Environmental adaptation of crops is essential for reliable agricultural production and an important breeding objective. Genebanks provide genetic variation for the improvement of modern varieties, but the selection of suitable germplasm is frequently impeded by incomplete phenotypic data. We address this bottleneck by combining a Focused Identification of Germplasm Strategy (FIGS) with core collection methodology to select soybean (Glycine max) germplasm for Central European breeding from a collection of >17,000 accessions. By focussing on adaptation to high-latitude cold regions, we selected an “environmental precore” of 3,663 accessions using environmental data and compared the Donor opulation of Environments (DPE) in Asia and the Target Population of Environments (TPE) in Central Europe in the present and 2070. Using single nucleotide polymorphisms, we reduced the precore into two diverse core collections of 183 and 366 accessions to serve as diversity panels for evaluation in the TPE. Genetic differentiation between precore and non-precore accessions revealed genomic regions that control maturity, and novel candidate loci for environmental adaptation, demonstrating the potential of diversity panels for studying adaptation. Objective-driven core collections have the potential to increase germplasm utilization for abiotic adaptation by breeding for a rapidly changing climate, or de novo adaptation of crops to expand cultivation ranges.  相似文献

Increasing temperatures resulting from climate change dramatically impact rice crop production in Asia. Depending on the specific stage of rice development, heat stress reduces tiller/panicle number, decreases grain number per plant and lower grain weight, thus negatively impacting yield formation. Hence improving rice crop tolerance to heat stress in terms of sustaining yield stability under high day temperature (HDT), high night temperature (HNT), or combined high day and night temperature (HDNT) will bolster future food security. In this review article, we highlight the phenological alterations caused by heat and the underlying molecular-physiological and genetic mechanisms operating under different types of heat conditions (HDT, HNT, and HDNT) to understand heat tolerance. Based on our synthesis of HDT, HNT, and HDNT effects on rice yield components, we outline future breeding strategies to contribute to sustained food security under climate change.  相似文献

Global warming increasingly challenges thermoregulation in endothermic animals, particularly in hot and dry environments where low water availability and high temperature increase the risk of hyperthermia. In birds, un-feathered body parts such as the head and bill work as ‘thermal windows’, because heat flux is higher compared to more insulated body regions. We studied how such structures were used in different thermal environments, and if heat flux properties change with time in a given temperature. We acclimated zebra finches (Taeniopygia guttata) to two different ambient temperatures, ‘cold’ (5 °C) and ‘hot’ (35 °C), and measured the response in core body temperature using a thermometer, and head surface temperature using thermal imaging. Birds in the hot treatment had 10.3 °C higher head temperature than those in the cold treatment. Thermal acclimation also resulted in heat storage in the hot group: core body temperature was 1.1 °C higher in the 35 °C group compared to the 5 °C group. Hence, the thermal gradient from core to shell was 9.03 °C smaller in the hot treatment. Dry heat transfer rate from the head was significantly lower in the hot compared to the cold treatment after four weeks of thermal acclimation. This reflects constraints on changes to peripheral circulation and maximum body temperature. Heat dissipation capacity from the head region increased with acclimation time in the hot treatment, perhaps because angiogenesis was required to reach peak heat transfer rate. We have shown that zebra finches meet high environmental temperature by heat storage, which saves water and energy, and by peripheral vasodilation in the head, which facilitates dry heat loss. These responses will not exclude the need for evaporative cooling, but will lessen the amount of energy expend on body temperature reduction in hot environments.  相似文献

Abiotic factors' effects on species are now well-studied, yet they are still often difficult to predict, especially for strongly interacting species. If these altered abiotic factors and species interactions occur as discrete events in time, such complications may occur because of the events’ relative timing. One such discrete abiotic factor is the short-duration, large magnitude increase in temperature called a heat shock. This study investigates how the timing of heat shocks affects the successful attack and reproduction of a parasitoid wasp (Aphidius ervi) attacking its host, the pea aphid (Acyrthosiphon pisum). We tested three relative timings: 1) heat shock before the wasp attacks hosts, 2) heat shock while the wasp is foraging, and 3) heat shock after the wasp has attacked hosts. In each scenario we compared wasp mummy production (pupal stage) with and without a heat shock. Our results showed that a heat shock had the largest effect when it occurred while wasps actively foraged, with fewer mummies produced when exposed to a heat shock compared to the no heat shock control. Follow-up behavioral tests suggest this was caused by wasps becoming inactive during heat shocks. In contrast, when heat shocks were applied three days before or after foraging, we found no difference in mummy production between the heat shock treatment and no heat shock control. These results show the potential importance of timing when considering the ramifications of an altered abiotic factor, especially with relatively discrete abiotic events and interactions.  相似文献

The purpose of this study was to determine local sweat rate (LSR) and sweat composition during heat acclimation (HA). For ten consecutive days of HA, eight participants cycled in 33 °C and 65% relative humidity at an intensity such that a rectal temperature of 38.5 °C was reached within ~40 min, followed by a 60-min clamp of this rectal temperature (i.e., controlled hyperthermia). Four participants extended HA by a 28-day decay period and five consecutive days of heat re-acclimation (HRA) using controlled hyperthermia. Sweat from the upper arm and upper back was collected three times during each heat exposure session. LSR and sweat sodium, chloride, lactate, and potassium concentrations were determined. Relative to HA day 1, LSR was increased at the final day of HA (day 10) (arm: +58%, P < 0.001; back: +36%, P < 0.05). Concentrations of sodium, chloride, and lactate significantly (P < 0.05) decreased to ~60% at HA day 10 compared to day 1 on the arm and back. Potassium concentration did not significantly differ on HA day 10 compared to day 1 (arm: +11%, P > 0.05; back: +8%, P > 0.05). The induction patterns of the sudomotor adaptations were different. Whilst LSR increased from HA day 8 on the arm and from HA day 7 on the back, sodium and chloride conservation already occurred from HA day 3 on both skin sites. Lastly, the sweat lactate reduction occurred from HA day 6 on the arm and back. Initial evidence is provided that adaptations were partly conserved after decay (28 days) and that a 5-day HRA may be sufficient to restore HA adaptations. In conclusion, ten days of exercise-induced HA using controlled hyperthermia led to increases in LSR and concomitant reductions of sweat sodium, chloride, and lactate concentrations, whilst potassium concentrations remained relatively constant.  相似文献

Hsp16.3, a molecular chaperone, plays a vital role in the growth and survival of Mycobacterium tuberculosis inside the host. We previously reported that deletion of three amino acid residues (142STN144) from C-terminal extension (CTE) of Hsp16.3 triggers its structural perturbation and increases its chaperone activity, which reaches its apex upon the deletion of its entire CTE (141RSTN144). Thus, we hypothesized that Arg141 (R141) and Ser142 (S142) in the CTE of Hsp16.3 possibly hold the key in maintaining its native-like structure and chaperone activity. To test this hypothesis, we generated two deletion mutants in which R141 and S142 were deleted individually (Hsp16.3ΔR141 and Hsp16.3ΔS142) and three substitution mutants in which R141 was replaced by lysine (Hsp16.3R141K), alanine (Hsp16.3R141A), and glutamic acid (Hsp16.3R141E), respectively. Hsp16.3ΔS142 or Hsp16.3R141K mutant has native-like structure and chaperone activity. Deletion of R141 from the CTE (Hsp16.3ΔR141) perturbs the secondary and tertiary structure, lowers the subunit exchange dynamics and decreases the chaperone activity of Hsp16.3. But, the substitution of R141 with alanine (Hsp16.3R141A) or glutamic acid (Hsp16.3R141E) perturbs its secondary and tertiary structure. Surprisingly, such charge tampering of R141 enhances the subunit exchange dynamics and chaperone activity of Hsp16.3. Interestingly, neither the deletion of R141/S142 nor the substitution of R141 with lysine, alanine and glutamic acid affects the oligomeric mass/size of Hsp16.3. Overall, our study suggests that R141 (especially the positive charge on R141) plays a crucial role in maintaining the native-like structure as well as in regulating subunit exchange dynamics and chaperone activity of Hsp16.3.  相似文献

Background

The recent morphological studies on chaperonins have revealed that nearly equivalent amount of symmetric GroEL–(GroES)2 (football-shaped) and asymmetric GroEL–GroES (bullet-shaped) complexes coexist during the chaperonin reaction cycle, which prompted us to reexamine the equatorial split observed for chaperonin from Thermus thermophilus by implementing semi-empirical molecular orbital (MO) calculations, since it is now believed that the symmetric formation is a precursor to the equatorial split.

Methods

Semi-empirical MO calculations were employed to investigate the intersubunit interactions within the bullet-shaped T. thermophilus chaperonin capturing the substrate of folding intermediates. Interaction energies between each cis-GroEL subunit and closely related remaining subunits in cis-GroEL ring, or in trans-GroEL ring across the equatorial plane, and further, interaction energies between each GroES subunit and adjacent subunits in the same GroES ring and in cis-GroEL ring were simulated.

Results

Anisotropic intensities and energy distribution of the subunits were revealed by the calculations, which are consistent with two conformations of the subunits forming cis-GroEL ring as revealed by X-ray crystal structure, and with an anisotropic critical binding site on cis-GroEL ring for chaperonin functioning.

Conclusions

This is the first application of semi-empirical MO calculations to the macromolecular complex of the native bullet-shaped chaperonin (GroEL–GroES–ADP homolog) from T. thermophilus.

General significance

The results also appear to support the occurrence of the equatorial split for T. thermophilus chaperonin observed via electron microscopy, but has not yet been fully observed for Escherichia coli GroEL–GroES system.  相似文献
