高温胁迫对不同热敏型辣椒同工酶及DNA甲基化的影响   总被引:1,自引:0,他引:1  
分别以耐热型晚熟紫色辣椒‘7036’和热敏型早熟绿色辣椒‘9050’为材料,研究了40℃高温胁迫对其POD同工酶以及DNA甲基化表达的影响。结果显示:(1)在POD同工酶表达方面,未经高温胁迫的辣椒‘7036’比其它处理多出1条带,而同工酶活性在2种辣椒中呈现不同的变化趋势。(2)随着高温胁迫时间延长,辣椒‘7036’DNA无甲基化和全甲基化比率增加,半甲基化和总甲基化比率降低,而辣椒‘9050’只有高温处理7d(T_7)的甲基化水平变化与之类似,但半甲基化和全甲基化比率的绝对值远低于辣椒‘7036’。(3)高温处理7d的DNA甲基化模式中,辣椒‘7036’去甲基化C型条带比率较高,辣椒‘9050’以超甲基化B型条带为主。实验表明,高温胁迫下紫色辣椒‘7036’的POD同工酶活性恢复,半甲基化水平大幅下降和全甲基化水平快速上升以及去甲基化等变化可能与其耐高温胁迫特性相关。  相似文献

The aim of this study was to investigate the effect of CO2-induced water acidification (pH values: 8.1 ? control, 7.5 and 7.0) on the hemolymph osmolality and total metabolic rate of amphipod Gammarus oceanicus under short-term exposure. The hemolymph osmolality was measured using a vapor pressure osmometer. The metabolic rate was determined based on the heat dissipation measurements using a Calvet-type isothermal twin calorimeter. The hemolymph osmolality remained unchanged after exposure to pH 7.5 and it increased in the lowest treatment. Acidification did not significantly affect the resting or active metabolic rate. The observed results indicate that high osmolality maintained during hypercapnia did not generate additional physiological costs. It may therefore be concluded that the studied species is preadapted for this factor. The long-term adaptation to hypoosmotic conditions in the brackish Baltic Sea might also have preadapted it to hypercapnia because similar physiological mechanisms are responsible for the resistance to both factors.  相似文献

为研究海南产白唇竹叶青蛇分类地位,捕捉来自海南岛各地的12个白唇竹叶青蛇样本,并从GenBank下载65个竹叶青蛇的样本序列,以瘤鼻蝮蛇为外群,建立16S系统发育树。系统发育树显示,竹叶青蛇分为多个属的观点具有很大的合理性,白唇竹叶青蛇隶属于帝汶(岛屿)竹叶青蛇属。进一步验证了经典形态学和生物地理学的观点,海南岛产白唇竹叶青与其它地方产白唇竹叶青蛇不存在生殖隔离。  相似文献

Solar desalination is a promising and sustainable solution for water shortages in the future. Interfacial solar‐to‐heat conversion for desalination has attracted increasing attention in the past decades, due to the heat localization induced high thermal efficiency, simple structure, and low cost. In this review, the authors summarize and analyze the critical processes involved in such a solar desalination system, including the thermal conversion and transport, salt dissipation, and vapor manipulation. Mathematical models of heat transfer and salt dissipation are also built for quantitative analysis of systematic performance relative to properties of employed materials and system designs. Recent efforts devoted to improving the overall thermal efficiency, salt rejection, and water yield are then summarized. Based on the analysis and previous results, opportunities for further interfacial solar desalination development are highlighted.  相似文献

MicroRNA393 (miR393) has been implicated in plant growth, development and multiple stress responses in annual species such as Arabidopsis and rice. However, the role of miR393 in perennial grasses remains unexplored. Creeping bentgrass (Agrostis stolonifera L.) is an environmentally and economically important C3 cool‐season perennial turfgrass. Understanding how miR393 functions in this representative turf species would allow the development of novel strategies in genetically engineering grass species for improved abiotic stress tolerance. We have generated and characterized transgenic creeping bentgrass plants overexpressing rice pri‐miR393a (Osa‐miR393a). We found that Osa‐miR393a transgenics had fewer, but longer tillers, enhanced drought stress tolerance associated with reduced stomata density and denser cuticles, improved salt stress tolerance associated with increased uptake of potassium and enhanced heat stress tolerance associated with induced expression of small heat‐shock protein in comparison with wild‐type controls. We also identified two targets of miR393, AsAFB2 and AsTIR1, whose expression is repressed in transgenics. Taken together, our results revealed the distinctive roles of miR393/target module in plant development and stress responses between creeping bentgrass and other annual species, suggesting that miR393 would be a promising candidate for generating superior crop cultivars with enhanced multiple stress tolerance, thus contributing to agricultural productivity.  相似文献

Heroin is a highly abused opioid that has reached epidemic status within the United States. Yet, existing therapies to treat addiction are inadequate and frequently result into rates of high recidivism. Vaccination against heroin offers a promising alternative therapeutic option but requires further development to enhance the vaccine’s performance. Hsp70 is a conserved protein with known immunomodulatory properties and is considered an excellent immunodominant antigen. Within an antidrug vaccine context, we envisioned Hsp70 as a potential dual carrier-adjuvant, wherein immunogenicity would be increased by co-localization of adjuvant and antigenic drug hapten. Recombinant Mycobacterium tuberculosis Hsp70 was appended with heroin haptens and the resulting immunoconjugate granted anti-heroin antibody production and blunted heroin-induced antinociception. Moreover, Hsp70 as a carrier protein surpassed our benchmark Her-KLH cocktail through antibody-mediated blockade of 6-acetylmorphine, the main mediator of heroin’s psychoactivity. The work presents a new avenue for exploration in the use of hapten-Hsp70 conjugates to elicit anti-drug immune responses.  相似文献

Sexual reproduction in flowering plants depends on the fitness of the male gametophyte during fertilization. Because pollen development is highly sensitive to hot and cold temperature extremes, reliable methods to evaluate pollen viability are important for research into improving reproductive heat stress (HS) tolerance. Here, we describe an approach to rapidly evaluate pollen viability using a reactive oxygen species (ROS) probe dichlorodihydrofluorescein diacetate (i.e. H2DCFDA‐staining) coupled with flow cytometry. In using flow cytometry to analyze mature pollen harvested from Arabidopsis and tomato flowers, we discovered that pollen distributed bimodally into ‘low‐ROS’ and ‘high‐ROS’ subpopulations. Pollen germination assays following fluorescence‐activated cell sorting revealed that the high‐ROS pollen germinated with a frequency that was 35‐fold higher than the low‐ROS pollen, supporting a model in which a significant fraction of a flower's pollen remains in a low metabolic or dormant state even after hydration. The ability to use flow cytometry to quantify ROS dynamics within a large pollen population was shown by dose‐dependent alterations in DCF‐fluorescence in response to oxidative stress or antioxidant treatments. HS treatments (35°C) increased ROS levels, which correlated with a ~60% reduction in pollen germination. These results demonstrate the potential of using flow cytometry‐based approaches to investigate metabolic changes during stress responses in pollen.  相似文献

目的:观察人参皂苷Rg2对慢性坐骨神经损伤大鼠痛觉敏化、抑郁状态的影响。方法: 将50只 SD 大鼠随机分为 5组(n=10): 空白对照组(Normal+生理盐水腹腔注射)、假手术组(手术但不结扎+生理盐水腹腔注射) 、坐骨神经慢性压迫损伤(CCI)组(CCI +生理盐水腹腔注射) 、人参皂苷Rg2低剂量组(CCI+ Rg2 5 mg/kg腹腔注射)、人参皂苷Rg2高剂量组(CCI+ Rg2 10 mg/kg 腹腔注射)。CCI模型建立后,药物通过注射器进行腹腔内注射 5 ml/kg,每天1次,连续14 d。分别在术前1 d和术后 1、3、5、7、10、14 d测定大鼠的机械性缩足反射阈值(MWT)和热缩足潜伏期(TWL);术前1 d和术后第14日时检测明暗箱实验和强迫游泳试验。 结果:与假手术组比较,CCI组术后14 d机械痛阈值和热痛潜伏期明显缩短(P<0.01),明箱内停留时间明显缩短(P<0.01),穿梭次数明显减少(P<0.01),游泳潜伏期明显延长(P<0.01)。与CCI组比较,人参皂苷Rg2组术后14 d机械痛阈和热痛潜伏期明显增加(P<0.01),大鼠在明箱内时间明显延长(P<0.01),穿梭次数明显增多(P<0.01),且游泳潜伏期明显缩短(P<0.01)。结论:人参皂苷Rg2能抑制 CCI 大鼠的机械痛敏和热痛敏,同时改善其抑郁状态。  相似文献
