The fluorescence emission spectrum of Synechocystis sp. PPC6803 cells, at room temperature, displays: i) significant bandshape variations when collected under open (F0) and closed (FM) Photosystem II reaction centre conditions; ii) a marked dependence on the excitation wavelength both under F0 and FM conditions, due to the enhancement of phycobilisomes (PBS) emission upon their direct excitation. As a consequence: iii) the ratio of the variable and maximal fluorescence (FV/FM), that is a commonly employed indicator of the maximal photochemical quantum efficiency of PSII (Φpc, PSII), displays a significant dependency on both the excitation and the emission (detection) wavelength; iv) the FV/FM excitation/emission wavelength dependency is due, primarily, to the overlap of PSII emission with that of supercomplexes showing negligible changes in quantum yield upon trap closure, i.e. PSI and a PBS fraction which is incapable to transfer the excitation energy efficiently to core complexes. v) The contribution to the cellular emission and the relative absorption-cross section of PSII, PSI and uncoupled PBS are extracted using a spectral decomposition strategy. It is concluded that vi) Φpc, PSII is generally underestimated from the FV/FM measurements in this organism and, the degree of the estimation bias, which can exceed 50%, depends on the measurement conditions. Spectral modelling based on the decomposed emission/cross-section profiles were extended to other processes typically monitored from steady-state fluorescence measurements, in the presence of an actinic illumination, in particular non-photochemical quenching. It is suggested that vii) the quenching extent is generally underestimated in analogy to FV/FM but that viii) the location of quenching sites can be discriminated based on the combined excitation/emission spectral analysis.  相似文献

A quantitative evaluation of stream temperature alterations due to a commercial forest harvesting practice and a research treatment is presented. Summer maximum stream temperatures averaged 1 ° C higher in the commercial clearcut and 9 °C higher in the clearcut-herbicided watershed than in the forested control. The largest average monthly temperature increase on the commercial clearcut (2.2 °C) occurred during April; on the clearcut-herbicided basin it occurred during June (10.5 °C). Significant changes in stream temperature were observed on both watersheds as early as February and as late as November. Changes in minimum stream temperatures are presented in detail along with the impact on diel temperature fluctuations. Changes in the stream temperature regimes of the clearcut watersheds from the headwaters to the mouth of the watersheds are also given. Potential impacts of the stream temperature alterations on aquatic ecosystems are summarized in relation to stress limits for brook trout and other organisms. Climate fluctuations and human exploitation are causing global changes in nutrient enrichment of terrestrial and aquatic ecosystems and declining abundances of apex predators. The resulting trophic cascades have had profound effects on food webs, leading to significant economic and societal consequences. However, the strength of cascades–that is the extent to which a disturbance is diminished as it propagates through a food web–varies widely between ecosystems, and there is no formal theory as to why this should be so. Some food chain models reproduce cascade effects seen in nature, but to what extent is this dependent on their formulation? We show that inclusion of processes represented mathematically as density‐dependent regulation of either consumer uptake or mortality rates is necessary for the generation of realistic ‘top‐down’ cascades in simple food chain models. Realistically modelled ‘bottom‐up’ cascades, caused by changing nutrient input, are also dependent on the inclusion of density dependence, but especially on mortality regulation as a caricature of, e.g. disease and parasite dynamics or intraguild predation. We show that our conclusions, based on simple food chains, transfer to a more complex marine food web model in which cascades are induced by varying river nutrient inputs or fish harvesting rates.  相似文献

Iriartea deltoidea grows abundantly from Nicaragua to Bolivia. The stem is harvested for construction, furniture making, and handicrafts. The objective of my study was to understand the potential for sustainable harvesting of this palm. Ethnographic fieldwork about market patterns and decision-making among harvesters revealed the nature of harvesting pressures near settled areas. The study also demonstrates methods for assessing social and economic influences on harvesting.  相似文献

In‐situ captive rearing of endangered passerines for reintroduction has rarely been used as a conservation tool. Nests of Mauritius fodies threatened with predation by introduced mammalian predators were harvested from the wild, and chicks were reared to independence for release onto an offshore, predator‐free island. The daily probability of the survival was higher in captivity than in the wild, and 69 chicks were reared to fledging of which 47 would have been expected to fledge in the wild. Harvesting of nests probably had little impact on the wild population. Captive breeding trials on Mauritius fodies showed that large numbers of individuals could be produced for a release program from a small number of pairs if enough space was provided. Artificial incubation of passerine eggs and rearing of chicks can be used to increase the productivity of endangered taxa. The intracellular ATP and amino acid concentrations were determined in human fibroblast cultures reaching confluence. The values obtained were very different, depending on the cell harvesting method: trypsinization or scraping. Trypsinization appeared to be the better method for measuring the ATP concentrations (21.25 +/- 0.96 nmol per mg cell protein), this level being much lower with scraping. On the contrary, scraping was the most appropriate method for amino acid measurement. This work underlines the importance of harvesting methods for metabolic studies in human cell cultures.  相似文献

A wide range of stressors can cause a dramatic and sudden rise in the death rate in populations, typically resulting in what is referred to as crisis mortality. Here we present a method to standardize the assessment of identifying moments of crises. A modification of the mortality Z‐score methodology which is combined with time series analysis was used to investigate mortality events over the course of nearly two centuries for two populations: Gibraltar and Malta. A benefit of this method is that it situates the yearly death rate within the prevailing mortality pattern, and by doing so allows the researcher to assess the relative impact of that event against the norm for the period under investigation. A series of threshold values were established to develop levels of mortality to distinguish moments of lower mortality than expected, background mortality, a crisis, and a catastrophe. Our findings suggested that within defined periods, a limited number of events constituted moments of excessive mortality in the range of a crisis or higher. These included epidemics (yellow fever and influenza in Gibraltar only, and cholera) and casualties associated with World War II. Episodes of lower than expected mortality were only detected (although not significant) in the 20th century in Malta, and at the micro level, the harvesting effect appears to have occurred following cholera epidemics in both locations and influenza in Gibraltar. Non‐photochemical quenching (NPQ) of the light energy absorbed is one of the main photoprotective mechanisms evolved by oxygenic photosynthetic organisms to avoid photodamage, at a cost of reduced photosynthetic efficiency. Tuning of NPQ has been reported as a promising biotechnological strategy to increase productivity in both higher plants and unicellular microalgae. Engineering of NPQ induction requires the comprehension of its molecular mechanism(s), strongly debated in the last three decades with several different models proposed. In this work, the molecular details of NPQ induction was investigated at intramolecular level by in vitro and in vitro site‐specific mutagenesis on chlorophyll binding sites of the Light‐Harvesting Complex Stress‐Related 3 (LHCSR3) protein, the pigment binding complexes identified as the quencher during NPQ induction in the model organism for green algae Chlamydomonas reinhardtii. The results obtained demonstrate a correlation between the quenching activity of LHCSR3 variants in vitro and the NPQ phenotypes observed in vivo. In particular, multiple quenching sites in LHCSR3 cooperatively dissipating the excitation energy were revealed with a peculiar role of Chl 613, a chromophore located a close distance to carotenoid binding site L1.  相似文献

Understanding the relative impacts of harvesting across an area such as a marine park is vital if the goals of fisheries management are to be met. Given their accessibility, densities of targeted intertidal turbinids should be relatively simple to quantify; however, natural spatial and temporal variability in these populations has hampered this effort. This study aimed to quantify short-term population dynamics of Turbo militaris in relation to current zoning regulations and accessibility. While our results reflected the variability found by other studies, we also detected significantly lower densities at a headland where harvesting is known to occur, and found a trend towards denser aggregations at more remote locations. Importantly, we found that densities at some locations were so low that current fisheries bag limits may be ineffective for protecting populations at local scales. Comparisons between study sites suggest a combination of no-take zoning and inaccessibility may provide the most effective protection for this species. However, a greater understanding of the wider impacts of harvesting, and processes affecting recovery, are essential to ensure sustainable management of this fishery.  相似文献

The central California coast is visited by millions of people every year, many of whom collect intertidal gastropods for food, bait or recreation in near-shore marine environments. The harvesting of these animals has caused a decline in body size because humans preferentially take the largest individuals. Marine protected areas (MPAs) may serve to protect gastropods from this impact. The average body size and frequency of four gastropod species from eight study locations along the central California coast were determined. Five study locations were in MPAs and three study locations were in non-MPA zones. Gastropods in four California museums were measured and compared to specimens collected in the field. Three of the four species studied were larger and/or more frequent in MPAs than outside MPAs, including both harvested species and one unharvested species. For three of the four species, the average size of individuals from MPA sites was significantly smaller than museum specimens, but this may be due, in part, to our sampling procedure. These findings indicate that MPAs can be effective at reducing human impacts for some species, but that further study is needed to make MPAs as effective as possible for more species.  相似文献
