体表现为明显的增加趋势;平均温度、热量指数的年代际变化特征明显,中晚、晚熟区的上升趋势均小于早、中熟区;冷害频率总体表现为明显的减小趋势;气候变暖对两个熟型区域4个生育阶段的冷害影响并不一致,早、中熟区的冷害平均频率最大值均出现在20世纪60年代,中晚、晚熟区的冷害平均频率最大值均出现在70年代,两个熟型区域的最小值均出现在21世纪初。研究结果可为未来东北地区调整玉米种植制度和生产布局,为适应气候变化和趋利避害提供科学依据。  相似文献

The mediatory role of kinins in both acute and chronic inflammation within nervous tissues has been widely described. Bradykinin, the major representative of these bioactive peptides, is one of a few mediators of inflammation that directly stimulates afferent nerves due to the broad expression of specific kinin receptors in cell types in these tissues. Moreover, kinins may be delivered to a site of injury not only after their production at the endothelium surface but also following their local production through the enzymatic degradation of kininogens at the surface of nerve cells. A strong correlation between inflammatory processes and neurodegeneration has been established. The activation of nerve cells, particularly microglia, in response to injury, trauma or infection initiates a number of reactions in the neuronal neighborhood that can lead to cell death after the prolonged action of inflammatory substances. In recent years, there has been a growing interest in the effects of kinins on neuronal destruction. In these studies, the overexpression of proteins involved in kinin generation or of kinin receptors has been observed in several neurologic disorders including neurodegenerative diseases such Alzheimer's disease and multiple sclerosis as well as disorders associated with a deficiency in cell communication such as epilepsy. This review is focused on recent findings that provide reliable evidence of the mediatory role of kinins in the inflammatory responses associated with different neurological disorders. A deeper understanding of the role of kinins in neurodegenerative diseases is likely to promote the future development of new therapeutic strategies for the control of these disorders. An example of this could be the prospective use of kinin receptor antagonists.  相似文献

Defense costs provide a major explanation for why plants in nature have not evolved to be better defended against pathogens and herbivores; however, evidence for defense costs is often lacking. Plants defend by deploying resistance traits that reduce damage, and tolerance traits that reduce the fitness effects of damage. We first tested the defense-stress cost (DSC) hypothesis that costs of defenses increase and become important under competitive stress. In a greenhouse experiment, uniparental maternal families of the host plant Arabis perennans were grown in the presence and absence of the bunch grass Bouteloua gracilis and the herbivore Plutella xylostella. Costs of resistance and tolerance manifest as reduced growth in the absence of herbivory were significant when A. perennans grew alone, but not in the competitive environment, in contrast to the DSC hypothesis. We then tested the defense-stress benefit (DSB) hypothesis that plant defenses may benefit plants in competitive situations thereby reducing net costs. For example, chemical resistance agents and tolerance may also have functions in competitive interactions. To test the DSB hypothesis, we compared differentially competitive populations for defense costs, assuming that poorer competitors from less dense habitats were less likely to have evolved defenses that also function in competition. Without competitive benefits of defenses, poorer competitors were expected to have higher net costs of defenses under competition in accordance with DSB. Populations of A. perennans and A. drummondii that differed dramatically in competitiveness were compared for costs, and as the DSB hypothesis predicts, only the poor competitor population showed costs of resistance under competition. However, cost of tolerance under competition did not differ among populations, suggesting that the poor competitors might have evolved a general stress tolerance. Although the DSC hypothesis may explain cases where defense costs increase under stress, the DSB hypothesis may explain some cases where costs decrease under competitive stress.  相似文献

The family of Nha antiporters mediating the efflux of alkali metal cations in exchange for protons across the plasma membrane is conserved in all yeast species. Yarrowia lipolytica is a dimorphic yeast, phylogenetically very distant from the model yeast Saccharomyces cerevisiae. A search in its sequenced genome revealed two genes (designated as YlNHA1 and YlNHA2) with homology to the S. cerevisiae NHA1 gene, which encodes a plasma membrane alkali metal cation/H+ antiporter. Upon heterologous expression of both YlNHA genes in S. cerevisiae, we showed that Y. lipolytica antiporters differ not only in length and sequence, but also in their affinity for individual substrates. While the YlNha1 protein mainly increased cell tolerance to potassium, YlNha2p displayed a remarkable transport capacity for sodium. Thus, Y. lipolytica is the first example of a yeast species with two plasma membrane alkali metal cation/H+ antiporters differing in their putative functions in cell physiology; cell detoxification vs. the maintenance of stable intracellular pH, potassium content and cell volume.  相似文献
